In vitro Translation and Methylation of Iso-1-Cytochrome C from Saccharomyces Cerevisiae

  • The gene for iso-1-cytochrome c for Saccharomyces cerevisiae was recloned into a pSP65 vector containing an active bacteriophage SP6 promoter. The iso-1-cytochrome c gene was cloned as an 856 bp Xho 1-Hind III fragment. When the resulting plasmid was digested at the Hind 111 site 279 bases downstream from the termination codon of the gene and transcribed in vitro using SP6 RNA polymerase, full length transcripts were produced. The SP6 iso-1-cytochrome c mRNA was translated using a rabbit reticulocyte lysate system and the protein products analyzed on SDS polyacrylamide gels. One major band was detected by autofluorography. This band was found to have a molecular weight of 12,000 Da and coincided with the Coomassie staining band of apocytochrome c from S. cerebisiae. The product was also shown to be identical with that of standard yeast apocytochrome c on an isoelectric focusing gel. The in vitro synthesized iso-a-cytochrome c was methylated by adding partially purified S-adenosyl-L-methionine . protein-lysine N-methyltransferase (Protein methylase III; EC 2.1.1.43) from S. cerevisiae along with S-adenosyl-L-methionine to the in vitro translation mixtures. The methylation was shown to be inhibited by the addition of the methylase inhibitor S-adenosyl-L-homocysteine or the protein synthesis inhibitor pu omycin. The methyl derivatives in the protein were identified as $\varepsilon$-N-mono, di and trimethyllysine by amino acid analysis. The molar ratio of methyl groups incorporated to that of cytochrome c molecules synthesized showed that 23% of the translated cytochrome c molecules were methylated by protein methylase III.

On-farm evaluation of dietary animal and plant proteins to replace fishmeal in sub-adult olive flounder Paralichthys olivaceus

  • Background: High demand and low supply of fishmeal due to overexploitation of fisheries resources have resulted in a dramatic increase in the price of this ingredient. Olive flounder (Paralichthys olivaceus) commercial feed contains approximately 60% fishmeal and limited success has been achieved in identifying sustainable alternative protein sources for this species. Methods: An on-farm feeding trial was conducted to compare a basal diet containing 65% as the control (CONT) with two experimental diets replacing 10% of fishmeal by animal protein (AP10) or 20% of fishmeal by animal and plant protein (APP20). Sub-adult olive flounder averaging 327 ± 9.3 g (mean±SD) were fed one of the three diets in triplicate groups for 16 weeks. Results: Weight gain, specific growth rate, feed efficiency, protein efficiency ratio, and survival were not significantly different among fish fed all the experimental diets (P > 0.05). Also, non-specific immune responses (superoxide dismutase and lysozyme activity), serum biochemical parameters, and intestinal villi length were not significantly different among fish fed all the experimental diets (P > 0.05). Conclusions: Therefore, based on growth performance, non-specific immune responses, serum biochemical parameters, and intestinal histology, dietary animal and plant protein mixtures could replace up to 20% of fishmeal in the diet of sub-adult olive flounder.

Effect of Feeding Bypass Protein with Urea Treated Grass on the Performance of Red Kandhari Calves

  • An experiment was carried out to assess the effect of feeding concentrate mixtures varying in bypass protein levels with urea-treated or untreated grass on the performance of twelve Red Kandhari calves (14 months of age and 78.15 kg body weight) for a period of 75 days. Dry grass was treated with 4 percent urea solution and ensiled for 30 days. The CP ($N{\times}6.25$) content in urea treated grass increased from 3.96 to 8.89 percent. Two iso-caloric and iso-nitrogenous concentrate mixtures (CM-I and CM-II) varying in RDP to UDP ratio viz., 65:35 and 55.45 were prepared. The calves in control group ($T_1$) were fed concentrate mixture-I with ad libitum untreated dry grass and those in experimental group ($T_2$) were fed concentrate mixture-II with ad libitum urea treated dry grass. The dry matter consumption in group $T_2$ was significantly (p<0.01) higher as compared to group $T_1$. The total DMI in $T_1$ and $T_2$ was 146.92 and 166.95 kg respectively, whereas the DMI per day and per 100 kg body weight was 1.94 and 2.22 and 1.90 and 2.35 kg, respectively. The average total gain in body weight (kg) and average daily gain (g) of calves in $T_2$ was significantly (p<0.01) higher as compared to those in $T_1$ the values being 28.66, 18.33 and 382.16, 244.44, respectively. Feed efficiency in terms of kg DM per kg gain in body weight was significantly (p<0.01) lower in group $T_1$ than in $T_2$. The cost of feed per kg gain in body weight for $T_2$ and $T_1$ group was Rs. 21.14, 28.22, respectively. The digestibility coefficients of DM, CP, EE, CF, NFE, NDF and ADF were 59.60, 57.50, 53.00, 65.04, 45.82, 48.48, 52.48 and 55.73 for $T_1$ group. The coressponding values were 68.78, 67.80, 59.83, 71.41, 49.93, 53.37 and 57.81, respectively for $T_2$ group. The digestibility coefficients for all the proximate principles in $T_2$ were significantly (p<0.01) higher as compared to $T_1$. However, NDF and ADF digestibilities were not significantly different. Nutritive value determined in terms of DCP and TDN for The experimental ration was significantly (p<0.01) higher than control ration, the values being 7.32 and 47.34 and 9.39 and 52.40% respectively. The blood urea nitrogen levels at 0, 3 and 6 h interval after feeding were significantly (p<0.01) lower in calves fed experiment ration as compared to control. The overall results indicated that in Red Kandhari calves an optimum growth can be economically achieved by feeding 4 percent urea treated dry and mature grass as basal roughage supplemented with a concentrate mixture containing 20 percent CP, 70% TDN and 45% UDP/bypass protein.

돈육에서 추출한 염용성 단백질에 수용성 키토올리고당 및 키토산을 첨가한 혼합액과 가열 겔의 물성특성 (Rheological Properties of the Mixture and Heat-induced Gel Prepared from Pork Salt Soluble Protein in Combined with Water Soluble Chitooligosaccharide and Chitosan)

  • 본 연구는 돈육후지에서 추출한 염용성 단백질을 키토올리고당과 키토산의 분자량과 첨가량을 달리하여 pH와 점도를 측정하였으며, 가열 겔의 pH와 수분함량 및 경도를 측정하여 키토산첨가가 육제품의 물성에 어떤 영향을 주는지 알아보기 위하여 실시하였다. 키토산과 염용성 단백질 혼합물의 pH는 저분자 키토올리고당의 경우 0.45% 첨가 시 pH가 높아졌으나, 중분자 키토산을 0.45% 첨가 시 또는 고분자 키토산을 0.3% 이상을 첨가하였을 경우 pH가 오히려 낮았다(p<0.05). 점도는 첨가량이 0.3% 이상의 수준에서 중분자와 고분자 키토산 처리구가 저분자 키토올리고당 처리구나 대조구보다 높은 값을 나타냈다(p<0.05). 반면 가열 겔의 pH, 수분함량 및 경도에서는 키토산의 분자량과 첨가량에 따른 효과가 나타나지 않았다(p>0.05). 이상의 결과를 종합하면 키토산과 염용성 단백질의 혼합물에서 중분자나 고분자 키토산을 0.3% 이상 첨가 시 증가되었던 점성이 가열처리에 의해 겔이 형성됨으로써 경도의 차이가 미미해졌으나 다양한 첨가물과 가열공정에 의한 실제적인 육제품에서는 그 결과가 달라질 수 있을 것으로 판단된다. 앞으로의 연구에서 키토산을 실질적인 육제품에 첨가했을 때 어떠한 효과를 나타내는지 모델연구와 비교 및 평가가 요구된다.

  • 가소제로서 솔비톨 혼합물의 첨가가 분리대두단백 필름의 수분흡습특성, 수분투과도, 색, 인장강도 및 신장률, 그리고 용해 특성에 미치는 영향을 조사하였다. 솔비톨 혼합물은 액상형과 결정형의 상태로 다양한 비율로 혼합하여 첨가하였고, 가소제로 글리세린을 첨가한 대조구 필름의 물리적 특성과 비교하였다. 솔비톨이 첨가된 필름은 대조구 필름에 비해 높은 인장강도를 갖는 반면 낮은 수분투과도 및 신장률을 보였다. 한편 솔비톨 첨가 형태에 따른 영향은 필름 신장률에는 유의적 차이는 보였으나 기타 수분투과도, 색, 인장강도 등의 필름특성에는 커다란 차이는 보이지 않았다. 솔비톨 첨가된 필름은 높은 수분차단 효과 및 필름강도를 갖는 대신 높은 용해성을 보여, 상업적 활용성을 증진시키기 위해서는 이를 극복하는 연구가 필요할 것으로 사료된다.

혼파유형이 목초의 수량과 품질에 미치는 영향 (Effect of Mixture Type on the Herbage Yield and Quality)

  • 휴경지에서의 집약적인 단기간(3∼6년) 초지이용체계에 적합한 혼파유형을 제시하고자 orchardgrass (OG) 단파, orchardgrass(OG) + red clover(RC) 단순혼파 및 orchardgrass + tall fescue + Kentucky bluegrass + ladino clover에 의한 다초종 혼파(cormplex mixtures, C. Mix) 등의 처리를 두어 이들 혼파유형이 목초의 수량과 품질에 미치는 영향에 대하여 3년간(1989∼'91) 시험하였다. 3년간의 ha당 평균 건물수량은 OG구(10,709kg)나 C. Mix구(11,371kg)에 비하여 OG+RC구(11,849kg)가 높았다(P<0.05). 조단백질함량은 처리간에 차이가 없었으나 3년간 평균 가소화 유기물함량은 OG구보다 OG+RC구가 높았다(P<0.05). ha당 조단백질수량은 OG구(1,816kg)나 C. Mix구(2,053kg)에 비하여 OG+RCf(2.231kg)가 높았고(P<0.01), 가소화 유기물수량도 OG구(6,116kg)나 C. Mix구(6,817kg)에 비하여 OG+RC구(7,147kg)가 높았다(P<0.05). 한편 체중 kg당 건물섭취량은 처리간에 차이가 없었으나 두당 유기물 섭취량은 OG구(377g)보다 OG+RC구(388g)와 C. Mix구(392g)에서 높았다(P<0.01). 이상의 결과를 검토할 때 OG+RC 단순혼파가 단기간의 집약적인 초지이용에 가장 적합하다고 사료된다.

Effect of Cassoy-urea Pellet as a Protein Source in Concentrate on Ruminal Fementation and Digestibility in Cattle

  • Four male crossbred native beef cattle (average body weight of 427.7 kg) were randomly allocated to four types of cassoy-urea pellet as a source of protein in concentrate according to a $4{\times}4$ Latin square design to determine effect of diets on ruminal fermentation and nutrient digestibility. The four types of cassoy-urea pellets contained cassava hay, soybean meal, urea and binding agent at 79.2:19.8:0:1 (27.9% CP dry matter), 78.4:19.6:1:1 (30.4% CP), 77.6:19.4:2:1 (33.0% CP) and 99:0:0:1 (23.8% CP) for dietary treatments; 1, 2, 3 and 4, respectively. All four concentrate mixtures contained similar crude protein levels (11% CP) and were fed to animals in two equal parts (0.5% of body weight per day) while urea-treated rice straw (5% urea) was given ad libitum. The experiment revealed that dietary concentrate treatments had no effect on dry matter intake while digestibilities of neutral-detergent fiber and crude protein were higher (p<0.05) in cattle fed dietary treatments 1, 2 and 3 than in cattle fed dietary treatment 4. Ruminal ammonia-nitrogen ($NH_3$-N), was higher and acetic acid concentration (C2) and ratio of C2 to propionic acid (C3) were lower (p<0.05) in cattle fed dietary treatments 1, 2 and 3 than in those on treatment 4. It is concluded that use of cassoy-urea pellet as a protein source in concentrates for cattle resulted in improvement of digestibility, ruminal fermentation and rumen ecology. Further research using cassoy-urea pellet in feeding trials with milking cows and fattening beef should be undertaken.

제주지역 초지에서 혼파조합 및 질소수준이 식생구성 및 사초생산성에 미치는 영향 (Effects of Seed Mixture and Nitrogen Levels on Botanical Composition and Forage Productivity for Pasture in Jeju)

  • 본 연구는 친환경축산의 실천 및 유기조사료 생산을 위해 방목초지에서 혼파조합 및 질소시비 수준이 식생구성과 사초생산성에 미치는 영향을 평가하기 위하여 수행되었다. 본 시험은 2003년 8월부터 2005년 12월까지 난지농업연구소 시험포장에서 실시하였다. 시험처리는 혼파조합을 주구로 질소 시비수준을 세구로 하는 9처리 3반복 분할구배치법으로 배치하여 사초생산성과 식생구성변화를 조사하였다. 목초의 총 건물생산량은 두과혼파에 의해 높은 증가(P<0.01)를 보였으며, 질소시용 수준간에는 뚜렷한 차이를 보이지 않았으며 식생구성비율은 1년차 봄에 화본과의 비율은 80% 정도의 비율을 차지하였으나 여름과 가을에는 두과 비율이 94% 이상의 높은 비율을 나타냈다. 혼파조합에 따른 조단백질 함량은 화본과 단파구(T1) 보다 두과목초 혼파구가 14.9%에서 18.0%로 평균 3.1% 높게 나타났으며 질소시비 수준에 따라서는 16.6~17.5%로 뚜렷한 차이를 보이지 않았다.

Effect of Feeding Bypass Protein with Urea Treated Jowar Kadbi (Sorghum Straw) on Performance of Cross Bred (HF × DEONI) Calve

  • A study was made of the efficiency of ammonia N retention by Jowar kadbi (sorghum straw), initially 6.41% crude protein (CP), treated with 4% urea solution. After 30 days the CP in straw that was unchaffed and had been left uncovered was 10.02, and in chaffed straw that had been covered with a polythene sheet was 10.9%. The two treated straws were each fed to six crossbred (HF$\times$Deoni) calves, initially $12{\pm}2$ months old and $86.7{\pm}3.2kg$ bodyweight. They were also given two isocaloric (70% TDN) and isonitrogenous (20% CP) concentrate mixtures differing in calculated Rumen Degradable to Undegradable Dietary Protein ratio (RDP:UDP). Those fed the unchaffed uncovered treated straw (treatment C) received 65 RDP:35UDP and the other group (T1) received concentrate with a 55:45 ratio. The T1 group had the higher DM intake (p<0.01) in total (306 vs 268 kg), per day (4.1 vs 3.6 kg) and per unit bodyweight. Digestibility of DM, OM, CP and NDF, but not ADF, was higher in T1 and that group had the higher daily gain (517 vs 333 g) and higher total gain (38.8 vs 25.0 kg) over the 75 d of the feeding trial. It is concluded that chaffing and covering of Jowar kadbi treated with urea, not likely to be adopted by farmers because of financial constraints, does not confer important benefits. A concentrate supplement (estimated 45% of the CP as UDP) to calves given the treated straw has a beneficial effect on their growth and development.

당-단백질 가열반응 시에 생성되는 향기성분에 미치는 지질의 영향 (Effect of Lipid Mediated Glucose-Protein Reaction on Thermal Flayer Generation)

  • 당-단백질의 가열반응 시에 생성되는 향기성분의 형성에 지질의 산화생성물이 어떠한 영향을 미치는가를 알아보고자 casein과 glucose(CG) 그리고 지질을 첨가한 사료(CGL)를 6$0^{\circ}C$에서 각각 2, 4주씩 저장한 후에 pressure reactor에 넣어 16$0^{\circ}C$에서 60분 가열 반응시킨 시료에서 향기성분을 추출하였다. Casein, glucose, 지질(CGL) 시료에서 단백질과 환원당의 반응물이 지질의 산화생성물과 반응하여 생성된 함질소 헤테로 고리화합물인 pyrazine, methylpyrazine과 2,5-dimethylpyrazine, 2-ethyl-5-methylpyrazine과 2-acetylpyrrole이 확인되었다. 또한 지질의 가열산화 분해성분인 3-methyll-butanol, heptanal, nonanal, 2-heptanone과 짧은 사슬의 지방산인 heptanoic acid, octanoic 가 검출되었다. 또 다른 화합물로 2-acetylfuran이 검출되었으며 glucose의 분해산물인 5-hdroxymethyl-2-furfural은 aems 시료에서 가장 큰 peak로 그 함량이 현저하게 많았다. 이상의 결과로 보아 시료에 첨가된 지질은 당-단백질 가열반응 시 생성되는 향기성분의 수와 그 함량을 증대시키는 작용을 하였다고 할 수 있다
