Distribution Types of the Relict Conifer Community and the Approach for the Ecological Management in Ulleung-Island (울릉도에 자생하는 침엽수류 유존군락의 분포유형과 생태적 관리방안에 대한 연구)

  • Distribution types of native conifers (Juniperus chinensis, Pinus parviflora, Tusga sieboldii and Taxus cuspidata var. latifolia) were studied by phytosociological investigation and ZM method in Ulleung Island, South Korea. Two main types were divided maritime vegetation (Juniperus chinensis forest) and mountain vegetation (Taxus cuspidata var. latifolia forest and Pinus parviflora-Tusga sieboldii forest). The former was divided into sea cliff distribution (J-SC) and sea ridge distribution (J-SR) type. The latter was classified 7 distribution types; Taxus cuspidata var. latifolia forest was rock distribution (Ta-R) and mountain slope distribution (Ta-MS) type, and Pinus parviflora-Tusga sieboldii forest was rock distribution (P T-R), upper and ridge distribution (P T-UR, 3 units sub-types:1sub, 2sub, 3sub), and Mountain slope distribution (P T-MS) type. It was considered that J-SC, Ta- R, and P T-R were maintained by topographic climax, but J-SR, Ta-MS, P T-UR and P T-MS were the process of vegetation succession. Distribution types of topographic climax are entrusted to process of vegetation succession. Types in the process of vegetation succession will be needed tending of forest to promote saplings growth and seedlings germination. Especially in order to restore Tsuga sieboldii forest should be afforest and make forest gap because It is mid shade tolerant tree and purity percentage of its seed is 1~2%. It was considered that the composition of group mixture forest constituted Pinus parviflora, Tsuga sieboldii, Taxus cuspidata, Camellia japonica, Machilus thunbergii and Acer okamotoanum, etc. will be able to restore native vegetation, after take the form of forest gap by strong thinning and pruning of Pinus thunbergii forest.

Monitoring for optimum antioxidant extraction condition of Gugija (Lycium chinensis Mill) extract (구기자 추출물의 최적 항산화 추출조건 모니터링)

  • This study optimized the extraction of antioxidants from Gugija (Lycium chinensis Mill). To determine operational parameters, including ethanol concentration ($X_1$, 0~80%) and extraction time ($X_2$, 1~5 hr), response surface methodology was applied to monitor yield, anthocyanins, flavonoids and DPPH radical scavenging activity. Coefficients of determinations ($R^2$) of the models were range of 0.8645~0.9859 (p<0.01~0.1) in dependant parameters. Yield of Gugija extracts was maximized 23.12% in extraction conditions of 4.22 h at 8.25% ethanol. Anthocyanins was maximized 1.43 (OD in 530 nm) in extraction conditions of 3.06 h at 79.98% ethanol. Flavonoids was maximized $3,100{\mu}g/100g$ in extraction conditions of 3.37 h at 67.02% ethanol. DPPH radical scavenging activity was maximized 96.93% in extraction conditions of 1.67 h at 69.81% ethanol. Optimum extraction conditions (2.5 h extraction at 70% ethanol) were obtained by superimposing the contour maps with regard to anthocyanins, flavonoids and DPPH radical scavenging activity of Gugija. Maximum values of anthocyanins, flavonoids and DPPH radical scavenging activity in optimum extraction condition were 1.0080 (OD in 530 nm), $3,145{\mu}g/100g$, 96.96%, respectively. But values of anthocyanins, flavonoids and DPPH radical scavenging activity in water extraction condition (1 h at water) were 0.4652 (OD in 530 nm), $1,633{\mu}g/100g$, 86.98%, respectively.

Inhibition of Nitric Oxide Production and Hyaluronidase Activities from the Combined Extracts of Platycodon grandiflorum, Astragalus membranaceus, and Schisandra chinensis (길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과)

  • In this study, the optimal extraction conditions for three medicinal herbs as functional sources against inflammatory and arthritic diseases were developed. Traditional medicinal herbs were screened for their inhibition of hyaluronidase (HAse) activity and nitric oxide (NO) synthesis. For the screening of anti-inflammatory properties, ethanolic extracts of 53 species of traditional medicinal herb were examined. We confirmed that Astragalus membranaceus (A.R.), Schisandra chinensis (S.F.), and Platycodon grandiflorum (P.G.) inhibit NO production. For extraction from all three herbs simultaneously, an ethanol concentration of 95%, a 1:2:1 mixture ratio, and at 50 rpm mixing speed, for over 12 h and at $30^{\circ}C$ was the best condition for optimal extract yield and NO inhibition effects. HAse inhibition from the three herb extraction was three fold higher than single samples. The ethanol extracts were fractionated with various solvents (n-hexane, chloroform, ethyl acetate, n-butanol, and water). The ethyl acetate-soluble fraction of the herb mixture showed the highest extract yield (13%) and NO inhibition effects (73%). In conclusion, this study provides experimental evidence that a mixture of P.G., A.R., and S.F. could be used as a source of antioxidant ingredients in the food industry.

Monitoring of antioxidant activities with dried Gugija (Lycium chinensis Mill) extraction (건조 구기자의 추출에 따른 항산화 효능 모니터링)

  • Thise study aimed to determine the optimum antioxidant extraction conditions of dried Gugija (Lycium chinensis Mill). To determine the operational parameters, including ethanol concentration ($X_1$, 0~80%) and extraction time ($X_2$, 1~5 hr), a response surface methodology was applied to monitor brown color intensity, total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity. Coefficients of determinations ($R^2$) of the models were 0.8486~0.9214 (p<0.05~0.1) in dependent parameters. Brown color intensity of Gugija extracts reached a maximum of 0.75 (OD in 420 nm) under extraction conditions of 2.88 hr in 78.10% ethanol. Total phenolic compounds reached a maximum of $2,355{\mu}g$ under extraction conditions of 4.94 hr in 30.17% ethanol. ABTS radical scavenging activity was 13.83% at 4.61 hr and 16.21% ethanol. $Fe^{2+}$ chelating activity showed a maximum of 58.54% under extraction conditions of 3.39 hr in 0.76% ethanol. Optimum extraction conditions (5 hr extraction in 15% ethanol) were obtained by superimposing the contour maps with regards to total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity of dried Gugija. Maximum values of total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity under optimum extraction condition were $2,397{\mu}g$, 15.62% and 54.78%, respectively.

Growth and Medical Constituents of Saururus chinensis Baill as Affected by Different Amounts of Nitrogen Fertilizer Application (질소 시비량이 삼백초 생육 및 성분함량에 미치는 영향)

  • This study was conducted to investigate the selected chemical properties of soils in Saururus chinensis Baill (Chinese lizard's tail) cultivation fields to provide optimal fertilizer application rates and to examine the growth and pharmaco-consitituents of Saururus chinensis Baill as influenced by different amounts of nitrogen (N) fertilizer applications. Based on the results of selected soil chemical properties in 37 cultivation sites of the plant, soil pH, organic matter content, and exchangeable $K^{+}$ concentration were lower than optimal values for cultivating general medicinal crops even though relatively high standard deviations were found in some of the values. At the harvesting stage of the plant aerial parts, soil pH, electrical conductivity (EC), available $P_2O_5$, and exchangeable $Ca^{2+}$, $Mg^{2+}$, $Na^{+}$ decreased as comparing with those before transplanting the plant, whereas the concentration of exchangeable $K^{+}$ increased in the plot treated with N 100% and compost. Fresh weight of the plant aerial parts were highest, 492.5kg/10, in the N 100% treatment plot. Correlation equation between N levels treated (X) and yield of the plant aerial parts (Y) presented as $Y=-2.1609X^2+30.082X+344.12$($R^2= 0.7113$) and the optimal rate of N fertilizer application for the plant was 6.6kg/10a. Carbon concentrations in the plant were not different among the different N levels applied. N and K concentrations in the plant were highest in the plot of N 100% with compost applications, the highest P concentration was in N 100% plot, and the highest Ca and S concentrations were in N 200% plot. Quercetin and quercitrin were highest in the N 150% plot and tannin was highest in N 100% or N 100% with compost application plot.

Multi-loci Molecular Characterisation of Endophytic Fungi Isolated from Five Medicinal Plants of Meghalaya, India

  • The phylogenetic relationships of the most dominant and morphologically cryptic endophytic fungal isolates from each of five selected medicinal plants, namely Potentilla fulgens, Osbeckia stellata, Osbeckia chinensis, Camellia caduca, and Schima khasiana of the biodiversity rich state of Meghalaya, were assessed with random amplification of polymorphic DNA and PCR-restriction fragment length polymorphism profiles. Sequencing of the internal transcribed spacer 1, small subunit rRNA and partial ${\beta}$-tubulin gene fragments was also conducted to determine the phylogenetic relationships of these isolates with fungal sequences available in Genbank, NCBI. The identity of the fungal isolates is suggested based on the molecular phylogenetic data.

Phytogeographical Distribution and Characteristics of Korean-native Anacardiaceae (한국산 옻나무과의 지리적 천연분포와 종의 특징)

  • This study was conducted to clarify not only the phytogeographical distribution but also species characteristics of Korean-native Anacardiaceae. 6 species of I genus were ascertained from herbarium specimens and field survey of natural population. Rhus verniciflua have been mainly cultivated in Wonju region, Kangwon-do and Hamyang region, Kyeongsangnam-do, could be cultivated in all parts of Korea, especially more in cold area. R. trichocarpa was distributed in all parts except for some far-islands of South in Korea. Petiole was especially reddish. Fruits are matured in June-July, and have stiff trichomes. R. sylvestris was widely distributed in the subtropical-temperate and temperate zone, and mainly distributed in the seashore and islands under 36$^{\circ}$ in Korea. Leaflets and petiole was reddish, and fruits were glabrous. R succedanea was only distributed in Jeju-do and several islands which is subtropical zone in Korea. Leaflets were lanceolate to oblong, characterized by little trichomes. R.chinensis is a ubiquitous tree which is widely distributed in the temperate zone, in Korea, and ranged perpendicularly to 800-900m. Inflorescense was characterized by apical site. Fruit was covered with dense short hairs and sometimes milky latex. R. ambigua was only distributed in subtropical zone, native in Kwang-do and Sangbaek-do, Yeochon-gun, Chunlanam-do in Korea. Especially, this species was a vine, trifoliolate and most poisonous. Leaflets were coriaceous, glabrous, and revolute type.

Vaccination of Shrimp (Penaeus chinensis) against White Spot Syndrome Virus (WSSV)

  • Two structural protein genes, VP19 and VP466, of white spot syndrome virus (WSSV) were cloned and expressed in Sf21 insect cells using a baculovirus expression system for the development of injection and oral feeding vaccines against WSSV for shrimps. The cumulative mortalities of the shrimps vaccinated by the injection of rVP19 and rVP466 at 15 days after the challenge with WSSV were 50.2% and 51.8%, respectively. For the vaccination by oral feeding of rVP19 and rVP466, the cumulative mortalities were 49.2% and 89.2%, respectively. These results show that protection against WSSV can be generated in the shrimp, using the viral structural protein as a protein vaccine.

Quality characteristic of Omija (Schizandra chinensis Baillon) seed oils by roasting conditions and extraction methods (볶음 조건 및 추출 방법에 따른 오미자씨유의 품질 특성)

  • The influence of different roasting temperatures, times and extraction methods on the quality characteristics of Omija (Schizandra chinensis) seed oils was investigated. Roasted Omija seeds were divided into five groups based on roasting temperature-time conditions: no roasting (Raw) and roasting [R11: $150^{\circ}C$, 10 min, R12: $150^{\circ}C$, 20 min, R21: $250^{\circ}C$, 10 min, R22: $250^{\circ}C$, 20 min (R22)]. Oils from each of the raw and roasted Omija seeds were obtained by solvent (n-hexane) and press (machine) extraction. The $L^*$ values decreased, but the $a^*$ and $b^*$ values increased with increasing the roasting temperature and time. The $L^*$ values were lower in the press-extracted oils than in the solvent-extracted oils. The peroxide value (POV) of Omija seed oils decreased with increasing the roasting temperature-time values. The POV value was higher in the press-extracted oils than in the solvent-extracted oils. ABTS (2, 2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)) radical inhibition of Omija seed oils was higher in the solvent-extracted oils than in the press-extracted oils, but there were no significant differences between the two oils. The four major kinds of fatty acid methyl esters detected in Omija seed oils were methyl butyrate, methyl hexanoate, methyl arachidate, and methyl eicosanoate. In conclusion, Omija seed oils obtained by solvent extraction and at higher roasting temperature-time values were more effective antioxidants.

The Composition of Dietary Fiber on Brassica Vegetables (Brassica 쌈샐러드 채소류의 일반성분과 식이섬유소에 관한 연구)

  • This study was conducted to determine the several structural carbohydrate of 9 kinds of brassica vegetables. The samples were dried at 6$0^{\circ}C$ for 24 hrs and ground to pass a 0.5 mm screen. The crude protein and crude fat contents of brassica vegetables were 2∼3 times higher than those of grain as dry matter basis. However, the crude ash content of brassica vegetables was 7 times higher than that of grain. Total dietary fiber (TDF) was ranged from 24.26% (Narinosa) to 47.33% (Chinensis) as dry matter basis. Insoluble dietary fiber (IDF) was ranged from 17.75% (Narinosa) to 26.81% (Toscana) as dry matter basis. Soluble dietary fiber (SDF) was ranged from 3.20% (Toscana) to 23.45% (Narinosa) as dry matter basis. The correlation of brassica vegetables was r=0.30 between TDF and IDF, r=0.89 between TDF and SDF (p<0.01), r =0.25 between TDF and CHO, and r=0.29 between DFi and NDF (p<0.05), respectively.
