In vitro Root Induction from Shoot Explants of Pear (Pyrus spp.)

  • The main objective of this study was to identify the most appropriate condition for root formation of in vitro micropropagated pear (Pyrus spp.) plants. In vitro propagation was induced on Murashige and Skoog (MS) medium with 2.0 mg/L of N6-benzyladenine (BA) and 0.2 mg/L of Indole-3-butyric acid (IBA) medium. The short pre-treatment of explants with a high concentration (1 mg/L) of NAA and IBA (R0 medium) in dark for three days, followed by transfer to five different media (R1 to R5) resulted in good rooting responses in the pear 'Oharabani (P. pyrifolia × P. communis)' genotype. For the rooting experiments, the highest rooting percentage (83.3 ± 8.3%), average root length (3.6 ± 1.9 mm), total root number (31 ± 4.0), and average root number per plant (2.6 ± 2.1) were obtained on half strength (1/2) of MS medium supplemented with 30 g/L sucrose without hormones and activated charcoal (AC) (R1 medium). The highest rooting percentage was obtained at 83.3% from explants on R1 and R3 media. The rooting procedure described in this study resulted in good root formation and significantly shorting the root induction time to within 14 days of culture. Further studies are underway to test the suitability of the protocol developed in this study for other pear genotypes.

사후시간 경과가 한우 거세우 배최장근의 pH, 온도, R-value 및 단백질 분해효소 활성에 미치는 영향 (Changes in pH, Temperature, R-values and Calpain Activity of M. longissimus from Hanwoo Steer during Rigor Development)

  • 본 연구는 한우고기의 품질에 영향을 미치는 사후 대사변화를 구명하고자 사후 1, 3, 9, 24시간별에 따라 한우 거세우 배최장근의 pH, 도체온도, R-value 및 단백질분해효소 활성측정을 수행하였다. 도체 pH와 온도는 사후 시간이 경과함에 따라 유의적으로 감소하였고(p<0.05), 사후 3시간에 6.50($31.99^{\circ}C$)을 나타내었다. $R_{248}$$R_{250}$ 값은 사후 9시간부터 유의적으로 증가하였지만, $R_{258}$ 값은 사후 9시간부터 감소하였다(p<0.05). Calpain I 활성은 사후 3시간부터 유의적으로 감소하였고, calpastatin 활성은 사후 9시간부터 유의적으로 감소하였다(p<0.05). pH와 도체온도는 $R_{258}$, calpain I, calpastatin에서 0.95이상의 높은 정의 상관관계를 나타내었지만, $R_{248}$$R_{250}$에서는 -0.97 이상의 부의 상관관계를 보였다. 이상의 결과를 종합하면, 사후9시간 이후에 대사 변화가 급격히 발생하는 것으로 나타났고, pH와 도체온도의 상관도가 높아 잠재적인 연화능력의 예측이 가능할 것으로 사료되지만, 사후 3시간부터 9시간 사이의 대사변화에 대하여 좀 더 심도 깊은 연구할 필요가 있을 것으로 생각된다.

Comparison of Glucosinolate Contents in Leaves and Roots of Radish (Raphanus spp.)

  • Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.

토종닭과 실용계에서 TYR 및 MC1R 유전자의 변이 분석 (Investigation of TYR and MC1R polymorphisms in Korean native chickens and the commercial chickens)

  • The commercial Korean native chickens (WR_CC) was developed by crossing a few native chicken breeds in Korea. In order to investigate the breed identification markers, SNPs from TYR gene and MC1R gene, which are associated with skin and feather colors respectively, were initially identified. In case of 3 identified SNPs in the TYR gene, yellow shank color was identified in Loss, Harvard, AA, RIR and CC, which have the fixed SNPs in most of the animals. On the other hand, SNP variations were observed in KNC_RB, C_B, WR_CC and HH_CC, which have the black, yellow and mixed color with black and yellow shank colors. Also, the investigation of 3 SNPs in the MC1R gene indicated that there were associations between shank and feather colors in RIR, SF, KNC_B, C_B and RIR. However, these results are not consistent among breeds. These SNP type inconsistencies within breeds suggested that the selection was performed based on the phenotypes, which is not include the genotype information. Thus, selection based on genetic information is required in the future.

젖소분뇨 퇴비화에 대한 부숙퇴비 혼합 효과 (The Mixing Effect of Decomposed Manure as Bulking Agent in Composting of Dairy Cow Manure)

  • According to the volumetric mixing rate of dairy cow manure (DCM) and moisture control materials such as decomposed manure (DM) and sawdust (S), 6 reactors (DCM only (R1), DCM : DM = 1:1 (R2), DCM : DM = 1.5:0.5 (R3), DCM : DM = 0.5:1.5 (R4), DCM : DM:S = 1:0.5:0.5 (R5) and DCM : S = 1:1 (R6)) were used for composting of dairy cow manure. Among the composting reactors, composting reactor of R5 was shown the highest temperature of the compost as a $66^{\circ}C$ during composting period. After 3 weeks composting, moisture content of R5 and R6 were 51% and 51.3%, respectively. These values were satisfied with the moisture content standard of livestock manure compost of Korea. We concluded that decomposed manure may be a good moisture control material for dairy cow manure composting when it is used in mixture with sawdust. The optimum volumetric mixing ratio of dairy cow manure and moisture control materials was 50% of livestock manure, 25% of decomposed manure and 25% of sawdust.

제주재래돼지와 듀록 참조축군에서 Melanocortin 4 Receptor (MC4R) 유전자형과 지방산 조성간의 관련성 분석 (Association of Melanocortin 4 Receptor (MC4R) Genotypes with Fatty Acid Compositions in an Intercross Population between Duroc and Jeju Native Pigs)

  • 본 연구에서는 제주재래돼지와 듀록 품종 사이에서 생산된 F2 참조축군에서 melanocortin-4-receptor (MC4R) 유전자형과 지방산 조성간의 관련성을 연구하였다. 전체 290 여두의 F2 자손을 이용하여 14개의 지방산 조성을 측정하였다. Taq I PCR-RFLP 방법을 이용하여 MC4R c.1426A>G (p.Asp298Asn)의 단일염기다형성(single nucleotide polymorphism, SNP)을 확인하였다. MC4R 세 가지 유전자형(AA, AB, BB)이 모두 발견되었고, 그 빈도는 각각 0.299, 0.542, 0.159로 확인되었다. AA 유전자형을 가진 개체에서 palmitic acid (C16:0, p<0.05), stearic acid (C18:0, p<0.01), eicosenoic acid (C20:1n9, p<0.05), saturated fatty acid (SFA, p<0.01) 함량이 GG 유전자형을 가진 개체보다 더 높은 것으로 확인되었다. 반면에 GG 유전자형을 가진 개체는 linoleic acid (C18:2n6, p<0.001), linolenic acid (C18:3n3, p<0.001), linolenic acid (C18:3n6, p<0.001), arachidonic acid (C20:4n6, p<0.001)과 같은 불포화지방산(unsaturated fatty acid) 함량이 AA 유전자형을 가진 개체보다 더 높은 것을 확인하였다. 제주재래돼지와 듀록 품종 사이에서 생산된 F2 참조축군에서 MC4R GG 유전자형이 포화지방산은 낮추고, 불포화지방산은 높이는 것으로 확인되었다. 본 연구 결과 MC4R의 유전적 다형성이 듀록과 제주재래돼지 교배 프로그램에서 육질 향상과, 고기내의 지방산 함량을 조절할 수 있는 유전적 표지 인자로 활용될 수 있을 것이라 사료된다.

  • 본 연구는 한우 inositol 1,4,5-triphosphate receptor type1(IP3R1) 유전자를 대상으로 SNP를 발굴하고, 도체형질과의 관련성 분석을 위하여 수행하였다. PCR 및 염기서열 결정법을 통해 IP3R1 유전자내 3개의 SNP를 발굴하였고, 이중 intron 29에 위치하는 SNP의 경우 미 보고된 신규 SNP로 확인되었다. 발굴된 3개의 SNP를 대상으로 표현형 기록치를 보유한 후대검정우 583두에 대하여 유전자형 분석 및 관련성 분석을 수행하였다. 분석결과 3개의 SNP 중 g.1428617A>G SNP가 생시체중(P<0.05) 및 도체중(P<0.01)과 유의적인 상관관계가 있음을 확인할 수 있었다. 이러한 결과는 추후 한우 개량을 위한 유전자 마커로 활용이 가능할 것으로 판단된다.

Effects of Planting Densities and Maturing Types on Growth and Yield of Soybean in Paddy Field

  • Field studies were conducted in the southeastern Korea ($36^{\circ}$N) on a commerce silt loam soil at paddy field. Seed were manually planted on 16 July 2003. Plants were planted with plant densities of 70${\times}$10 cm (row width x plant spacing), 50 x 10 cm, and 30 ${\times}$10 cm. Two seedlings per hill were taken prior to V3 stage. Fertilizer was applied prior to plant at a rate of 30-30-34 kg (N-$\textrm{P}_2\textrm{O}_5$-$\textrm{K}_2\textrm{O}$) per ha. Experimental design was a randomized complete block in a split plot arrangement with three replications. Yield from different planting densities responded similarly in three soybean cultivars and increased when planting density increased. Somyeongkong showed the highest increasing rate of yield about 26% by 338 g $\textrm{m}^{-2}$ at 30 x l0 cm compared to yield of conventional planting density (70 x 10 cm). Also, the planting density significantly affected pod and seed number and seed weight, but not seed per pod. The tallest plant appeared at 30${\times}$10 cm. The change of leaf area according to days after emergence showed differently in soybean cultivars. The highest and lowest total dry matter production per square meter appeared at 30 x 10 cm and at 70 x 10 cm, respectively. Crop growth rate (CGR) showed greater at R3∼R4 stages compared with V7∼R2 or R2∼R3 growth stages and showed the greatest at 30 x 10 cm in three soybean cultivars. As late planted soybean, there was a significant relation between seed yield and CGR, and leaf area index (LAI) according to planting densities at before and after the flowering stage. Relationship between seed yield and CGR in three planting densities showed a highly significant positive relation ($\textrm{R}^2$=0.757) at R3 to R4 stages, and significant relations ($\textrm{R}^2$=0.505, 0.617) at V7 to R2 and V2 to V3. Also, there was a highly significant positive difference between seed yield and LAI during R3 to R4 and R2 to R3 stages.

총체 벼 사일리지용 미생물의 발효능력 평가 (Evaluation of Fermentation Ability of Microbes for Whole Crop Rice Silage Inoculant)

  • 본 시험은 사료용 총체 벼 사일리지 전용 미생물 첨가제 개발을 위하여 2003년부터 2005년까지 축산연구소에서 수행되었다. 새로운 총체 벼 전용 첨가제 개발을 위해 수집된 28점의 총체 벼 사일리지로부터 미생물을 선발하여 전체 5개 균주(R4-1, R7-1, R7-2, R10-1 및 R12-1)를 분리하였다. 분리된 균주중 4종(R4-1, R7-1, R7-2 및 R10-1)은 Lactobacillus plantarum으로 동정되었고 R12-1은 L. pentosus로 동정되었다. 황숙기에 수확된 총체 벼는 사일리지를 제조할때 선발된 5개 균주 이외에 시중에서 판매되는 첨가제 3종을 접종을 하여 60일간 실온에서 저장한 후 평가한 결과는 다음과 같다. 저장된 사일리지의 pH와 초산 함량은 첨가제를 처리한 구에서 낮았고 젖산 함량은 높았다(p<0.05). 특히 R7-1은 초산 함량이 가장 낮았고 젖산 함량이 가장 높았다. 조단백질 함량은 R7-2 첨가제 처리구에서 가장 높았으며 ADF 및 NDF 함량은 R7-1에서 가장 낮았다(p<0.05). 선발된 균주들은 대부분 사일리지 품질개선효과를 나타내었으나 R7-1이 그 중에서 가장 효과적이었다. 본 미생물은 한국농용미생물 협회에 NLRI 401로 등록되었다.
