Cloning and Characterization of a Gene Cluster for Cyclohexanone Oxidation in Rhodococcus sp. TK6

  • A gene cluster for cyclohexanone oxidation was cloned from Rhodococcus sp. TK6, which is capable of growth on cyclohexanone as the sole carbon source. The 9,185-bp DNA sequence analysis revealed seven potential open reading frames (ORFs), designated as ssd-chnR-chnD-chnC-chnB-chnE-partial pcd. The chnBCDE genes encode enzymes for the four-step conversion of cyclohexanone to adipic acid, catalyzed by cyclohexanone monooxygenase (ChnB), $\varepsilon-caprolactone$ hydrolase (ChnC), 6-hydroxyhexanoate dehydrogenase (ChnD), and 6-oxohexanoate dehydrogenase (ChnE). Furthermore, the presence of a regulatory element in the downstream region of the chnD gene supports the notion that chnR is a putative regulatory gene. Among them, the activity of ChnB was confirmed and characterized, following their expression and purification in Escherichia coli harboring the modified chnB gene (chnB gene with 6 successive codons for His at the 3' terminus).

  • 울산광역시 울주군 두동면 천전리 306번지 일대에 건설되는 대곡댐은 높이 52m, 길이 190m의 표면차수벽형 석괴댐이다. 대곡댐 건설사업과 관련하여 여수로 좌안사면 굴착공사를 시행 중 2000. 7. 23 13:00~7. 24. 04:00 67.0mm의 강우가 있은 후 Sta. No. 1~2, EL. 137~134m 지점에 균열이 발생하였다. 안정대책을 수립하고자 지표지질조사 및 평사투영법에 의한 안정성을 검토하고 굴착 후 예상되는 불안정 요소를 고려하여 사면의 부분적인 추가절취 및 Pre-stress가 가능한 SSL PC Anchoring에 의한 억지공법을 수립, 시공하였다.

  • Some abnormal signals diagnostics and analysis through an important equivalent circuits modeling for passive elements under severe accident conditions have been performed. Unlike the design basis accidents, there are inherently some uncertainties in the instrumentation capabilities under the accident conditions. So, the circuit simulation analysis and diagnosis methods are used to assess instruments in detail when they give apparently abnormal readings as an accident alternative method. The simulations can be useful to investigate what the signal and circuit characteristics would be when similar to a variety of symptoms that can result from the environmental conditions such as high temperature, humidity, and pressure condition. In this paper, a new simulator through an analysis of the important equivalent circuits modeling under temperature accident conditions has been designed, the designed simulator is composed of the LabVIEW code as a main tool and the out-put file of the Multi-SIM code as an engine tool is exported to in-put file of the LabVIEW code. The procedure for the simulator design was divided into two design steps, of which the first step was the diagnosis method, the second step was the circuit simulator for the signal processing tool. It has three main functions which are a signal processing tool, an accident management tool, and an additional guide from the initial screen. This simulator should be possible that it could be applied a output signal analysis to some transient signal by variation of the resistance passive elements in the R-L-C equivalent circuit modeling under various degraded conditions in NPPs.

  • An original sensor system based on Fiber Bragg Gratings (FBG) for the strain monitoring of an adaptive wing element is presented in this paper. One of the main aims of the SARISTU project is in fact to measure the shape of a deformable wing for performance optimization. In detail, an Adaptive Trailing Edge (ATE) is monitored chord- and span-wise in order to estimate the deviation between the actual and the desired shape and, then, to allow attaining a prediction of the real aerodynamic behavior with respect to the expected one. The integration of a sensor system is not trivial: it has to fit inside the available room and to comply with the primary issue of the FBG protection. Moreover, dealing with morphing structures, large deformations are expected and a certain modulation is necessary to keep the measured strain inside the permissible measure range. In what follows, the mathematical model of an original FBG-based structural sensor system is presented, designed to evaluate the chord-wise strain of an Adaptive Trailing Edge device. Numerical and experimental results are compared, using a proof-of-concept setup. Further investigations aimed at improving the sensor capabilities, were finally addressed. The elasticity of the sensor structure was exploited to enlarge both the measurement and the linearity range. An optimisation process was then implemented to find out an optimal thickness distribution of the sensor system in order to alleviate the strain level within the referred component.

  • 슬로싱 하중은 LNG 화물창의 내부 구조물에 강한 충격을 유발시킨다. 이는 슬로싱 하중에 의해 평면이나 구석 부위에 순간 압력을 증가시키기 때문에 멤브레인 타입의 화물창에서는 매우 위험하다. 멤브레인 타입의 화물창은 선체구조에 비해 매우 유연하므로, 유체-구조 상호작용은 슬로싱 하중을 받는 화물창의 구조해석에서 매우 중요한 역할을 한다. 본 연구는 유한요소해석 기법을 이용하여 LNG 유체와 KC-1 단열시스템의 경계에 대시포트를 이용하여 유체-구조 상호작용을 고려한 해석 방법에 대해 다루었다. 결론적으로 표면 완충 효과를 적용하였을 경우, KC-1 단열시스템의 폴리우레탄폼에서 발생하는 최대응력이 약 1.5 배 낮아지는 효과가 있음을 알아내었다.

  • 본 논문은 지진발생시 빌딩프레임상의 각 부재의 흡수에너지율이 일정하도록 유도하는 다시말하면 각각의 부재의 손상지수값이 고루게 분포토록 하는 새로운 설계법인 손상제어 전산설계법의 유용성을 입증코져 하는 것이다. 이를 위하여 우선 사용된 기본적인 이력모델 및 손상모델의 정확성을 평가하기 위하여 one-bay one-stody 프레임의 실험적인 하중-변위곡선을 해석적으로 재생하여 비교분석하였다. 그리고 본 설계법을 각종 프레임에 적용한 결과 1) 구조물의 특징 및 사용된 지진형상에 관계 없이 손상제어 설계된 프레임은 일반적으로 종래의 방법으로 설계된 프레임보다 같은 지진하중하에서 더 작은 손상값으로 저항하였으며, 2) 손상제어 설계된 프레임의 하층부 부재들은 더 큰 항복강도를 나타내는 현상을 보였으며 상층부 부재들은 반대 현상을 보였다.

붉은 털 원숭이의 뇌조직에서 CCDC94 유전자 대체 전사체의 분자적 분석 (Molecular Analysis of Alternative Transcripts of CCDC94 Gene in the Brain Tissues of Rhesus Monkey)

  • 붉은 털 원숭이의 유전체는 인간의 것과 93% 정도로 동일하여, 진화적 연구 및 생물의학적 연구에 널리 활용되고 있다. 숙주 개체 내로 가동성 유전인자(TEs)의 삽입은 유전자 전사체의 다양성과 발현양상을 다르게 만든다. 본 연구에서는 붉은 털 원숭이의 뇌 조직으로부터 만든 cDNA 라이브러리에서 112개 전사체를 동정하여 분석하였다. 하나의 전사체 R54는 인간과 원숭이의 다양한 조직에서 유전자 발현양상을 비교분석 해 본 결과 서로 다른 패턴을 보여 주었다. 이러한 현상은 가동성 유전인자인 L2A의 삽입으로 인한 스플라이싱 도너 사이트가 변화된 것으로 생각된다. 따라서, 영장류의 진화과정에 있어 유전체 내로 TEs의 삽입은 전사체의 다양성과 유전자 발현 조절에 변화를 주는 것으로 시사된다.

Strengthening of Cutouts in Existing One-Way Spanning R. C. Flat Slabs Using CFRP Sheets

  • Openings in slabs are usually required for many different applications such as aeriation ducts and air conditioning. Opening in concrete slabs due to cutouts significantly decrease the member stiffness. There are different techniques to strengthen slabs with opening cutouts. This study presents experimental and numerical investigations on the use of Carbon Fiber Reinforced Polymers (CFRP) as strengthening material to strengthen and restore the load carrying capacity of R.C. slabs after having cutout in the hogging moment region. The experimental program consisted of testing five (oneway spanning R.C. flat slabs) with overhang. All slabs were prismatic, rectangular in cross-section and nominally 2000 mm long, 1000 mm width, and 100 mm thickness with a clear span (distance between supports) of 1200 mm and the overhang length is 700 mm. All slabs were loaded up to 30 kN (45% of ultimate load for reference slab, before yielding of the longitudinal reinforcement), then the load was kept constant during cutting concrete and steel bars (producing cut out). After that operation, slabs were loaded till failure. An analytical study using finite element analysis (FEA) is performed using the commercial software ANSYS. The FEA has been validated and calibrated using the experimental results. The FE model was found to be in a good agreement with the experimental results. The investigated key parameters were slab aspect ratio for the opening ratios of [1:1, 2:1], CFRP layers and the laminates widths, positions for cutouts and the CFRP configurations around cutouts.

La-Co 치환량에 따른 스트론튬 페라이트의 자기특성 (Magnetic properties of Sr-ferrite by La-Co substitution)

  • 스트론튬 페라이트에 포화자화를 높이기 위하여 Fe이온에 Co를 치환하고, 원소치환에 따른 전하보상차원으로 La를 동시 치환을 실시한 $_{l-x}$ La$Sr_{x}$ /$Fe_{12-x}$ $Co_{x}$ $O_{19}$ 조성에서 치환량 X=0에서 부터 X=0.5까지 0.1씩 변화하여 하소분말을 제조하여 하소분말 특성을 조사하고, 소결조제와 함께 미분쇄한 후 자장중 성형 및 소결하여 스트론튬 페라이트 자석을 제조하여 미세구조와 자기적 특성을 조사하였다. 스트론튬 페라이트에 La-Co 치환량이 증가할수록 하소 후 이방성자장과 하소분말의 보자력을 강하게 증가시키며, 하소후 결정크기가 작아지는 것으로 나타났다. 스트론튬 페라이트 자석은 La-Co의 치환량에 따라 회귀식 $B_{r}$ $≒0.097_{i}$ $H_{c}$ +4500의 함수적 관계를 나타내고 있으며, La-Co를 치환하지 않은 $SrFe_{12}$$O_{19}$ 조성에서 잔류자속밀도는 4090 G, 보자력은 3560 Oe를 나타내었으나, La-Co치환량 0.3인 $Sr_{0.7}$ $La_{0.3}$ $Fe_{11.7}$ $Co_{0.3}$ $O_{19}$ 조성에서 잔류자속밀도는 4080 G, 보자력은 4800 Oe 특성을 나타내었다.

Effect of Growell on Performance, Organ Weight and Serum Trace Element Profile of Broilers

  • Effect of Growell a herbomineral premix was evaluated on performance, organ weight, humoral immune response, tissue and serum trace element profile of boilers. Day old 50 Babcock broiler chicks were randomly divided in two groups (C and T) and reared on deep litter system for 6 weeks of age. Chicks from group C were given basal diet while chicks from T group were fed basal diet supplemented with Growell at the rate 0.35 g/Kg. The chicks were vaccinated with Lasota strain of NCDV at $4^{th}$ and $28^{th}$ day of age. The birds receiving Growell treatment had higher body weight with better feed conversion ratio as compare to that of control chicks. Growell treatment had significantly reduced per cent weight of spleen and kidney, whereas that of bursa was increased. There was no effect of treatment on relative weight of thymus. HI antibody titer against NCDV in Growell treated chicks were higher as compare to untreated chicks indicating better humoral immune status. Growell treatment had no effect on serum Fe and Zn concentration. Dietary supplementation of Growell had significantly increased iron content of liver, kidney and muscle; zinc content of kidney and muscle; copper content of kidney and muscle and Mn content of kidney. Growell treatment improved the body weight, FCR and humoral immune status of broilers. Similarly, deposition of trace minerals in various organs was also increased in comparison to control.
