Evaluating Nutritional Quality of Single Stage- and Two Stage-fermented Soybean Meal

  • This study investigated the nutritional quality of soybean meal (SBM) fermented by Aspergillus ($FSBM_A$) and/or followed by Lactobacillus fermentation ($FSBM_{A+L}$). Both fermented products significantly improved protein utilization of SBM with higher trichloroacetic acid (TCA) soluble true protein content, in vitro protein digestibility and available lysine content, especially in $FSBM_{A+L}$. Moreover, $FSBM_{A+L}$ produced a huge amount of lactic acid resulting in lower pH as compared to the unfermented SBM or soybean protein concentrate (SPC) (p<0.05). $FSBM_A$ and $FSBM_{A+L}$ raised 4.14% and 9.04% of essential amino acids and 5.38% and 9.37% of non-essential amino acids content, respectively. The ${\alpha}$-galactoside linkage oligosaccharides such as raffinose and stachyose content in $FSBM_A$ and $FSBM_{A+L}$ decreased significantly. The results of soluble protein fractions and distribution showed that the ratio of small protein fractions (<16 kDa) were 42.6% and 63.5% for $FSBM_A$ and $FSBM_{A+L}$, respectively, as compared to 7.2% for SBM, where the ratio of large size fractions (>55 kDa, mainly ${\beta}$-conglycinin) decreased to 9.4%, 5.4% and increased to 38.8%, respectively. There were no significant differences in ileal protein digestibility regardless of treatment groups. SPC inclusion in the diet showed a better protein digestibility than the SBM diet. In summary, soybean meal fermented by Aspergillus, especially through the consequent Lactobacillus fermentation, could increase the nutritional value as compared with unfermented SBM and is compatible with SPC.

Changes of Chemical Components During Seed Development in Black Soybean (Glycine max L.)

  • Changes in the level of metabolites in leaves and pods were examined with respect to the seed chemical composition in black soybean. There was no further increase in pod length after 42 days after flowering (DAF). Pod weight, however, persistently increase until 73 DAF, thereafter the weight was slightly lowered. The seed storage protein, however, increased drastically as the increasing rate of pod weight was lessened at 61 DAF. The accumulation of seed storage proteins was occurred conspicuously as the increasing rate of pod weight was slowed down. The chlorophyll content both in leaves and pods was drastically decreased after 50 DAF. The beginning of drastic reduction in chlorophyll content was occurred concomitantly with the reduction of soluble protein content in leaves. The sugar content in leaves showed similar tendency with chlorophyll and soluble protein content. The starch level in leaves, however, showed different changing pattern during seed development. The starch content in leaves was increased persistently until 66 DAF, thereafter the content was decreased drastically to about $55\%$ of maximal value at 66 DAF. Total phenolics content in leaves and the anthocyanins content in seeds were stable without noticeable increase until 66 DAF. The contents were increased dramatically after 66 DAF showing the synchronized pattern with the decrease in starch level in leaves. The levels of the selected metabolites in leaf and seed suggested that the accumulation of chemical components of black soybean seed is launched actively at 66 DAF. The profile of storage proteins was nearly completed at 61 DAF because there was no large difference in densitometric intensity among protein subunits after 61 DAF. In soybean, chemical maturation of seed begins around 61 to 66 DAF at which most metabolites in vegetative parts are decreased and remobilized into maturing seeds.

질소 시비조건에 따른 벼 품종의 미질과 식미특성 반응 (Varietal Response to Grain Quality and Palatability of Cooked Rice Influenced by Different Nitrogen Applications)

  • 질소수비 및 실비 시용량을 증시하여 미립내 단백질함량을 증가시킴으로써 이에 따른 식미저하와 쌀의 구조적 특성의 품종별 반응을 정밀하게 분석 검토하였으며, 그 결과를 요약하면 다음과 같다. 1. 현미 단백질함량은 질소 수비 실비의 증량에 따라 표준구(N6) 대비 $35{\sim}47%$까지 증가하였으며 식미가 낮은 품종일수록 증가 정도가 컸고, 동일한 질소 15kg/10a 수준으로 1,2차 수비가 충분히 준 조건에서도 1차 추비에 비해 실비를 준 경우 유메히까리와 레이호우는 현미 단백질함량이 유의하게 증가하였다. 2. 아밀로스함량은 질소 수비 실비의 증시에 따라 대체로 단백질함량과 반대로 변화정도는 품종간 차이보다 미미하지만 감소하는 경향을 나타내었다. 3. 식미 관능총평은 히노히까리 > 유메히까리 > 레이호우 순으로 높았는데 N 12 kg/10a 이상에서 질소 수비 실비 증시에 따라 유의하게 감소하는 경향을 보였으며 식미가 낮은 품종일수록 저하 정도가 컸고, 모든 식미관능 특성이 아밀로스함량보다 단백질함량 변화에 따라 크게 좌우되었으며 식미가 양호한 품종보다 식미가 낮은 품종에서 단백질함량 변화의 영향이 컸다. 4. 미립의 층위별 분포를 보아 외층에서 내층으로 갈수록 단백질함량은 현저히 감소하였고 아밀로스함량은 증가하였다. 식미가 양호한 품종이 낮은 품종에 비해 단백질함량의 분포가 겨층에서 높은 반면 백미층에서 크게 낮았으며 특히 아밀로스함량의 백미 외층 분포비율이 낮았다.

한외여과가 참깨박 농축단백질의 성분에 미치는 영향 (Effect of Ultrafiltration on the Components of Sesame Protein Concentrates)

  • Defatted sesame flour is the by-products obtained after oil extracting process. Although this flour has high quality and quantity of protein its use is limited only for animal feed and fertilization. Sesame seeds contain antinutrients such as oxalate, phytate and phenol compounds and these compounds lower their nutritive value. recently, ultrafiltration(UF) has been used to concentrate protein from various food sources. This study was carried out to examine the effects of UF with different membrane pore size on the components of sesame protein concentrates including antinutrients and to compare with that of conventional acid-precipitated sesame protein isolate. The protein contents of sesame protein concentrates prepared by JF using 10K, 30K, 100K were 84.2%, 82.7%, 76.4% and the protein yields were 36.44%, 34.69, 31.43% and the protein contents was 88.7% Alkali extraction process at pH 9.0 followed by UF technique reduced oxalate and phytate content. There were 85% and 94% reduction of oxalate and phytate content by UF with membrane pore size of 100K daltons, respectively. However, the content of total phenol compounds was not reduced by this method. About 99% of calcium and 50% of zinc were removed by UF with membrane of 100K daltons. total essential amino acid contents of sesame protein concentrates prepared by UF were decreased slightly when compared with acid-precipitated sesame protein concentrate.

인삼사포닌 분획이 단백부족 마우스의 면역체계에 미치는 영향 (Studies on the Effects of the Saponin Fraction of Panax Ginseng on the Immune System of the Mouse Fed with Protein Malnutritive Diet)

  • The thymus weight of the mouse was 54.1% in protein malnutritive diet group and 39.2% in group treated with saponin fraction of Panax ginseng in comparison to normal diet group. This decreasing effects of protein malnutritive diet and saponin fraction on the thymus weight practically disappeared after four weeks. The saponin fraction showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total peritoneal exudate cells, content of total serum protein and albumin content of the mouse, each 45, 8 and 10% respectively in comparision to that of normal diet group. And these values in protein malnutritive diet group were 61.2, 83.6 and 87.0% respectively in comparision to that of normal diet group, and recovered to the level of normal diet group by the supplement of the saponin fraction. The electrophoregram of the serum protein of the mouse fed with protein malnutritive diet was different from that of the mouse fed with normal diet, but this difference practically disappeared by the supplement of the saponin fraction.

장류제품의 아미노산 조성과 그 단백질 품질평가에 관한 연구 (Studies on the Amino Acid Composition of Korean Fermented Soybean Meju Products and the Evaluation of the Protein Quality)

  • 한국 재래식 장류 및 개량식 제품과 중간 가공물인 메주와 koji-mix 및 원료 대두에 대한 아미노산 조성과 available lysine 함량을 분석하고 이로부터 각 제품의 단백질 품질평가를 시도하였으며 Eggum의 단백질 chemical score와 biological value 와의 상관회귀 방정식을 이용하여 각 제품 단백질의 대략적인 biological value를 추산하였다. 메주용 대두의 amino-N 함량은 총 질소량의 85%에 달하나 장류 제품에는 총 질소량의 약 75%의 amino-N 함량으로 감소하는 반면 $NH_3-N$ 과 기타 N-compounds 함량이 증가한다. 이중 가내제조된 간장은 예외적으로 낮아 총 질소량의 53%에 불과한 amino-N을 함유하고 있다. 장류제조에서 재래식 방법과 개량식 제품 단백질의 질적변화의 차이는 크게 나타나지 않으며, 메주제조 단계까지는 원료 대두단백질의 영양가가 크게 손상되지 않으나 사임 담금이후 후숙 분리과정에서 단백가가 크게 손상되어 제품의 단백가는 원료 대두 단백질의 약 1/2에 불과한 chemical score를 나타내며 약 $1/3{\sim}1/2$에 미달되는 낮은 lysine 가용도를 갖게된다.

The Effects of Temperature, Starvation, and Kind of Foods on Growth, RNA/DNA Ratios and Protein Contents during the Early Life Stages of Olive Flounder (Paralichthys olivaceus)

  • The effects of temperatures, starvation, and kind of foods on growth, RNA/DNA ratio and protein contents during metamorphosis and early juvenile stage of olive flounder (Paralichthys olivaceus) were examined. During metamorphosis, warm-acclimated fish showed higher RNA and DNA content than those of the cold-acclimated fish, excepting H stage (28 DAH) at which the ratio was higher at cold temperature. RNA/DNA ratio during metamorphosis showed similar values at two temperatures tested. However, after 42 DAH warm-acclimated juveniles had higher DNA content compared with cold-acclimated fish, resulted in marked decreases in RNA/DNA ratios. Higher RNA content at H stage of cold-acclimated fish was consistent with an increase in protein content. Growth of fish rearing at warm temperature was higher than those of fish at cold temperature during all experiments. In starvation experiment, contents of DNA, RNA and protein significantly decreased. Even though there were no significant differences in total length (TL) and body weight between the live mysid-fed and artificial pellet-fed fish at 35 mm TL, both RNA/DNA and protein/DNA ratios of the former group was significantly higher than those of the latter due primarily to lower DNA content of the live mysid-fed group. The results from this study suggest that temperature, starvation and kind of foods should be considered when RNA/DNA ratio applied to assessing the cultured larval and juvenile fish condition.

Response of Grain Yield and Milled-Rice Protein Content to Nitrogen Topdress Timing at Panicle Initiation Stage of Rice

  • Response of grain yield and milled-rice protein content to nitrogen topdress (N) timing at panicle initiation stage (PIS) is critical for quantifying real-time N requirement for target grain yield and milled-rice protein content. Two split-split-plot experiments with three replications, one in 2004 and the other in 2005, were conducted in Experimental Farm, Seoul National University, Suwon, Korea. The experiments included three N rates at tillering stage (TS), three N timing treatments at panicle initiation stage (PIS) and two rice cultivars. The N rates at TS, N timing at PIS, and rice cultivars were randomly assigned to main plot, sub plot, and sub-sub plot, respectively. Results showed that the delayed N application at PIS reduced grain yield in 2004 and increased milled-rice protein content in both years significantly at 0.05 probability level. The calculated optimum N timing at PIS from pooled data by N rates and rice cultivars in two years was at 28 days before heading (DBH). However, real-time of N timing at PIS was dependent on plant growth and N status around PIS that in turn was dependent on applied N rates at TS. The optimum N timing at PIS was at 30 DBH for no N treatments at TS while at 27 DBH for 3.6 and 7.2 kg N/10a treatments and at 27 and 29 DBH for Hwaseongbyeo and Daeanbyeo, respectively. In general, earlier applied N at PIS resulted in lower milled-rice protein content but the highest grain yield was expected to be obtained when N topdress at PIS was applied at the time when shoot N concentration started to drop below about 23 mg/g due to dilution effect after transplanting. In conclusion, the results of our experiments imply that the currently recommended N topdress time (24DBH) at PIS in Korea should be reconsidered for the higher grain yield and the better quality of rice.

단백질 함량 향상을 위한 치킨스톡 제조 조건의 최적화 및 성분 분석 (Optimization of Chicken Stock Manufacturing Conditions to Improve Protein Content and Analysis of Its Components)

  • 본 연구는 다양한 식물성 원료의 첨가에 따른 치킨스톡의 영양성분 향상 및 단백질 용출을 증가시키기 위한 목적으로 수행되었다. 유기산에 의한 단백질 용출 증가를 목적으로 사과를 첨가한 치킨스톡(CSA, chicken stock added apple)을 제조하여 사과를 첨가하지 않은 치킨스톡(CS, chicken stock)과 비교하였다. 시료의 당도 분석 결과 CSA군이 CS군에 비해 비교적 높은 당도를 나타냈다. CSA군의 조단백 함량을 측정한 결과, CS군과 비교하여 유의적으로 높게 나타났다. 사과 첨가로 당도가 높게 나타나 사과의 양을 줄이고 유기산 함량이 높은 토마토를 첨가한 군(CSA-T, chicken stock added apple and tomato)과 토마토 및 레몬을 첨가한 군(CSA-TL, chicken stock added apple, tomato and lemon)을 추가하여 성분을 비교하였다. 치킨스톡의 조단백 함량을 측정한 결과, CSA-TL군의 조단백질 함량은 CSA-T군에 비해 35% 증가하였다. 이러한 결과를 바탕으로 CS군, CSA-TL군 및 시판용 치킨스톡(CCS, commercial chicken stock)군의 영양소 분석을 하여 비교하였다. 조단백질의 경우 CSA-T군은 CS군에 비해 약 79% 향상되었으며, 시판용 치킨스톡에 비해 약 6.8배 정도의 단백질을 함유하고 있는 것으로 나타났다. 아미노산 분석 결과 CS군에 비해 CSA-TL군에서 glutamic acid, aspartic acid 및 각종 필수아미노산의 함량이 증가하였다. 본 연구에서 개발한 치킨스톡은 기존 제품을 대체할 수 있는 건강지향적 제품으로 활용될 수 있을 것으로 기대된다.

남빙양 크릴육중에 함유된 불소의 감량에 관한 연구 (Decrease of Fluoride Content of Antarctic Krill)

  • 생 크릴근육 및 자비한 크릴근육에 이온화되어 있는 불소는 각각 총 불소함량이 70%, 49%이었으며 자비한 크릴근육의 총 불소함량이 생크릴근육에서 보다 높은 것으로 나타났다. 크릴근육의 가용성 단백질에 결합된 불소함량은 생크릴과 자비한 크릴 모두 pH4-6범위에서 높은 수준을 보였고 pH10 부근에서 가장 낮은 값을 보였다. 생 크릴근육의 가용성 단백질에 결합된 불소의 함량은 1가이온인 Na 염의 이온강도가 0.05, pH11일 때 약 8 ppm으로 감소되어 불소함량에 효과적인 조건인 것으로 조사되었다.
