Study on Extraction of Mucopolysaccharide-protein Containing Chondroitin Sulfate from Chicken Keel Cartilage

  • The objective of this study was to investigate technical methods for extraction of mucopolysachharide-protein containing chondroitin sulfate from keel cartilage of chickens. The chemical composition of chicken keel cartilage was determined. For the preparation of mucopolysaccharide-protein from lyophilized chicken keel cartilage, hot water extraction and alcalase hydrolysis methods were examined. Results showed that the optimum condition of hot water extraction was incubation for 120 min with a yield of 40.09% and chondroitin sulfate content of 28.46%. For alcalase hydrolysis, the most effective condition was 2% alcalase in 10 volumes of distilled water for 120 min. The yield of hydrolysate was 75.87%, and chondroitin sulfate content was 26.61%. For further separation of chondroitin sulfate from the alcalase hydrolysate, which has a higher yield than that of hot water, 60% ethanol precipitation was performed. The yield of the ethanol precipitate was 21.41% and its chondroitin sulfate content was 46.31%. The hot water extract, alcalase hydrolysate and ethanol precipitate showed similar electrophoretic migration with standard chondroitin sulfate (chondroitin sulfate A), using cellulose acetate membrane electrophoresis. These results indicated that a significant amount of mucopolysaccharide-protein containing chondroitin sulfate could be acquired form chicken keel cartilage. Therefore, keel cartilage in chicken may provide an inexpensive source of chondroitin sulfate for commercial purposes.

Effects of sea horse (Hippocampus abdominalis)-derived protein hydrolysate on skeletal muscle development

  • Hippocampus abdominalis, the big belly sea horse, is widely known for its medicinal value in Chinese folk medicine. In this study, extract obtained by proteolytic degradation of this species was investigated for its effects on skeletal muscle development, both in vitro and in vivo. Muscle cell lines ($C_2C_{12}$ and $L_6$) treated with the bioactive peptide did not have any detrimental effects on the cell viability, which was above 80%. Optical microscopy analysis on the morphology of the sea horse extract (SHE)-treated cells showed enhanced differentiating ability with myotube formation. Moreover, cells incubated with the hydrolysate displayed decreased proliferation rate, as recorded by the electric cell substrate impedance sensing system, thereby supporting enhanced differentiation. For a period of 12 weeks, mice models were fed with SHE and simultaneously subjected to treadmill exercise, which increased the expression of Myogenin, a key myogenic regulatory factor. In addition, there was an increase in the expression of AMPK- and Cytochrome C, both of which are important in mitochondrial biogenesis. Thus, the SHE from Hippocampus abdominalis can be a promising candidate as protein supplement aiding muscle development.

Peptide Inhibitor for Human Immunodeficiency Virus Type 1 (HIV-1) Protease from a Thermolysin Hydrolysate of Oyster Proteins

  • A peptide that inhibits HIV-1 protease was isolated from a hydrolysate of oyster (Crassostrea gigas) proteins digested with thermolysin. The peptide was using membrane filtration, gel permeation chromatography, ion exchange chromatography, and reverse-phase high performance liquid chromatography. Amino acid sequence of the peptide was determined to be Val-Phe-Glu-Leu. Chemically synthesized Val-Phe-Glu-Leu showed an $IC_{50}$ value of 106 ${\mu}M$.

Porcine Splenic Hydrolysate has Antioxidant Activity in vivo and in vitro

  • The antioxidant capacity of porcine splenic hydrolysate (PSH) was studied in vitro and in vivo. Peptide hydrolysates were prepared, using the proteolytic enzyme $Alcalase^{(R)}$. The molecular weights of PSH were 37,666, 10,673, 6,029, and 2,918 g/mol. Rats were fed a 5% (w/v) PSH diet, instead of a casein diet, for 4 wk. The food intake, body weight gain, and liver weight of rats in the PSH group were similar to those in the control (CONT) group. There were no differences in the serum total cholesterol, triglyceride, total protein, or albumin levels between PSH and CONT groups. However, the level of in vivo hepatic lipid peroxidation in PSH group was significantly lower than that in CONT. In vivo hepatic catalase and glutathione peroxidase activities in the PSH group were significantly higher than those in the control group. The in vitro protein digestibility of PSH was lower than that of casein. The in vitro trolox equivalent antioxidant capacity of PSH was significantly higher than that of the peptide hydrolysate from casein. The in vitro radical scavenging activities of PSH were significantly higher than those of the peptide hydrolysate from casein. The present findings suggest that porcine splenic peptides improve the antioxidant status in rats by enhancing hepatic catalase and GSH-Px activities, and indicate a potential mechanism of radical scavenging activity during gastrointestinal passage.

Characterization of antioxidative peptide purified from black eelpout (Lycodes diapterus) hydrolysate

  • The functional peptides from protein hydrolysates of various fishery sources have been identified such as antioxidant activity. The main intention of this study was purification and characterization of antioxidative peptide from black eelpout muscle. The antioxidative peptides were purified from black eelpout (Lycodes diapterus) muscle using different proteases. Antioxidant activity of black eelpout hydrolysates was evaluated using DPPH radical scavenging activity. Among six hydrolysates, the pepsin hydrolysate had the highest antioxidant activity compared to the other hydrolysates. Therefore, it was further purified and a peptide with seven amino acid residues of DLVKVEA (784 Da) was identified by amino acid sequence analysis. The EC50 value for scavenging DPPH radicals by purified peptide was 688.77 μM. Additionally, the purified peptide exhibited protective effect against DNA damage induces by oxidation in mouse macrophages (RAW 264.7 cells). The results of this study suggest that black eelpout muscle protein hydrolysate could potentially contribute to development of bioactive peptides in basic research.

The Novel Angiotensin I Converting Enzyme Inhibitory Peptide from Rainbow Trout Muscle Hydrolysate

  • The purpose of this study was the purification and characterization of an angiotensin I converting enzyme (ACE) inhibitory peptide purified from enzymatic hydrolysates of rainbow trout Oncorhynchus mykiss muscle. After removal of lipid, the approximate composition analysis of the rainbow trout revealed 24.4%, 1.7%, and 68.3% for protein, lipid, and moisture, respectively. Among six hydrolysates, the peptic hydrolysate exhibited the highest ACE inhibitory activity. We attempted to purify ACE inhibitory peptides from peptic hydrolysate using high performance liquid chromatography on an ODS column. The $IC_{50}$ value of purified ACE inhibitory peptide was $63.9{\mu}M$. The amino acid sequence of the peptide was identified as Lys-Val-Asn-Gly-Pro-Ala-Met-Ser-Pro-Asn-Ala-Asn, with a molecular weight of 1,220 Da, and the Lineweaver-Burk plots suggested that they act as a competitive inhibitor against ACE. Our study suggested that novel ACE inhibitory peptides purified from rainbow trout muscle protein may be beneficial as anti-hypertension compounds in functional foods.

분리대두단백질 가수분해물의 Angiotensin-I Converting Enzyme 저해효과 (Effect of Angiotensin-I Converting Enzyme Inhibitory from Hydrolysate of Soybean Protein Isolate)

  • 분리대두단백질의 가수분해물을 제조 시, 최적 가수분해 조건을 설정하기 위하여 총 7종의 단백질 가수분해 효소를 이용하여 효소선정, 효소농도, 가수분해시간 및 분리대두단백질분말의 첨가량에 따른 단백질의 함량 정도를 나타내는 280 nm의 흡광도 측정과 수율을 대신한 brix 측정, 가수분해물의 ACE 저해활성 등을 조사하였다. 분리대두단백질 분말에 증류수를 5%의 분산농도로 하여 분해효소 Alcalase 2.4 L을 1%(v/w) 첨가하여 $65^{\circ}C$에서 4시간 동안 가열하여 가수분해하였다. 가수분해를 마친 후 효소의 반응정지를 위하여 $80^{\circ}C$에서 5분간 열처리한 후, $4^{\circ}C$로 냉각하여 원심분리(7,000 rpm, 40 min) 하였다. 원심분리 한 상등액을 취하여 여과한 후, $60^{\circ}C$에서 농축하였다. 농축액을 냉동시켜, 진공 동결건조 하여 분리대두단백질 가수분해물을 제조하였다. 분리대두단백질 가수분해물의 수율은 38.32%를 나타냈으며, ACE 저해활성($IC_{50}$)은 $79.94 {\mu}g/mL$를 나타내었다.

효소 단백 가수분해물의 항균 활성 (Antimicrobial activity of protein hydrolysate by protease)

  • 밀 단백질에 효소가수분해 할 때 생산되는 peptide의 항균활성과 천연항균제로서의 이용가능성을 검토하기 위하여 실험을 행하였다. 밀 단백질에 7종의 단백질가수분해효소를 작용시켜 생성된 가수분해물의 항균활성을 측정하고 한외여과, membrane filtration, HPLC를 이용하여 항균성 peptide를 분리 정제한 후 분자량과 아미노산 결합순서를 측정한 결과는 다음과 같다. 밀 단백질에 7종의 단백질 분해효소를 적용시켜 제조한 가수분해물중 Asp. saito protease를 적용시켜 얻어진 peptide 만이 항균활성을 나타내었다. Asp. saito protease는 $37^{\circ}C$, pH 6.0에서 작용시킨 경우에 항균활성이 가장 높았으며, $50^{\circ}C$ 이상에서는 활성을 나타내지 않았다. 밀단백 효소가수분해물은 membrane filtration에 의하여 분자량 1,000~3,000 에서 항균활성이 나타났다. Membrane filtration으로 얻어진 항균활성분획을 HPLC로 분리한 결과 retention time 31.1~31.8 min에서 항균활성을 나타내었다. 밀단백 효소가수분해물은 $121^{\circ}C$에서 15분간 가열하여도 효소활성이 유지되는 매우 안정한 화합물이었다. 항균활성분획을 MALDI-mass로 질량을 분석한 결과 1,633이었다. 항균성 peptide의 아미노산 결합순서는 cysteine, glycine, prolin, prolin, prolin, valine, valine, alanine, alanine, arginine 의 순서였다.

Plant Proteins Differently Affect Body Fat Reduction in High-fat Fed Rats

  • This study examined the effects of corn gluten (CG), wheat gluten (WG), and soybean protein isolate (SPI), as well as their hydrolysates, on weight reduction in rats fed a high-fat diet. Eight-month-old male Sprague-Dawley rats (n=70) were fed a high-fat diet (40% of the calories were fat) for 4 weeks. Rats were then randomly divided into seven groups and were fed isocaloric diets with different protein sources for 8 weeks. The protein sources were casein (control group), intact CG (CG group), CG hydrolysate (CGH group), intact WG (WG group), WG hydrolysate (WGH group), intact SPI (SPI group), and SPI hydrolysate (SPIH group). Body weight gain, adipose tissue weights, lipid profiles in plasma and liver; and hepatic activities of carnitine palmitoyl transferase, fatty acid synthase (FAS), malic enzyme, and glucose-6-phosphate dehydrogenase were assessed. The CGH group showed significant weight reduction compared with the other groups. Epididymal fat pad and plasma triglycerides in the CGH group were the lowest and were significantly different than those in the control group. FAS activity in the CGH group was significantly lower than that in the other groups. In conclusion, the CGH diet of these experimental animals demonstrated a weight-reducing effect by lowering the adipose tissue weight and by affecting the activities of hepatic lipogenic enzymes.

팔당호에서 종속영양 활성도의 계절적 변화 및 세균의 세포외 효소활성 (Seasonal Fluctuations of Heterotrophic Activity and Bacterial Extracellular Enzyme Activity in Paldang Lake)

  • 수계생태계에서 유기물질의 순환을 이해하기 위하여 팔당호에서 종속영양 활성도와 세균세포의 효소활성의 계절절 변화를 연구하였다. 팔당호 I 의 glucose 전환시간은 수층, 퇴적토에서 2-1,300 시간, 17-170 시간, protein hydrolysate 는 5-900 시간, 15-240 시간, acetic acid 는 4-350 시간, 15-230 시간으로 계절적인 변화를 나타냈다. Glucose, protein hydrolysate, acetate 각각의 호흡율은 수층에서 23-32%, 38-41%, 22-28%로 나타났고 퇴적토에서는 34%, 61% and 41% 로 나타났다. 이 결과로 3가지 유기물질 종류 모두가 수층보다 퇴적토에서는 높은 율로 호흡됨을 알 수 있었다. 한편 세균의 $\alpha$-glucosidase, $\beta$-glucosidase, N-acetyl-$\beta$-D-glucosaminidase, aminopeptidase 활성력을 살펴본 결과 수층에서는 효소 각각에 대해 32-44%, 31-32%, 18-34% 61-67% 의 범위를 나타내었고 퇴적토에서는 34%, 40%, 23% 65%로 나타났다.
