Analysis of genetic characteristics and development of substrate for cultivation in brown strains of Flammulina velutipes (갈색팽이버섯의 유전적 특성검정 및 배지재료 개발)

  • Analysis of genetic characteristics of Flammulina velutipes showed that strains have a range of 85% in genetic distribution diagram. According to this result, we divided these strains into five groups. In experiment of determinating the optimum media and condition in cultivating F. velutipes, we found the optimum temperature and pH range for hypha growth were $25^{\circ}C$ and 6.0 to 7.0, respectively. in addition, the best media for growth of that in plate was MCM (Mushroom Complete Media) which have a growth length from 68 to 83 mm. In vivo test, we observed that fast growth and good density of hyphae in mixture media of douglas fir sawdust, cotton seed meal and beet pulp (6:2:2 V/V). Also when we cultivated F. velutipes in this media, we harvested high yield of fruiting body.

Acute toxicity response caused by mixture or tank mix of several insecticides (몇 가지 살충제의 혼용 및 혼합 시 독성반응)

  • Tank mixing application of pesticides has been used to reduce labor and to control wide spectrum of pests, but it may cause significant pesticide poisoning on human and animals due to carelessness. The toxic response of pesticides for the tank mixtures and mixtures was investigated to determine acute toxicity and enzymatic change using experimental animals. Acute oral toxicity and acute dermal toxicity were tested by RDA test guideline. The $LD_{50}$ was calculated by probit analysis method and cholinesterase was measured with automatic analyzer. The toxicities were generally higher than estimated toxicities in tank mixing and mixture. Serum cholinesterase activity was inhibited more than expected at the dose levels of 1/5, 1/10 and 1/20 of $LD_{50}$. Therefore, the results of this study showed that acute toxicity caused by the pesticide mixtures should be considered before the tank mixing method is applied.

Adjustment of heterogeneous variance by milk production level of dairy herd (젖소군의 유생산 수준별 이질성 분산 보정)

  • This experiment was conducted to compare heterogeneity for the variance in dairy cattle population and to induce homogeneity of variance using 502,228 performance test records of dairy cattle. The estimates of heritability for milk yields, fat yields and protein yields were 0.28, 0.26 and 0.24, respectively and the estimate of average breeding value by birth year was lower in HV (heterogenous variance) model than in animal model, collectively. The average breeding values of milk yields, fat yields and protein yields for 545 sire bulls applicable to the criteria of interbull MACE programme were 453.54kg, 10.75kg and 14.33kg, respectively and when the heterogeneity was adjusted they were 432.06kg, 10.15kg and 13.40kg, respectively, which were lower in all milk traits collectively. In animal model, coefficients of phenotypic correlation between dataset I and II were 0.839 in milk yields, 0.821 in fat yields, and 0.837 in protein yields, while in HV model, they were 0.841 in milk yields, 0.820 in fat yields, and 0.836 in protein yields, showing similar results in 2 models. When compared using animal model and HV model, the regression coefficient for ratio of number of daughters by calving year of milk yields increased from 15.157 to 16.105 and that of fat yields increased from =0.227 to =0.196, but that of protein yields decreased from 0.630 to 0.586.

Occurrence of Anthracnose Caused by Glomerella cingulata on Eucaly trees in Korea (Glomerella cingulata에 의한 유카리나무 탄저병 발생)

  • Since 2003, anthracnose symptoms on Eucalyptus globulus were observed in farmer's field at Jisepo, Ilwoon-myon, Geoje city, Gyeongnam province, Korea. Typical symptoms of dark brown to black spot appeared on the leaves, twigs, and stems. Infected young trees were wilted, blighted and died eventually. The pathogen isolated from the typical symptom formed gray to dark gray colony on potato dextrose agar and showed optimum growth at $30^{\circ}C$. Conidia were single celled, colorless, cylindrical with obtuse ends, and $9{\sim}22{\times}3{\sim}6{\mu}m$ in size. Appressoria were dark brown, ovate to obovate, and $6{\sim}18{\times}4{\sim}10{\mu}m$ in size. Perithecia were black and globose in shape and $76{\times}274{\mu}m$ in size. Asci were clavate to cylindrical in shape and $42{\sim}76{\times}8{\sim}12{\mu}m$ in size. Ascospores were cylindrical, fusiform, slightly curved at the center, and $10{\sim}23{\times}4{\sim}6{\mu}m$ in size. On the basis of mycological characteristics and pathogenicity test on E. globulus, the pathogen was identified as Glomerella cingulata. This is the first report of the anthracnose on E, globulus caused by G. cingulata in Korea.

Pathogenicity of Entomopathogenic Nematode, Steinernema carpocapsae Pocheon Strain Against Anomia mesogona and Anomis commoda(Lepidoptera: Noctuidae) (무궁화잎밤나방(Anomis mesogona)과 큰붉은잎밤나방(Anomis commoda)에 대한 Steinernema carpocapsae 포천 계통의 병원성)

  • Pathogenicity of entomopathogenic nematde, Steinernema carpocapsae Pocheon strain (ScP) was evaluated against different larval stages(2nd, 3-4th and 5th) of Anomis commode and Anomis mesogona(Lepidoptera: Noctuidae) in petri dish and pot. The $LC_{50}$ values were increased in proportion to larval stage of A. commoda and A. mesogona. $LC_{50}$ value of ScP against 2nd instar of A. commoda and A. mesogona was 9.7 and 4.5, respectively. The 2nd instar of both species was also susceptible to ScP in pot test. That is, mortality of 2nd instar of A. commode and A. mesogona was higher representing $72.5\%\;and\;87.5\%$ 5 days later after treatment, respectively, when ScP was applied at the ,ate of 90,000 infective juveniles(Ijs) per $pot(=1\times10^9\;Ijs/ha)$. However, susceptibility was decreased from 3rd instar. Mortality of 5th instar of A. commoda and A. mesogona was $5.0\%\;and\;10.0\%$, respectively, at the rate of 90,000 Ijs/pot. When ScP was applied into pot including mixed larval stages from 2nd to 5th instar(10 larvae far 2nd instar, 10 larvae for 3rd - 4th instars, and 10 larvae for 5th instar) at the rate of 90,000 Ijs/pot, mortality of A. commoda and A. mesogona was $69.2\%\;and\;50.0\%$, respectively.

Anti-Toxoplasmosis Effect of the Halophyte Suaeda maritime (염생식물 해홍나물의 항톡소포자충 효과)

  • Toxoplasmosis is an important cause of foodborne, inflammatory illnesses, as well as congenital abnormalities. Currently available therapies are ineffective for persistent chronic disease and congenital toxoplasmosis or have severe side effects which may result in life-threatening complications. There is an urgent need for safe and effective therapies to eliminate or treat this cosmopolitan infectious disease. The aim of this study was to investigate the in vitro anti-Toxoplasma activities of Suaeda maritime, one of the halophytes, using tachyzoit of T. gondii RH strain infected HeLa cells. As the results, the selectivity of Suaeda maritime extract was 6.63, which was higher than Sulfadiazine selectivity (2.06). Also, we perfomed the cell proliferation inhibition test and the morphological study to evaluate the anti-T. gondii activity of Suaeda maritime extract with HeLa cells. As the results, the inhibition rate of the Suaeda maritime extract was high inhibition rate. This indicates that the Suaeda maritime extract may be used for new anti-T. gondii agent.

Suppression of VCAM-1 Expression in Human Aortic Smooth Muscle Cells Treated with Ethanol Extracts of Cynanchum wilfordii Radix, Arctium lappa L., and Dioscorea opposita (백수오, 우엉, 마 추출물 혼합비율에 따른 혈관부착인자 VCAM-1의 발현억제 효과)

  • Cynanchum wilfordii Radix (CWR), Arctium lappa L (ALL), and Dioscorea opposite (DO) have been known to improve blood lipid profile, blood pressure, and inflammation. To find the optimal combination ratio of CWR, ALL, and DO in terms of vascular health improvement, we compared the effects of various combinations on gene expression of Vascular cell adhesion protein 1 (VCAM-1) in human aortic smooth muscle cells (HASMC). VCAM-1 mediates endothelial leukocyte adhesion and is upregulated in atherosclerosis. Cells was stimulated by TNF-α (10 ng/㎖, 2h) and treated with various combinations for 24 h. A combination (CADM5, CWR:ALL:DO = 2:1:1) showed the strongest suppression of VCAM-1 so that CADM5 was chosen for further experiments. We performed cell viability test with CADM5 (0, 3.125, 12.5, 25, 50, and 100 ㎍/㎖) and no cytotoxicity was found. We also investigated the effect of CADM5 on protein expression of VCAM-1, ICAM-1, Nrf-2, and HO-1 using western blotting. We found that CADM5 diminished the expression of VCAM-1 and increased the expression of Nrf-2 and HO-1. Therefore, we concluded that CADM5 (CWR:ALL:DO = 2:1:1) effectively improves vascular health by regulating the expression of VCAM-1.

Impact of phosphorus application on the indigenous arbuscular mycorrhizal fungi, soybean growth and yield in a 5-year phosphorus-unfertilized crop rotation

  • Arbuscular mycorrhizal fungi (AMF) are particular soil fungi that benefit many crops and require a symbiosis with plant roots to survive. In our previous study, there was a positive correlation between AMF root colonization and soybean grain yield in a four-year consecutive winter cover crop-soybean rotational system without phosphorus fertilizer. It is suggested that higher AMF root colonization can be a better solution for improving soybean growth and grain yield in P-limited soil. Our purpose in this study was to test the hypothesis that a P application is the main factor improving soybean growth, P nutrition and grain yield, and the benefit from AMF to soybean P uptake and growth in a P-limited soil. Impact of a P application on AMF root colonization and communities in soybean roots and their potential contribution to soybean growth and P nutrition under a five-year P-unfertilized crop rotational system were investigated over two-years. In this study, four cover crop treatments included 1) wheat (Triticum aestivum); 2) red clover (Trifolium pratense); 3) rapeseed (Brassica napus); and 4) fallow in the crop rotation. The amount of triple superphosphate as a P fertilizer applied rate after cultivation of cover crops was 120 and $360k\;ha^{-1}$ in 2014 and 2015, respectively. Soybean roots were sampled at full-flowering and analyzed for AMF communities using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) and quantitative real-time PCR (qPCR) techniques. The AMF root colonization in the soybean roots at full bloom stage was significantly influenced by cover crop and P application throughout the two-year rotation. The two-year rotation of different cover crops or fallow impacted the molecular diversity of AMF communities colonizing roots of soybean. Redundancy analysis (RDA) indicated that AMF communities colonizing roots of soybean were significantly different among cover crop rotations. The AMF communities colonizing roots of soybean were clearly influenced by a P application in the two-year trial. Moreover, a P application may have positively impacts on the AMF communities under P-deficit soil due to the continuous cover crop-soybean rotational system without a P fertilizer.

Structure-Activity Relationships of Polyhydroxyursane-type Triterpenoids on the Cytoprotective and Anti-inflammatory Effects

  • Eleven polyhydroxyursane triterpenoids (PHUTs) were tested to determine their cytoprotective, immunosuppressive and anti-inflammatory effects. To compare the bioactivities of $19{\alpha}$-hydroxyursane-type triterpenoids {23-hydroxytormentic acid (6), its methyl ester (7), tormentic acid (8), niga-ichigoside $F_1$ (9),euscaphic acid (10) and kaji-ichigoside $F_1$ (11)} of the Rosaceae crude drugs (Rubi Fructus and Rosa rugosae Radix) with PHUTs possessing no $19{\alpha}-hydroxyl$ of Centella asiatica (Umbelliferae), the four PHUTs, asiaticoside (1), madecassoside (2), asiatic acid (3), and madecassic acid (4) were isolated from C. asiatica and 23-hydroxyursolic acid (5) from Cussonia bancoensis. Cytoprotective effects were assessed by measuring cell viabilities against cisplatin-induced cytotoxocity in $LLC-PK_1$, cells (proximal tubule, pig kidney) to determine whether these agents have protective effects against nephrotoxicity caused by cisplatin. The inhibitory effect of 11 PHUTS on nitric oxide (NO) and prostaglandin $E_2\;(PGE_2)$ were evaluated by measuring nitrite accumulation in lipopolysaccharide (LPS)-induced macrophage RAW 264.7 cells, and their anti-inflammatory effects were tested in 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse ear edema model. Six MHUTs (compounds 1, 2, 4, 6, 10, and 11) exhibited higher cell viabilities during cisplatin-induced cytotoxicity testing even at a concentration of $200\;{\mu}g/ml$ than cisplatin only-treated group, suggesting that ese compounds have the potentcytoprotective efffcts. Compounds 1 and 3 of the C. asiatica and niga-ichigoside $F_1$ exhibited no inhibitory effect on NO and/or $PGE_2$ production whereas other PHUTs produced mild to significant NO and/or $PGE_2$ production.The four compounds (2, 5, 9, and 10) potently inhibited mouse ear edema induced by TPA whereas two compounds (1 and 3) had no activity in this test. These results suggest that many PHUTs are potentchemopreventives. Structure-activity relationship (SAR) was also discussed in each assay with regard to the significant role of OHs at the position of 2, 3, 6, 19, and 23 and to the glycoside linkage at the 28-carboxyl.

Brown Leaf Spot of Doonggulle (Polygonatum odoratum) Caused by Phyllosticta cruenta (Phyllosticta cruenta에 의한 둥굴레 갈색점무늬병)

  • Brown leaf spot caused by Phyllosticta cruenta from 2007 to 2009 occurred severely in an herbs exhibition on Doonggulle (Polygonatum odoratum) at Gangju pond in Jeongchon-myon, Jinju city, Gyeongnam province, Korea. The typical symptoms of this disease were appeared on the brown leaf spot of leaves. Lesions on the leaves were appeared elliptical or irregular with 2~8 mm in size. Infected leaves were wilted, blighted and eventually died. Pycnidiospores and pycnidia of the fungus were abundantly formed on PDA. The optimum temperature for mycelial growth was $20{\sim}{25^{\circ}C}$. Pycnidiospores were not readily formed on PDA, colorless in color, globose~subglobose in shape, $10{\sim}18{\times}5{\sim}12{\mu}m$ in size and appendage hanged $4{\sim}15{\mu}m$ in size. Pycnidia were globose~subglobose in shape, dark brown in color and $60{\sim}162{\times}70{\sim}174{\mu}m$ in size. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Phyllosticta cruenta (Kunze: Fries) Kickx. This is the first report on the brown leaf spot of Doonggulle caused by P. cruenta in Korea.
