담자균류의 약효 성분의 개발에 관한 연구

  • 말징버섯 Calvatia craniformis의 culture broth 40 liter를 여과하여 얻은 균사채를 열수 추출하여 진한 갈색 건조분말(Fr. A) 13.1g을 분리하였다. Fr. A에 대하여 DEAE-cellulose ion chromatography를 시행하여 중성분획인 흰색 분말 (Fr. B) 2.50g을, 산성분획인 갈색 분말 (Fr. C) 3.50g을 각각 분리하였다. Fr. B 500mg에 대하여 Sepharose CL-4B gel filtration chromatography를 시행하여 흰색분말 Fr. D(Calvatan) 350mg을 분리하였다. Calvatan 350mg에 대하여 Con A-Sepharose 4B affinity chromatography를 적용하여 미흡착 분획인 Fr. F($\alpha$-form)와 흡착 분획인 Fr. E ($\beta$-form)로 정제하였다. 항암작용의 기전을 구명하고자 마우스에 대하여 면역학적 실험을 시행하였다. macrophage의 활성에 대한 영향을 조사하였던 바, 투여군의 활성화된 macrophage에 의해 유리되는 superoxide anion의 양은 대조군에 비해 1.4배 증가하였고 Calvatan 투여군의 용혈반 형성세포(PFC)는 대조군에 비해 3.1배 증가하였다. 화학 분석에 의해, Calvatan은 다당체 87.2%, 단백질 1.8% 및 hexosamine 1.3%로 구성되어 있었다. 따라서 항암성 분획들은 protein-bound polysaccharide임을 알 수 있었다. 또한 각 분획의 다당체를 구성하고 있는 단당류는 glucose, galactose, mannose 및 xylose 였으며 단백질 부분은 16종의 아미노산으로 구성되어 있었다. IR 스펙트럼은 3300-3400 $cm^{-1}$에서 0-H stretching frequency, 2900 $cm^{-1}$ 에서 C-H stretching frequency, 1630 $cm^{-1}$ 에서 C-0 stretching frequency. 1000-1100 $cm^{-1}$에서 C-H 및 C-0 bending frequency를 나타내는 다당체의 전형적인 특성을 보여주었다.을 보여주었다.

Effect of Fucoidan on Expression of Diabetes Mellitus Related Genes in Mouse Adipocytes

  • Fucoidan (fucan sulfate) is a fucose-containing sulfated polysaccharide from brown algae such as Fucus vesiculosus, Ecklonia kurome, and Cladosiphon okamuranus. The aim of this study was to investigate the effect of fucoidan on the expression of diabetes-related genes in mouse cell line 3T3-L1. 3T3-L1 adipocytes were cultured for 48 hr with or without fucoidan (10, 100, and 500 ppm) on a 60 mm dish. Reverse transcription polymerase chain reaction (RT-PCR) was used for measurement of peroxisome proliferators activated receptor ${\gamma}\;(PPAR{\gamma})$, CCAAT/enhancer binding protein ${\alpha}\;(C/EBP{\gamma})$, and glucose transporter 4 (GLUT4) RT-PCR analysis revealed that expression level of GLUT4, $PPAR{\gamma}$, and $C/EBP{\alpha}$ mRNAs increased with fucoidan treatment from 10 to 500 ppm in a dose-dependent manner. Fucoidan appears to enhance insulin sensitivity by increasing the expression level of diabetes-related genes in 3T3-L1 adipocytes. Therefore, fucoidan is potentially useful as a natural therapeutic material for hyperglycemia in type II diabetes patients.

Inhibitory Effects of Red Ginseng on Passive Cutaneous Anaphylaxis and Scratching Behavior Reactions in Mice

  • To evaluate the antiatopic effect of Korea Red Ginseng (RG, steamed root of Panax ginseng C.A. Meyer, Family Araliaceae), its inhibitory effect on passive cutaneous anaphylaxis reaction and itching in mice was measured. RG and its ingredient saponin fraction (SF) potently inhibited passive cutaneous anaphylaxis (PCA) reaction and scratching behaviors. RG at a dose of 100 mg/kg and SF at a dose of 50 mg/kg significantly inhibited the scratching frequency by 32% and 38%, respectively. RG and SF also inhibited the degranulation and protein expression of tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-4 of RBL-2H3 cells induced by IgE-antign complex. However, polysaccharide fraction of RG did not inhibit it. Based on these findings, RG can improve allergic skin disorders atopic dermatitis and contact dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and their membrane stabilization.

MBR-RO 공정에서 MBR 운전조건에 따른 막오염 특성 및 RO 공정 막오염 평가 (Evaluation of membrane fouling by MBR operation conditions in MBR-RO)

  • This study compares characteristic of membrane fouling in MBR-RO systems. In lab. scale MBRs test, MBRs were operated at different Flux(10, 20, 30 & 40 LMH) and temperature(10, 15, 20, 25 & $30^{\circ}C$). The results show that MBR permeate was measured lower amounts of organic substances in Higher flux and lower temperature and led to lower RO fouling rates. The main cause was that due to cake fouling formed on membrane surfaces in MBRs. Under both cases, Cake layer of membrane surfaces formed in MBRs removed RO fouling factors, polysaccharide and protein, because of cake layer attached on membrane surfaces greater amounts of organic substances. This study implies that optimization of MBR with operating conditions is a crucial strategy to RO membrane fouling control.

Electron Beam 조사가 인삼분말의 사포닌 및 생리화학적 특성에 미치는 영향 (Effects of Electron-Beam Irradiation on Saponins and Physio-chemical Properties of Ginseng Powders)

  • 인삼분말에 대한 전자선(electron beam)의 이용가능성을 연구하고자, 방사선 살균처리(2.5∼15 kGy)가 백삼 및 홍삼분말의 사포닌 함량 및 패턴, 페놀함량, 산성다당체 함량, amylase activity, 단백질 패턴 등에 미치는 영향을 감마선 조사와 비교하였다. 사포닌 함량과 패턴을 TLC 및 HPLC로 분석해 본 결과, 조사선량과 에너지원에 따른 변화는 확인되지 않았으며, 실온 저장 4개월 이후에도 안정된 경향을 보였다. 총페놀성분 및 산성다당체 함량은 홍삼이 백삼 보다 높은 함량을 보였으며, 산성다당체 함량은 조사선량의 증가로 높은 추출량을 보였다. 백삼분말의 amylase activity는 방사선조사 직후 증가하였으나, 4개월 이후에는 대조구 보다 낮은 간을 보였으며, 추출단백질의 전기영동 패턴은 15 kGy의 전자선 및 감마선구에서도 대조구와 차이가 없었다.

수용액 및 연고기제중의 상피세포 성장 인자의 안정화 (Stabilization of Epidermal Growth Factor in Aqueous Solution and Ointment Base)

  • Epidermal growth factor (EGF) is a mitogen which activate the proliferation of basal cells in skin, which implicate the wound healing in severe skin damage such as burn. To carry out the preclinical test for the pharmacological action of EGF, EGF in transdermal delivery system must be stable. Since EGF is a protein susceptible to proteolysis and unstable in aqueous solution, in vitro stabilization of EGF is prerequisite for the formulation. In this study, effect of additives on the stability of EGF is investigated in vitro. The stability of EGF in aqueous solution was enhanced with the various water-soluble polysaccharides such as HPMC, sorbitol, mannitol and dextrin. EGF was successfully extracted from the ointment with 5% HPMC solution, and EGF in aqueous solution and ointment was also successfully stabilized with 5% HPMC. The ointments prepared with different amount of EGF were applied on the damaged dorsal skin of rats for the determination of optimal concentration of EGF. The ointment with EGF $(10\;{\mu}g/g)$ showed good wound healing action on the damaged skin of rats.

ERK Activation by Fucoidan Leads to Inhibition of Melanogenesis in Mel-Ab Cells

  • Fucoidan, a fucose-rich sulfated polysaccharide derived from brown seaweed in the class Phaeophyceae, has been widely studied for its possible health benefits. However, the potential of fucoidan as a possible treatment for hyperpigmentation is not fully understood. This study investigated the effects of fucoidan on melanogenesis and related signaling pathways using Mel-Ab cells. Fucoidan significantly decreased melanin content. While fucoidan treatment decreased tyrosinase activity, it did not do so directly. Western blot analysis indicated that fucoidan downregulated microphthalmia-associated transcription factor and reduced tyrosinase protein expression. Further investigation showed that fucoidan activated the extracellular signal-regulated kinase (ERK) pathway, suggesting a possible mechanism for the inhibition of melanin synthesis. Treatment with PD98059, a specific ERK inhibitor, resulted in the recovery of melanin production. Taken together, these findings suggest that fucoidan inhibits melanogenesis via ERK phosphorylation.

C/N비 변화가 호기성 그래뉼 슬러지의 안정성에 미치는 영향 (Effects on the Stability of Aerobic Granular Sludge (AGS) at Different Carbon/Nitrogen Ratio)

  • In this study, the effect on the stability of Aerobic Granular Sludge (AGS) with different Carbon/Nitrogen (C/N) ratios was investigated. The C/N ratios were controlled to 10.0, 7.5, 5.0, and 2.5 using the sequencing batch reactor, and the results showed that the removal efficiency of organic matter and total nitrogen decreased simultaneously with the decrease of C/N ratio. The removal efficiency of organic matter and total nitrogen at C/N ratio of 2.5 was 70.7% and 52.3% respectively. In addition, the AGS/mixed liquor suspended solids (MLSS) ratio showed a tendency to decrease from 85.7% to 73.7%, while the sludge volume index showed a tendency to increase from 82 mL/g to 102 mL/g as the C/N ratio decreased. At the same time, the apparent deviation of polysaccharide (PS) content in extracellular polymeric substances was observed, and polysaccharides/protein (PS/PN) ratio decreased from 0.62 to 0.31 as the C/N ratio decreased. Optical microscope observations showed that the reduction in C/N ratio caused the growth of filamentous bacteria and significantly affected the stability of AGS.

Rhamnogalacturonan I-rich fractions from cherry tomatoes stimulate phagocytosis in RAW 264.7 macrophages

  • Tomato (Lycopersicon esculentum) is widely known for its beneficial effects on human health. To investigate the beneficial effects of polysaccharides from cherry tomato, cherry tomato polysaccharides (CTP) were prepared, the component sugars were analyzed, and the immunomodulatory activities in RAW 264.7 macrophages were assessed. CTP mainly contained arabinose (Ara) and galactose (Gal), suggesting that CTP might be enriched with an arabinogalactan (AG) moiety. The Ara and Gal present in CTP are likely components of AG-II (35.4%), namely $arabino-{\beta}-(3,6)-galactan$. To investigate the immunomodulatory activity of CTP, cytokine levels and iNOS2, COX-2, and $NF-{\kappa}B$ protein levels were analyzed, and $NF-{\kappa}B$ nuclear translocation and phagocytosis were observed by immunofluorescence. CTP significantly increased the levels of $TNF-{\alpha}$, MCP-1, and IL-6. CTP also increased iNOS2 and COX-2 expression as well as $NF-{\kappa}B$ nuclear translocation in RAW 264.7 cells. CTP significantly stimulated phagocytosis activity. These results showed that CTP stimulates macrophage activity, which can boost the innate immune response. CTP with high AG-II content could be used as a prebiotic to strengthen immunity.

고령소비자를 위한 칼슘 강화 식단 개발 (Development of Calcium Enriched Menu for the Aged)

  • The objectives of this study were to develop calcium enriched menu for the aged and to investigate physicochemical characteristics and consumer acceptances of the menus. four kinds of the menu were developed with emphasis on calcium enriched food materials of red pepper leaves seasoned vegetables for menu1, shrimp for menu2, anchovy for menu3, seaweed for menu4. one portion of calorie, polysaccharide, protein, fats and calcium for the aged were calculated as 567kcal, 92.8g, 21.3g, 12.6g 223.1mg, respectively based on the recommended daily allowances for adults. Calcium content increased in the lowest value of all(p<0.05). The hunter lightness(L) values of barely rice in menu 1, radish soup in menu 1, chopped roast chicken in menu 4 and radish salad in menu 2 had significantly the highest values of all(p<0.05). The calcium enriched menus for the aged were successfully developed and these data could be used as basic informations for the improvement in health and life of the aged.
