Effects of Aqueous Extracts from Naturalized and Korean Wild Plants on Seed Germination and Seedling Growth of Zoysiagrass (자생식물과 귀화식물의 수용 추출액이 Zoysiagrass의 발아와 유근생장에 미치는 영향)

  • The soil pH favored by Korean wild plants is 5.33~7.20, while naturalized invader species prefer soils of pH 3.95~6.48. The germination rate of Zoysia japonica was inhibited sharply, up to 60%, in extracts of naturalized plants of concentrations over 50%. Erigeron canadensis extract most strongly inhibited germination, while the Korean wild species, Cassia mimosoides var. nomame increased germination rates in concentrations of 30% and 50%. The seedling growth of Zoysia japonica in extracts of Korean wild species and naturalized species did not show differences, but was inhibited strongly, up to 60%, in the extract of Cassia mimosoides var. nomame concentration of over 15%. Analysis of the extract from Cassia mimosoides var. nomame identified procatechuic acid, $\rho$-hydroquinone, $\rho$-coumaric acid and ferulic acid at 254nm; and vanillic acid, hydroquinone, benzoic acid and cinnamic acid were discovered at 284nm. The seed germination and seedling growth of Zoysia japonica were investigated under different concentrations of phenolic compounds. Ferulic acid and vanillic acid were associated with an increased germination rate, while $\rho$-coumaric acid appeared to inhibit seedling growth.

Cytotoxic Constituents from Solanum Lyratum

  • Activity-guided fractionation of the ethanol extract of the whole plant from Solanum lyratum resulted in the isolation of a new pregnane derivative glycoside, 16-dehydropregnenolone 3-O-${\alpha}$-L-rhamnopyranosyl-($1{\to}2$)-${\beta}$-D-glucopyranosid uronic acid (2), as well as other six known compounds: 16-dehydropregnenolone (1), allopregenolone (3), protocatechuic acid (4), vanillic acid (5), caffeic acid (6), and scopoletin (7). The structures of the isolated compounds were elucidated on the basis of their spectral data and chemical evidences. Compounds 1, 3, 4 were isolated for the first time from this plant. Cytotoxic activities of the isolated compounds were evaluated. Compound 1 exhibited significant cytotoxic activity against A375-S2, HeLa, SGC-7901, and Bel-7402 with $IC_{50}$ values of $13.1{\pm}0.9,\;21.5{\pm}1.0,\;40.2{\pm}0.7$, and $49.8{\pm}1.2\;{\mu}g/mL$, respectively.

  • The flowers of Coreopsis lanceolata were extracted with 80% aqueous MeOH and the concentrates were partitioned into EtOAc, n-BuOH, and H2O fractions. The repeated silica gel (SiO2) and octadecyl silica gel column chromatographies for the EtOAc fraction led to isolation of one flavonol and one benzoyl compounds. The chemical structures of the compounds were respectively determined as melanoxetin (1) and protocatechuic acid methyl ester (2) based on spectroscopic analyses including NMR, IR, and MS. These two compounds were isolated for the first time from C. lanceolata flowers in this study. All fractions and the isolated compounds were evaluated for 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activities.

Comparison of antioxidant activities of pearled and wholegrain barley harvested in Jeju (도정 유무에 따른 제주산 보리의 항산화 활성 비교)

  • Barley contains health-beneficial compounds, such as phenolic acids, flavonoids, lignans, and tocopherols. The antioxidant activities of pearled and wholegrain hull-less, premature, and black barley harvested in Jeju were investigated by measuring total phenolic content (TPC), total flavonoid content (TFC), phenolic compounds, tocopherols (T), and tocotrienols (T3), along with their in vitro antioxidant activities. Consequently, TPC and TFC in wholegrain barley groups were higher than those in pearled barley. Gallic acid (1.55-2.98 ㎍/g) and protocatechuic acid (0.67-2.84 ㎍/g) were the predominant phenolic compounds in barley. Total T and T3 concentrations of wholegrain barley groups were significantly higher than those of pearled barley (p<0.05). Except for the metal chelating effect and reducing power, the in vitro antioxidant activities of wholegrain barley were significantly higher than those of pearled barley. These results indicate that wholegrain Jeju barley can be used as a natural antioxidant source in the food industry.

Study on DPPH Free Radical Scavenging and Lipid Peroxidation Inhibitory Activities of Vietnamese Medicinal Plants

  • Among 90 Vietnamese medicinal plant extracts investigated for their antioxidant activity by DPPH assay at various concentrations from $10-100{\mu}g/mL$, 67 showed an inhibition rate over 50% at $100{\mu}g/mL$; 47 had greater than 50% inhibition at $50{\mu}g/mL$; 17 showed over 50% inhibition at $25{\mu}g/mL$. 8 extracts which exhibited strong inhibitory activity more than 50% inhibition at $10{\mu}g/mL$ were further tested for lipid peroxidation inhibition by TBA assay. They displayed activity with $IC_{50}$ values from 30.6 to $158.9{\mu}g/mL$. Until now, this is the first report on antioxidant activity of the female flower of Borassus flabellifer, and the stem of Combretum latifolium, Embelia ribes, Spatholobus parviflorus, and Tetrastigma erubescens. Fractionations of the EtOAc extract prepared from S. parviflorus led to the isolation of protocatechuic acid (1), ferulic acid (2), epicatechin (3), and gallic acid (4). These compounds showed significant DPPH inhibitory activity with $IC_{50}$ values from 6.5 to $23.6{\mu}M$.

Determination of Total Phenol Content and Selected Phenolic Metabolites Analysis of Rice (Oryza sativa L.) Genetic Resources

  • The study of total phenol (TP) content from 700 rice varieties was evaluated using the UV-Vis spectrophotometric method. The calibration curve of serial diluted gallic acid as a standard of this study showed the acceptable performances (R2 = 0.999, mean accuracy 90%) and the mean of % relative standard deviation (%RSD: 0.07%) within the range of 7.8 to 1000 ppm concentrations. The mean value of total phenol content from 700 rice varieties was 2723.15 ㎍/g ranged from 55.48 ㎍/g to 9922.23 ㎍/g and the mean %RSD was 2.5%. Furthermore, this study aim was to analyze and profile individual phenolic compounds in the rice genetic resources to construct an integrative database for development of new rice variety with high functionality for health and understanding of phenolics characteristics in the rice grain. Herein, we analyzed selected 100 rice varieties based on high TP content and identified total 15 phenolic compounds by LC-ESI-MS/MS. Among selected 100 rice genetic resources, the phenolic metabolites consisted of higher amount of flavonoid (catechin) and phenolic acid mainly protocatechuic acid. Further research of more selected rice genetic resources would be continued to provide for an integrative phenolics profile of different rice genetic resources.

A Comparative Study on Anthocyanin and Polyphenol Contents in Colored Agricultural Products (유색 농산물 중 안토시아닌과 폴리페놀 함량 비교 연구)

  • In this study, 17 kinds of polyphenols and 5 kinds of anthocyanins were analyzed to compare the contents of polyphenols and anthocyanins in 76 colored agricultural products. A total of 17 polyphenols were analyzed simultaneously by 9 phenolic acids (gallic acid, protocatechuic acid, chlorogenic acid, vanillic acid, caffeic acid, syringic acid, p-coumaric acid, t-ferulic acid, t-cinnamic acid) and 8 flavonoids ((+)catechin, syringic aldehyde, rutin, epicatechin gallate, naringin, luteolin, naringenin, kaempferol) and 5 anthocyanins (delphinidin-3-glucoside, delphinidin-3-rutinoside, cyanidin-3-galactoside, cyanidin-3-glucoside, cyanidin-3-arabinoside) were simultaneously analyzed. The total content of 17 polyphenols was determined as seoritae $255.1{\pm}7.5{\mu}g/g$, seomoktae $275.8{\pm}5.3{\mu}g/g$, black rice $78.5{\pm}4.6{\mu}g/g$, black sesame $75.8{\pm}3.2{\mu}g/g$, blueberry $143.3{\pm}5.5{\mu}g/g$, aronia $195.2{\pm}4.9{\mu}g/g$ and blackcurrent $131.6{\pm}3.2{\mu}g/g$, the highest content was found in the order of seomoktae > seoritae > aronia > blueberry > blackcurrant > black rice > black sesame. The total content of 5 anthocyanins was determined as seoritae $82.4{\pm}17.2{\mu}g/g$, seomoktae $95.2{\pm}6.1{\mu}g/g$, black rice $74.1{\pm}9.7{\mu}g/g$, black sesame were not detected, blueberry $110.8{\pm}1.9{\mu}g/g$, aronia $218.9{\pm}6.1{\mu}g/g$ and blackcurrent $209.7{\pm}4.0{\mu}g/g$, the highest content was found in the order of aronia > blackcurrant > blueberry > seomoktae > seoritae > black rice. These results indicated that seomoktae and aronia possessed the high level of functional components and further study will be needed to develop high value-added foods based on the colored agricultural products.

  • Peroxynitrite ($(ONOO^-)$ is a cytotoxic species formed from nitric oxide and superoxide anion, which are highly implicated in the pathogenesis of oxidative stress-mediated diseases. The aim of this study was to investigate the scavenging effects of Phellinus linteus on authentic $ONOO^-$, and further phytochemical studies are planned that will attempt to identify the active principles. From the active EtOAc fraction, a mixture of fungisterol and 5-dihydroergosterol (1), a mixture of betulin and 1,2-benzenedicarboxylic acid bis (2-methyl heptyl) ester (2), protocatechualdehyde (3), protocatechuic acid (4), cirsiumaldehyde (5), hispidin (6), caffeic acid (7), phelligridin D (8), uracil (9), gallic acid (10), 2,5-dihydroxybenzoic acid (11), ferulic acid (12), 2,3-dihydroxybenzaldehyde (13), arbutin (14), isoferulic acid (15), guanosine (16), and ellagic acid (17) were isolated, and their structures were characterized based on spectroscopic data. All compounds except 3, 6, 7 and 16 were isolated for the first time from P. linteus. Compounds 3, 4, 6-8, 10-15, and 17 showed potent scavenging activity on $ONOO^-$, with $IC_{50}$ values of $2.06\;{\pm}\;0.10$, $3.45\;{\pm}\;0.57$, $0.71\;{\pm}\;0.05$, $2.78\;{\pm}\;0.36$, $5.42\;{\pm}\;0.26$, $1.13\;{\pm}\;0.02$, $1.82\;{\pm}\;0.17$, $0.91\;{\pm}\;0.19$, $1.59\;{\pm}\;0.09$, $1.88\;{\pm}\;0.07$, $1.22\;{\pm}\;0.37$, and $2.01\;{\pm}\;0.02\;{\mu}M$, respectively, as compared to the positive control, DL-penicillamine, with an $IC_{50}$ value of $5.04\;{\pm}\;0.06\;{\mu}M$.

Phytochemical Contents and Antioxidant Activities of Opuntia ficus-indica var. saboten (보검선인장의 Phytochemical 함량 분석 및 항산화 활성)

  • The aim of this study was to evaluate key properties of the prickly pear cactus (Opuntia ficus-indica var. saboten (OFI) ie, levels of key chemicals (carotenoids, flavonoids and polyphenolic compounds as well as phenolic acid) and its antioxidative potential, depending on where the plant had been cultivated in Korea. The levels of flavonoids and polyphenolic compounds in OFI were 55.45~65.32 mg (+)-catechin/g and 149.00~181.15 mg gallic acid/g, respectively. Protocatechuic acid was the most abundant phenolic acid in the ON1 and ON2 (161.90 and $196.25{\mu}g/g$ DW (dry weight)). Nineteen flavonoids were identified and analyzed by LC-ESI-MS in cladodes from OFI. Narcissin was the most abundant flavonoid in all of the samples ($1,241.89{\sim}1,775.10{\mu}g/g$ DW). Capxanthin and zeaxanthin were the most abundant carotenoids in OFI (64.88~128.08 and $48.10{\sim}93.82{\mu}g/g$ DW). The level of DPPH radical and ABTS radical scavenging activities in OFI were 10.78~25.35 and 16.85~34.16 mg AA eq/100 g, respectively. OFI by cultivar has different kind of phenolic acid, flavonoids, and carotenoids. Therefore, dietary intake of cladodes from OFI may be helpful for improving human health.

Assessment of Antioxidant and Phenolic Compound Concentrations as well as Xanthine Oxidase and Tyrosinase Inhibitory Properties of Different Extracts of Pleurotus citrinopileatus Fruiting Bodies

  • Cellular damage caused by reactive oxygen species has been implicated in several diseases, thus establishing a significant role for antioxidants in maintaining human health. Acetone, methanol, and hot water extracts of Pleurotus citrinopileatus were evaluated for their antioxidant activities against ${\beta}$-carotene-linoleic acid and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, reducing power, ferrous ion-chelating abilities, and xanthine oxidase inhibitory activities. In addition, the tyrosinase inhibitory effects and phenolic compound contents of the extracts were also analyzed. Methanol and acetone extracts of P. citrinopileatus showed stronger inhibition of ${\beta}$-carotene-linoleic acid compared to the hot water extract. Methanol extract (8 mg/mL) showed a significantly high reducing power of 2.92 compared to the other extracts. The hot water extract was more effective than the acetone and methanole extracts for scavenging DPPH radicals. The strongest chelating effect (92.72%) was obtained with 1.0 mg/mL of acetone extract. High performance liquid chromatography analysis detected eight phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, ferulic acid, naringenin, hesperetin, formononetin, and biochanin-A, in an acetonitrile and hydrochloric acid (5 : 1) solvent extract. Xanthine oxidase and tyrosinase inhibitory activities of the acetone, methanol, and hot water extracts increased with increasing concentration. This study suggests that fruiting bodies of P. citrinopileatus can potentially be used as a readily accessible source of natural antioxidants.
