Isolation of Marine Bacteria Killing Red Tide Microalgae II. Isolation and Algicidal Properties of Pseudomonas sp. LG-2 Possessing Killing Activity for Dinoflagellate, Prorocentrum micans (적조생물 살조세균 탐색 II. 적조생물 Prorocentrum micans 살조세균 Pseudomonas sp. LG-2의 분리와 살조특성)

  • We have isolated a bacterial strain that tends to kill P. micans from the mixed culture of p. minns plus seawater filtrate (poresize, 0.8 $\mu$m) collected at Masan bay in July 1996, in which the mixed culture grown in the f/2 medium. According to the experimental results of the isolated bacterium such as fatty acids analysis, morphological and biochemical characteristic tests, the strain was supposed to be a Pseudomonas and then it was named as Pseudomonas sp. LG-2. The killing effect of Pseudomonas sp. LG-2 against P. micans was proportionally increased with the concentrations of culture filtrate (pore size, 0.8 $\mu$m) is well as with the number of bacterium inoculated. In the mixed culture inoculated with $1.3\times10^6$ cells/ml of Pseudomonas sp. LG-2, the number of P. micans (2,000 cells/ml) was gradually decreased and then killed below 100 cells/ml within 7 days. In addition, the culture filtrate with $30\%$ of final concentration revealed a significant killing effect against P. micans around 3 days after culture. In the relationship between killing effects and growth stage of Pseudomonas sp. LG-2, the culture filtrate at lag phase has little effects on P. micans. In constant, the culture filtrate at mid-log phase showed the killing effect by decreasing P. micans to 112 in number within 5 days. In particular, the culture filtrate at stationary phase showed a significant killing effect against P. micans in which the majority of it was killed after 3 day culture. The species specificity of killing effects of Pseudomonas sp. LG-2 against 5 species of dinoflagellate was only found in P. micans and Scrippsiella trochoidea.

  • Protein profiles of a common mixotrophic dinoflagellate, Prorocentrum micans, growing autotrophically and mixotrophically (fed on the cryptophyte Rhodomonas salina) were compared using two-dimensional gel electrophoresis (2-DE) to determine if they vary in different trophic modes. Approximately 2.3% of the detected proteins were differentially expressed in the different trophic modes. Twelve proteins observed only in the mixotrophic condition had lower pI value (<5) than the fifteen proteins observed only in the autotrophic condition (>5). When the internal amino acid sequences of five selected proteins differentially expressed between autotrophic and mixotrophic conditions were analyzed using matrix-assisted laser desorption time-of-flight (MALDI-TOF) mass spectrometry, two proteins that were specifically expressed in the autotrophic condition showed homology to glyceraldehyde-3-phosphatase dehydrogenase (GAPDH) and a bacterial catalase. Three mixotrophy-specific proteins showed homology to certain hypothetical proteins from an insect and bacteria. These results suggested the presence of certain gene groups that are switched on and off according to the trophic mode of P. micans.

Algal Bloom and Distribution of Prorocentrum Population in Masan-Jinhae Bay (마산-진해만에서 Prorocentrum 개체군의 발생양상과 분포)

  • Genus Prorocentrum of dinoflagellate has been known as representative causative algae of red tide in Masan-Jinhae Bay. Prorocentrum populations- P. dentatum Stein, p. micans Ehrenberg, P. minimum (Pavillard) Schiller and P. triestinum Schiller- were monitored from January 1990 to August 1997, Prorocentrum populations usually have bloomed during the water stratification periods from June to August. Water temperature ranged from 24$^{\circ}C$ to 28$^{\circ}C$ and salinity from 24$\textperthousand$ to 34$\textperthousand$ during the Prorocentrum blooms. Bloom magnitude of Prorooentrum populations gradually increased from offshore to inshore area of coastal embayment and this populations tended to concentrate from surface to 2.5m depth. Prorocentrum algal blooms have become more frequent and intense than those of 1980s and early 1990s. P. minimum was the most persistent species in terms of bloom frequency and cellular abundance.

  • The causative organism of red tied occurred mainly in embayments along southern coast of Korea mostly composes of dinoflagellate species. Among them Genus Prorocentrum is predominant in Masan Bay and it has already reported eight Prorocentrum species from this area. The authors have identified five species of Genus Prorocentrum during the year of 1982 in the surveyed area and each species was described by means of the observation with light microscope as well as scanning electron microscopy. An additional species, p.dentatum is new to Korean waters.

Reduction in CO2 uptake rates of red tide dinoflagellates due to mixotrophy

  • We investigated a possible reduction in $CO_2$ uptake rate by phototrophic red tide dinoflagellates arising from mixotrophy. We measured the daily ingestion rates of Prorocentrum minimum by Prorocentrum micans over 5 days in 10 L experimental bottles, and the uptake rates of total dissolved inorganic carbon ($C_T$) by a mixture of P. micans and P. minimum(mixotrophic growth), and for the predator P. micans (phototrophic growth; control) and prey P. minimum (phototrophic growth; control) alone. To account for the effect of pH on the phototrophic growth rates of P. micans and P. minimum, measurements of $C_T$ and pH in the predator and prey control bottles were continued until the pH reached the same level (pH 9.5) as that in the experimental bottles on the final day of incubation. The measured total $C_T$ uptake rate by the mixture of P. micans and P. minimum changed from 123 to $161{\mu}mol\;C_T\;kg^{-1}\;d^{-1}$ over the course of the experiment, and was lower than the $C_T$ uptake rates shown by P. micans and P. minimum in the predator and prey control bottles, respectively, which changed from 132 to $17{\mu}mol\;C_T\;kg^{-1}\;d^{-1}$ over the course of the experiment. The reduction in total $C_T$ uptake rate arising from the mixotrophy of P. micans was 7-31% of the daily $C_T$ uptake rate seen during photosynthesis. The results suggest that red tide dinoflagellates take up less $C_T$ during mixotrophy.

  • Food items of the five species of marine cladocerans were reported to be largely confined to centric diatoms. Reexamination of the prey items, however, revealed that flagellar organisms were also well fed by cladocerans. This additional finding suggests that marine cladocerans might extend their food items to weakly motile flagellates e.g., Prorocentrum micans, Protoperidinium sp. and unidentified flagellates.

  • Identified 7 species of the genus Prorocentrum which have been obtained from the southern coast area for 4 years from 1990 to 1994 can be summarized as followed. P.balticum is rare species, causing a red tide, and P.dentatum, P.micans, P.minimum, P.triestinum are cosmopolitan species often causing a red tide in the study area. P.gracile and P.lima are very rarely showed up, the former is recorded at first in domestic and later is benthic attached species which has diarrheic shellfish poison.

  • The ability of a red tide species to take up nutrients is a critical factor affecting its red tide dynamics and species competition. Nutrient uptake by red tide species has been conventionally measured by incubating nutrient-depleted cells for a short period at 1 or 2 light intensities. This method may be applicable to certain conditions under which cells remain in oligotrophic water for a long time and high nutrients are suddenly introduced. Thus, a new method should be developed that can be applicable to the conditions under which cells are maintained in eutrophicated waters in healthy conditions and experience light and dark cycles and different light intensities during vertical migration. In this study, a new repletion method reflecting these conditions was developed. The nitrate uptake rates of the red tide dinoflagellate Prorocentrum micans originally maintained in nitrate repletion and depletion conditions as a function of nitrate concentration were measured. With increasing light intensity from 10 to $100{\mu}E\;m^{-2}s^{-1}$, the maximum nitrate uptake rate ($V_{max}$) of P. micans increased from 3.6 to $10.8 pM\;cell^{-1}d^{-1}$ and the half saturation constant ($K_{s-NO3}$) increased from 4.1 to $6.9{\mu}M$. At $20{\mu}E\;m^{-2}s^{-1}$, the $V_{max}$ and $K_{s-NO3}$ of P. micans originally maintained in a nitrate repletion condition were similar to those maintained in a nitrate depletion condition. Thus, differences in cells under nutrient repletion and depletion conditions may not affect $K_{s-NO3}$ and $V_{max}$. Moreover, different light intensities may cause differences in the nitrate uptake of migratory phototrophic dinoflagellates.

Molecular Phylogeny of Phytoplakton Isolated from Red Tides in Southern Coast of Korea (국내 남해안에 발생한 적조원인생물들의 24S rRNA 유전자 염기서열분석)

  • Cultured isolates of Prorocentrum minimum, P. micans, P. triestinum, P. balticum, Gymnodinium sanguineum, Alexandrium catenella, Scrippsiella trochoidea, and Heterosigma akashiwo from red tides in southern coast of Korea were phylogenetically compared on the basis of their 24S rRNA genes. For each isolate approximately 700 bp of 24S rDNA, encompassing D1 and D2 hypervariable domains, was amplified using the polymerase chain reaction and sequenced. The sequences were aligned with those reported in Genbank by using ClustalW program and phylogenetic tree was generated to show that morphospecies designations in the Alexandrium and Prorocentrum species are congruent with terminal taxa defined by phylogenetic analysis of the 24S rRNA fragment. Our results suggest that the molecular phylogeny approach could facilitate identification of domestic dinoflagellates which have ambiguous morphologies.

Singlet Oxygen Quenching by Deoxygadusol and Related Mycosporine-Like Amino Acids from Phytoplankton Prorocentrum micans

  • Deoxygadusol (DO) and structurally related mycosporine-like amino acids, i.e. mycosporine glycine (MO) and mycosporine taurine (MT), were isolated from phytoplankton Prorocentrum micans and studied for the reactivity toward singlet oxygen. These water-soluble compounds with a cyclohexenone chromophore were all shown to be highly effective in quenching singlet oxygen ($^1$ $O_2$), with the efficiencies being significantly larger compared with histidine, a well-known $^1$ $O_2$ quencher. The $^1$ $O_2$ reaction rate constant ( $k_{Q}$) of DG was determined to be 5.4 ${\times}$ 10$^{7}$ $M^{-1}$ $s^{-1}$ by a steady state method based on competitive inhibition of rubrene oxidation. The feasibility of this method was confirmed by estimating the $k_{Q}$ values for MG and two other quenchers, furfuryl alcohol and 1,4-diazabicyclo [2,2,2]octane, and comparing with those values determined by the time-resolved $^1$ $O_2$ decay method in the previous work.ork.
