Effect of Intracellular Calcium Level on the Hybridoma Cell Growth and Monoclonal Antibody Production (세포내 calcuim 농도가 하이브리도마 세포 성장 및 단일클론항체 생산에 미치는 영향)

  • The effect of intracellular Ca2+ level on the hybridoma cell growth and monoclonal antibody(MAb) production was examined. For the manipulation of intracellular Ca2+ concentration, the cells were treated with A23187, ryanodine, and thapsigargin at about 1x106 cells/mL. The treated cells were recultivated by using the Iscove's Modified Dulbecco's Medium(MDM) containing 1.49mM CaCl2. The ryanodine-treated cells showed better cell growth, MAb concentration, and specific MAb productivity than others. In comparison with control, the maximum cell concentration, MAb concentration, and specific MAb productivity were increased by 40.6%, 48.1% and 83.3%, respectively. Confocal microscopic images of Fura-2/AM loaded cells indicate that the increase in intracellular Ca2+ level can enhance the MAb productivity by allowing the calcium influx into the endoplasmic reticulumn.

Studies of the Forest Structure and Productivity in Korea -Models of Maximum Productivity and Optimum Cutting Time of the Forests by Annual Ring Growth analyses- (韓國의 森林構造에 따른 物質生産力에 관한 分析 - 年齡生長 分析에 의한 森林 樹木의 最高生産力期 및 最適伐採期 推定모델 -)

  • For the maximun yield of the forest trees in the forest management, the growth of annual ring area of the major forest trees was analysed in the four areas in South Korea. The time to the maximum productivity and the optimum cutting time for the maximum yield were estimated. The growth curve of annual ring area showed sigmoid like that of other organisms. Only the growth coefficient among the areas between Fraxinus rhynchophylla and Pinus koraiensis represented significance (5% level). The growth coefficient among forest trees between Pinus densiflora and Abies holophylla, Larix kaempferi and Carpinus laxiflora, Larix kaempferi and Quercus mongolica, Larix kaempferi and Quercus serrata, Larix kaempferi and Pinus koraiensis, and Larix kaempferi and Abies holophylla represented significance (5% level). Among eight forest trees, the time to maximum productivity (tm) of Larix kaempferi was the earliest (21.4 year), and Abies holophylla was the latest (91.9 year). The optimum cutting time for the maximum yield (tc) of L. kaempferi was the earliest (29.4 year) and that of A, holophylla was the latest (122.2 year) of all communities. The optimum cutting time for the maximum yield was 1.33 times as late as the time to the maximum productivity. If the growth of annual ring area as the forest tree for wood is regarded, L. kaempferi and P. densiflora are thought to be more economical than A. holophylla and P. koraiensis.

Effect of Acorn Powder on the Biomass Productivity of Microalgae (도토리 가루가 미세조류 증식에 미치는 영향)

  • The focus of this study was to observe the growth of Chlorella vulgaris and Botryococcus braunii under mixotrophic conditions (i.e., added acorn) with the aim of increasing biomass and triacylglycerols (TAGs) content. The result of investigation indicated that the acorn contains a lot of carbonate (87.29%) and glucose (97.99 mg%). A significant growth of biomass was obtained when grown in acorn rich environment comparing to autotrophic conditions. 3 g/L acorn yielded the highest biomass concentration for these strains. Thus, the biomass productivity with 3 g/L acorn was obtained 2.31 times and 2.10 times higher than that of authotrophic conditions for Chlorella vulgaris and Botryococcus braunii, respectively. The maximum amount of TAGs was reached 14.35% and 18.41% for Chlorella vulgaris and Botryococcus braunii, respectively, in the growth medium with 5 g/L acorn. The effect of acorn could enhance the investigated microalgae growth, biomass productivity and TAGs content. This provides a feasible way to reduce the cost of bioenergy production from microalgae.

Production of Yeast Diet for Aquaculture in Batch Fermenters

  • Production of yeast diet has been carried out in batch fermenters under optimum culture conditions. The fermentation of Candida utilis on a $2\%$ complex medium resulted in 1.22 g/L/h productivity and $65\times10^8$ viable cells/ml, and the addition of $15{\mu}M$ zinc to the medium increased both the productivity and the number of viable cells just a little more. In the case of the fermentation of Kluyveromyces fragilis, the highest value of the biomass productivity, 1.94 g/L/h, was obtained on a $2.5\%$ fructose medium with $70\times10^8$ viable cells/ml, and $1\%$ peptone was found to be a growth factor in this fermentation. When $3.5\%$ NaCl was added to the given medium, both the biomass productivity and the number of viable cells decreased significantly in each fermentation, but this may be considered to preserve yeast diet long without osmotic lysis.

Growth Accounting Analysis of Korean Port-Logistics Industry (한국의 항만물류산업의 성장회계 분석)

  • The purpose of this study is to analyze contribution factors of economic growth through growth accounting analysis in Korean port-logistics industry. Comparing with the average level of entire industry for 1990-2003, the contributions of total factor productivity and labor in port-logistics industry were high, but that of capital stock was very low. The pattern of growth in Korean port-logistics industry has greatly changed before and after Korean financial crisis. Before the 1997 financial crisis, the economic growth rate of port-logistics industry was 14.1%, which is higher than that of the whole industries, 7.7% for 1990-1998. Main contribution factors of the economic growth rate were the growth of capital stock and productivity, but ratios of their contributions were relatively low and did not come up to that for the whole industry. After the financial crisis, annualized growth rate of GDP in port-logistics industry had rapidly declined at 5.4% for 1998-2003, which did not get to that of the entire industry (10.1%). The main contribution factors of the economic growth rate over the 1998-2003 period were capital stock 13.1%, labor 57.0 %, and total factor productivity 29.9 %, Such growth pattern as excess dependence on growth of labor brought reduction of the rate of economic growth with degradation of productivity growth in the Korean port-logistics industry.

Effects of Light Environment on Dry Matter Production and Growth of Zoysia japonica (광환경이 한국들잔디(Zoysia japonica)의 물질생산과 생장에 미치는 영향)

  • This study was carried out to investigate the interaction between productivity and light condition and to analyze the material productivity and productive charactaristics under different hours of light in Zoysia japonica. 1. Rate increasement of leaf number and total leaf length was remarkably high at the early growth stage in the control and 9 hour light treatment. The rate gradually decreased as growth proceeded. But 3 hour treatment was very low in the rate from its early growth stage through the whole test period. 2. The increasing rate of leaf area ratio (LAR) in all the experimental plots was remarkably high at the early growth stage after transplanting the grass. The shorter photoperiod resulted remarkable lower increasement of LAR. 3. The rhizome growth rate of the 9 hour photoperiod was high contrast to the 3 hour photoperiod treatment. The increasing rate of node number was also showed similar trend. 4. Chlorophyll content was very high at the 36 days after transplanting and then decreased gradually. Chlorophyll content on 3 hours photoperiod plot severely decreased. The ratio of chlorophyll a to b was decreased significantly at short day treatment. 5. The content of soluble sugar was increased at shorter photoperiod. The highest ratio of sugar content was on August, 28 by HPLC method. Such a result was attributed to accumulation of sugar in spite of low synthesis of ploysacchrides, translocation by low consumption. 6. The increasing rate of standings in all light treatment was high at the early growth stage after transplanting. Short photoperiod treatment by 3hour showed especially low prganic productivity on Zoysia japonica.

A Study on Source of Total Factor Productivity Growth of Trade Ports in Korea (한국무역항만 총요소생산성 증가의 원동력에 관한 연구)

  • This paper selects length of berth, area of yard, unloading capacity and number of berth as the input indexes, and cargo turnover as output index to research the source of TFP(Total Factor Productivity) growth of 23 main ports in Korea. The major conclusions are as follows. The TFP of the trade ports in Korea is at the fluctuating stage, but it generally displays a rising trend, and it’s growth originate from the growth of technical efficiency. The Growth rates of TFP of trade ports in the different areas are different, and the sources of growth are also different, but the changing trends are basically the same. Four major factors to the increase of TFP are following: competition between ports, reform of property system, harbor-hinterland economic and international trade, modeling, imitation and innovation in management, technology and system.

Effect of Waterlogging Duration on Growth Characteristics and Productivity of Forage Corn at Different Growth Stages Under Paddy Field Conditions

  • The purpose of this study was to determine the effect of waterlogging duration on the growth characteristics and productivity of forage corn at different growth stages under paddy field conditions. Treatments consisted of waterlogging at two growth stages (V7 or V14) for four waterlogging durations (no waterlogging, 48 hours, 72 hours, and 96 hours, respectively). The V14 growth stage was more vulnerable to waterlogging than the V7 stage. Among the waterlogging durations, the lodging score increased at 48 hours. The stem height of forage corn decreased with the increase in waterlogging duration at the different growth stages (V7 and V14). Increase in waterlogging duration reduced the stem dry matter yield, ear dry matter yield, and total dry matter yield at both growing stages (V7 and V14). The waterlogging treatments at the V14 stage affected ear dry matter yield more than those at the V7 growing stage. Thus, the management of forage corn under paddy field conditions must be strengthened during early (V7) and grain fill stages (V14). When waterlogging occurs, surface and subsurface drainage should be implemented within 48 hours to control (no waterlogging) the groundwater level and, thus, minimize economic losses due to forage corn damage.

Impact of Railroads on Local Economies: Evidence from U.S. History

  • Purpose - The introduction and expansion of the railway network since the 19th century brought revolutionary changes in economic activities, performance, and structure. The purpose of this study is estimating the impact of railroads on the local agricultural and manufacturing structures in the 19th century USA. Research design, data, and methodology - To identify the impact of railroads on local economic structure, county-level panel data from the U.S. census were analyzed using a panel fixed-effect differences-in-differences regression. The empirical investigation focuses on whether railroads changed the overall volume and sectoral composition of the local agricultural sector, and whether they contributed to the growth of the local manufacturing industry and its productivity. Results - The railroad introduction led to the relative decline of the agricultural sector, while encouraging the growth of market-oriented gardening. As such, manufacturing productivity increased by the introduction of railroads, although manufacturing inputs and home manufactures were unaffected. Conclusions - The findings imply that railroads contributed to the growth of market-oriented farming in rural areas, and the rise of productivity in the local manufacturing sector. Meanwhile, evidence of railroad-driven growth for the entire agricultural sector or a massive reallocation of resources from agriculture to manufacturing were not found.

How Productive Are Life Insurance Institutions in Malaysia? A Malmquist Approach

  • The purpose of this study is to investigate empirical evidences of productivity of life insurance institutions in Malaysia. Therefore, this study attempts to explore the productivity of the life insurance institutions in Malaysia. The overall findings show that the total factor productivity (TFP) has progressed by 2.5% per year during the study period from 2012 to 2016 in the Malaysian insurance industry. However, TFP change has declined from 2012 to 2015 and observed a negative growth in 2015-16 (3.3%). The highest productivity progress was documented during 2012-13 at a rate of 11.7% while the minimum productivity progress was during 2014-15 (only 0.2%). The results also indicate that the decomposition of TFP found that overall progress could mainly be attributed to technological change (TC). However, technical efficiency change (TEC) and pure technical efficiency change (PTE) have negative impact on TFP. The findings also show that most of the insurance companies have a steady growth. Therefore, this study will contribute new insights for the policy makers and insurance institutions to take appropriate steps in developing relevant policies for increasing productivity of insurance institutions in Malaysia.
