품종과 보존제의 처리가 춘계수확 연맥의 특성과 사료가치에 미치는 영향 (Effect of Cultivar and Preservative on Characteristics and Quality of Spring Harvested Oat Hay)

  • An experiment was carried out to determine effect of cultivar and preservative on the characteristics and quality of baled oat(Avena sativa L.) hay after 60 days of storage. Ihe main plots consisted of the cultivars such as 'Foothill' and 'Swan', and the subplots consisted of preservatives such as commercial propionic acid(CA), lactic bacteria inoculant(LB) and control(C0). Bale temperature of 'Foothill' oat hay maintained higher than that of 'Swan' oat hay during storage. Bale temperature of oat hay treated with CA remained lowest during the earlier storage period and no significant difference was found among preservatives during the latter storage period. Visual assessment of bale of 'Swan' oat hay was better in the evaluation of mold, odor and color than that of 'Foothill' oat hay after storage. DM content of Foothill oat hay increased markedly than that of 'Swan' oat hay through the storage period. CFU(Colony fodng unit) of total viable micmrganism of oat hay bales was not quite different between preservatives, but that of lactic bacteria of 'Swan' oat hay and 'Foothill' was different by over 16 and lo4, respectively. Total nitrogen content of 'Swan' and 'Foothill' oat hay was 1.45 and 2.22%, respectively(P<0.05) and effect of preservatives on total N content was higher in the order of LB, CA, and CO.   • 디올 성분으로 폴리(테트라메틸렌 글리콜)(PTMG)과 함께 말단에 히드록시에틸기를 갖는 폴리디메틸실록산(PDMS-OH)을 혼합하여 사용하면서, 이소시아네이트 화합물로 이소포론 디이소시아네이트를, 수분산성을 부여하기 위해 디메틸올 프로피온산을, 가교제로 트리메틸올프로판을, 사슬연장제로 부탄디올을 사용하여 실록산 성분을 포함하는 선형 및 가교형 수분산성 폴리우레탄(WPU-Si)을 합성하였다. PTMG와 PDMS-OH 혼합 비율을 달리하여 합성한 선형 및 가교형 WPU-Si 에멀젼의 안정성을 분석하였고, 이들을 필름으로 제조하여 표면 특성과 $T_g$, 인장 강신도 변화를 분석하였다. PDMS 함량이 많아질수록 시료의 소수성이 증가하고 $T_g$가 높아졌으며, 파단 인장강도는 증가하고 파단 신장률은 감소하였다.

Purification and Properties of Extracellular Lipases with Transesterification Activity and 1,3-Regioselectivity from Rhizomucor miehei and Rhizopus oryzae

  • Rhizomucor miehei NRRL 5282 and Rhizopus oryzae NRRL 1526 can produce lipases with high synthetic activities in wheat bran-based solid-state culture. In this study, the purification and biochemical characterization of the lipolytic activities of these lipases are presented. SDS-PAGE indicated a molecular mass of about 55 and 35 kDa for the purified R. miehei and Rh. oryzae enzymes, respectively. p-Nitrophenyl palmitate (pNPP) hydrolysis was maximal at $40^{\circ}C$ and pH 7.0 for the R. miehei lipase, and at $30^{\circ}C$ and pH 5.2 for the Rh. oryzae enzyme. The enzymes showed almost equal affinity to pNPP, but the $V_{max}$ of the Rh. oryzae lipase was about 1.13 times higher than that determined for R. miehei using the same substrate. For both enzymes, a dramatic loss of activity was observed in the presence of 5 mM $Hg^{2+}$, $Zn^{2+}$, or $Mn^{2+}$, 10 mM N-bromosuccinimide or sodium dodecyl sulfate, and 5-10% (v/v) of hexanol or butanol. At the same time, they proved to be extraordinarily stable in the presence of n-hexane, cyclohexane, n-heptane, and isooctane. Moreover, isopentanol up to 10% (v/v) and propionic acid in 1 mM concentrations increased the pNPP hydrolyzing activity of R. miehei lipase. Both enzymes had 1,3-regioselectivity, and efficiently hydrolyzed p-nitrophenyl (pNP) esters with C8-C16 acids, exhibiting maximum activity towards pNP-caprylate (R. miehei) and pNP-dodecanoate (Rh. oryzae). The purified lipases are promising candidates for various biotechnological applications.

선형과 가교형 수분산 폴리우레탄 엘라스토머의 합성 및 물성 (Synthesis and Properties of Linear and Crosslinkable Polyurethane Elastomers)

  • 선형 수분간 폴리우레탄과 가교형 수분산 폴리우레탄을 2,4-톨루엔다이이소시아네이트, 다이메틸올프로피오닉에시드 및 폴리옥시프로필렌글리콜 또는 폴리옥시프로필렌글리세린를 사용하여 합성하였다. 합성된 중합체의 구조는 $^1H-NMR$ 및 FT-IR을 이용하여 분광학적 방법으로 측정하였고, 열적 성질은 DSC 및 TGA로 기계적 특성은 Instron, 입자의 형태는 AFM등으로 조사하였다. 수분산 폴리우레탄의 합성에서 폴리올의 분자량이 증가할수록 에멀젼의 입자 크기가 가교도가 증가할수록 에멀젼의 입자 크기, 점도 및 유리전이온도가 증가하였다. 폴리옥시프로필렌글리콜에 폴리옥시프로필렌글리세린을 혼합하여 합성하는 가교형 수분산 폴리우레탄에서는 폴리옥시프로필렌글리세린의 함량이 15% 이상부터는 수분산이 이루어지지 않았다. 따라서 본 연구에서는 폴리옥시프로필렌글리세린의 함량을 5%, 8%, 13% 및 15%로 한정하여 합성하였다.

Survey of ERETIC2 NMR for quantification

  • The ERETIC (Electronic REference To access In vivo Concentrations)2 method is a new qNMR experimental technique to measure analytes based on the signal of the reference compound without additional hardware equipment. In this study, ERETIC2 method was validated, and we sought to identify whether it would be possible to apply this method to a specific compound analysis of metabolites in plant. The $90^{\circ}$ pulse value (P1) and spin-lattice relaxation time ($T_1$) of each compound were measured for ERETIC2. The $9^1H$ of 3-(trimethylsilyl) propionic-2,2,3,3-$d_4$ acid (TSP) was used as a reference peak for ERETIC 2, and then, a suitable solvent and pulse sequence for each compound were selected. Under the NOESY-presat sequence, the relative accuracy error for quantitative analyses of primary metabolites was within the range of 5%, with the exception of glucose, which showed ${\geq}$ 55% error due to saturation. It showed excellent results for the quantification of glucose by using a $30^{\circ}$ pulse sequence, which did not suppress the water peak. In addition, the quantitative accuracy for secondary metabolites was extremely accurate, with an error ${\leq}$5% when considering the purity of the standard sample. The ERETIC2 method showed outstanding linearity, precision, and accuracy.

Castor oil based hyperbranched polyester/bitumen modified fly ash nanocomposite

  • A low cost environmentally benign surface coating binder is highly desirable in the field of material science. In this report, castor oil based hyperbranched polyester/bitumen modified fly ash nanocomposites were fabricated to achieve the desired performance. The hyperbranched polyester resin was synthesized by a three-step one pot condensation reaction using monoglyceride of castor oil based carboxyl terminated pre-polymer and 2,2-bis (hydroxymethyl) propionic acid. Also, the bulk fly ash of paper industry waste was converted to hydrophilic nano fly ash by ultrasonication followed by transforming it to an organonano fly ash by the modification with bitumen. The synthesized polyester resin and its nanocomposites were characterized by different analytical and spectroscopic tools. The nanocomposite obtained in presence of 20 wt% styrene (with respect to polyester) was found to be more homogeneous and stable compared to nanocomposite without styrene. The performance in terms of tensile strength, impact resistance, scratch hardness, chemical resistance and thermal stability was found to be improved significantly after formation of nanocomposite compared to the pristine system after curing with bisphenol-A based epoxy and poly(amido amine). The overall results of transmission electron microscopic (TEM) analysis and performance showed good exfoliation of the nano fly ash in the polyester matrix. Thus the studied nanocomposites would open up a new avenue on development of low cost high performing surface coating materials.

수분산성 폴리우레탄의 합성 및 물성에 관한 연구 (Study on Synthesis and Properties of Water-born Polyurethane)

  • 폴리우레탄 프리폴리머들이 폴리올과 디이소시아네이트로부터 제조되었다. 이온화되지 않은 폴리우레탄 프리폴리머들은 폴리프로필렌 글리콜(PPG, MW:1000), 디메치롤 프로피온산(DMPA)과 이소포론 디이소시아네이트(IPDI)를 사용하여 합성되었다. 폴리올과 디이소시아네이트의 몰비를 변화시키면서 합성된 폴리우레탄 프리폴리머들은 물에 분산된 후 물성을 측정하였다. 실험결과는 프리폴리머가 하드세그먼트 40%, NCO 3.43%, [NCO]:[OH]=1.5:1.0의 조건에서 가장 안정한 분산액을 보여 주었고, 30%의 고형분으로 물에 분산되었다. 폴리우레탄 프리폴리머들은 또한 PPG와 DMPA의 몰비를 변화시키면서 합성되었다. DMPA의 몰비가 높을 때 폴리우레탄 수분산액의 입경은 감소하였다 3가지 종류의 말단기 차단제(blocking agent)를 사용하여 제조된 폴리우레탄 프리폴리머들은 사용된 차단제에 따라 각각 다른 초기 해리 온도를 나타내었고 3가지 종류의 차단제에 대한 해리의 시작은 모두 30분 안에 일어났다.

Microsized Corn-Oil Droplet (MOD)의 Trichloroethylene (TCE) 생물학적 탈염소화 분해 자연저감 완효성 촉진제 적용성 평가 (Feasibility of Natural Attenuation for TCE Anaerobic Reductive Dechlorination Using Microsized Corn-Oil Droplet as an Activator)

  • Recently, enhanced in situ bioremediation using slow substrate release techniques has been actively researched for managing TCE-contaminated groundwater. This study conducted a lab-scale batch reactor experiment to evaluate the feasibility of natural attenuation for TCE dechlorination using microsized corn-oil droplet (MOD) as an activator considering the following three factors: 1) TCE dechlorination in the presence or absence of MOD; 2) TCE dechlorination in the presence or absence of inactivator of native microbial activity; and 3) MOD concentration effects on TCE dechlorination. Batch reactors were constructed using site groundwater and soil in which Dehalococcoides bacteria were present. Without MOD, TCE was decomposed into dichloroethylene (DCE). However, other by-products of TCE dechlorination were not detected. With MOD, DCE, vinyl chloride (VC), and ethylene (ETH) were sequentially observed. This result confirmed that MOD effectively supplied electrons to complete dechlorination of TCE to ETH. However, when an excess of MOD was provided, it formed unfavorable conditions for anaerobic digestion because dechlorination reaction did not proceed while propionic acid was accumulated after DCE was generated. Therefore, if an appropriate amount of MOD is supplied, MOD can be effectively used as a natural reduction activator to promote biodegradation in an aquifer contaminated by TCE.

혹서기 무창계사에서 육계의 혈액지질 및 짧은 사슬지방산에 관한 역전점등과 냉각수 효과 (Effect of cooling water and inverse lighting on short chain fatty acid and blood lipid of broiler chickens in closed poultry house during hot weather)

  • This experiment evaluated the interaction effect of extreme heat diet(EHD), inverse lighting, and cool water on the growth performance of broiler chickens under extreme heat stress. There were 4 experimental groups (T1: EHD 1, 10:00-19:00 dark, 19:00-10:00 light, cold water $9^{\circ}C$; T2: EHD 2, 10:00-19:00 dark, 19:00-10:00 light, cold water $9^{\circ}C$; T3: EHD 1, 09:00-18:00 dark, 18:00-09:00 light, cold water $14^{\circ}C$; T4: EHD 2, 09:00-18:00 dark, 18:00-09:00 light, cold water $14^{\circ}C$), each group composed of 25 broilers and the experiment was repeated 3 times. EHD 1 contained soybean oil, molasses, methionine and lysine. EHD 2 contained all nutrients of EHD 1 and vitamin C additionally. As a result, T1 and T2 displayed higher body weight increase and diet intake compared to T3 and T4 (p<0.05). The weights of their liver and gizzard were similar but the weights of the thymus and bursa F were higher for T1 and T2 compared to that of T3 and T4 (p<0.05). It was observed that T1 and T2 displayed higher concentrations of blood triglyceride, total cholesterol, HDL-C and blood sugar compared to that of T3 and T4 but LDL-C level was higher for T3 and T4 compared to that of T1 and T2 (p<0.05). T1 and T2 displayed higher levels of immunity substances such as IgG, IgA and IgM compared to T3 and T4 but the blood level of corticosterone displayed to be lower for T1 and T2 compared to T3 and T4 (p<0.05). The T1 and T2 contained a higher amount of fecal lactobacillus compared to that of T3 and T4 but the T3 and T4 contained a higher amount of fecal E. coli, total aerobic bacteria, coliform bacteria compared to that of T1 and T2 (p<0.05). T1 and T2 displayed higher concentrations of cecal acetic acid, propionic acid and total short chain fatty acids compared to T3 and T4 but T3 and T4 displayed higher concentrations of butyric acid, isobutyric acid, valeric acid and isovaleric acid compared to T1 and T2 (p<0.05). These results have been observed that broiler chickens exposed to extreme heat stress with feeding EHD, inverse lighting and cold water would improve blood lipid, and elevate the production of immunity substance, beneficial microorganisms, and short chain fatty acids. This provision would also reduce the blood sugar consumption rate as energy sources and these effects will improve the growth performance of the broilers exposed to extreme heat.

한국재래산양(韓國在來山羊)의 사양개선(飼養改善)에 관(關)한 연구(硏究) - 제일위((第一胃)에서의 영양소(營養素) 흡수(吸收)에 대(對)하여 - (Nutritional Studies for Improvement of Feeding on Korean Native Goat - Absorption of Nutrients in Rumen -)

  • 식량(食糧)으로서 단백질자원(蛋白質資源)의 개발문제(開發問題)는 동남(東南)아시아에 있어서 특(特)히 중요(重要)하며 한국(韓國)에 있어서도 섭취량(攝取量)이 부족(不足)한 현상(現狀)이다. 이 문제(問題)를 개선(改善)하는 방법으로 특(特)히 영양가(營養價)가 높은 동물성(動物性) 단백질(蛋白質)의 증산(增産)을 목적(目的)으로 한국재래산양(韓國在來山羊)의 사육(飼育)에 관(關)해서 연구(硏究)를 행하였다. 이 실험(實驗)은 우선 한국고유(韓國固有)의 재래산양(在來山羊)을 육자원(肉資源)으로 개발(開發)할 가치(價値)가 있는가를 확인(確認)하기 위해서 소형(小型) 제(第)1위법(胃法)으로 탄수화물(炭水化物)과 휘발성지방산(揮發性脂肪酸)의 흡수(吸收)를 실험(實驗)하였고 한편 in vivo법(法)에 의해서 제(第)1위(胃)의 아미노산 흡수(吸收)에 관해서 실험(實驗)을 했다. 이와 같은 실험(實驗) 결과(結果)에서 한국재래산양(韓國在來山羊)의 사육(飼育) 개선(改善)에 관해서 요약(要約)하면 다음과 같다. 1. 소형(小型) 제(第)1위법(胃法)에 의해서 재래산양(在來山羊)의 영양흡수(營養吸收)에 관하여 실험(實驗)한 결과(結果)는 영양소(營養素)를 주입(注入) 한 후 0.5, 1, 2시간(時間)의 간격(間隔)으로 측정(測定)한 V F A의 흡수율(吸收率)이 푸로피온산 70~86%, 초산 74~87%, 낙산 76~89%였다. 유기물(有機物)의 흡수율(吸收率)은 0.5%의 에칠 알콜이 29~89%였으며, 0.1M의 젖산이 12~27% 흡수(吸收)되었다. 2. 제(第)1위내(胃內)에 유리(遊離) L형(型) 아미노산을 투여(投與)하고 그 흡수(吸收)를 측정(測定)한 결과(結果)는 제(第)1위(胃) 정맥(靜脈)의 아미노산 함량(含量)에 있어서 전요(全要)아미노산구(區)에 비하여 메치오닌 결핍구(缺乏區)의 흡수율(吸收率)이 적었고 메치오닌 3배구(倍區)와 요소첨가구(尿素添加區)의 아미노산 흡수율(吸收率)이 높고 특(特)히 메치오닌을 결핍(缺乏)시켜도 점막층(粘膜層)의 함량(含量)에는 변화(變化)가 없었다. 3. 제(第)1위내(胃內)에 근육층(筋肉層)의 아미노산 흡수(吸收)도 점막(粘膜)과 같은 경향(傾向)을 보였으며 메치오닌 결핍(缺乏)에 의한 영향은 없고, 메치오닌을 3배(倍)로 첨가(添加)하면 전요(全要)아미노산구(區)에 비(比)하여 메치오닌과 글루타민의 함량이 147.44배(倍) 이상으로 증가(增加)되며 또 요소첨가구(尿素添加區)는 글루타민, 프롤린, 발린의 흡수율(吸收率)이 증가(增加)되었다. 4. 제(第)1위(胃) 정맥혈장(靜脈血漿) 중의 아미노산 농도(濃度)는 제(第)1위(胃) 점막(粘膜) 중이나 근육(筋肉) 중에서 보다 일반적(一般的)으로 낮으며 메치오닌의 결핍(缺乏)에 의해서 아미노산의 흡수율(吸收率)이 저하(低下)되고 메치오닌 3배구(倍區)나 요소첨가구(尿素添加區)는 아미노산 흡수(吸收)를 증가(增加)시키는 등 아미노산 조성분(組成分)에 따라서 그 흡수상태(吸收狀態)에 변화가 생긴다. 5. 요소첨가구(尿素添加區)는 제(第)1위(胃) 근육층(筋肉層)의 암모니아태질소(態窒素)와 아미노태질소(態窒素) 및 요소(尿素)의 함량(含量)을 증가(增加)시켰다. 재래산양(在來山羊) 제(第)1위(胃)안을 완전(完全)하게 깨끗이 할 수 없으므로 남아있는 세균(細菌)을 조사(調査)하였으며 암모니아태질소(態窒素)의 변화(變化)는 이러한 조건(條件)의 영향을 받았다. 6. 아미노산의 흡수차(吸收差)에 의한 제(第)1위(胃) 조직(組織)의 변화(變化)를 일반(一般) 현미경(顯微鏡)과 형광(螢光) 현미경(顯微鏡)으로 관찰(觀察)한 결과(結果)는 메치오닌 3배구(倍區)에 있어서 점막(粘膜)의 단층엔주상피(單層円柱上皮)와 고유층(固有層)이 약간 엷어졌으며 요소첨가구(尿素添加區)는 두껍게 변화(變化)하였는데 점막하조직(粘膜下組織)이나 근층(筋層)에 있어서는 각(各) 구간(區間)에 큰 차이(差異)가 인정(認定)되지 않았다.
