Antioxidant Enzymes in Postpartum Anoestrus Buffaloes Supplemented with Vitamin E and Selenium

  • The 15 buffaloes were divided into three groups, viz. group 1: normal cyclic buffaloes; group 2: postpartum anoestrus buffaloes and group 3: post partum anoestrus buffaloes supplemented with intramuscular injections of Vit. E.-care Se containing 500 mg $\alpha$-tocopheryl acetate and 15 mg selenium at weekly intervals for two months. The postpartum anoestrus buffaloes had significantly higher levels of erythrocytic lipid peroxidation, superoxide dismutase and glucose-6 phosphate dehydrogenase activities but lower glutathione peroxidase activity as compared to normal cyclic buffaloes. The supplementation of vitamin E and selenium lowered the level of erythrocytic lipid peroxidation, superoxide dismutase and glucose-6 phosphate dehydrogenase activities but it had no effect on whole blood selenium and erythrocytic gluathione peroxidase activity. All the animals in group 3 became cyclic and showed 60% conception rate.

  • Different bacteria wwere seperated from saliva and teeth of cariogenic patients and identified by a variety of morphological and biochemical tests. Extracts of green tea strongly inhibited Escherichia coli, Streptococcus salivarius and Streptococcus mutans. The antibacterial effect of green and black tea extracts were compared with those of amoxicillin, cephradine and eugenol.

  • In this research article, we introduce certain concepts, including domination, total domination, strong domination, weak domination, edge domination and total edge domination in m-polar fuzzy graphs. We describe these concepts by several examples. We investigate some related properties of certain dominations in m-polar fuzzy graphs. We also present a decision making method based on domination in m-polar fuzzy graphs.

  • Urban heat island (UHI) is one of the most important climatic implications of urbanization and thus a matter of key concern for environmentalists of the world in the twenty-first century. The relationship between climate and urbanization has been better understood with the introduction of thermal remote sensing. So, this study is an attempt to understand the influence of urbanization on local temperature for a small developing city. The study focuses on the investigation of intensity of atmospheric and surface urban heat island for a small urbanizing district of Punjab, India. Landsat 8 OLI/TIRS satellite data and field observations were used to examine the spatial pattern of surface and atmospheric UHI effect respectively, for the month of April, 2018. The satellite data has been used to cover the larger geographical area while field observations were taken for simultaneous and daily temperature measurements for different land use types. The significant influence of land use/land cover (LULC) patterns on UHI effect was analyzed using normalized built-up and vegetation indices (NDBI, NDVI) that were derived from remote sensing satellite data. The statistical analysis carried out for land surface temperature (LST) and LULC indicators displayed negative correlation for LST and NDVI while NDBI and LST exhibited positive correlation depicting attenuation in UHI effect by abundant vegetation. The comparison of remote sensing and in-situ observations were also carried out in the study. The research concluded in finding both nocturnal and daytime UHI effect based on diurnal air temperature observations. The study recommends the urgent need to explore and impose effective UHI mitigation measures for the sustainable urban growth.

A Retrospective Analysis on the Cystic Echinococcosis Cases Occured in Northeastern Punjab Province, Pakistan

  • A retrospective study was performed to report the case occurrence of cystic echinococcosis (CE) in three hospitals of north-eastern region in Punjab Province, Pakistan. We reviewed retrospectively the clinical records of patients in 4 hospitals which were diagnosed with CE during 2012-2017. A total of 198 cases, 82 (41.4%) male and 116 (58.6%) females were detected as CE. The most Highest incidence was revealed in${\ldots}$. 21-30 years-old group (24.2%) followed by 41-50 (22.7%), 31-40 (16.2%), 11-20 (13.6%), 51-60 (8.1%), below 10 (5.5%), over 71 (5.1%) and 61-70 year-old group (4.5%). CE was detected in various organs of infected individuals. However, most of CE cases were detected in the liver (47.4%) and lung (18.6%). The present study indicated that CE is more or less prevalent in surveyed areas and one of the most important public health problems in Punjab Province, Pakistan.

In-sacco Degradability of Dietary Combinations Formulated with Naturally Fermented Wheat Straw as Sole Roughage

  • Twelve dietary combinations were prepared using 70 parts of fermented wheat straw (FWS) as the sole roughage supplemented with 30 parts of either the low protein concentrate mixture (Conc.-I), high protein concentrate mixture (conc.-II), maize grains (M), solvent extracted mustard cake (DMC), deoiled rice bran (DRB), uromol bran mixture (UBM), deep stacked poultry litter (DSPL), dried poultry droppings (DPD), M-DMC mixture (50:50), M-UBM mixture (50:50), M-DPD mixture (50:50) or M-UBM-DPD mixture (50:25:25) and evaluated by in-sacco technique. The above dietary combinations were also evaluated by changing the roughage to concentrate ratio to 60:40. The digestion kinetics for DM and CP revealed that FWS:DPD had the highest, whereas, the FWS:M-DMC had the lowest rapidly soluble fraction. The potentially degradable fraction was found to be maximum in FWS:M and minimum in FWS:DPD dietary combinations. The higher degradation rate of FWS:DRB and FWS:UBM combinations was responsible for their significantly (p<0.05) higher effective degradability as compared to other combinations. The highest undegradable fraction noted in FWS:M-UBM-DPD followed by FWS:DMC was responsible for high rumen fill values. The FWS:DRB, FWS:UBM and FWS:DPD combinations had higher potential for DM intake. The dietary combination with higher concentrate level (60:40) was responsible for higher potentially degradable fraction, which was degraded at a faster rate resulting in significantly higher effective degradability as compared to the corresponding dietary combination with low concentrate level (70:30). The low undegradable fraction in the high concentrate diet was responsible for low rumen fill values, which predicted of high potential for DM intake. Out of 24 dietary combinations, FWS with either of UBM, DRB, DMC, Maize, M-DMC or DPD in 70:30 ratio supplemented with minerals and vitamin A in comparison to conventional feeding practice (roughage and concentrate mixture) could be exploited as complete feed for different categories of ruminants.

Expression and Purification of Transmembrane Protein MerE from Mercury-Resistant Bacillus cereus

  • Mercury-resistant ($Hg^R$) bacteria were isolated from heavy metal polluted wastewater and soil collected near to tanneries of district Kasur, Pakistan. Bacterial isolates AZ-1, AZ-2 and AZ-3 showed resistance up to $40{\mu}g/ml$ against mercuric chloride ($HgCl_2$). 16S rDNA ribotyping and phylogenetic analysis were performed for the characterization of selected isolates as Bacillus sp. AZ-1 (KT270477), Bacillus cereus AZ-2 (KT270478) and Bacillus cereus AZ-3 (KT270479). Phylogenetic relationship on the basis of merA nucleotide sequence confirmed 51-100% homology with the corresponding region of the merA gene of already reported mercury-resistant Gram-positive bacteria. The merE gene involved in the transportation of elemental mercury ($Hg^0$) via cell membrane was cloned for the first time into pHLV vector and transformed in overexpressed C43(DE3) E. coli cells. The recombinant plasmid (pHLMerE) was expressed and the native MerE protein was obtained after thrombin cleavage by size exclusion chromatography (SEC). The purification of fusion/recombinant and native protein MerE by Ni-NTA column, dialysis and fast protein liquid chromatography (FPLC/SEC) involved unfolding/refolding techniques. A small-scale reservoir of wastewater containing $30{\mu}g/ml$ of $HgCl_2$ was designed to check the detoxification ability of selected strains. It resulted in 83% detoxification of mercury by B. cereus AZ-2 and B. cereus AZ-3, and 76% detoxification by Bacillus sp. AZ-1 respectively (p < 0.05).

Pathogenicity Determinants of Sclerotinia sclerotiorum and Their Association to Its Aggressiveness on Brassica juncea

  • White rot or stem rot caused by Sclerotinia sclerotiorum is one of the most destructive fungal diseases that have become a serious threat to the successful cultivation of oilseed Brassicas. The study was designed with an aim to investigate the association between the pathogenic aggressiveness and pathogenicity determinants of this pathogen specifically in Brassica for the first time. For this, a total of 58 isolates of S. sclerotiorum from different geographical regions were collected and purified. These isolates were inoculated on a Brassica juncea cv. RL-1359 and they exhibited high level of variation in their disease progression. The isolates were grouped and then 24 isolates were selected for the biochemical analysis of pathogenicity determinants. The isolates varied significantly with respect to their total organic acids, oxalic acid production and pectin methyl esterase and polygalacturonase activity. The oxalic acid production corresponded to the disease progression of the isolates; the isolates with higher oxalic acid production were the more aggressive ones and vice-versa. This is, in our knowledge, the first study to establish a correlation between oxalic acid production and pathogenic aggressiveness of S. sclerotiorum on B. juncea. However, the pectinases' enzyme activity did not follow the trend as of disease progression. These suggest an indispensable role of oxalic acid in pathogenicity of the fungus and the potential to be used as biochemical marker for preliminary assessment of pathogenic aggressiveness of various isolates before incorporating them in a breeding program.

Coconut shell waste as an alternative lightweight aggregate in concrete- A review

  • This review article highlights the physical, mechanical, and chemical properties of coconut shells, and the fresh and hardened properties of the coconut shell concrete are summarized and were compared with other types of aggregates. Furthermore, the structural behavior in terms of flexural, shear, and torsion was also highlighted, with other properties including shrinkage, elastic modulus, and permeability of the coconut shell concrete. Based on the reviewed literature, concrete containing coconut shell as coarse aggregate with normal sand as fine showed the 28-day compressive strength between 2 and 36 MPa with the dried density range of 1865 to 2300 kg/m3. Coconut shell concretes showed a 28-day modulus of rupture and splitting tensile strength values in the ranges of 2.59 to 8.45 MPa and 0.8 to 3.70 MPa, respectively, and these values were in the range of 5-20% of the compressive strength. The flexural behavior of CSC was found similar to other types of lightweight concrete. There were no horizontal cracks on beams which indicate no bond failure. Whereas, the diagonal shear failure was prominent in beams with no shear reinforcements while flexural failure mode was seen in beams having shear reinforcement. Under torsion, CSC beams behave like conventional concrete. Finally, future recommendations are also suggested in this study to investigate the innovative lightweight aggregate concrete based on the environmental and financial design factors.
