혈부축어탕이 대동맥 평골근 세포에서 NO 생성에 미치는 영향 (Effects of Hyeolbuchukeo-tang (Xiefuzhuyutang) on NO Production in Aortic Vascular Smooth Muscle Cells)

  • Objective : This study was designed to investigate the effect of Hyeolbuchukeo-tang (HCT) on NO production and the molecular mechanism of NO production modulated by HCT in the primary VSMC (vascular smooth muscle cells). Method : Primary VSMC was established from aorta and cultured VSMC used in this study. NO production of VSMC was assayed by Griess reagent and the expression of iNOS gene was assayed by Western, RT-PCR. Result : $TNF-{\gamma}$ induced NO production, but $IFN-{\gamma}$ or HCT alone did not induce NO production in cultured VSMC. However, $IFN-{\gamma}$ or HCT potentiated NO production in $TNF-{\gamma}-treated$ VSMC in a time- and dose-dependent manner. $TNF-{\gamma}$ induced the iNOS gene expression corresponding to NO production in $TNF-{\gamma}-treated$ VSMC. HCT potentiated NO production in $TNF-{\gamma}-treated$ VSMC by about 20%, but HCT did not increase the level of iNOS mRNA in $TNF-{\gamma}-treated$ VSMC. HCT slightly increased the level of iNOS protein in $TNF-{\gamma}-treated$ VSMC. Calcium ionophore A23187 decreased NO production in $TNF-{\gamma}-treated$ VSMC, but HCT attenuated the effect of A23187. Conclusion : As NO is deeply involved in the development of arteriosclerosis and dilation of blood vessels, drugs or chemicals modulating NO production in VSMC could be used for preventing and treating arteriosclerosis. Considering the effect of HCT on the modulation of NO production in VSMC, MCT has a potential capacity for preventing and treating diseases of the circulation system including arteriosclerosis.

염의 삼투압이 Torula sp.의 증식과 Erythritol 생산에 미치는 영향 (Effect of Osmotic Pressure of Salts on Growth of Torula sp. and Erythritol Production)

  • To investigate the effect of salts on the production of erythritol by Torula sp., cells were grown on the media containing various concentrations of KCl or NaCl. Cell growth and glucose consumption rates decreased when KCl or NaCl concentration increased from 0.0 to 0.5M. The production of erythritol, however, was maximal at 0.3M aCl. The erythritol concentration of 54.3g/l in the medium containing 0.3M NaCl and 200g/l glucose was obtained after 120h. The production of erythritol decreased in cultures above 0.3M NaCl or 0.4M KCl due to the inhibition of cell growth. To elucidate the effect of salts more quantitatively, KCl and NaCl concentrations were converted to osmotic pressure. As the osmotic pressure increased, the yield of erythritol from glucose increased regardless of the kinds of salts and the yield of erythritol was approximately 49% at the osmolality of 2.4Os/kg. When the osmotic pressure increased to 2.5Os/kg, the specific growth rate of cells decreased but the production rate of erythritol increased. For the effective production of erythritol, osmotic pressure must be adjusted not to inhibit markedly the growth rate of cells and to stimulate the production rate of erythritol by supplementing salt.

수은(水銀)이 Lactobacillus bulgaricus의 젖산 생성(生成)과 생육조해작용(生育阻害作用)에 대한 양파 추출물(抽出物)의 효과 (Effect of Onion Extracts on the Growth and lactic Acid Production of Lactobacillus bulgaricus in Medium Containing Mercury)

  • Onion extracts were tested for effects on the growth and lactic acid production of Lactobacillus bulgaricus in the medium containing mercury. The media containing mercury were added with onion extracts and inoculated with the bacterium and then incubated at $39^{\circ}$ for four days. All of the onion extracts examined increased the growth and lactic acid production of the bacterium in the medium containing mercury. At the addition of the edible portion extract of onion to the medium containing 5 ppm, 10 ppm and 20 ppm mercury, the higher the concentration of the onion extract was added, the greater the increasing effect on the growth and lactic acid production. The brown peel extract of onion increased the growth and lactic acid production of the bacterium in the medium containing mercury at all concentrations. The higher the concentration of the extract added, the more effective the increasing effect. The onion solutions of edible portion and brown peel extracted at $90^{\circ}\;to\;100^{\circ}$ showed more desirable effects than those extracted at the room temperature. Among four kinds of the onion extracts, the brown peel solution extracted at $90^{\circ}\;to\;100^{\circ}$ was the most effective in increase of the growth and lactic acid production in the medium containing mercury.

Inhibitory Effect of Scutellaria baicalensis Root Extract on Chemical Mediator Release and Immune Response

  • Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha $(TNF-{\alpha})$ production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of $interferon-{\gamma}$. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.

Micromonospora inyoensis에 의한 Sisomicin 생산에서 Sodium Acetate의 영향 (Effect of Sodium Acetate on the Production of Sisomicin by Micromonospora inyoensis)

  • This study was carried out to enhance the production of sisomicin by Micromonospora inyoensis ATCC 276000. The effect of various sugars and organic acids as the supplements to a basal mineral medium was tested for the sisomicin production by fully grown mycelium. Among the substances tested, acetate and citrate greatly increased the production of sisomicin at the sixth day of culture. Especially, in the basal mineral medium containing 0.1M sodium acetate, sisomicin production was 6.7 times more than that in the same medium with glucose. When 0.1M sodium acetate was added in the fermentation medium initially, the cell growth was inhibited by sodium acetate, although specific productivity was higher than that in the same medium without sodium acetate. On the contrary, when sodium acetate was added to the culture after three days, the sisomicin production and specific productivity were 1.6 times higher than those in the same medium without sodium acetate. The results suggested that sodium acetate was a stimulating substance of sisomicin production by M. inyoensis.

  • 본 연구는 용존산소의 양에 따른 MnP의 생산양상을 체계적으로 연구하였으며, 과량의 산소 공급 시 MnP 생산이 저해될 수 있음을 산소공급에 따른 과량의 $H_2O_2$생산 측면에서 설명하고 있다. 또한 보다 높은 MnP 생산을 위한 산소공급방법을 제시하고자 하였다.

효과적인 VFX 수중 폭발효과 구현을 위한 유체 시뮬레이션 제어 (Fluid Simulation Control for Effective VFX Underwater Explosion Effects)

  • The explosion effect of computer graphics Visual Effects(VFX) used in films and animations is an important element that determines the completeness of the film, and its usage is getting extended. The realistic explosion effect of VFX should be made according to observations and analysis of various factors of actual explosion in real world. This experimental research would suggest the efficient production guideline for the technical characteristics of underwater explosion of VFX. For this research process, first, the comparison of actual explosion and VFX explosion effect, classification of actual explosion, and characteristics of underwater explosion effect will be addressed. Second, based on the literature reviews, the four steps of experimental production analysis tool will be derived. Third, the experimental research will be processed in along with technical factors four steps of the underwater explosion effect, (1)realistic creation and emission of fluid, (2)fluid expansion control by water pressure, (3)bubble effect, and (4)motion of bubble & dissipation of fluid. The effective method of fluid simulation production will be verified through experimental studies based on the characteristics of the actual explosion process. This experimental study suggested the VFX production technique is expected to be used as the basic data for related research field.

Effect of Culture Conditions on Growth and Production of Docosahexaenoic Acid (DHA) using Thraustochytrium aureum ATCC 34304

  • Environmental and medium factors were investigated as basic data for optimizing DHA production when using Thraustochytrium aureum. To study the effect of environmental conditions, the rotation speed and culture temperature were changed. Plus the trend of the growth characteristics, lipid content in the biomass, and DHA content in lipids were evaluated according to various initial glucose concentrations. The biomass, lipid, and DHA analyses showed that the physiological characteristics of T. aureum were closely related with the environmental and medium conditions, as in the case of other marine microorganisms. For example, a low rotation speed of 50 rpm lowered the cell growth rate as well as the DHA content in the lipids. A low temperature had a negative effect on the cell growth, yet a positive effect on the lipid content in the biomass. Different initial glucose concentrations had no effect on the lipid content in the biomass or DHA content in the lipids, yet did affect the cell growth. Accordingly, these results show that environmental and medium factors must be synthetically considered in order to optimize DHA production when using T. aureum.

주요 콩 생산국의 생산변동 조정유형과 무역전이효과 분석 (Adjusting Patterns of Production Fluctuation and Trade Transmission Effect of Major Soybean Production Countries on World Market)

  • 본 연구에서는 우리나라의 대표적 수입 곡물인 콩의 주요 교역국들의 생산, 소비, 무역 및 재고 동향과 각국의 생산변동에 따른 조정유형 분석을 수행한 후, 이를 바탕으로 각국의 국제시장 전이효과를 계측하였다. 분석결과 최근 콩의 국제시장 전이효과가 주요 생산국인 브라질, 파라과이, 캐나다 등을 중심으로 증대되고 있으며, 대부분의 국가가 재고보다는 소비와 무역을 통해 생산변동을 조절하고 있는 것으로 나타나 국제 콩시장의 불안정성 증가가 예상된다. 따라서 대규모 콩 수입국인 우리나라는 일정량의 국내생산기반 유지, 적절한 재고정책의 활용, 선물시장의 활용, 수입선 다변화 등과 같은 해외조달능력의 향상을 통한 안정적인 국내 콩 수급 방안 마련이 시급한 것으로 나타났다.

Effect of Dietary Sodium Nitrate Consumption on Egg Production, Egg Quality Characteristics and Some Blood Indices in Native Hens of West Azarbaijan Province

  • The aim of the study was to evaluate the effect of sodium nitrate consumption on egg quality and quantity, and some blood parameters of native breeder hens of West Azerbaijan province. One hundred native hens were used from wk 25 to 32 of age. These birds were divided into two groups. One group was fed the control diet (CD) but the other fed the same diet supplemented with 4.2 g/kg sodium nitrate (ND). After 2 wks of adaptation, eggs were collected daily and egg mass and egg production were measured weekly for five weeks. To assess the egg quality parameters, two eggs from each replicate pen were collected for three consecutive days each week. At the end of experimental period (wk 32 of age), blood samples of 5 birds per replicate were collected from the wing vein into anticoagulant tubes. Dietary sodium nitrate didn't affect the egg production, shell stiffness, shell thickness and Haugh unit (p>0.05) but it decreased the both egg production and egg mass during the last three weeks (wks 30, 31 and 32) (p<0.05). Furthermore, a treatment effect was observed for yolk colour (p<0.05). Both the egg production and egg mass were increased over time (p<0.05). No significant treatment${\times}$time interaction was observed for egg weight, egg production and egg mass (p>0.05). No effect of time or treatment${\times}$time were observed for shell stiffness (p>0.05). Over time, shell thickness was decreased while Haugh unit increased (p<0.05). None of the blood TP and TG or the activity of ALT, AST and LDH enzymes were affected by dietary consumption of sodium nitrate at wk 32 of age (p>0.05). Sodium nitrite decreased both the TAC and TC at wk 32 of age (p<0.001). It was concluded that the lower body antioxidant capacity of nitrate fed birds resulted in the lower performance (egg weight, egg production and egg mass).
