Protective Effect of Korean Panax ginseng against Chromium VI Toxicity and Free Radicals Generation in Rats

  • Earlier studies have demonstrated that chromium (Cr) VI compounds have been shown to be more toxic and carcinogenic than other chromium compounds. The aim of the present work was to evaluate the antioxidant effects of red ginseng against chromium VI -induced toxicity and free radical generation. Sixty adult male rats were divided into six equal groups include: control group, group received Cr VI alone (50 mg/kg b.w.), group treated with Korean ginseng (K. ginseng) alone (20 mg/kg b.w), group treated with Cr VI for 15 days then received K. ginseng for other 15 days, group treated with Cr VI and K. ginseng at the same time for 15 days, and group treated with K. ginseng for 15 days then Cr VI for other 15 days. The results revealed that Cr VI caused significant increase in ALT, AST, ALP, G-GT, urea, creatinine, and acid phosphatase. Whereas, it caused significant decrease in TP, albumin, testosterone, GPX, and SOD indicating a stress for liver, kidney and testes. K. ginseng alone caused significant increase in GPX and SOD activities in healthy animals and this result suggests a prophylactic role for this herb in protection against the damaging impact induced by free radical species. Furthermore, the other biochemical parameters measured after K. ginseng administration were comparable to the control values. Treatment with Cr VI followed by K. ginseng, Cr VI and K. ginseng or K. ginseng followed by Cr VI resulted in significant improvement in all tested parameters towards the normal values of the controls. However, this improvement was pronounced in the group pre-treated with K. ginseng for 15 days before Cr VI administration. It could be concluded that K. ginseng exhibited a protective action against the toxic effects of Cr VI and it had the ability to scavenge free radicals resulted from Cr VI intoxication.

Effect of dietary supplementation of garlic powder and phenyl acetic acid on productive performance, blood haematology, immunity and antioxidant status of broiler chickens

  • Objective: The effect of garlic powder (GP) and phenyl acetic (PA) acid throughout the fattening period of broiler chickens on performance, blood parameters, immune, and antioxidant parameters as well as carcass traits was evaluated. Methods: A total of 210 day-old Cobb broiler chicks were randomly distributed into seven dietary treatments having five replications with six chicks per replicate. The first group (control) fed a basal diet without supplements, whereas the 2nd, 3rd, and 4th group were fed basal diet plus 0.25, 0.50, and 0.75 g GP/kg diet, respectively and the group 5th, 6th, and 7th were fed on the basal diet plus 0.25, 0.50, and 0.75 g PA/kg diet. Results: Broiler body weight and gain at 21 and 42 days were increased (p<0.05) with diets supplemented with GP and PA. Red blood cells and hemoglobin were improved in chickens fed diets enriched with GP. Broiler chickens received diets containing either GP or PA recorded the higher values (p<0.05) of total protein, globulin, high-density lipoprotein, immunoglobulin M (IgM), and IgG, superoxide dismutase and total antioxidant capacity; while, blood total cholesterol, low-density lipoprotein, aspartate-aminotransferase, and malondialdehyde were lowered (p<0.05) compared to control-diet. Liver and immune-related organs weight were improved (p<0.05) in broilers fed diet supplemented with GP and PA. Conclusion: Feeding of GP or PA in diet had positive effects on performance traits and immunological, antioxidant and physiological status of broilers. Thus, the use of tested feed additives as an eco-friendly alternative to antibiotics produced a positive effect on animal health.

Effects of feed intake and water hardness on fluralaner pharmacokinetics in layer chickens

  • Background: Fluralaner is a novel drug belonging to the isoxazoline class that acts on external parasites of domestic animals. It is used systemically via drinking water, especially against red poultry mite in layer chickens. Fluralaner is frequently used in layers infected with D. gallinae. However, no study to date has investigated the effects of feed intake and water hardness. Objectives: This study aimed to investigate the effects of variable water hardness and feed intake on the pharmacokinetic profile of fluralaner. Methods: Layer chickens were divided into four groups (n = 8): fed + purified water (Group 1), feed restricted + purified water (Group 2), feed restricted + hard water (Group 3), and feed restricted + soft water (Group 4). After administering a single dose of the drug with drinking water, the blood samples were collected for 21 days. Fluralaner concentrations in plasma samples were determined by liquid chromatography/tandem mass spectrometry. The maximum plasma concentration (Cmax), time to reach maximum plasma concentration (tmax), area under the concentration-time curve values (AUC0-21d), half-life (t1/2), and other pharmacokinetic parameters were calculated. Results: Although the highest maximum plasma concentration (Cmax) was determined in Group 1 (fed + purified water), no statistically significant difference was found in the Cmax, tmax, t1/2, MRT0-inf_obs, Vz/Fobs, and Cl/F_obs parameters between the experimental groups. Conclusions: It was concluded that the feed intake or water hardness did not change the pharmacokinetic profile of fluralaner in layer chickens. Therefore, fluralaner could be used before or after feeding with the varying water hardness in poultry industry.

A Multicenter Pilot Study of Biliary Atresia Screening Using Digital Stool Color Imaging

  • Purpose: The presence of alcoholic stool in biliary atresia (BA) patients is the basis of a stool color card (SCC), a screening tool that has led to more patients receiving Kasai portoenterostomy earlier. This study aimed to evaluate the color image processing of stool images captured using smartphones. We propose that measuring digital color parameters is a more objective method for identifying BA stools and may improve the sensitivity of BA screening. Methods: A prospective study was conducted in five hospitals in Thailand between October 1, 2020, and December 31, 2021. Stools from infants presenting with jaundice, acholic stool, or dark-colored urine were photographed. Digital image color analysis was performed, and software was developed based on the color on the original SCC. Sensitivity and specificity for predicting BA stools were compared between the SCC and the software. Results: Of 33 infants eligible for data collection, 19 were diagnosed with BA. Saturation and blue were two potential digital color parameters used to differentiate BA stools. The receiver operating characteristic curve was used to determine the optimum cutoff point of both values, and when saturation ≤56 or blue ≥61 was set as a threshold for detecting BA stool, high accuracy was achieved at 81.8% and 78.8%, respectively. Conclusion: Digital image processing is a promising technology. With appropriate cutoff values of saturation in hue, saturation, value and blue in red, green, blue color models, BA stools can be identified, and equivocal-colored stools of non-BA patients can be differentiated with acceptable accuracy in infants presenting with jaundice.

Red Beet의 모상근 배양을 이용한 천연색소인 Betacyanin 생산의 최적화 (Optimization of Betacyanin Production by Red Beet (Beta vulgaris L.) Hairy Root Cultures.)

  • Red beet(Beta vulagris L.) 모상근의 회분배양을 이용한 천연색소인 betacyanin 생산 최적화를 위해 광도, C/N ratio, 인산의 농도를 각각 변화시켜 세포생육과 색소 생산성에 관한 동력학적 분석을 실시했다. 광도변화에 따른 배양 결과 3 klux의 경우 0.3(1/day)의 최대 비 생육속도와 0.11(mg/g-dry cell/day)의 최대 비 생산속도 그리고 14 k1ux에서 0.242(1/day)의 최대 비 생육속도와 0.125(mg/g-dry cell/day)의 최대 비 생산속도를 나타냈다. 광도와 균체의 생육관계를 검토한 결과 광도에 따른 세포 생육은 photoinhibition model이 적용됨이 확인되었다. Red beet 모상근으로부터 betacyanin의 생산은 partially growth related process임이 입증됐다. 이에 따른 세포당 최대 betacyanin 생산을 나타내는 $\alpha$는 0.3756 (mg/cell)이며, 최대 생산속도를 나타내는 $\beta$는 0.001 (mg/g-cel1/day)로 측정됐다. C/N ratio에 따른 실험결과 42.1(w/w)에서 0.26(1/day)의 최대 비생육 속도를 나타내었으나 최대 비 생산속도는 31.6(w/w)에서 0.075(mg/g-cell/day)를 나타냈다. 인 농도에 대한 균체의 생육 및 물질 생산성의 관계를 검토한 결과 1.25mM에서 0.31(1/day)의 비생육 속도와 0.134(mg/g-dry cell/day) 비생산 속도를 나타내었다. 최적 조건을 결정하기 위한 response surface methodology(RSM)결과 세포 생육과 betacyanin의 최대 생산을 위한 최적 광도는 5.5 (klux),최적 C/N ratio와 인의 농도는 27(w/w), 1.25 (mM)로 결정됐다. 그리고 0.1 $\mu$M kinetin 첨가시 대조구에 비해 비생산성이 0.085(mg/g-dry cell/day)로 증가함이 입증됐다. Normal조건과 optimum조건의 비교결과 세포의 농도인 X(g-dry wt./L)가 8와 16, betacyanin의 생산량인 P(mg/L)가 4.48과 12.5, 그리고 optimum 조건에서 최대비 생육속도인 $\mu$$_{max}$ 가 0.375와 그리고 최대비 생산속도인 q$^{max}$ $_{p}$ 는 0.134로 약 2배로서 최적화가 되었다.

맹종죽순 장아찌의 유통기한 설정 (The Prediction of Shelf-life of Pickle Processed from Maengjong bambo)

  • 맹종죽순을 이용하여 제조한 고추장 장아찌 및 간장 장아찌의 저장온도($25^{\circ}C$, $35^{\circ}C$$45^{\circ}C$)에 따른 미생물수, pH 및 산도, 향, 맛, 색 및 전체기호도 등의 품질 및 관능적 변화를 검토하였다. 맹종죽순 고추장 장아찌 및 간장장아찌 모두 저장온도와 상관없이 전체 30일간의 분석 기간 동안 미생물, pH 및 산도의 변화는 크게 나타나지 않았다. $25^{\circ}C$에서 저장할 경우, 맹종죽순 고추장 장아찌의 향, 맛, 색 및 전체적인 기호도 등의 관능적 품질의 변화는 30일의 저장기간 동안 유의적인 차이를 보이지 않았다. $35^{\circ}C$$45^{\circ}C$에서 저장할 경우, 맹종죽순 고추장 장아찌의 향, 맛 및 색은 크게 변화지 않았으나, 전체적인 기호도는 저장 초기부터 유의적인 차이를 나타내었다. 맹종죽순 간장 장아찌의 향, 맛 및 전체적인 기호도는 $25^{\circ}C$, $35^{\circ}C$$45^{\circ}C$에서 저장하는 동안 유의적인 차이를 보이지 않았으나, 색의 경우 $45^{\circ}C$에서 저장한 경우 25일차부터 유의적인 차이를 보였다. 맹종죽순 고추장 장아찌와 간장 장아찌에 대하여 각각 전체적인 기호도 및 색을 지표로 정하여 유통기한을 산출한 결과, $20^{\circ}C$에서의 상온 유통을 기준으로 각각 308일(10개월)과 447일(14개월)로 계산되었다.

Effects of Brazilin on Induction of Immunological Tolerance by Sheep Red Blood Cells in C57BL/6 Female Mice

  • Brazilin was examined for its effects on the induction of immunological tolerance. Brazilin was administered to C57BL/6 female mice for 2 consecutive days before the immunization with high dose SRBC (109 cells) which can produce immunological tolerance. Delayed type hypersensitivity, IgM plaque forming cells, ConA induced IL-2 production and mitogen- or antigen-induced proliferation of lymphocytes were measured as evaluation parameters. Administration of brazilin prior to immunization could keep the DTH and IL-2 production almost optimaly immunized levels. Brazilin also inhibited the elevation of non-specific suppressor cell activity. ConA induced proliferation of splenocytes in high dose SRBC immunized mice was significantly decreased by pretreatment of brazilin. And this might be one of the reason for augmentation of DTH by brazilin. However, IgM plaque forming cells were not affected by the treatment of brazilin. These results indicate that brazilin prevents the induction of mmunological tolerance caused by high dose SRBC by suppressing the elevation of suppressor cell activity and by inhibiting the decrease in IL-2 production in C57BL/6 female mice.

On-the-go Nitrogen Sensing and Fertilizer Control for Site-specific Crop Management

  • In-field site-specific nitrogen (N) management increases crop yield, reduces N application to minimize the risk of nitrate contamination of ground water, and thus reduces farming cost. Real-time N sensing and fertilization is required for efficient N management. An 'on-the-go' site-specific N management system was developed and evaluated for the supplemental N application to com (Zea mays L.). This real-time N sensing and fertilization system monitored and assessed N fertilization needs using a vision-based spectral sensor and controlled the appropriate variable N rate according to N deficiency level estimated from spectral signature of crop canopies. Sensor inputs included ambient illumination, camera parameters, and image histogram of three spectral regions (red, green, and near-infrared). The real-time sensor-based supplemental N treatment improved crop N status and increased yield over most plots. The largest yield increase was achieved in plots with low initial N treatment combined with supplemental variable-rate application. Yield data for plots where N was applied the latest in the season resulted in a reduced impact on supplemental N. For plots with no supplemental N application, yield increased gradually with initial N treatment, but any N application more than 101 kg/ha had minimal impact on yield.

Photoinhibition Induced Alterations in Energy Transfer Process in Phycobilisomes of PS II in the Cyanobacterium, Spirulina platensis

  • Exposure of algae or plants to irradiance from above the light saturation point of photosynthesis is known as high light stress. This high light stress induces various responses including photoinhibition of the photosynthetic apparatus. The degree of photoinhibition could be clearly determined by measuring the parameters such as absorption and fluorescence of chromoproteins. In cyanobacteria and red algae, most of the photosystem (PS) II associated light harvesting is performed by a membrane attached complex called the phycobilisome (PBS). The effects of high intensity light (1000-4000 ${\mu}mol$ photons $m^{-2}s^{-1}$) on excitation energy transfer from PBSs to PS II in a cyanobacterium Spirulina platensis were studied by measuring room temperature PC fluorescence emission spectra. High light (3000 ${\mu}mol$ photons $m^{-2}s^{-1}$) stress had a significant effect on PC fluorescence emission spectra. On the other hand, light stress induced an increase in the ratio of PC fluorescence intensity of PBS indicating that light stress inhibits excitation energy transfer from PBS to PS II. The high light treatment to 3000 ${\mu}mol$ photons $m^{-2}s^{-1}$ caused disappearance of 31.5 kDa linker polypeptide which is known to link PC discs together. In addition we observed the similar decrease in the other polypeptide contents. Our data concludes that the Spirulina cells upon light treatment causes alterations in the phycobiliproteins (PBPs) and affects the energy transfer process within the PBSs.

Implementation and Validation of Earth Acquisition Algorithm for Communication, Ocean and Meteorological Satellite

  • Earth acquisition is to solve when earth can be visible from satellite after Sun acquisition during launch and early operation period or on-station satellite anomaly. In this paper, the algorithm and test result of the Communication, Ocean and Meteorological Satellite (COMS) Earth acquisition are presented in case of on-station satellite anomaly status. The algorithms for the calculation of Earth-pointing attitude control parameters including those attitude direction vector, rotation matrix, and maneuver time and duration are based on COMS configuration (Eurostar 3000 bus). The coordinate system uses the reference initial frame. The constraint calculating available time-slot to perform the earth acquisition considers eclipse, angular separation, solar local time, and infra-red earth sensor blinding conditions. The results of Electronics and Telecommunications Research Institute (ETRI) are compared with that of the Astrium software to validate the implemented ETRI software.
