Cytotoxicity and Genotoxicity of Newly Developed Calcium Phosphate-based Root Canal Sealers (신개발 인산칼슘계 근관 봉함재의 세포독성 및 유전독성에 관한 연구)

  • The purpose of this study was to compare the cytotoxicity by MTT test and genotoxicity by Ames test of new calcium phosphate-based root canal sealers (CAPSEAL I, CAPSEAL II) with commercially available resin-based sealers (AH 26, AH Plus) , zinc oxide eugenol-based sealers (Tubliseal EWT, Pulp Canal Sealer EWT), calcium hydroxide-based sealer (Sealapex), and tricalcium phosphate based sealers (Sankin Apatite Root Canal Sealer I, II, III). According to this study, the results were as follows : 1. The extracts of freshly mixed group showed higher toxicity than those of 24 h set group in MTT assay (p<0.001). 2. CAPSEAL I and CAPSEAL II were less cytotoxic than AH 26, AH Plus, Tubliseal EWT, Pulp Canal Sealer EWT Sealapex and SARCS II in freshly mixed group (p<0.01). 3. AH 26 in freshly mixed group showed mutagenicity to TA98 and TA100 with and without S9 mix and AH Plus extracts also were mutagenic to TA100 with and without S9 mix. 4. Tubliseal EWT, Pulp Canal Sealer EWT and Sealapex in freshly mixed group were mutagenic to TA100 with S9 mix. 5. Among those of 24 h set groups the extracts of SARCS II were mutagenic to TA98 with and without S9 mix and AH 26 showed mutagenic effects to TA98 with S9 mix. 6. No mutagenic effect of CAPSEAL I and CAPSEAL II was detected. 7. There is no statistically significant difference between CAPSEAL I and CAPSEAL II at MTT assay and Ames test in both freshly mixed group and 24 h set group.

Automatic Color Recognition System for Stockigt Sizing Test (II) - Application of the automatic recognition principle of red coloration for developing the novel automatic system - (스테키히트 시험용 자동발색인지 시스템 개발을 위한 기초 연구(II) -자동 발색 인지 원리를 적용한 발색 자동인지시스템-)

  • Stockigt sizing test, which is readily affected by individual tester's bias as well as testing conditions in recognizing red coloration, had to be modified to improve its reliability and reproducibility. The novel testing system with the automatic recognizing program of red coloration was developed with the auxiliary equipments including an automatic liquid dispenser and a specimen shifter. The analysis program used a hue value of a droplet image in recognizing a point of time on red coloration instead of RGB values that are not similar to human perception of color. Hue was more sensitive in recognizing the red coloration of a droplet than the other two factors, Saturation and Value. During the test, the program records the time consumed up to a specific hue value of a droplet on a specimen. Differently from the conventional test, the automatic test could obtain a reliable and reproducible sizing degree with a minor error. Furthermore, the Stockigt sizing degree measured by the automatic system showed great correlations with contact angle and Hercules sizing degree. It means that such great correlations will contribute to the development of an integrated measuring system capable of predicting contact angle, surface tension, surface energy and Hercules sizing degree of paper and paperboards through the Stbckigt sizing test. It was meaningful to note that the automatic system for Stbckigt sizing test might be able to used to predict contact angle, Hercules and Cobb sizing degree, based upon the high correlation coefficients.

Comparison of Partial Pulpotomy in Permanent Molars Using Different Pulp Capping Agents and Restoration Materials (치수복조재와 최종수복재에 따른 대구치 부분치수절단술의 비교)

  • The purpose of this study was to compare the success rates of two pulp capping materials, Biodentine™ and RetroMTA®, used for partial pulpotomy in permanent molars after carious exposures and to compare the final restorative outcome of using composite resin to that of using a stainless steel crown. We studied children who were diagnosed with dental caries of permanent molars and underwent partial pulpotomy. The patients were followed up for more than 1 year. Clinical and radiographic evaluation were used to evaluate the success of each treatment. Fisher's exact test was used to compare the outcomes of two groups. For pulp capping agents, the success rate of using RetroMTA® was lower than that of using Biodentine™(p < 0.05). Final restoration with composite resin was less successful than restoration with an stainless steel crown(p < 0.05). In combination of pulp capping agents and final restoration material, RetroMTA®-composite resin shows the lowest success rate(p < 0.05).

Biobleaching of Softwood Kraft Pulp Using Recombinant Xylanase and Cellulase (재조합 Xylanase와 Cellulase의 연속처리에 의한 침엽수 Kraft 펄프의 Biobleaching)

  • To know the effect of enzymatic pre-treatment on softwood Kraft pulp, two xylanse-encoding genes, named xynl and xynll were isolated from Thrichoderma ressei. Structural genes of xylanase (XYNI, XYNII) and cellulase (EGIV-CBDII) were isolated from T. ressei and Rumicoccus albus respectively, and expressed in E. coli. bacterial culture. The specific activity of purified recombinant XYNI is higher than XYNII. The brightness of XYNI treated softwood Kraft pulp increased to 29.9%. On further sequential treatment with EGIV-CBDII and XYNI the brightness of softwood Kraft pulp were improved to 9.1 and 73% respectively. As expected the Kappa number of softwood Kraft pulp also decreased 8.1, 4.6 and 3.2% respectively. Results further indicate that this sequential combination of enzyme treatment has synergic effect for improving bleaching of softwood Kraft pulp.

GENE EXPRESSION ANALYSIS OF THE DENTAL PULP IN HEALTHY AND CARIES TEETH (치아 우식증에 따른 치수내 유전자 발현 변화에 관한 분석)

  • Deep caries may induce pulpitis and the pulpal tissue interacts with microbial invasion. The immune response to protect the pulpal tissue can be mediated by cellular signal molecules produced by the pulpal cells. The understanding of these processes is important to find future therapeutic method for the diseased pulp. The pulp tissue from sound teeth was set as control group (n=30) and the pulp tissue from decayed teeth was set as test group (n=30). Total RNA was extracted from the pulp of each group and it was used for cDNA microarray and reverse transcriptase-polymerase chain reaction(RT-PCR). The expression of TGF-${\beta}1$ was studied by immunohistochemistry. The results were as follows: 1. cDNA microarray analysis identified 520 genes with 6-fold or greater difference in expression level with 143 genes more abundant in health and 377 genes more abundant in disease. 2. The RT-PCR analysis was done for randomly selected 14 genes and the results supported the result of cDNA microarray assay. 3. TGF-${\beta}1$ was highly expressed in the carious pulp and it was found in odontoblast by immunohistochemistry. In conclusion, many cytokines were found to be significantly changed their expression in the diseased pulp(/M/>1.6).

DIRECT PULP CAPPING WITH BONDING RESIN (접착용 레진을 이용한 유치의 직접 치수복조술에 관한 연구)

  • A direct pulp capping with different medicaments has been attempted for a long time. The most commonly used among those is the calcium hydroxide. In primary teeth, however, a success rate of direct pulp capping with calcium hydroxide has been reported to be lower than that of pulpotomy. The disappointing results of calcium hydroxide have prompted the search for other capping materials. Lately, several researchers suggested an application of adhesive resin-based composite systems as a capping material. They claimed that when an exposed vital pulp is capped directly with bonding resin, the pulp tissue is free of inflammation or necrosis without clinical symptoms. The aim of this study was to compare short-term effects of the bonding resin which was applied on the mechanically exposed vital pulp tissue and those of direct pulp capping with calcium hydroxide. The second objective was to compare success rates of the primary teeth which already underwent physiologic root resorption and those of the teeth which had not undergone physiologic root resorption yet, in each capping material groups. The vital, healthy pulp of forty-one primary teeth were exposed mechanically during a cavity preparation. They were divided into two groups: Group 1(n=21) underwent capping with bonding resin, and group 2(n=20) underwent capping with calcium hydroxide. Then these two groups were subdivided into two groups in each : the teeth which show physiologic root resorption and the teeth without root resorption. All of the sample teeth were restored with composite resin. Clinical evaluations such as percussion test, ice test, EPT, were recorded and also before- and after- standard x-ray films were compared and evaluated to decide whether the case was successful or not. Evaluation was performed at least 3 months after the capping materials. The results were as follows 1. There was no difference in success rate between group 1 and group 2. 2. Success rate of the teeth with physiologic root resorption was higher than that of the teeth without physiologic root resorption in group 1 and group 2. 3. There was no difference in success rate between anterior teeth and posterior teeth.

ANTIMICROBIAL EFFECT OF MINOCYCLINE EMBEDDED IN POLYCAPROLACTONE ON NECROTIC CANAL (괴사치수 근관에서 항생제 국소약물송달제제의 항균효과에 관한 연구)

  • Therapeutic use of antibiotics in the pulpal disease should reduce the clinical symptoms and pathogenic microflora in the pulp. The purpose of this study was focused on local drug delivery into the root canal in endodontic therapy. Monolithic films from polycaprolactone and polyethylene glycol preparations with minocycline were prepared, and then the antimicrobial effect of these films on necrotic canal was tested. A total 17 necrotized pulp teeth were sampled before and 1 week after insertion of film into the pulp. Bacterial culture were performed with anaerobical condition and seeded in the 5 selective and non - selective media for 7 days in $37^{\circ}C$ of anaerobic chamber. Bacterial identification were performed with Gram staining, biochemical test, and API kit There was significantly decreased of anaerobic and aerobic microflora of 13 among 17 patients after therapy. Among the identified microflora, Streptococcus species, black - pigmented species and Fusobacterium species were significantly reduced 1 week after treatment with local delivery antibiotics. Furthermore, clinical symptoms included in pain and local swelling were continuousely reduced after therapy. These results suggest that local drug delivery with minocycline is effeicient to treat the endodontic involved teeth for releasing clinical symptoms and microbiological shifting.

  • This study were performed for the effects of Panax ginseng on the side effect of Cyclophosphamide to splenic tissue in the Rats. The experimental animals were divided into control group, test group by the way of the method treatment of the drug. Each group was sacrificed and stained in accordance with the schedule and observed under light microscope. The results of this study were as follows : 1. After the ginseng extract administration rat's weight and volume were similarly increased than normal and control group. 2. The decrease of the numbers of the splenic tissue after treatment of cyclophosphamide(CP) were recovered with prescription of the ginseng extract; the decrease of white pulp, red pulp, marginal zone were recovered. 3. Increased macrophage in red pulp of splenic tissue with administration of CP were disapeared after treatment of ginseng extract.

Survival rate of stainless steel crown restorations according to depth of caries in primary molars (유구치에서 우식의 깊이에 따른 기성 금속관 수복물 생존율 연구)

  • The purpose of this retrospective study was to compare outcome of preformed stainless steel crown restorations according to depth of caries in primary molars. The study subjects consisted primary molars having dentin caries extending at least one-fourth of dentin without signs and symptoms of irreversible pulpitis. A total 295 primary molars received preformed stainless steel crown restorations without any base materials were included in this study. Survival rates were compared using a Kaplan-Meier analysis test. There was no significant difference between depth of caries and survival period in primary molars. And also there was no significant difference with or without pulp therapy. The results of this study suggest that neither the amount of caries nor pulp therapy affect the survival period of primary molars significantly. Proper restoration is more important to achieve sealing for stopping caries progression.

  • As a mood-altering drug, long-term alcohol consumption have significant harmful effects on the human body and people's mental functioning. This study observed that the suppression of cell mediated immunity induced in spleen of ICR mouse by long-term alcohol administration. After 8% alcohol voluntary administered for 120 days, the splenic tissue irnmunohistochemically stained by following ABC method that used monoclonal antibody including L3T4(CD4), Ly-2(CD8), IL-2 receptor(CD25R) and NK-1.1(CD56) after embedding with paraffin. The results were as follows. 1. The size of marginal zone in splenic white pulp was diminished and the number of macrophage in marginal zone was decreased in test group than control group. 2. After alcohol administration, the number of Helper T lymphocyte, cytotoxic T lymphocyte, and IL-2 receptor were decreased in periarterial lymphatic sheaths of white pulp and penicilla artery of red pulp and the degree of CD4, CD8, and CD25R positive reaction were soften. 3. In test group, the number of NK cell were decreased. These results indicated that the secretion of lymphokine as IL-2 was inhibited by long-term alcohol administration and subsequently prevent to activate and proliferate splenic T lymphocytes and NK cells as cell mediated immunity component.
