Apple Virus Diagnosis Using Simplified RNA Extraction Method (사과바이러스 간편 진단을 위한 RNA추출법 개선)

  • Kyungsan nursery complex which has a vast area for the production of various species of fruit tree stocks is in a high demand of virus-free saplings. Apple tree stocks, the most important products, urgently need more rapid and reliable viral diagnosis. In this study, a bead beater was tested because of convenience in dealing with large number of samples. Also, industrial glass bead abrasive (0.4 mm in diameter) at very low cost was used in a disposable way. For bead beater-aided RNA extraction from apple stem tissues, the guanidine thiocyanate method was confirmed to be very reliable. Silca membrane filter tube in connection to vacuum filtering device was strongly suggested for simplifying RNA capture and washing steps. Apple virus detection was confirmed by RT-PCR.

Anti-inflammatory Effect of Ishige foliacea in RAW 264.7 Cells (넓패추출물에 의한 RAW 264.7 세포에서의 항염효과)

  • This study was carried out to identify the anti-inflammatory effects of Ishige foliacea (I. foliacea) extract on skin using RAW 264.7 cells. The anti-inflammatory effects of I. foliacea extract on RAW 264.7 cells were assessed by cell viability assay, mRNA expressions, and nitric oxide (NO)/prostaglandin E2 (PGE2) productions. The anti-inflammatory effects of I. foliacea extract were elucidated by analysis of IL-1α/IL-1β/IL-6/TNFα gene expressions and PGE2/NO production. Quantitative real-time polymerase chain reaction showed that I. foliacea extract decreased the gene expression levels of iNOS/COX2/IL-1α/IL-1β and IL-6. Furthermore, PGE2/NO production also revealed that I. foliacea extract exhibited anti-inflammatory properties. These results suggest that I. foliacea extract is an anti-inflammatory compound. It could be a potent cosmeceutical material for anti-inflammatory effects. Further studies on the anti-inflammatory mechanisms of broadleaf extracts are expected to help identify pharmacological mechanisms related to inflammation in addition to cosmeceuticals.

Effects of Various Garlic (Allium sativum) Extracts on Cholesterol Synthesis in HepG2 Cells (다양한 마늘 추출물이 HepG2 세포에서 콜레스테롤 합성에 미치는 효과)

  • This study was performed to investigate the effects of various garlic extracts on cholesterol synthesis in HepG2 cells. Raw garlic, grilled garlic, and freeze dried garlic were subjected to cold water extraction, and extracts were incubated at room temperature for 1 min or 60 min. The extracts were treated to HepG2 cells for 4 h, and cholesterol synthesis and mRNA expression level of HMG-CoA reductase were investigated. The alliin contents were reduced when garlic was incubated at room temperature for 60 min. Raw garlic extracts showed lower intracellular cholesterol contents compared to that of the control group. However, raw garlic extracts incubated for 60 min showed no differences compared to the control group. Freeze-dried garlic extract showed minimum intracellular triglyceride and cholesterol contents. Relative mRNA expression level of HMG-CoA reductase, a rate-limiting enzyme of cholesterol synthesis, decreased in the garlic extracts. Compared with 60 min, garlic extracts incubated for 1 min showed a reduced level of HMG-CoA reductase mRNA expression. The freeze-dried garlic extract reduced mRNA expression level of HMG-CoA reductase in a dose-dependent manner in cells treated with 5% of 0.2, 0.5, 0.8, 1.0, and 1.5 mg/mL in medium, and the effect was maxed out at dose of 5% garlic extract at 1.0 mg/mL in medium.

  • Ribonucleic acid contents and mononucleotides distribution from the mycelium and fruit bodies of Ganoderma lucidum were studied. P.E.I. cellulose TLC and HPLC were applied in this study. The obtained results are as follows; The levels of ribonucleic acids from the young basidiocarp mycelium were higher than those of mature basidiocarp. Guanosine 5'-monophosphate and xanthosine 5'-monophosphate were found in both young basidiocarp mycelium and mature basidiocarp. The levels of guanosine 5'-monophosphate and xanthosine 5'-monophosphate from the young basidiocarp were higher than those of the mature basidiocarp. However, inosine 5'-mono­phosphate was not detected.

  • Small subunit ribosomal RNA (SSU rRNA) gene nucleotide sequences of bovine ReiLerin isolates from Korea (KLS and KCB) and japan (JHS) were determined. The genes from each isolate were amplified by the polymerase chain reaction and the approxi- mately 1.8 kb product cloned and sequenced by a modified dideoxynucleotide method. Overlapping gene segments produced with a series of primers were sequenced, resoRting in a complete DNA sequence for both forward and reverse strands of the SSU rRNA genes of each isolate. SSU rRNA gene sequences (termed Type A) were identical among the bovine ReiLeri,n isolates from Korea and the isolate from Japan. A GenBank data library homolo- gy search showed the sequence to be the same as that listed as leiLeyia buKeLi isolated from cattle in Marula, Kenya.

MMP-1 and PIP Expressions from Ethanol Extract of Hydnocarpus anthelmintica Pierre in Human Fibroblast Cells (사람유래 섬유아세포에서 대풍자 에탄올 추출물의 MMP-1과 PIP의 발현에 대한 연구)

  • This study aims to evaluate the effects of antioxidant activities, protein and mRNA expressions of matrix metalloproteinase (MMP) -1 and procollagen type I C-peptide (PIP) in 70% ethanol extract from Hydnocarpus anthelmintica Pierre (HE). DPPH and ABTS+ radicals scavenging assays were measured for antioxidant activities and HE had 73.5% and 74.4% of scavenging activities at 1,000 ㎍/ml concentration, respectively. And we investigated the inhibition of collagenase by HE, and the result was a 78.8% inhibition effect on concentrations of 1,000 ㎍/ml. In addition, an MTT assay was performed to confirm the toxicity of the CCD-986sk fibroblasts to the HE, and as a result, the cell viability rate was about 91.7% at a concentration of 50 ㎍/ml or less, and subsequent cell experiments were performed at a concentration of 50 ㎍/ml or less. We treated the cells with UVB (20 mJ/cm2) for stimulation, treated HE at various concentrations, and performed ELISA tests and RT-PCR experiments. And HE increased the PIP and mRNA in a dose-dependent manner and showed an expression rate of about 64.2% and 83.4%, respectively, at a concentration of 50 ㎍/ml compared with Cont (50.3% and 45.8%, respectively). And HE suppressed the MMP-1 protein and mRNA in a dose-dependent manner and showed a low expression rate of about 48.7% and 35.9%, respectively, at a concentration of 50 ㎍/ml. These results can be applied to developing anti-wrinkle materials for functional food and cosmetics with HE.

  • 최근 MicroRNA(miRNA)가 질병 발생과 밀접한 연관성이 있다고 밝혀진 이래, 이와 관련된 연구가 활발히 진행되고 있다. 하지만 각종 질병 관련 miRNA의 기능과 역할 그리고 질병 발생 메카니즘 등이 명백히 밝혀진 것이 없는 실정이다. 본 논문에서는 여러 종류의 miRNA 데이터베이스(miRecords, miRTarBase, miR2Disease 등)를 통합하고, 본 논문에서 새로이 제안하는 scoring 방법과 특정 질병과 관련된 miRNA의 순위결정과정을 통하여 질병과 연관성이 높은 miRNA을 밝혀내는 방법을 제안한다. 새로이 제안하는 방법을 바탕으로 miRNA와 특정 질병과의 연관성을 효과적으로 밝혀냈다.

Effect of Ginseng on Cerebral Nucleic Acid Content of Mice (고려인삼이 마우스의 대뇌조직 핵산 함유량에 미치는 영향)

  • A study 6was Planned to see if administration of ginseng extract has any influence upon cerebral nucleic acid content of mice. Thirty male mice $(body\;weight:\;18{\sim}20\;gm)$ were divided into the ginseng and the saline groups. Once a day for S days they received subcutaneously 0.05 ml/10 gm body weight of ginseng extract solution (4 mg of ginseng alcohol extract in 1 ml of saline), and the same amount of saline, respectively. On the 5th experimental day, all animals were sacrificed 2 hours after the last medication and their cerebral tissue was removed. Cerebral RNA and DNA contents were measured using the chemical method of Schmidt-Thannhauser-Schneider. Following results were obtained: 1. Cerebral RNA and DNA contents were significantly higher in the hippocampal group than in the saline group. 2. The RNA/DNA ratio was lower in the ginseng group compared with that in the saline group, because the cerebral DNA increased more remarkably than the RNA did following administration of ginseng. The ginseng is inferred to augment cerebral RNA and DNA content of mice.

  • It was found that petroleum ether extract of White Ginseng had a very slightly stimulating effect on the RNA biosynthesis of Baker's yeast, while both ethanol extract of White Ginseng(WGpet-alc) and Red Ginseng Extract(RGE) showed a significant stimulating effect on the RNA biosynthesis and its activity to change mononucleotide composition of the yeast RNA was noticeable; RGE induced a remarkable decrease of AMP, CMP and especially of UMP, however a marked increase of GMP, and it was observed that the molar ratio of GMP was 78.5 % of all mononucleotides of the yeast RNA. It is of interest that these results are closely related with the stimulating effect of protein biosynthesis of Baker's yeast cells.

Protective Effects of Akebia quinata Fruit Extract on Acute Alcohol-induced Hepatotoxicity in Mice (급성 알코올 간독성을 유발한 생쥐에 있어서 으름 열매 추출물의 간 기능 보호효과)

  • We studied the effects of Akebia quinata fruit extract (AQ) on acute alcohol-induced hepatotoxicity in mice. AQ (30-1,000 mg/kg body weight (BW) per day) was orally administered to the study group, once daily for 1 week. On the last day of AQ treatment, ethanol (6 mg/kg BW) was orally administered to induce acute liver injury. The AQ-treated group showed significantly lower levels of alanine aminotransferase and aspartate aminotransferase, compared to the only ethanol-treated group (ETG). The glutathione level in the AQ-treated group elevated up to 20.6%, compared to that observed in the ETG. The mRNA expression of glutathione synthetic enzymes was also higher in the AQ-treated group, compared to the ETG. The AQ-treated group also exhibited lower levels of expression of NADPH oxidase 4 and tumor necrosis factor alpha mRNA. Thus, these results show that AQ treatment can be a potential method to reduce oxidative stress and inflammation in ethanol-treated mouse liver and also that AQ can be a useful therapeutic agent for acute alcohol-induced hepatotoxicity.
