Gas Chromatographic Analysis and Cholinesterase Activity of the Essential Oil from Korean Agastache rugosa (기체크로마토그래피에 의한 한국산 배초향의 정유 분석과 Cholinesterase 억제활성)

  • The herb of Agastache rugosa (Lamiaceae) called Korean mint as a spice or Agastache Herba as a crude drug is known to contain highly fragrant volatile substances. This research aimed to establish the quantitative gas chromatography (GC) method on the essential oil of A. rugosa using the three standard compounds, estragole, methyleugenol, pulegone, and to find whether the essential oil has anti-Alzheimer's activity. The GC quantification method was established by determining the linearity of calibration curve ($R^2$), linear range, and both limit-of-detection (LOD) and limit-of-quantification (LOQ). The $IC_{50}$ of the essential oil on the activities of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) were determined to be $69.06{\pm}0.26$ and $76.71{\pm}0.58{\mu}g/ml$, respectively.

  • Partially ranked data refers to the situation in which there are p distinct objects; however each judge specifies only first s (s < p) choices. The group theoretic formulation for partially ranked data analysis was set up by Critchlow (1985). We propose a graphical method for partially ranked data by quantifying objects and judges. In a plot for judges, the interpoint distances can be interpreted as Spearman or Kendall distances between two rankings given by respective judges. Similarly, we also construct a plot for objects with a sensible relationship to the previous plot for judges. This study extends the Han and Huh (1995) quantification method of fully ranked data using Gabriel's (1971) biplot technique for multivariate data matrix.

  • The shear strength of jointed rock is influenced by effective normal stress, joint wall compressive strength, joint roughness and so on. Since joint roughness makes considerable influences on shear strength of jointed rock, many studies tried to get quantitative joint roughness parameter. Until now, Joint Roughness Coefficient, JRC proposed by Barton has been prevalently used as a rock joint roughness parameter In spite of its disadvantages. In this study, a quantification of rock joint roughness is performed using surface roughness parameter, Rs. Proposed method is applied to rock core specimens, field joint surfaces, and JRC profiles. The scale of fluctuation is introduced to extend the suggested method to the large scale field joint surface roughness. Based on the quantification of joint surface roughness, joint shear tests are performed with the portable shear box. The relationship between joint surface roughness and joint shear strength is investigated and finally, a rock joint shear strength equation is derived from these results. The equation has considerable credibility and originality in that it is obtained from laboratory tests and expressed with quantified parameter.

Bioavailability of Clonazepam in human plasma using a simple HPLC

  • We aimed at determining bioavailability of clonazepam, an anxiolytic drug, and developing a simple analysis in human blood using HPLC. A rapid and sensitive HPLC method was developed and validated using reverse-phase C18 column with retension time and limit of quantification of clonazepam being 2.58 min and 5hg/ml, respectively. Quantification was performed at 235 nm with p-hydroxybenzoic acid ethyl ester as internal standard. The method involved a simple extraction. In order to study blood level profile in time, sight volunteers were enrolled and orally took 6 mg clonazepam once. (omitted)

A rapid and sensitive HPLC method was developed and validated using reverse-phase C18 column with retension time and limit of quantification of procainamide HCl being 2.58 min and 50ng/ml, respectively. Quantification was performed at 275 nm with caffeine as internal standard. The method involved a simple extraction. In order to study blood level profile in time, eight volunteers were enrolled and orally took 250 mg procainamide HCl once. (omitted)

Modifier parameters and quantifications for seismic vulnerability assessment of reinforced concrete buildings

  • In recent years, some studies have identified and quantified factors that can increase or decrease the seismic vulnerability of buildings. These modifier factors, related to the building characteristics and condition, are taken into account in the vulnerability assessment, by means of a numerical estimation resulting from the quantification of these modifiers through vulnerability indexes. However, views have differed on the definition and the quantification of modifiers. In this study, modifier parameters and scores of the Risk-UE Level 1 method are adjusted based on the Algerian seismic code recommendations and the reviews proposed in the literature. The adjusted modifiers and scores are applied to reinforced concrete (RC) buildings in Boumerdes city, in order to assess probable seismic damage. Comparison between estimated damage and observed damage caused by the 2003 Boumerdes earthquake is done, with the objective to (i) validate the model involving influence of the modifier parameters on the seismic vulnerability, and (ii) to define the relationship between modifiers and damage. This research may help planners in improving seismic regulations and reducing vulnerability of existing buildings.

  • In this study, we used NIR spectrum method instead of conventional HPLC method to shorten the analysis and manufacturing time of the loxoprofen products. Loxoprofen mixtures with other pharmaceutical additives were prepared and evaluated by the NIR spectrometer and the HPLC system. Validation of both methods was performed for specificity, accuracy and precision. NIR spectrometer method was validated and revealed proper results for the in-process quality control in the pharmaceutical field. In conclusion, NIR spectrometry can be replaced by HPLC method.

Selection of Calibration Approaches and Their Impact on the Quantification of Unknown Samples: Case Study on Reduced Sulfur Gases (환원황화합물의 분석과 검량기준의 선택에 따른 오차발생의 특성)

  • In this study, different calibration approaches for reduced sulfur compounds (RSCs) were investigated by using thermal desorption coupled with gas chromatography (GC) and pulsed flame photometric detection (PFPD). To evaluate the effects of calibration procedures, gaseous standards of 4 RSCs ($H_2S$, $CH_3SH$, DMS, and DMDS) prepared at 10 ppm level were analyzed at 7 loading injection volumes (40, 60, 80, 100, 160, 240, and 320 ${\mu}L$). The results were then compared with calibration curves made with the Z (zero offset) and N (non-zero offset) method. The concentrations of unknown samples were then quantified by using R (ratio) method in which the slope values are compared between standards and samples. Secondly, in A (average) method, results obtained from a multi-point analysis of unknown samples were also averaged to extract representative values for each sample. Results of both experiments showed that analytical error of low molecular weight components (such as $H_2S$ and $CH_3SH$) was greatly expanded with the Z method. In conclusion, the combined application of N-A method was the more realistic approach to reduce biases in the quantification of RSCs.
