$H_2$ Production by a Purple Sulfur Bacterium Blooming in Lake Kaiike (카이이케호에서 농밀하게 분포하는 Purple Sulfur Bacterium의 수소생산)

  • $H_2$ production by Chromatium sp., a large purple sulfur bacterium blooming in lake Kaiike, under various environmental conditions was examined. Chromatium sp. produced $H_2$ only in the presence of light and $H_2$. Maximum $H_2$ production ($0.01\;{\mu}mol/hr/(mg\;dry\;cell\;weight)$) was obtained in the solution of 20 mg $H_2S-S/l$ under low light intensity (1000 lux) at $30^{\circ}C$. $H_2$ production was severely inhibited by the presence of $N_2\;or\;NH_4^+$. The rate observed for Chromatium sp. was relatively low compared to that of other phototrophic bacteria. Chromatium sp. is probably a most potent Ha producing species in lake Kaiike, since the bacterium readily produced $H_2$ photoautotrophically even at low light intensities by the application of suboptimal $H_2$ concentrations. Based on the photoautotrophic characteristics of bacterial $H_2$ production, it is suggested that Chromatium sp. can be an economic and practical species for biological $H_2$ production system, particularly in temperate region.

  • 한국 서해안과 남해안 47개소의 해수와 mud 시료로부터 광합성세균으로 유추되는 13개의 균주를 분리하였고, agar-shake tube method와 RAPD-PCR을 이용하여 4개 서로 다른 균주를 순수분리 하였다. 4개의 균주 중 폐수분해능이 가장 뛰어난 균을 선정하여, 형태학적, 배양적, 생화학적 특성 및 16S rRNA sequencing에 의한 동정결과 purple, sulfur bacteria 쪽에 가까웠으나 형태학적, 배양학적, 생화학적 특성이 purple, non-sulfur bacteria의 Rhodobacter 속에 가장 근접하여, Rhodobacter sp. EGH-24로 명명하였다.

  • A species of facultative photo-organotrophic, purple, non-sulfur bacterium was isolated from the 47 point at west and south coast of Korea in September 2001. Separated 13 samples of changes with red color under 28-32$^{\circ}C$, 3000 lux, anaerobe conditions for 7 days cultivated in basal medium. For pure isolation from 13 samples, we used agar-shake tube method (0.4 % agar) and separated 5 strains through 13-repetition test. EGH-24 and EGH-30 was identified as the same strain through the RAPD(Random Amplified Polymorphic DNA)-PCR of strain EGH-9, EGH-13, EGH-23, EGH-24, EGH-30. Four isolates cultivated in synthesis wastewater for wastewater biodegradation test. EGH-24 was selected with efficient wastwater treating strain. Based on the results obtained from morphology, nutrient requirements, major bacteriochlorophyll content, 16S-rDNA phylogenetic analysis, EGH-24 strain may be identified as a new strain of the genus Rhodobacter and named Rhodobacter sp. EGH-24.

  • Thiocapsa roseopersicina NCIB 8347은 purple sulfur bacteria이며 광합성종속영양 조건에서는 nitrogenase 효소계가 유도되어 질소를 고정하며, 수소를 발생한다. 또한 광합성독립영양 조건에서는 hydrogenase 효소계가 유도되어 3~4개 종류의 특성이 다른 hydrogenase가 membrane에 결합되어 있거나, cytoplasma에 존재하며, 이 중의 일부는 산소농도와 온도의 상승에도 비교적 안정하다. 본 연구에서는 T. roseopersicina NCIB 8347이 광합성종속영양 조건에서 수소를 생산할 수 있는 제반 배양조건을 최적화하고, nitrogenase와 일부 hydrogenase역가를 측정하여 purple non-sulfur bacteria, Rhodobacter sphaeroides KD131의 nitrogenase와 비교하여 수소생산을 최적화하였다. 할로겐램프를 8-9 $Klux/m^2$로 조사할 때와 배양온도 $26{\sim}30^{\circ}C$, 배양시간 72시간에서 균체 성장과 수소생산이 가장 높았다. T. roseopersicina NCIB 8347는 광합성 독립영양, 종속영양 조건에서 모두 성장 할 수 있었다.

For a pure isolation from 13 samples it used agar-shake tube method (0.4 % agar) and it separated 5 strains through 13-repetition test. The RAPD(Random Amplified Polymorphic DNA)-PCR result of strains (EGH-9, EGH-13, EGH-23, EGH-24, EGH-30) that EGH-24 and EGH-30 was same strain. For wastewater biodegradation test that 4 isolation strains cultivated in synthesis wastewater in 7 days. EGH-24 was high 63000 mg/L (CODcr) to 43400 mg/L (CODcr). EGH-24 was selected with efficient wast water treated strain. Based on the results obtained from morphology, nutrient requirements, major bacteriochlorophyll content, 165-rDNA phylogenetic analysis, this strain may be identified as a new strain of the genus Rhodobacter and named Rhodobacter sp. EGH-24.

Purification of Hydrogenases from Purple Sulfur Bacterium Thiocapsa roseopersicina Using Various Applications of Chromatography (홍색 유황세근 Thiocapsa roseopersicina로 부터 생산되는 Hydrogenase의 각종 크로마토그래피에 의한 정제)

  • Crude cytoplasmic fraction of phototrophic purple sulfur bacterium, Thiocapsa roseopersicina NCIB 8347, were initially prepared and purified by sonication, ultracentrifugation, ammonium sulfate fractionation and heat-treatment and it has been previously reported. Using various applications of chromatography far the purification of membrane-bound and soluble hydrogenases from heat-treated enzyme fraction were studied at present report. When the heat-treated enzyme preparation was applied to the anion column chromatography using Q-sepharose, Fraction I and II, which were extracted with the KCl 0-0.5 M gradient, showed the specific evolution hydrogenase activity 3.86 and 2.27 U/mg-protein respectively. Specific hydrogenase activitys of Fraction I and II were further increased to 4.35 and 7.46 U/mg-protein for Fraction I and to 2.49 and 4.41 U/mg-protein fur Fraction II respectively, when hydrophobic interaction column, Phenyl superose, and anion exchange column, Mono-Q, were applied. Size exclusion chromatography using superdex 200 concentrated the hydrogenase Fraction I and II to 9.19 and 7.84 U/mg-protein respectively at the final step of purification.

Characterization of the purple nonsulfur bacterium, rhodopseudomonas palustris strain P-1, degrading ferulate

  • Photosynthetic bacteria which can utilize ferulate as a sole carbon source for their metabolic activities were isolated from soils by liquid enrichment culture technique. The strain P-1 was selected by the highest capability of degrading ferulate in aerobic and anaerobic conditions. The strain P-1 was rod-shaped with its motility, strained gram negatively and could not utilize sulfur compounds. This strain has the bacteriochlorophyll a group I carotenoid and membrane structures like lamellae. As the results of physiological, morphological and cultural charactderistics, the isolate was identified as Rhodopseudomonas plaustris, one of the purple nonsulfer bacteria. The strain P-1 utilized 2mM/day in aerobic condition and 0.86 mM/day in anaerobic condition.

Isolation and Characterization of Purple Non-Sulfur Bacteria, Afifella marina, Producing Large Amount of Carotenoids from Mangrove Microhabitats

  • This study determined the effect of light intensity and photoperiod on the dry cell weight and total amount of carotenoids in four isolates of purple non-sulfur bacteria obtained from shaded and exposed microhabitats of a mangrove ecosystem in Kota Kinabalu, Sabah, Malaysia. The initial isolation of the bacteria was carried out using synthetic 112 medium under anaerobic conditions (2.5 klx) at $30{\pm}2^{\circ}C$. On the basis of colony appearance, cell morphology, gram staining, motility test, and 16S rRNA gene sequencing analyses, all four bacteria were identified as Afifella marina. One of the bacterial isolates, designated as Af. marina strain ME, which was extracted from an exposed mud habitat within the mangrove ecosystem, showed the highest yield in dry cell weight ($4.32{\pm}0.03g/l$) as well as total carotenoids ($0.783{\pm}0.002mg/g$ dry cell weight). These values were significantly higher than those for dry cell weight ($3.77{\pm}0.02g/l$) and total carotenoid content ($0.706{\pm}0.008mg/g$) produced by the isolates from shaded habitats. Further analysis of the effect of 10 levels of light intensity on the growth characteristics of Af. marina strain ME showed that the optimum production of dry cell weight and total carotenoids was achieved at different light intensities and incubation periods. The bacterium produced the highest dry cell weight of 4.98 g/l at 3 klx in 72 h incubation, but the carotenoid production of 0.783 mg/g was achieved at 2.5 klx in 48 h incubation. Subsequent analysis of the effect of photoperiod on the production of dry cell weight and total carotenoids at optimum light intensities (3 and 2.5 klx, respectively) revealed that 18 and 24 h were the optimum photoperiods for the production of dry cell weight and total carotenoids, respectively. The unique growth characteristics of the Af. marina strain ME can be exploited for biotechnology applications.

Purification of Hydrogenase from Thiocapsa roseopersicina : Effect of Ammonium Sulfate Precipitation and Heat-Treatment (홍색 유황세균 Thiocapsa roseopersicina가 생산하는 수소생산 효소의 정제 : Ammonium sulfate 침전 및 열처리의 효과)

  • Effect of $(NH_4)_2SO_4$ precipitation and heat-treatment on hydrogenase which was extracted from the cytoplasmic fraction of the phototrophic purple sulfur bacterium Thiocapsa roseopersicina NCIB 8347 was studied. Crude enzyme extract was prepared by centrifugation($28,000{\times}g$, $400,000{\times}g$) after sonication of cells grown under photosynthetic condition for 96 hrs. Various conditions of $(NH_4)_2SO_4$ precipitation and heat-treatment were examined and the effect of protein concentration was analyzed by SDS-electrophoresis between the treatments. Optimum conditions for $(NH_4)_2SO_4$ precipitation and heat-treatment for evolution hydrogenase activity were 40-60% saturation and $60^{\circ}C$ for 20 min, respectively, which exhibited the specific hydrogenase activity of 0.78 U/mg-protein. Specific hydrogenase activity was decreased to 31.6% when the heat-treatment at $60^{\circ}C$ increased from 20 min to 5 hrs.

Comparison of Hydrogenases from Clostridium butyricum and Thiocapsa roseopersicina: Hydrogenases of C. butyricum and T. roseopersicina

  • The properties related to the temperature and oxygen stability of the cytoplasmic hydrogenases from the fermentative strict anaerobic bacterium, Clostridium butyricum NCIB 9576 (Cl. butyricum), and purple sulfur phototrophic bacterium, Thiocapsa roseopersicina NCIB 8347 (T. roseopersicina), were compared. The optimum temperatures for the growth of Cl. butyricum and T. roseopersicina were 37$^{\circ}C$ and 25$^{\circ}C$, respectively, whereas those for the H$_2$ evolution of the cytoplasmic hydrogenases prepared from Cl. butyricum (C-H$_2$ase) and T. roseopersicina (T-H$_2$ase) were 45$^{\circ}C$ and 65$^{\circ}C$, respectively. The T-H$_2$ase was more thermostable than the C-H$_2$ase and retained its full activity for 5 h at 50$^{\circ}C$ under anaerobic conditions and 90% of its activity at 60$^{\circ}C$, whereas the C-H$_2$ase lost its activity drastically at 50$^{\circ}C$. The optimum pHs for H$_2$ oxidation of the C-H$_2$ase and T-H$_2$ase were 9.0 and 7.5, respectively. Both enzymes showed a maximum H$_2$ evolution activity at pH 7.0. Under aerobic conditions, 80% of the T-H$_2$ase activity was retained for 10 h at 30$^{\circ}C$, and 50% of the activity remained after 6 days under the same experimental conditions. However, the C-H$_2$ase was labile to oxygen and lost its activity immediately on exposure to air. Therefore, these properties of the T-H$_2$ase are expected to be advantageous for application in in vitro biological H$_2$ production systems.
