Effects of the Components and Extracts of Some Edible and Medicinal Plants on the Formation of Lipid Peroxide in Rat Liver Homogenate (식용 및 약용식물 추출물과 화합물이 흰쥐의 간장조직에서 과산화지질 생성에 미치는 영향)

  • We have studied the effect of 31 extracts and 10 components from some edible and medicinal plants on the formation of lipid peroxide in the liver homogenate of rat in vitro. The 70% acetone extracts of Allium tuberosum, Beta vulgaris var. cicla and Brassica juncea var. integrifolia, and methanol extract of Capsicum annuum decreased the formation of lipid peroxide by 33%, 58%, 62% and 56% at the concentration of 1mg/ml, respectively. And these four extracts inhibited the lipid peroxidation at the concentration of $10^{-1}$mg/ml by 17%, 46%, 49% and 45% respectively. Among the component tested, quercetin, quercitrin and isorhamnetin reduced the formation of lipid peroxide by 45%, 15% and 28% respectively at the concentration of $10^{-2}$mg/ml.

Phenolic Compounds from Japanese Anise (Illicium anisatum L.) Leaves

  • Japanese anise (Illicium anisatum L.) leaves were collected and ground after drying, then immersed with 50 % aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze dried after concentration. A portion of EtOAc (3.12 g) and $H_2O$ (6.08 g) soluble fractions were chromatographed on a Sephadex LH-20 column with various aqueous MeOH solution to isolate the compounds. Compound 1 ((+)-catechin) was isolated from EtOAc soluble fraction. Compounds 2 (quercetin), 3 (quercitrin) and 4 (2''-O-rhamnosylvitexin) were isolated from $H_2O$ soluble fraction. For the first time, quercitrin (3) and 2''-O-rhamnosylvitexin (4) of the isolated compounds were obtained from the extracts of japanese anise leaves.

Flavonol Glycosides from the Wood of Platycarya strobilacea

  • This study was carried out to investigate the constituents of Platycarya strobilacea (Juglandaceae) wood. To isolate compounds, wood was extracted with ethanol (EtOH) and then partitioned with petroleum ether, diethyl ether (Et2O) and ethyl acetate (EtOAc) successively. After partitioned, diethyl ether fraction was subjected to column chromatography with various solvent system in silica gel and/or Sephadex LH-20. Structures were elucidated by spectroscopic methods including MS, 1H, 13C and 2D-NMR experiments. Three compounds were isolated from the wood and identified as kaempferol 3-O-𝛼-L-rhamnopyranoside (afzelin, I), quercetin 3-O-𝛼-L-rhamnopyranoside (quercitrin, II), myricetin 3-O-𝛼-L-rhamnopyranoside (myricitrin, III).

Antimutagenic Effects and Isolation of Flavonoids from Humulus japonicus Extract (환삼덩굴(Humulus japonicus) 추출물의 항돌연변이 효과와 Flavonoid 성분의 분리)

  • To investigate the physiological activities of Humulus japonicus, we examined the mutagenic and antimutagenic effects of the extract and isolated the flavonoid compounds. The water and methanol extracts of Humulus japonicus did not show the mutagenicity by Ames test in the 0.5-4 mg/plate. The methanol extract showed the antimutagenic effect aganinst Salmonella typhimurium TA98 and TA100 induced by 4-nitro-o-phenylenediamine(NPD) and 4-nitroquinoline-N-oxide(NQO). The hexane fraction showed comparatively higher antimutagenic effect than other solvent fractions from the methanol extract. The 50% inhibition concentration($IC_{50}$) of hexane fraction were 2.0 mg/plate, 0.5 mg/plate in TA98 and TA100 respectively. Quercitrin and luteolin were identified in the ethyl acetate fraction from methanol extract of Humulus japonicus by TLC and HPLC.

Antiinflammatory Effect of Bioactive Compounds from Geranium nepalense (Notes) (현초에서 분리한 생리활성물질의 항염증효과 (단보))

  • The Geranium nepalense has been used traditionally for treatment of various diseases. However, the molecular studies on the effect of Geranium nepalense have not been carried out. In the present study, Quercetin, quercitrin, and afzelin were isolated from the methanol extract of Geranium nepalense were tested for their anti-inflammatory effect. The anti-inflammatory effect of the compounds was studied in lipopolysaccharide(lps)-treated mouse macrophage cells, RAW 264.7. RAW 264.7 cells were pre-incubated with isolated compounds(0, 5, 10, 20, 40, $50\;{\mu}g/ml$) for 4h and treated with $1\;{\mu}g/ml$ lps for 18h, and then the anti-inflammatory effects of compounds were determined. The results are as follows: Quercetin at various concentration inhibited the viability of Raw 264.7 from 7% to 45%, quercitrin from 25% to 80%, and afzelin from 13% to 52%. Isolated compounds showed a significant decrease in iNOS (inducible nitric oxide synthase) and COX-2 (cyclooxygenase-2). These results suggest that these compounds can be used as stable anti-inflammatory materials.

Analysis of Flavonoid Components from Machilus thunbergii Leaves (후박나무 잎 Flavonoid 성분의 분석)

  • Six flavonoid compounds from the ethyl acetate fraction of Machilus thunbergii leaves were identified by HPLC. Separation by reversed phase chromatography on u-Bondapak C18 column was achieved by isocratic elution. The contents of the major flavonoids, guaijaverin and quercitrin were about 0.48% (w/w) and 0.98% (w/w) for the methanol extract, respectively.

Phenolic Compounds from Japanese anise (Illicium anisatum L.) Twigs

  • Japanese anise (Illicium anisatum L.) twigs were collected and ground after drying, then immersed with 50% aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze-dried after condensation. A portion of EtOAc soluble fraction (5.7 g) was chromatographed on a Sephadex LH-20 column with various aqueous $MeOH-H_2O$. Compound 2 and compound 3 were isolated from fraction 8 and 5, respectively. Compound 1 and compound 4 were isolated after rechromatography of fraction 7. The isolated compounds were elucidated as (+)-catechin (1), taxifolin (2), taxifolin-3-O-${\beta}$-D-(+)-xylopyranose (3) and quercitrin (4) by spectral and literature data, and by comparison with the authentic samples. Of the isolated compounds, taxifolin (2), taxifolin-3-O-${\beta}$-D-(+)-D-xylopyranose (3) and quercitrin (4) were isolated, for the first time, from the extracts of japanese anise twigs.
