Ginsenoside-Rb1 Acts as a Weak Estrogen Receptor Agonist Independent of Ligand Binding.

  • Ginseng is a medicinal herb widely used in Asian countries, and its pharmacological effects has been demonstrated in various systems such as cardiovascular, central nervous, and endocrine systems. Its effects are mainly attributed to the ginsenosides. We hypothesize that a component of Panax ginseng, ginsenoside-Rbl, acts by binding to estrogen receptor. We have investigated the estrogenic activity of ginsenoside-Rbl in a transient transfection system using estrogen receptors ${\alpha}$ or ${\beta}$ with estrogen -responsive luciferase plasmids in COS monkey kidney cells. Ginsenoside-Rbl activated both estrogen receptors ${\alpha}$ and ${\beta}$ in a dose-dependent manner (0.5 -100 M ). Activation was inhibited by the specific estrogen receptor antagonist ICI 182,780, indicating that the estrogenic effect of ginsenoside-Rbl is estrogen receptor dependent. Next, we evaluated the ability of ginsenoside-Rbl to induce estrogen-responsive progesterone receptor gene by semi-quantitative RT-PCR assays. MCF-7 cells treated with l7${\beta}$-estradiol or ginsenoside- Rb1 exhibited an increased expression of progesterone receptor mRNA. However, ginsenoside-Rbl failed to displace the specific binding of [3H]17${\beta}$-estradiol to estrogen receptor in MCF-7 cells as examined by whole cell ligand binding assays, suggesting that there is no direct interaction of ginsenoside-Rbl with estrogen receptor. Our results indicate that estrogen-like activity of ginsenoside-Rbl is independent of direct estrogen receptor association.

Diffusion-Weighted MRI for the Assessment of Molecular Prognostic Biomarkers in Breast Cancer

  • This study systematically reviewed the role of diffusion-weighted imaging (DWI) in the assessment of molecular prognostic biomarkers in breast cancer, focusing on the correlation of apparent diffusion coefficient (ADC) with hormone receptor status and prognostic biomarkers. Our meta-analysis includes data from 52 studies examining ADC values in relation to estrogen receptor (ER), progesterone receptor (PgR), human epidermal growth factor receptor 2 (HER2), and Ki-67 status. The results indicated significant differences in ADC values among different receptor statuses, with ER-positive, PgR-positive, HER2-negative, and Ki-67-positive tumors having lower ADC values compared to their negative counterparts. This study also highlights the potential of advanced DWI techniques such as intravoxel incoherent motion and non-Gaussian DWI to provide additional insights beyond ADC. Despite these promising findings, the high heterogeneity among the studies underscores the need for standardized DWI protocols to improve their clinical utility in breast cancer management.

흰쥐의 발정주기동안 난소내 Leptin 및 Leptin 수용체 발현의 주기적 변화에 관한 연구 (Study on the Cyclic Change of Leptin and Its Receptor Expression during the Estrous Cycle of Rat)

  • 비만유전자 산물인 leptin은 비만뿐만 아니라 여성의 생식 생리와 관련이 있는 것으로 보이나, 아직 이러한 leptin이 난소에 직접적으로 작용하는지 정확하게 밝혀지지 않고 있다. 따라서 본 연구에서는 흰쥐 난소에서 leptin과 leptin 수용체의 발현을 면역조직화학방법으로 확인하고 발정주기에 따른 leptin과 leptin 수용체의 발현 양상을 RT-PCR 방법으로 조사하고자 하였다. 면역조직화학적 염색방법 결과 흰쥐 난소내에서 leptin은 협막세포와 폐쇄 난포의 일부 과립세포에 염색되었고, leptin 수용체는 협막세포, 간질세포와 난포강이 형성되지 않은 난포의 난자에 염색되었다. 특히 폐쇄 난포에서는 leptin과 leptin 수용체가 정상 난포에 비해 강하게 염색되었다. 흰쥐의 발정주기 동안 혈청내 estradiol, progesterone leptin의 농도는 ELISA 방법으로 측정하였고, 난소내 leptin과 leptin 수용체의 mRNA 발현 양상은 RT-PCR 방법으로 조사하였다. 혈중 leptin 농도를 측정한 결과 estrous 시기에 비하여 metestrous 시기에 유의하게 증가하였고, 이 시기에 progesterone 농도가 함께 증가하는 것을 관찰할 수 있었다. Leptin mRNA는 모든 발정주기에서 발현되지 않았지만 leptin 수용체 mRNA는 diestrous 시기를 제외한 다른 발정주기에 모두 발현되었다. 이러한 결과는 leptin이 흰쥐 난소의 기능을 조절하는데 직접적으로 관여할 수 있다는 것을 제시하고 있다.

프로게스테론의 비유전자 수준 작용 : 포유류 난소에서의 신호 전달 경로를 중심으로 (Non-Genomic Actions of Progesterone : Focussed on the Signaling Pathways in the Mammalian Ovary)

  • 본 논문은 포유동물 난소에서의 신속한 프로게스테론(P4) 선호 전달경로에 관해 현재 통용되는 지식을 요약하였다. P4는 안드로겐과 에스트로겐 합성 과정에서의 중요한 중간 산물이면서 그 자체로도 배란, 난포폐쇄(atresia), 황체형성과정(luteinization)에서 결정적인 역할을 하며, 모든 포유동물의 초기 임신 유지에 필수적이다. 이와 같은 생리적인 중요성에도 불구하고 포유동물 난소에서의 정확한 P4 작용기작은 아직까지도 완전히 알려져 있지 않다. 이러한 관점에서 볼 때, 비유전자 수준이면서 전사와 무관한 P4의 세포내 작용을 매개하는 수용체의 실체에 관해 오래 동안 계속된 의문과 논란은 과학적인 흥미를 유발하는 생식생리학의 주요 관심사이다. 포유류 난소에서 P4는 1) 잘 알려진 유전자 수준의 경로(genomic pathway)인 호르몬이 소위 고전적인 세포 내의 수용체에 결합하고, 이어 리간드-호르몬 복합체가 전사조절물질로 작용하여 표적 유전자 발현을 조절하거나, 2) 유전자에 직접 작용하지 않기 때문에 비유전자 수준이라 불리우는 경로(non-genomic pathway)로 작용한다. P4의 비유전자 수준 작용의 주요한 특징은 (i) 신속하고, (ii) 전사억제제에 반응하지 않고, (iii) 세포막과 연관된 물질들에 의해 신호가 전달된다. 아마도 난소에서 P4의 비유전자 수준 작용은 (a) 세포막 또는 그 근처에 위치한 고전적인 P4 수용체(PGR), (b) 세포막 프로게스틴 수용체(membrane progestin receptors; MPR $\alpha$, MPR $\beta$ and MPR $\gamma$) 패밀리, (c) progesterone receptor membrane component I(PGRMC1), 그리고 (d) serpine I mRNA binding protein(SERBP1)의 세포막 복합체에 의해 매개되는 것으로 추정된다. 포유류 난소에서의 P4 작용에 대한 완전한 이해를 위해서는 향후 많은 연구가 필요할 것이다.

The Effects of Sex Hormones on the Expression of ODF/OPG in Human Gingival Fibroblast and Periodontal Ligament Cell at Serum Concentration During Pregnancy

  • Periodontitis is a chronic infectious disease that leads to the destruction, one of the major cause of tooth loss in human. Osteoclast Differentiation Factor(ODF), also called as Receptor activator of NF-${\kappa}B$ ligand(RANKL), a surface-associated ligand on bone marrow stromal cells and osteoblasts, activates its cognate receptor RANK on osteoclast progenitor cells, which leads to differentiation of these mononucleated precursor cells. Osteoprotegerin(OPG), a decoy receptor, is released from stromal cells and osteoblasts to inhibit the interaction between RANKL and RANK. The experiment for the effect of pregnancy on gingival health showed greater gingival inflammation and edema during pregnancy, despite similar plaque index. There should be many factors affecting the periodontal health in pregnancy. In this experiment, we examined the direct effects of sex hormones(estrogen and progesterone) on the ODF/OPG expression in human gingival fibroblasts and periodontal ligament cells at the serum concentration of pregnancy. The ratio was high in the 1st trimester of pregnancy by estrogen and in the late 2nd trimester by progesterone. Therefore, the local periodontal destruction might be accelerated by these hormonal effect on the periodontal cells.

Expressional Correlation of Human Epidermal Growth Factor Receptor 2, Estrogen/Progesterone Receptor and Protein 53 in Breast Cancer

  • Background: This study aimed to show the localization of estrogen / progesterone receptors, human epidermal growth factor receptor 2 (Her-2) and protein 53 (p53) by immunohistochemistry in a series of consecutive breast cancer patients. Materials and Methods: The study covered invasive breast cancers from 299 patients presenting at the Oncogenetic Clinic and Pathology Centers of Ahwaz Jondishapour University of Medical Sciences Hospital in Iran during the time period from 2009 to 2011. The Scarff-Bloom Richardson scoring method was used. Results: Of the 299, 27% (80/299) were <40, 33% (100/299) were 41-50, and the remaining 40% (119/299) were>50 years old. The highest incidence of breast cancer in this study population was in the group of more than 50 year age, and the most common histological type of breast cancer was the invasive ductal carcinoma, which accounted for 68% (203/299) of the cases. Out of possible total of 207, 6% (13/207), 41% (85/207), and 53% (109/207) were scored as grade I, II, III, respectively. Conclusion: Our findings demonstrated a lack of association between labeling for the markers studied and tumor size and age of the patients. We confirmed an association between ER labeling and nuclear grade of breast cancer. The conflicting results obtained compared with the literature be because of differences in the immunohistochemical techniques applied in the various studies and to the scoring systems used.

자궁내막증과 progesterone receptor gene polymorphism (PROGINS)과의 연관성에 관한 연구 (A study on association of progesterone receptor gene polymorphism (PROGINS) with Endometriosis)

  • 목 적:자궁내막증은 유전적 연관성이 명확히 규명되지 않은 에스트로겐 의존성 질환이다. 그 연관성에 대하여 명확히 밝혀진 것은 없으나 일부 유전적 연구 결과들을 살펴보면 자궁내막증이 프로게스테론 수용체 유전자의 특정 다형성 부위와 연관되어 있음을 알 수 있다. 본 연구는 프로게스테론 수용체 유전자의 다형성(PROGINS)을 분석하여 자궁내막증 환자와 대조군에 차이가 있는지를 알고자 고안되었다. 방 법:수술 및 병리조직 소견에서 자궁내막증을 확진받은 100명의 환자와 건강검진을 시행 받은 일반 인구군 110명을 대상으로 하였다. 대상 환자들의 말초 혈액에서 DNA를 추출한 후 프로게스테론 수용체의 유전자 다형성 검사를 PCR-SSP (Sequence specific primer)방법으로 시행하였고 자궁내막증 환자군과 건강대조군에 있어서 특정 유전자 다형성의 빈도에 차이가 있는지 $x^2$ 검정을 통하여 통계 분석하였다. 결 과:자궁내막증군과 대조군에서 각각 1명에서만 PROGINS 다형성 양상이 T1/T2 이형접합자로 확인이 되었고 나머지 개체에서는 모두 T1/T1 동형접합자임이 확인되었다. 결 론:자궁내막증과 프로게스테론 수용체의 유전자 다형성(PROGINS)간의 연관성은 밝혀지지 않았으나 대규모의 연구 및 다른 유전자의 연구가 자궁내막증의 병인을 규명하는데 유용할 것으로 생각된다.

Correlation of Hormone Receptor and HER-2/neu Expression with Clinicopathologic Parameters in Primary Breast Tumors

  • Background: Breast cancer (BC) is a major health issue worldwide as well as in Pakistan. All women belonging to any race, ethnicity or lineage are in danger of developing breast cancer. Significant factors influencing the development of breast malignancies are the genetic background, environmental conditions, reproductive parameters, the consequences of female hormones both intrinsic and extrinsic, alteration of immune status, and biologic determinants. Materials and Methods: Overall 150 biopsy proven patients were included in the study. Samples were submitted for histopathology and determination of estrogen and progesterone receptor expression and HER-2/neu status. Associations with other characteristics like age, tumor stage, node involvement, histological grade were also studied. Results: Mean age at presentation was 46.7 years. The majority had invasive ductal carcinoma, 100 (84.7%), and were in stage pT3, 54 (45.7%). Important relationships (P<0.05) were found among ER, PR positivity, and Her 2 neu overexpression. However, no noteworthy link was identified amongst ER, PR, Her 2 neu and tumor grade, stage, age, lymph node involvement except for the menopausal status. Conclusions: In summary, breast cancer patients featured an advanced stage of disease, more lymph node involvement, and moderately high grade tumors and with more estrogen, progesterone receptor and HER-2 positive tumors.

Role of Diacyl Glycerol (DAG) in Caprine Sperm Acrosomal Exocytosis Induced by Progesterone

  • Capacitated goat spermatozoa generated diacyl glycerol (DAG) when suspended in Krebs-Ringer bicarbonate medium and induced by progesterone or $Ca^{2+}$ ionophore A23187. We have added Sn-1-oleoyl-2-acetyl glycerol externally, to study the effect of DAG in goat sperm acrosomal exocytosis. Addition of neomycin abolished the DAG generating capacity of progesterone in a dose dependent manner, suggesting the involvement of a phosphoinositidase C activated phospholipase C system in the process. The level of increase in phosphatidic acid was considerably low and was produced well after the DAG generation thereby suggesting the involvement of a DAG kinase which phosphorylates DAG to produce PA. The inhibition of progesterone mediated effect by inhibitors of $GABA_A/Cl^{-}$ channel and $Ca^{2+}$ channels further supports the evidence that the events of binding of agonist to the receptor(s), opening of $Ca^{2+}$ channels and the activation of phospholipase C are reconciled to perform the function of acrosome reaction in capacitated goat spermatozoa.

Triple Negative Breast Cancer

  • Triple-negative breast cancers (TNBC), characterized by absence of the estrogen receptor (ER) and progesterone receptor (PR) and lack of overexpression of human epidermal growth factor receptor 2 (HER2), have a poor prognosis. To overcome therapy limitations of TNBC, various new approaches are needed. This mini-review focuses on discovery of new targets and drugs which might offer new hope for TNBC patients.
