Characterization of the RNA binding protein-1 gene promoter of the silkworm silk grands (누에 견사선에서 분리한 RNA binding protein-1 유전자 프로모터 분석)

  • We isolated highly-expressed genes in the posterior silk glands of silkworm on a previously study, which one of these was identified as RNA binding protein-1 homologue (RBP-1) gene. In this study, we investigated gene expressional characteristics of the RBP-1 depending on silkworm development stages and several tissues of the larvae, respectively. Northern blot hybridization analysis showed that the RBP-1 gene was expressed high in larval and pupal periods, and highly expressed than endogenous internal control gene (BmA3) on all tested larval tissues. In addition, we isolated and analyzed a phage DNA having 1,660 bp-long promoter region of the RBP-1 gene from a genomic DNA library. To study the RBP-1 gene promoter activity, RBP-1 (-740/+ 30) was amplified by PCR and subcloned into a pGL3 basic vector to generate pGL-RBP1. A luciferase report vector carrying RBP-1 gene promoter (770 bp) was tested by luciferase assay in Sf9 cells. In the result, the RBP-1 gene promoter was more efficient than constitutive promoter (BmA3) by approximately ten percent.

Ovarian Tumors in Rbp9 Mutants of Drosophila Induce an Immune Response

  • The Drosophila protein, Rbp9, is homologous to human Hu, which is reported to be involved in small cell lung cancer. Rbp9 functions in cystocyte differentiation, and mutations in Rbp9 cause ovarian tumors. Here we show that the antimicrobial peptide, Attacin, is upregulated in Rbp9 mutants, especially in ovaries where tumors form. Upregulation seems to result from activation of the NF-${\kappa}B$ pathway since we detected nuclear localization of Relish in Rbp9 mutant ovaries but not in wild type ovaries. Inactivation of NF-${\kappa}B$ in the Rbp9 mutant allows prolonged survival of malformed egg chambers. We conclude that Drosophila initiates an anti-tumor defense response via activation of NF-${\kappa}B$.

Effect of 4-Nonylphenol on the Gene Expression of Retinol-Binding Protein in the Rockfish, Sebastes schlegeli (조피볼락(Sebastes schlegeli)의 Retinol-Binding Protein의 유전자 발현에 미치는 4-Nonylphenol의 영향)

  • Retinol-binding protein(RBP) plays an important role in the specific transport of retinol to target cells through the blood stream in higher vertebrates. In order to clarify the effects of 4-nonylphenol(4-NP) on RBP mRNA expression in the rockfish, Sebastes schlegeli which is common in coastal waters of Korea and commercially important species, the cDNA library was constructed from the liver, and a partial fragment of the RBP gene was cloned. The deduced amino acid sequence from the RBP mRNA showed a high homology to the amino acid sequence from Sparus aurata(80%), Oncorhynchus mykiss(72%) or Anguilla anguilla(78%). Effects of 4-NP on RBP and vitellogenin(VTG) mRNA expression level in rockfish were examined by the northern blot analysis. In female and male rockfish injected with 4-NP(10 mg/kg BW, lower dose), there was no changes in the levels of VTG mRNA expression in the liver. The RBP mRNA levels, however, decreased at 48 hours after the injection in male. In the rockfish injected with 4-NP(25 mg/kg BW, higher dose), the level of VTG mRNA expression increased after 24 hours, regardless of sex. The level of RBP mRNA expression decreased at 48 hours after the injection in both sexes. These data indicate that estrogenic mimics such as 4-NP exhibit a contrasting effect on RBP and VTG gene expression in rockfish.

Production of Retinol-binding Protein by Caprine Conceptus during the Time Period of Maternal Recognition of Pregnancy

  • The purpose of the study were to characterize the proteins secreted by elongating caprine conceptus, to identify a group of low molecular weight proteins as retinol-binding protein (RBP), to identify RBP cell-specific localization in conceptus tissue, and to demonstrate that the conceptuses secreted continuously RBP during the time period maternal recognition of pregnancy. Caprine conceptuses were removed from the uterus between days 16 and 22 of pregnancy, the time period maternal recognition of pregnancy. Isolated conceptuses were cultured in a modified minimum essential medium in the presence of radiolabeled amino acids. Proteins synthesized and secreted into medium were analyzed by fluorography of two-dimensional polyacrylamide gel electrophoresis and fluorography. At least five proteins showed consistently a grouping of spots with characteristic location on two-dimensional gels. A major low molecular weight protein consisted of two major isoforms (pI 5.3-6.0) of similar molecular mass (21 kDa) was identified as RBP by using antiserum against RBP. Presence of RBP in conceptus culture medium and uterine flushings between days 16 and 22 of pregnancy were determined by immunoprecipitation and Western blotting using anti-RBP serum. In immunocytochemical study, strong immunostaining for RBP was localized in trophectoderm and endoderm of conceptus. These results clearly demonstrated that the caprine conceptus was active in protein synthesis as early as day 16 of pregnancy. Secretion of RBP by caprine conceptuses (days 16-22) coincident with the rapid transformation of the conceptus from a spherical blastocyst to a filamentous structure. Production of RBP by the elongating conceptuses may be indicative of an important role for conceptus RBP in the transport, availability and metabolism of retinol during maternal recognition of pregnancy.

  • The translocation of ribose-binding protein (RBP) into the inverted membrane vesicles (IMV) of Escherichia coli and eukaryotic microsomes was studied using the in vitro translation/translocation system. It was found that RBP was translocated into heterologous eukaryotic microsomes co-translationally, as well as post-translationally However, RBP was translocated only past-translationally into IMV. Degradation fragments of RBP with the molar mass of 14 and 16 kDa were produced during the translocation into IMV However, the amount of the degradation products decreased and the mature form of RBP appeared in the presence of phenylmethylsulfonyl fluoride (PMSF). PMSF and GTP accelerated the translocation of RBF It was also found that SecB enhanced the post-translational translocation of RBP It appears that RBP is translocated via at least two targeting paths.

Protein Kinase A Increases DNA-Binding Activity of Testis-Brain RNA-Binding Protein

  • Testis brain RNA-binding protein (TB-RBP) is a DNA/RNA binding protein. TB-RBP is mainly expressed in testis and brain and highly conserved protein with several functions, including chromosomal translocations, DNA repair, mitotic cell division, and mRNA transport, stabilization, and storage. In our previous study, we identified TB-RBP as an interacting partner for the catalytic subunit $(C{\alpha})$ of protein kinase A(PKA) and verified their interaction with several biochemical analyses. Here, we confirmed interaction between $C{\alpha}$. and TB-RBP in mammalian cells and determined the effect of $C{\alpha}$. on the function of TB-RBP. The activation of $C{\alpha}$. increased the TB-RBP function as a DNA-binding protein. These results suggest that the function of TB-RBP can be modulated by PKA and provide insights into the diverse role of PKA.

Repeatability of New Phoria Test Using Color Chart and Color Filter (색 시표와 색 필터를 이용한 새로운 사위검사의 신뢰도)

  • Purpose: The repeatability of a new Red-Blue phoria chart test (Red-Blue phoria chart; RBP) was appreciated. Methods: Distance (5 m) and near (40 cm) heterophoria was measured in 38 visually normal subjects. Phoria tests using RBP, Howell phoria card (HP), and MIM card (MIM) were done and the repeatability of each phoria test was compared with one another. Results: The mean of horizontal deviations was that RBP was $-0.602{\pm}0.727{\Delta}$, HP was $-0.865{\pm}1.051{\Delta}$, and MIM was $-1.501{\pm}1.346{\Delta}$, at distance, and that RBP was $-2.566{\pm}2.352{\Delta}$, HP was $-2.804{\pm}2.411{\Delta}$, MIM was $-3.838{\pm}2.603{\Delta}$, at near. The repeatability was RBP>MIM>HP in distance tests and MIM>RBP>HP in near tests. Conclusions:RBP test is identified as a reliable phoria test having high repeatability.

RAGE-binding peptide-conjugated polyethylenimine as a dual-functional carrier: A RAGE-mediated gene carrier and an anti-angiogenic reagent

  • Receptor for advanced glycation end-products (RAGE) is overexpressed in various cancer cells. In this study, a RAGE-binding peptide (RBP) was conjugated to polyethylenimine (25 kDa, PEI). RBP-conjugated PEI (PEI-RBP) was characterized as a dual-functional reagent, a RAGE-mediated gene carrier and an anti-angiogenic reagent. As a gene carrier, PEI-RBP had higher transfection efficiency to the C6 glioblastoma cells than PEI. As an anti-angiogenic reagent, the pEmpty/PEI-RBP complex reduced RAGE expression on the surface of the C6 glioblastoma cells. Also, the complex reduced the VEGF expression and tube formation of endothelial cells. Therefore, PEI-RBP may be useful for development of glioblastoma therapy.

Repeatability and Reliability of a New Phoria Test Using Flashed and Auto-flashed Presentation

  • The purpose of this study was to assess repeatability and reliability of a new phoria test as compared to established phoria tests and to assess the possibility of mobile or online testing using the auto-flashed presentation. Near dissociated phoria was measured using the von Graefe method, the Howell card test, the modified Thorington test, and a new dichromatic card test (Red-Blue Phoria card test; RBP card test) on 109 subjects. The inter-examiner difference and test-retest difference were calculated. With the auto-flashed presentation of the RBP card test, near dissociated phoria was measured and the intraclass correlation coefficient and test-retest repeatability was assessed on 26 subjects. The variation in inter-examiner repeatability was the smallest for the modified Thorington test (+3.1/-2.6). The RBP card test was +3.0/-3.1, the Howell card test was +3.5/-2.7, and the von Graefe test was +6.2/-6.0. The variation in test-retest repeatability was the smallest for the RBP card test (+0.4/-1.4). The modified Thorington test was +1.3/-1.3, the Howell card test was +1.27/-1.45, and the von Graefe test was +1.59/-2.20. The ICC was 0.919 for the auto-flashed presentation of the RBP card test. The RBP card test is considered a highly repeatable method clinically and can be made a useful application for testing in mobile or online.
