Generally, it is known that the composition of the cation of the dog's RBCs is high in potassium(K) and low in sodium(Na). However, it is reported that certain kinds of dogs have HK, HG phenotype which contains a large amount of reduced glutathione(GSH) by the effect of Na-K pump on the cell membrane of RBC with high concentration of K and low concentration of Na. Although this HK phenotype is not regarded as a disease, it is supposed to be an important assignment to examine the distribution and the occurrence rate of the dogs that contain HK cell in their RBCs for the proper clinical treatments as these HK dogs are very sensitive to aromatic disul-fide or onions and have a tendency to cause hemolysis. Accordingly, present study was performed to measure the concentration of K, Na and GSH in the RBCs of Jindo dogs and that of Dosa dogs at the same time.

The purpose of this study is to investigate the effects of 80% ethanol grape leaf extract (VGLE) and resveratrol (VGLR) from Vitis romaneti on antioxidant of red blood cells (RBC) of rat and efficacy of blood flow improvement in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. RBC oxidative hemolysis and plasma lipid peroxidation induced by the aqueous peroxyl radical generator 2,2'-azobis(2-amidinopropane) dihydrochloride were significantly suppressed by VGLE or VGLR in a dose-dependent manner. The $FeCl_3$ treatment seriously damaged the carotid artery: the walls of the artery and blood flow. However, VGLE or VGLR administration has ameliorated the blood flow and suppressed thrombus in blood vessels. These results suggest that VGLE or VGLR ameliorates the oxidative stress of RBC and thrombosis against blood vessel damage.
Kaempferia parviflora, an herbaceous plant in the family Zingiberaceae, is popular in many tropical regions. It is called as black ginger or krachaidum in Thailand and Laos, and its raw or dried root have been used as spices and teas. The rhizomes also have been traditionally used to treat gastrointestinal disorders, ulcers, gout, dysentery, allergies and to improve physical work capacity. Recently, its anti-obesity, anti-oxidant, anti-inflammatory and blood clot-lysis activities were reported. In this study, the anti-thrombosis activity of black ginger was investigated, since improvement in blood fluidity leads to the prevention of various lifestyle-related diseases. The hot water and ethanol extract and their subsequent solvent fractions (hexane, ethylacetate, butanol fractions and water residue) were prepared, and their anti-coagulation and platelet aggregation inhibitory activities were determined, respectively. Among the black ginger extracts and their fractions, the ethylacetate fraction (EAF) of ethanol extract only showed significant extensions of blood coagulation time determined by thrombin time (TT), prothrombin time (PT), and activated partial thromboplastin time (aPTT). At 5 mg/ml concentration, TT, PT and aPTT were extended to 1.22, 1.49 and >15-folds compared to non-treatment. The EAFs of ethanol and hot water extract showed strong inhibitions against collagen-induced platelet aggregations, which are comparable to inhibitions of aspirin. Also the EAFs from black ginger did not show any hemolysis activity against human RBC up to 0.5 mg/ml. Our results suggest that the EAF of black ginger has a potential as novel anti-coagulation and ant-platelet aggregation agent. This report provides the first evidence of anti-coagulation activity of black ginger.
Pollen corrected from Quercus species is abundant in Korea and has been used to treat anemia and inflammation of the prostate gland. It is also used for hemostasis in oriental medicine. In this study, the ethanol extract of the acorn pollen and its subsequent organic solvent fractions using hexane, ethylacetate, butanol, and water residue were prepared, and their antioxidant, anti-thrombosis, and hemolysis activities were evaluated. The ethylacetate fraction of acorn pollen (EF-AP) showed the highest polyphenol content (225.0 mg/g) and strong antioxidant activity among the solvent fractions. The RC50 of EF-AP against DPPH, ABTS, and nitrite radicals was 72.2, 27.7, and 62.6 ㎍/ml, respectively. The blood coagulation activities of the all-solvent fractions determined by thrombin time, prothrombin time, and activated partial thromboplastin time were negligible up to 5 mg/ml. Platelet aggregation inhibitory activities were observed in the EF-AP, butanol fraction, and water residue, whereas hexane fraction induced strong platelet aggregation. The EF-AP has no hemolysis activity against human RBC up to 1 mg/ml. UPLC/MS/MS analysis of the EF-AP revealed that rutin, isoquercitrin, and astragalin are major compounds for antioxidant and anti-thrombosis activities. Our results suggest that EF-AP could be developed as a noble antioxidant and anti-thrombosis agent.

Dystaenia takesimana is an endemic plant found only in Korea, especially on Ulleung Island. The leaves and roots of D. takesimana have been used as food, forage, and oriental medicine. Anti-bacterial, anti-inflammation, antioxidant, and α-glucosidase inhibition biological activities have been reported in the plant's root extract. However, studies concerning the anti-thrombosis activities of D. takesimana are still in the rudimentary stage. In this study, the extracts of the leaf (DT-L), stem (DT-S), and root (DT-R) of D. takesimana were prepared using 70% ethanol, and their anti-thrombosis activities were evaluated. DT-L extracts (0.25 mg/ml) showed strong inhibitions against platelet aggregation, comparable to aspirin, with strong radical scavenging activities. Furthermore, the DT-L extract did not show any RBC hemolysis up to 1 m/ml. The ant-coagulation and antioxidant activities of the DT-S extract were ignorable. While the DT-R extract showed inhibitions against thrombin and blood coagulation factors, it also showed strong platelet aggregation. This is a first report of the anti-thrombosis activities of D. takesimana, and our results suggest that DT-L could be developed as a valuable bioresource for high value-added products.

To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.
This study was designed to develop the functional beverage using Salicornia herbacea L (SH). To evaluate the anti-oxidation and anti-thrombosis activities of SH, the ethanol extracts of leaf (LF), stem (ST) and seed (SD) of SH were prepared. Components analysis showed the total polyphenol of LF > ST > SD, and the total flavonoid of LF > SE > ST, respectively. Amounts of total sugar and reducing sugar of LF and ST were similar, but the SD had only 1/10 of sugar contents of LF. Anti-oxidation activities of each extract determined by calculated RC50 showed LF > SD > ST for DPPH anion, nitrite and reducing power, and LF = SD > ST for ABTS cation scavenging activity, respectively. In anti-coagulation assay, SD showed strong inhibition activity against thrombin and blood coagulation factors. All the extracts had no platelet aggregation activities against human platelet and no hemolysis against human RBC. Our results suggest that the SD from SH has a great potential as a new anti-coagulation agent.

Nuruk has been used as fermentation starter in the alcohol industry for some time in Korea. Various bioactivities, such as antiproliferative and anti-obesity, of R4 nuruk made from Rhizopus oryzae KSD-815 have been previously reported. In this study, the hot water and ethanol extract of R4 nuruk and their subsequent organic solvent fractions were prepared, and their antithrombosis activities were evaluated. The ethylacetate fraction showed strong anti-coagulation activity, and the ethylacetate and butanol fraction from hot water extract demonstrated platelet aggregation inhibitory activity, without hemolysis against human RBC. Our results suggest that R4 nuruk has the potential to act as a new anti-thrombosis agent.
Benzyl alcohol is known to have dual effect on the red blood cell shape change. At low concentration up to 50 mM benzyl alcohol transformed the shape from discocyte to stomatocyte by preferent binding to the inner hemileaflet, however, at higher concentratransformed the shape from discocyte to stomatocyte by preferential binding to the inner monolayer, however, at higher concentration above 50 mM benzyl alcohol transformed to echinocyte by affecting both monolayers. These results suggest that the effect of benzyl alcohol on the red blood cell shape and $Ca^{++}$ transport across cardiac cell membranes to assess the effects of the drug on the structures and functions of the biological cell membranes. The results are as follows: 1) Benzyl alcohol up to 40 mM caused progressive stomatocytic shap change of the red blood cell but above 50 mM benzyl alcohol caused echinocytic shape change. 2) Benzyl alcohol up to 40 mM inhibited both osmotic hemolysis and osmotic volume change of the red blood cell in hypotonic and hypertonic NaCl solutions, respectively. 3) Benzyl alcohol inhibited both Bowditch Staircase and Wood-worth Staircase phenomena at rat left auricle. 4) Benzyl alcohol at concentration of 5 mM increased $Ca^{++}-ATPase$ activity of red blood cell ghosts slightly but above S mM benzyl alcohol inhibited the $Ca^{++}-ATPase$ activity. 5) Benzyl alcohol at concentrations of 5 mM and 10 mM increased $Ca^{++}-ATPase$ activity slightly at rat gastrocnemius muscle S.R. but above 10 mM benzyl alcohol inhibited the $Ca^{++}-ATPase$ activity. Above results indicate that benzyl alcohol inhibit water permeability and $Ca^{++}$ transport across cell membranes in part via effects on the fluidity and transition temperatures of the bulk lipid by preferential intercalation into cytoplasmic monolayer and in part via other effect on the conformational change of active sites of the $Ca^{++}-ATPase$ molecule extended in cytoplasmic face.
Pre-analytical variables account for most laboratory errors and many factors affect the results from a patient. Type of tubes facilitated rapid separation and prevented hemolysis upon prolonged storage. However, there were some limitations associated with vacutainer conditions. To circumvent the problems, the comparability of complete blood cell count values was examined using various vacutainers. The results of the analysis showed a large coefficient variation of 0.24, 0.21 in the value of white blood cells and platelets, and significant correlation was observed between white blood cells, platelets, and the value of red blood cells (p<0.01). In each of the three tubes, compared to the value of platelets, white blood cells, the greatest coefficient variation was 0.27, 0.21. In correlation of the three companies, significant difference was observed in values of white blood cells, platelets, and platelet distribution width (p<0.01), however G and B, the value of platelets, and platelet distribution width were significantly lower (p<0.05). In conclusion, analysis of vacutainers showed that they were suitable for stability of these analytes under vacutainer conditions.
