Roughage Energy and Degradability Estimation with Aspergillus oryzae Inclusion Using Daisy In vitro Fermentation

  • The aim of this study was to predict the energy value and dynamic degradation of roughage in Taiwan using the $Daisy^{(R)}$. in vitro fermentation method to provide information on one of the very important nutrients for ration formulation. The second objective was to study the effects of Aspergillus oryzae (AFE) inclusion on nutrient utilization. Three ruminal fistulated dry dairy cows were used for rumen fluid and fifteen conventional forages used in dairy cattle were collected around this island. The degradability of these feedstuffs with and without AFE ($Amaferm^{(R)}$.) treatment was measured using the $Daisy^{(R)}$. in vitro method. The roughage energy values, including TDN and NEL, were calculated according to Robinson (2000). Results from the 30 h in vitro neutral detergent fiber (NDF) degradability and predicted energy evaluations showed that alfalfa (among the forages) contained the highest degradability and energy values, Bermuda straw having the lowest. Peanut vines and corn silage contained higher energy values and the lowest value found in Pangola and Napier grasses among the locally produced forages. Pangola and Napier grasses had lower values than most imported forages except Bermuda straw. Among the by-products, wheat middling contained the highest NDF degradability, while rice bran contained the richest energy value due to its high oil content. From the dynamic dry matter (DM), organic matter (OM), acid detergent fiber (ADF) and neutral detergent fiber (NDF) degradation, corn silage contained the highest effective degradation among the local forages; wheat middling (among the by-products) degraded the fastest in DM, OM, ADF and NDF and showed the highest effective degradability. AFE inclusion was inconsistent among the forages. Alfalfa hay showed significantly increased 30 h NDF degradability and energy values, Pangola hay, Napier grass and brewer's grains showed decreased degradability and energy values. AFE inclusion increased the DM, OM and NDF degradation rate in most forage, but only increased the DM degradation rate in sorghum distiller's grains, the OM degradation rate in bean curd pomace and the NDF and ADF degradation rates in soy pomace (among the by-products).

Rhizopus Soft Rot on Lily Caused by Rhizopus oryzae in Korea (Rhizopus oryzae에 의한 백합 무름병)

  • Rhizopus soft rot of lily (Lilium longiflorum) caused by Rhizopus oryzae was observed in the experimental field in Taean Lily Experiment Station in Korea, 2012. The typical symptoms were water-soaked lesions on bottom stem and leaf rot. The lesion rapidly expanded and the plant was softened totally. The fungus grew vigorously at an optimum temperature ($25^{\circ}C$) and brownish colony and black sporangia were formed on potato dextrose agar medium. Sporangiophores formed on end of sporangia were sub-globose, brownish and $6-10{\mu}m$ in size. Sporangia were globose, blackish and $87-116{\mu}m$ in size. Sporangiospores were irregularly oval and sub-globose, brownish $4-8{\mu}m$ in size. On the basis of mycological characteristics, analyzing sequences of internal transcribed spacer region of ribosomal DNA, and pathogenicity test on host plants, the causal fungus was identified as R. oryzae. This is the first report of Rhizopus soft rot on lily caused by R. oryzae in Korea.

  • The 98 and 116 isolates of Pyricularia oryzae collected from Gyeong-nam Province in 1982 and 1983 were differentiated by using Korean differential rice cultivars. Twelve races from98 isolates and 11 races from 116 isolates of P. oryzae were classified indicating that KJ races group was $87\%$ and KI group was $13\%$ in total. The races KJ301 and KJ 401 were ca. $50\%$ in frequency among races identified.

  • Two-component systems (TCSs) are critical to the pathogenesis of Xanthomonas oryzae pv. oryzae (Xoo). We mutated 55 of 62 genes annotated as responsive regulators (RRs) of TCSs in the genome of Xoo strain PXO99A and identified 9 genes involved in Xoo virulence. Four (rpfG, hrpG, stoS, and detR) of the 9 genes were previously reported as key regulators of Xoo virulence and the other 5 have not been characterized. Lesion lengths on rice leaves inoculated with the mutants were shorter than those of the wild type and were significantly restored with gene complementation. The population density of the 5 mutants in planta was smaller than that of PXO99A at 14 days after inoculation, but the growth curves of the mutants in rich medium were similar to those of the wild type. These newly reported RR genes will facilitate studies on the function of TCSs and of the integrated regulation of TCSs for Xoo pathogenesis.

Bioconversion of Cyanidin-3-Rutinoside to Cyanidin-3-Glucoside in Black Raspberry by Crude α-ʟ-Rhamnosidase from Aspergillus Species

  • Cyanidin-3-glucoside (C3G) has been known to be more bioavailable than cyanidin-3-rutinoside (C3R), the most abundant anthocyanin in black raspberry (Rubus occidentalis). The aim of this study was to enhance the bioavailability of anthocyanins in black raspberry by cleaving ʟ-rhamnose in C3R using crude enzyme extracts (CEEs) from Aspergillus usamii KCTC 6956, A. awamori KCTC 60380, A. niger KCCM 11724, A. oryzae KCCM 12698, and A. kawachii KCCM 32819. The enzyme activities of the CEEs were determined by a spectrophotometric method using ρ-nitrophenyl-rhamnopyranoside and ρ-nitrophenyl-glucopyranoside. The CEE from A. usamii had the highest α-ʟ-rhamnosidase activity with 2.73 U/ml at 60℃, followed by those from A. awamori and A. niger. When bioconversion of C3R to C3G in black raspberry was analyzed by HPLC-DAD, the CEEs from A. usamii and A. awamori hydrolyzed 95.7% and 95.6% of C3R to C3G, respectively, after 2 h incubation. The CEEs from A. kawachii and A. oryzae did not convert C3R to C3G in black raspberry.

Enhanced Anti-oxidant Activity Effects of Smilax china L. Rhizome Water Extracts Added with Its Fermented Leaf Water Extracts (발효 청미래덩굴잎 추출물의 혼합에 의한 토복령의 항산화활성 증진효과)

  • To evaluate the improving effects of antioxidant activity, we observed antioxidant capacities such as electron donating ability (EDA), Ferric reducing antioxidant power (FRAP), inhibitory activity of xanthine oxidase (XO) and aldehyde oxidase (AO), and sensory characteristics on mixture of Smilax china L. root water extract added with water extract of fermented S. china L. leaf by Aspergillus oryzae (FSCL). Those contents of mixture with higher ratio of FSCL were proportionally high. And OD475 of mixture with higher ratio of FSCL was almost proportionally high ($R^2=0.9850$). Antioxidant capacities of EDA and FRAP of the mixture was higher than that of non-mixture. In addition, XO inhibitory activity ($IC_{50}$) of A (1.19) was 59.80% higher than that of F (2.96), and the activity of mixture by the higher ratio of FSCL was proportionally low ($R^2=0.9490$). Taste acceptability of A was slightly higher than that of F, whereas that of C was highest. And color acceptability of 40-80% mixture was higher than those of A, F, and B. Overall acceptability of C and D was highest than those of others. Moreover, hot water extract of S. china L. leaf fermented with A. oryzae was maroon color, which looks like Puerh tea style, and mixture of S. china L. root extract added with hot water extract of S. china L. leaf was high acceptability of beverage. These results suggest that mixture of extract of S. china L. root and hot water extract of S. china L. leaf fermented with A. oryzae could improve antioxidant activities.

Analysis of Genomic Structure of an Aflatoxin Biosynthesis Homologous Gene Cluster in Aspergillus oryzae RIB Strains

  • To investigate non-aflatoxin-production of A. oryzae at the molecular level, an aflatoxin biosynthesis gene homolog cluster of RIB 40 was analyzed. Although most genes in the corresponding cluster exhibited from 97 to 99 % similarity to those of Aspergillus flavus, three genes shared 93 % similarity or less. In addition, although slight expression of aflR, positive transcriptional regulator gene, was detected in some A. oryzae strains having seven aflatoxin biosynthesis homologous genes, other genes related to aflatoxin production were not detected. RIB strains were mainly divided into group 1, having seven aflatoxin biosynthesis homologous genes (aflT, nor-i, aflR, norA, avnA, verB, and vbs), and group 2, having three homologous (avnA, verB, and vbs). Partial aflatoxin homologous gene cluster of RIB62 from group 2 was sequenced and compared with that of RIB40 from group 1. RIB62 showed a large deletion upstream of ver-1 with more than half of the aflatoxin homologous gene cluster missing including aflR, a positive transcriptional regulatory gene. Adjacent to the deletion of the aflatoxin homologous gene cluster, RIB62 has a unique sequence of about 8kb and a telomere. Southern analysis of A. oryzae RIB strains with four kinds of probe derived from the unique sequence of RIB62 showed that all group 2 strains have identical hybridizing signals. Polymerase chain reaction with specific primer set designed to amplify the junction between ver-1 and the unique sequence of RIB62 resulted in the same size of DNA fragment only from group 2 strains. Based on these results, we developed a useful genetic tool that distinguishes A. oryzae group 2 strains from the other groups' strains and propose that it might have differentiated from the ancestral strains due to chromosomal breakage.

A Study on Removal Efficiency of Cd by using Chitosan Complex isolated from Aspergillus oryzae (Aspergillus oryzae로부터 분리한 chitosan복합체에 의한 카드뮴 제거 효율에 관한 연구)

  • The removal efficiency of cadmium by chitosan complex isolated from Aspersillus oryzae was investigated through laboratory experiments. The results of the study are as follows. The adsorption kinetics of cadmium was reached the adsorption equilibrium in approximately 20 minutes and the removal efficiency was showed 95.8%. The effect of temperature on cadmium adsorption by chitosan complex shows that as the temperature increased, the amount of cadmium adsorption per unit weight of chitosan complex increased. The correlation between amount of cadmium adsorption per unit weight of chitosan complex and temperature was obtained through the coefficient of determination($R^2$). $R^2$ values was 0.854(p<0.05). A linearized Freundlich equation was used to fit the acquired experimental data. As a result, Freundlich constant, the adsorption intensity(1/n) was 0.550, and the measure of adsorption(k) was 2.181. So, it was concluded that adsorption of cadmium by chitosan complex is effective.

  • Twenty-six N-substituted-5-chloro-1,3-dimethylpyrazole-4-carboxamide were synthesized and their antifungal activity against Rhizoctonia solani, Pyricularia oryzae, Botrytis cinerea and Colletotricum gleosporiodes was compared. N-phenyl-5-chloro-l,3-dimethylpyrazole-4-carboxamides having electron releasing group at the meta position of phenyl ring demonstrated good fugicidal activity against R. solani.

Identification and Pathogenicity of Rhizoctonia spp. isolated from Turfgrasses in Golf Courses in Korea (골프장 잔디에 병을 일으키는 Rhizocatonia의 동정 및 병원성)

  • Turfgrass Rhizoctonia blight is a severe disease in golf courses in Korea. Attempts were made in 1989 to 1994 to identify the Rhizoctonia species associated with turfgrass blights and also to examine their epidemiology. A total of 120 Rhizoctonia isolates collected were identified as R. solani AG1, R. solani AG2-2, R. cerealis(AG-D) and R. oryzas from brown patch, large patch, yellow patch and white patch, re-spectively. R. solani AG1 was mostly associated with brown patch of cool-season grasses. and most frequently isolated in June through July and also in September. R. solani AG2-2 was isolated exclusively from zoysiagrasses from April to November, most frequently in June through July and October through November. R. cerealis was isolated frequently from both creeping hentgrass in March through April and in November, and zoysiagrass in April and July. Thermophilic R. oryzae was isolated only from creeping bentgrass in August, although with very low frequency. R. solani AG2-2 was strongly pathogenic specifically to Korean lawngrasses(Zoysia japonica, Z.matrella, Z. tenuifolia), but non-pathogenic to creeping bentgrass(Agrostis palustris), bermudagrass (Cynodon dactylon), Kentucky bluegrass(Poa pratensis), perennial ryegrass(Lolium prenne), and creeping red fescue(Festuca rubra subsp. ruhra L.). R. cerealis was strongly pathogenic to zoysiagrass and bentgrass only, but was isolate-specific i.e., from non-pathogenic to pathogenic, for other turfgrasses. The mycelial growth was optimum at relatively high temperature ranges of 25~30$^{\circ}C$ for R.solani AG1, AG2-2 and R. oryzae, while the mycelial growth of R. cerealis was initiated at $^{\circ}C$ and almost ceased at or above $^{\circ}C$.
