Cloning, Expression, and Characterization of Endoglucanase Gene egIV from Trichoderma viride AS 3.3711

  • Endoglucanase gene egIV was cloned from Trichoderma viride AS 3.3711, an important cellulose-producing fungus, by using an RT-PCR protocol. The egIV cDNA is 1,297 bp in length and contains a 1,035 bp open reading frame encoding a 344 amino acid protein with an estimated molecular mass of 35.5 kDa and isoelectronic point (pI) of 5.29. The expression of gene egIV in T. viride AS 3.3711 could be induced by sucrose, corn straw, carboxymethylcellulose (CMC), or microcrystalline cellulose, but especially by CMC. The transcripts of egIV were regulated under these substrates, but the expression level of the egIV gene could be inhibited by glucose and fructose. Three recombinant vectors, pYES2-xegIV, $pYES2M{\alpha}$-egIV, and $pYES2M{\alpha}$-xegIV, were constructed to express the egIV gene in Saccharomyces cerevisiae H158. The CMCase activity of yeast transformants $IpYES2M{\alpha}$-xegIV was higher than that of transformant IpYES2-xegIV or $IpYES2M{\alpha}$-egIV, with the highest activity of 0.13 U/ml at induction for 48 h, illustrating that the modified egIV gene could enhance CMCase activity and that $MF{\alpha}$ signal peptide from S. cerevisiae could regulate exogenous gene expression more effectively in S. cerevisiae. The recombinant EGIV enzyme was stable at pH 3.5 to 7.5 and temperature of $35^{\circ}C$ to $65^{\circ}C$. The optimal reaction condition for EGIV enzyme activity was at the temperature of $55^{\circ}C$, pH of 5.0, 0.75 mM $Ba^{2+}$, and using CMC as substrate. Under these conditions, the highest activity of EGIV enzyme in transformant $IpYES2M{\alpha}$-xegIV was 0.18 U/ml. These properties would provide technical parameters for utilizing cellulose in industrial bioethanol production.

Detection of Inflow Permeable Zones Using Fluid Replacement Conductivity Logging in Coastal Aquifer (공내수 치환 전기전도도검층을 이용한 연안지역 대수층의 탐지)

  • Fluid replacement and conductivity logging have been applied to three boreholes in coastal aquifer in order to identify permeable fractures and to estimate the origin of saline groundwater. Fluid replacement technique measures and monitors the change of borehole fluid conductivity with depth under ambient or pumping condition after replacing the original borehole fluid with different one (by pumping out original one and injecting simultaneously new one at the hole bottom). After the replacement of borehole fluid, the change of fluid conductivity can be the direct indicator of the intake flow of formation water through aquifer such as permeable fractures or porous formations. The conductivity profiles measured with times therefore indicate the locations of permeable zone or fractures within the open hole or the fully slotted casing hole. As a result of fluid conductivity logging for three boreholes at coastal area in Yeonggwang, Jeonam Province, it is interpreted that the seawater intrusion in this area is not by remnant saline groundwater after land reclamation but mainly by intrusion of saline water through fractured rock. This approach might be useful for assessing the characteristics of seawater intrusion, the design of optimal pumping, the mitigation of seawater intrusion using freshwater injection, and estimating the hydraulic characteristics in coastal aquifer.

Fluctuation Features and Numerical Model for Underground Temperature in Shallow Subsurface Soil (천층 토양 내 지중온도 변동 특성과 수치모델 평가)

  • This is conducted to observe underground temperature and to analyze its change affected by climate condition and soil infiltration in the mountainous area, Yesan region, Chungcheong-namdo province. Additionally, underground temperature change is also simulated using air temperature and soil thermal properties with a numerical model. Soil temperature monitoring data acquired from each depth, 20 cm, 50 cm, and 100 cm, indicates that the data within 50 cm in depth shows peak-shaped big fluctuation directly affected by air temperature and it at 100 cm has open-shaped small fluctuation. Underground temperature variation, a difference between high and low values, during monitoring period is weakly proportional to hydraulic conductivity of the sediment and it is assumed that water plays a part in delivering air temperature in soil. The underground temperature estimated by a numerical model is very similar to the observed data with an average value of 0.99 cross-correlation coefficient. From the result of this study, the aquifer unsaturated hydraulic conductivity of the soil and the groundwater recharge is likely to be able to estimate with underground temperature profile calculated using a numerical model.

Studies on Varietal Resistance and Chemical Control to the Wilt of Strawberry caused by Fusarium oxysporum (Fusaium oxysporum에 의한 양딸기 시들음병의 약제방제 및 품종저항성에 관한 연구)

  • The strawberry cultivar Hokowase showing rapid wilting and death around harvest time was first found in the fold at Woongcheon, Chungnam province in 1974. The fungus, Fusarium oxysporum was isolated frequently from the crown, petiole and root of strawberry plants collected from Woongcheon and was pathogenic to Hokowase. The fungus abundantly produced micro-and macro-conidia and chiamydospore on PSA. The size of micro conidia, macro conidia and Chlamydospores was $5.0\~13.0\times2.5\~3.0,\;12.8\~62.5\times2.5\times50\mu\;and\;7.5\~13.8\times5.5\~12.5\mu$, respectively. Mycelial growth of the fungus was best between $25\~30^{\circ}C$ on PSA. The disease at Woongcheon occurred from the end of March and incidence of the disease increased from the beginning of May reaching $40.2\%$ of diseased Plants at the 1st part of June. In laboratory and field evaluation tests with twelve fungicides, Difolatan, Benlate T and Tospin M showed some control effects against the disease although they did not show ignificant differences in effects compared with that of the non-treatment. The cultivar Yachiyo, Daehak 1, Line 10-2, and Senga Sengana were highly resistant, and Harunoka and Empire were moderate resistant whereas Northwest and Hokowase were highly susceptible to the fungus under field condition.

Environmentally-friendly Control Methods and Forecasting the Hatching Time Lycorma delicatula (Hemiptera: Fulgoridae) in Jeonnam Province (전남지역에서 꽃매미 부화시기 예측과 친환경 방제방법)

  • This study was conducted to predict the hatching time of eggs of Lycorma delicatula, to select an effective environmentally-friendly agriculture material (EFAM) and to evaluate the attraction effect of brown sticky traps for controling of Lycorma delicatula nymph and adults. Eggs hatched 55.9, 26.8, 21.6 days after incubation at 15, 20, $25^{\circ}C$ with 14L:10D condition and the hatching rates of egg were 61.9, 57.8, 30.4%, respectively. At high temperature conditions, egg development periods were shorter and the hatching rate was lower. The relationship between temperature and developmental rate was expressed by the linear equation Y=0.0028X-0.0228, $R^2$=0.9561. The low temperature threshold of eggs was $8.14^{\circ}C$ and the thermal constant required to reach larva was 355.4 DD. According to this relationship, the mean estimated hatching date was $22^{nd}$ May. The effective EFAM was natural plant extract, sophora extract, derris extract to nymph and natural plant extract, pyrethrum extract, sophora extract to adult. Among three colors of sticky trap : brown, blue and yellow, the brown sticky trap was the most attractive to nymphs and adults of L. delicatula over a 2 weeks trial period. It suggested that the brown sticky trap could be a very useful and environment-friendly control method for nymphs and adults of L. delicatula.

An Extended Benefit-Cost Analysis of Land Consolidation and On-farm Development Project -With Respect to Jigok Land Consolidation and On-farm Development Project in Seosan, Chungnam Province, Korea- (경지정리사업의 확장편익-비용분석 - 충남서산시 지곡지구를 중심으로 -)

  • The economic feasibility analysis including benefit-cost ratio and internal rate of return of a land consolidation and on-farm development project was mainly depended upon the direct benefits and costs arising during project life. Therefore the projects were neglected in allocating the government's financial funds and loans on account of the low economic rate of return of the project. Accordingly the extended benefit-cost analysis method should be introduced and adapted to cover not only the benefit s such as non-market values of environmental and food security fun ct ions of the project but also market values of the project outputs. The main purposes of this study are (1) to prepare a guide line for economic feasibility analysis based on the IBRD and ADB guidelines, (2) to estimate the benefits from productivity increase, labor saving, savings of O&M cost of farm mechanization and project facilities, savings of farmer's burden for their public health, increasing environmental and public functional value of paddy fields, improving food security condition and formation of peaceful and uncontroversial rural society by implementing the land consolidation and on-farm development, (3) to introduce the newly adapted analytical method as the extended benefit cost analysis which could make possible to be included the values of non-market goods such as the food security and the environmental public function of paddy fields. To carry out this study, the existing publications on the guidelines for economic agricultural projects were reviewed and consultation was made with a For the post evaluation study of the land consolidation and on-farm development project, field survey in Jigok and other comparative area were made to get available information. According to the project completion report, Jigok land consolidation and on-farm development project has 55ha of benefit area out of 69ha of gross area. The project was started in November 11th, 2000 and completed october 30th, 2001. The total project costs were amounted to 2,548 million won and the annual project benefits were estimated at 335million won evaluated by domestic market prices. The ERR(Economic Rate of Return) and SRR(Social Rate of Return) of the project based on the shadow pricing system were estimated at 4.4% and 16% respectively. On the other hand, the ERR and SRR based on the domestic market value system were amounted to 6.37% and 14.62%. In conclusion, Korean land consolidation and on-farm development projects have not carried out from the view point of economic rate of return under shadow pricing system but from the view point of domestic pricing system. For the future feasibility studies on land consolidation projects have to be carried out including the non-market values as environmental and food security function of the projects.

The Regional Folk Plants in Southern Inland Area of Gyeonggi-do (경기남부 내륙지역의 민속식물자원)

  • This study was carried out to clarify the regional folk plants in the southern inland area of Gyeonggi-do Province. According to the survey results, derived from 1,229 sheets of 90 residents at 35 places from 9 counties and cities between October 2009 and December 2010, the folk plants in the inland southern area of Gyeonggi-do consisted of a total of 259 taxa; 82 families, 200 genera, 221 species, 4 subspecies, 32 varieties, and 2 forma. The classification of the plants by its usage was; 214 as edible, 119 as medicinal, 34 as ornamental, 18 as timber, 12 as dye, 5 as oil, 3 as spice, 2 as fiber, 2 as aroma, and 22 as others uses. So, the edible and medicinal uses are higher than other uses. The most useful part was the leaf, followed by fruit, stem, root, and flower. The collection season was focused on spring and summer, because the leaf was mainly collected in an edible condition during those seasons. Traditional knowledge of folk plants was mostly passed down orally. The consistency comparison between the standard common name and the local name was the highest in the people in 50s and the lowest in those in 80s.

Comparison of the Self Confidence for Newborn Baby Care Before and After Clinical Convergence Baby Massage Training Program in Nursing students (임상융합 신생아마사지 프로그램 전·후 간호대학생의 신생아돌보기 활동 자신감 비교)

  • The purpose of this study was conducted to identify differences of the self confidence for newborn baby care before and after clinical convergence baby massage training program. Study subjects were 50 juniors in B nursing college located in Chungnam Province, modified baby massage training programs had been provided 2 hours a day, total 4 times from March 1 to March 25, 2016. Confidence for newborn baby care was measured pre and post program, collected data were analyzed using SPSS 21.0 program, paired t-test. The results of the study showed that the score of self confidence for newborn baby care(t=-7.209, p<.001), feeding(t=-6.025, p<.001), belching(t=-3.870, p<.001), bathing(t=-7.432, p<.001), after bathing hygiene(t=-5.892, p<.001), navel care(t=-3.843, p<.001), assessment of baby condition and baby care(t=-6.113, p<.001) were statistically significant differences before and after baby massage training program. Finally, the findings of this study, clinical convergence baby massage training program based on the necessary knowledge and practice in clinical filed will positively help to develop a confidence for newborn baby care for nursing students in nursing practice and clinical nursing care.

Building an Innovation System for Industrial Development in a Knowledge based Economy (산업의 지식집약화를 위한 혁신체제 구축 방향)

  • The purposes of this research are to examine the theoretical background and industrial policy issues with regard to building a Innovation System for encouraging industrial competitiveness and fostering regional industry in Korea. Knowledge has become the driving force of economic growth and the primary source of competitiveness in the world market. So since 1990s, Innovation Systems have been put emphasis on as new industrial development strategy in a knowledge-based economy. It can be understood that Innovation System is composed of National Innovation System(NIS) and Regional Innovation System(RIS) and interrelated the concept of clusters and networks, which are contribute to industry development throughout boosting innovation. As for the Korean industrial policy, when the former centralized policy decision making process became decentralized through the implementation of local autonomy, the role of local or state government in relation to regional industrial promotion intensified. But with the impotance of for fostering strategic industry in the region. new industrial policy issues in Korea are needed as follows; $\circled1$ Building a market-oriented support system for industrial cluster through providing the resource of innovation. $\circled2$ Establishing agency for regional industrial development. $\circled3$ Making a evolutionary vision for broader region including 2 or 3 province, $\circled4$ Fostering strategic industry which is selected in term of specialization and potential of the region. The RIS model for industry development is outlined in this paper but policy initiatives for building a RIS have to be extracted from further case studies.

Rural Tourism Village Building with Residents' Participation : In Case of Jinmok Village in Jangheung County (주민참여에 의한 농촌관광마을 만들기 : 장흥군 진목마을을 사례로)

  • This study focuses on the actual condition and level of residents' participation in the case of Jinmok village, located in Jeonnam province Jangheung-gun Hwejin-myeon, and presents suggestions to inspire residents' awareness and enhance participation. This village has been developing a Green Rural Tourism Village Building Project under management of the Ministry of Agriculture and Forestry. Consequently, the importance of residents recognizing they are a "community" has been emphasized as well as promoting the residents cooperation as a community. In addition, it was necessary to research ways to inspire the village to realize the ultimate goal of building rural tourism village. For this, in the beginning of the project, profitable work needed to be provided for the residents to participate in. Constructive participation by all residents is essential in each stage of the project s development. Operating a farm stay or running lodging facilities are options that could be considered. Moreover, it is imperative to immediately establish a awareness about the importance of participation in those who might have a negative or impartial opinion about the project, by providing education. The most efficient way to encourage residents' participation is to support various opportunities for education and field trips. Through these efforts, the residents are able to raise their level of education and develop their man-power as well. Once more, providing an incentive for residents' reliance is the kernel of the project. Considering the state of Korean rural areas, which are rapidly aging and becoming depopulated, green tourism can be a practical alternative for continuing rural development.
