Isolation and Determination of Phenolic Compounds from Tuber of Color-fleshed and White-fleshed Potatoes

  • Color-fleshed potatoes 'Hong-young' and 'Ja-young' were developed by RDA, and it has reported that they have high content of anthocyanin. Additionally they show higher radical scavenging activity compared to white or yellow fleshed potatoes. So it will be expected that the consumption of color-fleshed potatoes gradually increase by pre-peeled potatoes and color potato chips. This study was conducted to enhance the utilization of the tuber of color-fleshed potatoes. At first, we isolated four compounds from the organic solvents soluble layer in ethanol extract of tuber, and their structures were characterized by spectroscopic methods and by comparing their data to those in the literature. Their structures were characterized to be caffeic acid (1), chlorogenic acid (2), gallic acid (3) and protocatechuic acid (4) for the first time reported from this source. These compounds were already reported ingredients but considered to exhibit a high physiological activity. The quantitative determination on the four compounds in tuber of color-fleshed [Hong-young (HY) & Ja-young (JY)] and white-fleshed [Superior (SP)] potatoes samples were measured using HPLC. The concentration of caffeic acid in each total fractionations of HY (184.4 g/g) and JY (435.1 g/g) were higher than in total fractionation of SP (31.1). The concentration of gallic acid in each total fractionations of HY (282.1 g/g) and JY (315.2 g/g) were higher than in total fractionation of SP (143.3 g/g). The concentration of chlorogenic acid contents appeared to be highest in total fractionation of SP (954.2 g/g). The concentration of protocatechuic acid in total fractionation of HY (120.3 g/g) was higher than in each total fractionationss of JY (74.4 g/g) and SP (102.7 g/g). Overall, color-fleshed potatoes had higher amount of physicochemical properties than 'Superior'. Therefore, color-fleshed potatoes are expected to be highly valuable items for the development and applications of a functional food. In addition, these results will provide fundamental information for improving sitological value, and breeding of new cultivar.

약용식물(藥用植物)(음나무, 오가피)로부터 생리활성물질(生理活性物質) 검정(檢定) (Identification of Biologically Active Substances from Medicinal Plants)

  • 약용식물(藥用植物)을 대상(對象)으로 생리활성(生理活性)을 지닌 phenolic산(酸)과 지방산(脂肪酸) 및 유기산(有機酸) 등(等)을 조사(調査)한 결과(結果)는 다음과 같다. 음나무와 오가피의 EtOH 추출물(抽出物)은 상치의 발아(發芽)를 강하게 억제(抑制)하는 생리활성물질(生理活性物質)을 함유(含有)하고 있었다. 음나무와 오가피로부터 GLC에 의해 동정(同定)된 phenolic 산(酸)은 protocatechuic 산(酸) 외(外) 11종(種)이었으며 총(總) phenolic 산(酸)의 함량(含量)은 음나무가 1.7971mg/g, 오가피가 0.9567mg/g 이었다. HPLC로 검정(檢定)된 polyphenols은 ncochlorogenic, chlorogenic, scopoletin, rutin, kaempferolglycoside 였으며 chlorogenic 산(酸)의 함량(含量)이 음나무 23.7ppm, 오가피 13.0ppm으로 가장 많았다. 음나무는 지방산(脂肪酸)이 5.26mg/g, 유기산(有機酸)이 27.69mg/g, 오가피는 지방산(脂肪酸)이 3.22mg/g, 유기산(有機酸)이 9.80mg/g 함유(含有)되어 있었으며 음나무가 오가피보다 많은 지방산(脂肪酸)과 유기산(有機酸)을 함유(含有)하고 있었다. 지방산(脂肪酸) 가운데는 linoleic 산(酸)이 전체(全體)의 50% 이상(以上)을, 유기산(有機酸) 가운데는 oxalic 산(酸)이 80% 이상(以上)의 비율(比率)로 함유(含有)되어 있었다.

복분자와 오디의 항산화 특성 (Antioxidant Activities of Rubus coreanus Miquel and Morus alba L. Fruits)

  • 다양한 추출용매를 사용하여 얻은 복분자와 오디의 추출용액 중에서 가장 높은 항산화 활성을 보인 60% acetone과 이들의 분획물을 대상으로 측정한 항산화 활성에서 시료별로는 오디, 분획별로는 ethyl acetate 분획의 $EC_{50}$ 값이 낮게 나타났다. 복분자와 오디의 항산화 성분 중 총 페놀성 화합물과 총 플라보노이드 함량이 안토시아닌 함량에 비해 매우 높게 나타났다. 항산화 성분과 항산화 활성의 상관성은 총 페놀성 화합물과 총 플라보노이드에서는 높은 상관성을 나타냈으나 안토시아닌에서는 오디에서만 높은 상관성을 나타내었다. 복분자와 오디의 주된 phenolic acid는 protocatechuic acid, p-hydrobenzoic acid 및 salicylic acid이었으며 주된 안토시아닌은 cyanidin-3-glucoside이었다. 위의 결과들을 종합해보면 복분자와 오디의 항산화 성분은 추출에 사용된 용매의 종류에 따라 함량 및 조성이 변화하며 그 결과로서 항산화 활성에 영향을 주게 됨을 확인할 수 있었다. 또한 오디 ethyl acetate 분획물은 DPPH assay와 reducing power에서 BHT보다 각각 2.2와 1.2배 낮은 $EC_{50}$ 값을 보여 천연 항산화 소재로서의 가능성을 보였다.

폐광지역에서 분리한 quinoline 분해 세균인 Pseudomonas sp. NFQ-1의 특성연구 (Characterization of the Quinoline-Degrading Bacterium Pseudomonas sp. NFQ-1 Isolated from Dead Coal Pit Areas)

  • 폐광지역으로부터 quinoline (2,3-benzopyridine)을 유일한 탄소원, 질소원, 그리고 에너지원으로 이용하는 세균 NFQ-1을 농화 배양기법을 통하여 분리하였다. 분리된 세균은 그람음성의 간균으로서 BIOLOG 시험을 통하여 Pseudomonas nitroreducens로 동정되었으며, 본 연구에서는 Pseudomonas sp. NFQ-1으로 명명하였다. Quinoline의 분해는 호기적 조건하의 B-배지에서 Pseudomonas sp. NFQ-1를 이용하여 실시되었다. 균주 NFQ-1 세균은 2.5 mM quinoline을 9시간 이내 완전히 분해하였다. 배양기간 동안 quinoline 분해의 중간대사산물인 2-hydroxyquinoline이 일시적으로 생성되었다가 배양기간 후반부에 사라졌다. 배양의 초기 pH 8.0은 6.8로 감소하다가 배양이 진행됨에 따라 7.0이 되었다. 대상 기질로서 quinoline의 농도가 증가함에 따라 생장곡선에서 유도기가 길어졌으며, 고농도의 quinoline (>15 mM)은 주어진 조건에서 균주의 생장과 quinoline의 분해를 억제하였다. 부가 질소원으로 7.6 mM $(\textrm{NH}_{4})_{2}\textrm{SO}_{4}$의 첨가조건하에서 Pseudomonas sp. NFQ-1은 2-hydroxyquinoline, p-coumaric acid, benzoic acid, p-cresol, p-hydroxybenzoate, protocatechuic acid, catechol 등의 다양한 화합물을 이용할 수 있었으나 일부 화합물들 (예, 6-hydroxyquinoline, 8-hydroxyquinoline, coumarin, indoline, pyridine, lepidine, quinaldine, 4-bydroxycournarin, benzene, salicylic acid, phenol, phthalate)은 탄소원으로 이용되지 못하였다. euinoline의 분해경로를 규명하기 위하여 catechol dioxygenases의 specific activity를 결정하였다. 그 값은 catechol 1,2-dioxygenase에서 약 184.7 U/mg, 그리고 catechol 1,2-dioxygenase에서 약 33.19 U/mg이었다. 그 결과 균주 NFQ-1은 quinoline를 분해하기 위하여 주로 ortho-분해경로를, 그리고 부분적으로 meta-분해경로를 이용하는 것을 보여주었다.

흰쥐의 브로모벤젠대사계에 미치는 어성초의 영향과 페놀성 화합물의 분리 (The Effects of Houttuynia cordata on the Hepatic Bromobenzene Metabolizing Enzyme System in Rats and Isolation of Phenolic Compounds)

  • Effects of Houttuynia cordata on the level of lipid peroxide and the enzyme activities of the liver were investigated in bromobenzene-induced rats. Lipid peroxide content in liver was increased by bromobenzene. It was decreased when the methanol extract from the aerial parts of H. cordata was treated to the rat. The methanol extract reduced the activities of aminopyrine N-demethylase and aniline hydroxylase that increased by bromobenzene, however did not affect glutathione S-transferase activity. The methanol extract recovered the activity of epoxide hydrolase activity that decreased significantly by bromobenzene. We suggest that under our experimental conditions the extract might play an important play in the prevention of hepatotoxicity by reduction of aminopyrine N-demethylase and aniline hydroxylase activities as well as enhancement of epoxide hydrolase activity. Six phenolic compounds have been isolated from H. cordata and identified by means of spectral analysis as protocatechuic acid, quercetin, apigenin, afzelin, hyperoside and quercitrin.

Phytochemical Constituents of Bistorta manshuriensis

  • Phytochemical investigation of the MeOH extract of the aerial parts of Bistorta manshuriensis resulted in the isolation of two cerebrosides, two lactams, six phenolic compounds and seven flavonoids. Their chemical structures were characterized by spectroscopic methods to be pinelloside (1), soyacerebroside I (2), pterolactam (3), 5-hydroxypyrrolidine-2-one (4), vanillic acid (5), caffeic acid methyl ester (6), protocatechuic acid (7), caffeic acid (8), 3,5-di-O-caffeoyl quinic acid methyl ester (9), chlorogenic acid methyl ester (10), avicularin (11), afzelin (12), quercetin (13), isoorientin (14), quercetin 3-O-${\beta}$-D-glucoside (15), quercitrin (16), and luteolin (17). The isolated compounds (1 - 4, 7, 12, 14) were isolated for the first time from this plant source and the compounds 1 - 4, 9 and 10 were first reported from the genus Bistorta. Compound 17 exhibited moderate cytotoxicity and compound 6 exhibited weak cytotoxicity against four human cancer cell lines in vitro using an SRB bioassay.

이질풀 추출물의 항산화 효능에 관한 연구 (Study on the Antioxidant Activity of Geranium nepalense subsp. thunbergii Extract)

  • 본 연구는 이질풀 추출물의 항산화 효능에 관한 연구로, 항산화 및 항노화의 기능성 화장품 소재로서의 개발 가능성을 확인하고자 하였다. 이질풀의 주요 성분으로는 tannin, (-)epicatechin, kaempferitin, kaepferol-7-rhamnoside, brevifolin, corilagin, pyrogallol, ellagitannin, geraniin, gallic acid, succinic acid, quercetin, protocatechuic acid 등이 보고되어 있다. DPPH 라디칼 소거능 평가를 통한 이질풀 추출물의 항산화 효능을 측정한 결과, 대조군인 quercetin과 비교하여 우수한 소거 활성을 확인할 수 있었다. 동일 농도($50\;{\mu}g/mL$)를 처리하였을 때 이질풀 추출물의 DPPH 라디칼 소거능은 약 98.33 %로 측정되었으며, quercetin의 경우 78.05 %로 측정되었다. Human keratinocyte (HaCaT)에 이질풀 추출물을 처리하여 세포내 Reactive Oxygen Species (ROS) 생성 저해능을 측정한 결과, $IC_{50}$값은 이질풀 추출물과 quercetin에서 각각 43.22, $102.35\;{\mu}g/mL$로 측정되어 이질풀 추출물의 우수한 항산화 효능을 확인할 수 있었다. 또한 피부 자극 테스트 및 세포 독성 평가를통하여 이질풀 추출물이 피부에 자극을 유발시키지 않는 안전한 소재임을 확인하였다.

자소엽(Perillae Folium) 열수추출물의 식물화학성분 연구 (Phytochemical Study of Hot-Water Extract of Perillae Folium)

  • In this study, 15 compounds were elucidated from the hot-water extract of Perillae Folium. Fifteen isolates were determined to be protocatechuic acid (1), caffeic acid (2), (R)-rosmarinic acid (3), (S)-shisoflavanone A (4), luteolin-7-O-β-D-glucuronopyranoside (5), scutellarein-7-O-β-D-glucuronopyranoside (6), apigenin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (7), luteolin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (8), kelampayoside A (9), trans-N-feruloyloctopamine (10), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acrylamide (11), perilloside C (12), perilloside A (13), (6S,9R)-9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside (14) and (6S,9R)-roseoside (15) through spectroscopic evidences. The HPLC analysis revealed that hot-water extract of Perillae Folium contained caffeic acid, rosmarinic acid and glycosides of apigenin, luteolin and scutellarein as main constituents.

GC/MS에 의한 잎담배중 Phenolic Acid의 분석 (Analysis of Phenolic Acids in Tobacco Leaf by GC/MS)

  • A GC/MS method was developed to analyze phenolic acid extract from tobacco leaf. Extracted acids were converted to their methyl esters by refluxing with 3M hydrogen chloride in methanol, and the esters were reacted with his (trimethylsilyl) trifluoroacetamide plus 10% trimethylchlorosilane to silylate the phenolic groups. Derivatives of standard salicylic, p-hydroxybenzoic, vanillic, gentisic, p-coumaric, syringic, ferulic, and sinapic acids prepared by this procedure were analyzed by GC/MS on $20m{\times}0.2mm$ column of SE-54 glass capillary. GC/MS analysis of the extract from tobacco leaf revealed the presence of salicylic, p-hydtoxybenzoic, vanillic, gentisic, protocatechuic, p-coumaric, syringic, gallic, ferulic, caffeic, sinapic, and quinic acids, respectively. The quantitative analysis of these phenolic acids were achieved by using multiple ion selection technique.

Pinus rigida Allelochemicals가 차풀종자의 발아과정에서 동위효소의 활성에 미치는 영향 (Effects of Pinus rigida Allelochemicals on Isozyme Activities during Seed Germination of Cassia mimosoides var.nomame)

  • Eleven phenolic compounds including caffeic acid were identified through analyzing the aqueous extracts of Pinus rigida by HPLC. Among them, protocatechuic acid was the maximum amount of 6.84 ppm. Seed germination of Cassia mimosoides var. nomame was significantly stimulated by the extract of P. rigida leaves in the proportion ot concentration. However, root growth was elevalted at a threshold concentration below 25%, but it was inhibited at high concentrations. In 50% extract of P. rigida, upward root tip of C. mimosoides var. nomame showed negageotropism which the root end showed necrosis. New isozyme bands were induced indicating concentration activity of peroxidase from the extract of C. mimosoides var. nomame, especially in the cathodic region. Although it reduced the mumber of isozyme bands of esterase, esterase activities were stimulated in the anodic region of C. mimosoides var. nomame. The activity of amylase was not remarkably different between control and treatment.
