Analysis of the Effect of Media Types and Chromagenic Chemicals on the Detection of Extracellular Laccase Activity among Lentinula edodes Strains (표고 교잡균주들의 세포외 laccase 활성 검출에 미치는 배지성상과 발색반응 시약의 영향 분석)

  • Breeding of Lentinula edodes generates a number of hybrid strains that are subject to evaluation for good traits for the mushroom production. As an effort to understand biochemical properties of the hybrid strains, this study tried to develop a fast and easy method for comparison of the ability of producing extracellular laccase among hybrid strains of Lentinula edodes. For this aim, we estimated the effect of media types and chromagenic chemicals on the detection of extracellular laccase in seven hybrid strains of L. edodes. When Remazol Brilliant Blue R (RBBR) dye was used for chromagenic reaction, the detection of the enzyme activity was feasible both in the solid and liquid media containing not potato dextrose but malt extract as a nutrient component. When guaiacol was used for chromagenic reaction, the detection of the enzyme activity was feasible both in the solid and liquid media containing either potato dextrose or malt extract as a nutrient component. Malt extract-based liquid culture with RBBR or guaiacol in 2 ml microfuge tube allowed us to economically and quantitatively detect and compare the enzyme activity within 3 days among the tested hybrid strains of L. edodes.

Comparison of Carcass Composition of Iranian Fat-tailed Sheep

  • Most breeds of sheep in Iran are adapted to their agro ecological niches where it is likely that they were also artificially selected by their owners. In general, most of sheep breeds are multipurpose producing lambs, wool and milk. To compare the physical and chemical composition of the carcasses of ten Iranian native fat-tailed sheep breeds, 243 male lambs (6-7 months of age) of ten fattailed, Iranian breeds of sheep, Sanjabi (S), Ghezel (G), Afshari (A), Mehrabani (M), Lori (L), Lori Bakhtiari (LB), Kordi Khorasan (K), Sangesari (Sa), Baluchi (B) and Chal (C) were studied. Lamb breed group had a significant (p<0.05) effect on all the carcass traits measured. The LB and C lambs showed the same live weight which was significantly (p<0.05) higher than the other breeds. Carcass dressing- out percentage in S lambs was lowest (p<0.05), but not different from G lambs. K and Sa breeds showed the highest (p<0.05) carcass dressing-out percentage. The S lambs had the highest (p<0.05) lean meat percent. Lean meat percentage was not significantly (p>0.05) different in the G, A, M and C breeds. The Sa and K breeds showed the lowest lean meat percent. The S lambs showed the lowest (p<0.05) fat percent in their carcass, while K and Sa showed the highest (p<0.05). Subcutaneous fat in K, Sa and B was higher (p<0.05) than the other breeds. Lambs of S, G, A and M breeds had the lowest subcutaneous fat in their carcasses (p>0.05). Intramascular fat was significantly (p<0.05) lower in M, S and C despite the fact that this values were highest in B and K lambs. The K and Sa breeds had highest fat-tail percentage (p<0.05) in their carcass, whereas S and G showed lowest. Lambs of G, S and A breeds had higher bone percent than other breeds (p<0.05). Lowest bone percent (p<0.05) was seen in K and Sa lambs. The carcass moisture percent was not significantly (p>0.05) different in S, G, A, M, L and C breeds. M lambs showed the lowest crude protein percentage and S breed the highest (p<0.05). There were no significant (p>0.05) differences among other lamb breeds for this trait. Chemical fat percentage was the same in S, G, A, C and M breeds, but significantly (p<0.05) lower from LB, K, Sa and B. Ash percent in S, G and A had no significant (p>0.05) difference. According to higher lean meat and lower fat percentages in the carcass, the ranking of breeds would be S, G, A, M and C.

Effects of Dietary Alpha-lipoic Acid on Anti-oxidative Ability and Meat Quality in Arbor Acres Broilers

  • An experiment was conducted to evaluate the effects of dietary alpha-lipoic acid (LA) on growth performance, carcass characteristics and meat quality in Arbor Acres broilers. A total of 240 1-d-old male Arbor Acres broilers were randomly allocated to 4 dietary treatments (0, 300 ppm, 600 ppm, and 900 ppm dietary LA supplementation, respectively). Birds were slaughtered at 42 days old. Live body weight (BW), average daily gain (ADG), average feed intake (AFI), feed conversion ratio (FCR), dressing percentage, breast muscle percentage, thigh muscle percentage, abdominal fat percentage, muscle color (L*, a*, b*), pH values at 24 h postmortem, meat shear force value (SFV) and anti-oxidative ability were measured. Results showed that addition of 600 ppm or 900 ppm LA decreased BW (p<0.01), ADG (p<0.01) and AFI (p<0.05) compared with other diets. FCR was not affected by dietary LA content. LA had no marked effect on dressing percentage, breast muscle percentage or thigh muscle percentage. Abdominal fat percentage was lower (p<0.05) in the 900 ppm LA supplementation group than the control group. Dietary 900 ppm LA increased (p<0.05) breast and thigh muscle pH value at 24 h postmortem compared with the control treatment. Dietary LA increased thigh muscle a* value, though no significant difference was found in thigh muscle a* value among the treatments. Dietary LA significantly decreased breast muscle L* value (p<0.05), breast muscle b* value (p<0.01) and thigh muscle b* value (p<0.05). Broilers fed LA had higher breast muscle a* value (p<0.05) and thigh muscle L* value (p<0.05). All test groups had lower (p<0.05) breast muscle SFV than the control group. Dietary 600 ppm or 900 ppm LA both decreased (p<0.01) thigh muscle SFV compared with the control treatment. Dietary 900 ppm LA significantly increased (p<0.05) TAOC, SOD and GSHPx compared with no LA treatment. Broilers fed LA had lower (p<0.01) MDA compared with the control treatment. These results suggested that dietary LA enhanced the anti-oxidative ability and oxidative stability, and contributed to the improvement of meat quality in broilers.

Expression of genes related to lipid transport in meat-type ducks divergent for low or high residual feed intake

  • Objective: This study examined the effects of divergence in residual feed intake (RFI) on expression profiles of key genes related to lipid transport in the liver and duodenal epithelium and their associations with feed efficiency traits in meat-type ducks. Methods: A total of 1,000 male ducks with similar body weight (1,042.1±87.2 g) were used in this study, and their individual RFI was calculated from 21 to 42 d of age. Finally, the 10 highest RFI (HRFI) and 10 lowest RFI (LRFI) ducks were chosen for examining the expression of key genes related to lipid transport in the liver and duodenal epithelium using quantitative polymerase chain reaction. Results: In the liver, expression levels of albumin (ALB), CD36 molecule (CD36), fatty acid hydroxylase domain containing 2 (FAXDC2), and choline kinase alpha (CHKA) were significantly higher in LRFI ducks than in HRFI ducks (p<0.01); negative correlations (p<0.05) between expression levels of ALB, CD36, FAXDC2, and CHKA and RFI were detected in the liver. Additionally, ALB expression was strongly positively correlated (p<0.05) with CD36, FAXDC2, CHKA, and apolipoprotein H (APOH) expression in the liver. In duodenal epithelium, we found that mRNA levels of ALB, CD36, FAXDC2, and APOH were significantly higher in LRFI ducks than in HRFI ducks (p<0.01); RFI was strongly negatively correlated (p<0.05) with ALB, FAXDC2, and APOH expression, while ALB expression was strongly positively correlated with APOH expression (p<0.01) in duodenal epithelium. Furthermore, expression levels of both ALB and FAXDC2 genes were significantly associated with feed conversion ratio and RFI in both liver and duodenal epithelium (p<0.05). Conclusion: Our findings therefore suggest that ALB and FAXDC2 genes might be used as potential gene markers designed to improve feed efficiency in future meat-type duck breeding programs.

A Study of User-Oriented Storytelling Based on Enneagram (에니어그램을 활용한 사용자 중심 스토리텔링에 관한 연구)

  • The narratives of the digital age have attempted interactions, and the interactive storytelling represented by the Branch Narratives has a problem of the expensive production cost of many optional implementations and poor narrative compared to linear story. As an alternative to this, this study proposes a user-oriented storytelling using user's personality traits. Using the Enneagram, a model of human psyche, and the Actantial model of the semiotician Greimas, when stories and characters are the same, the story could be reconstructed by deriving different topics from the same story according to the user's Enneagram personality type. The theme is determined by defining the axis of the desire of the Actantial model by respectively setting the character with the user's Enneagram type as the subject and the core value in the type as the object. The axis of Power could be defined by the stress and security points in Enneagram. In this way, we can derive the themes of 9 Enneagram types and the corresponding Actantial model and make plots. The users will appreciate one of these reconstructed plots in different perspectives and themes, depending on their personality type. In this study, we applied the above methodology to the story of the pansori novel "Tokkijeon". User-centered storytelling is a new attempt to predict user's choice and reconstruct the story based on the user's personality and perspective.

  • Fishing villages have rich potential on account of serving as a base of fishing activities with various maritime ecosystem, culture and maritime resources as well. The object of the study is ports located in Gijang, Busan and it presents the overall development direction in ports within this area. Through systematic and well-planned developing small-sized ports, this research analyzes regional economic implications by improving settlement conditions for fishing villagers scattered around this area taking the ports' natural traits into consideration. The research correlates regional input output analysis by fishing port arrangement project with connection between inter-industries and inter-regions according to the model of input-output analysis and attempts to systematically analyze regional economic structure with the economic implication divided into production, employment and income, influenced upon by the change of political exogenous variable.

Adjustment of heterogeneous variance by milk production level of dairy herd (젖소군의 유생산 수준별 이질성 분산 보정)

  • This experiment was conducted to compare heterogeneity for the variance in dairy cattle population and to induce homogeneity of variance using 502,228 performance test records of dairy cattle. The estimates of heritability for milk yields, fat yields and protein yields were 0.28, 0.26 and 0.24, respectively and the estimate of average breeding value by birth year was lower in HV (heterogenous variance) model than in animal model, collectively. The average breeding values of milk yields, fat yields and protein yields for 545 sire bulls applicable to the criteria of interbull MACE programme were 453.54kg, 10.75kg and 14.33kg, respectively and when the heterogeneity was adjusted they were 432.06kg, 10.15kg and 13.40kg, respectively, which were lower in all milk traits collectively. In animal model, coefficients of phenotypic correlation between dataset I and II were 0.839 in milk yields, 0.821 in fat yields, and 0.837 in protein yields, while in HV model, they were 0.841 in milk yields, 0.820 in fat yields, and 0.836 in protein yields, showing similar results in 2 models. When compared using animal model and HV model, the regression coefficient for ratio of number of daughters by calving year of milk yields increased from 15.157 to 16.105 and that of fat yields increased from =0.227 to =0.196, but that of protein yields decreased from 0.630 to 0.586.

Effect of Pen Floor Condition Depending upon Housing Orientation on the PerFormance of Finishing Hanwoo Steers (우사의 향방에 따른 사육장 바닥면의 조건이 비육말기 거세한우의 생산성에 미치는 영향)

  • In order to investigate the effect of floor condition on the fattening performance of cattle, sixteen finishing Hanwoo steers of weighting 570kg were fed for 16 weeks to slaughter on two types of sawdust bedding pen oriented to the south(SP) and the north(NP) then obtained following results. The floor condition of SP roofed by transparent material was dry and flat during overall experimental period, whereas, that of NP roofed by solid slate was muddy and rough even in summer and fall, and then frozen in winter period resulted in interfering with the locomotion and resting behavior of steers. Average daily dry matter intake of steers was not different between tow treatments, however, the average daily gain of steers in NP and SP was 0.64kg and 0.75kg, respectively, meaning that gained 14.3% less for the NP group(p<0.05), accordingly, feed requirement(feed/gain) and TDN/gain increased by 15.9% and 15.5%, respectively. No significant differences were found in slaughter weight, carcass weight, dressing percentage, meat yield index and meat quality traits between two treatments. In conclusion, this study revealed that poor floor condition of feeding pen could decrease weight gain and feed efficiency of cattle by increasing energy consumption for locomotion and conductive energy loss while resting on the floor surface. These results indicates that housing orientation of cattle shed and the selection of roof material are important factors in beef cattle production.

Identification of Functional and In silico Positional Differentially Expressed Genes in the Livers of High- and Low-marbled Hanwoo Steers

  • This study identified hepatic differentially expressed genes (DEGs) affecting the marbling of muscle. Most dietary nutrients bypass the liver and produce plasma lipoproteins. These plasma lipoproteins transport free fatty acids to the target tissue, adipose tissue and muscle. We examined hepatic genes differentially expressed in a differential-display reverse transcription-polymerase chain reaction (ddRT-PCR) analysis comparing high- and low-marbled Hanwoo steers. Using 60 arbitrary primers, we found 13 candidate genes that were upregulated and five candidate genes that were downregulated in the livers of high-marbled Hanwoo steers compared to low-marbled individuals. A BLAST search for the 18 DEGs revealed that 14 were well characterized, while four were not annotated. We examined four DEGs: ATP synthase F0, complement component CD, insulin-like growth factor binding protein-3 (IGFBP3) and phosphatidylethanolamine binding protein (PEBP). Of these, only two genes (complement component CD and IGFBP3) were differentially expressed at p<0.05 between the livers of high- and low-marbled individuals. The mean mRNA levels of the PEBP and ATP synthase F0 genes did not differ significantly between the livers of high- and low-marbled individuals. Moreover, these DEGs showed very high inter-individual variation in expression. These informative DEGs were assigned to the bovine chromosome in a BLAST search of MS marker subsets and the bovine genome sequence. Genes related to energy metabolism (ATP synthase F0, ketohexokinase, electron-transfer flavoprotein-ubiquinone oxidoreductase and NADH hydrogenase) were assigned to BTA 1, 11, 17, and 22, respectively. Syntaxin, IGFBP3, decorin, the bax inhibitor gene and the PEBP gene were assigned to BTA 3, 4, 5, 5, and 17, respectively. In this study, the in silico physical maps provided information on the specific location of candidate genes associated with economic traits in cattle.

Changes of physiochemical properties of LOX-3 null rice lines stored at different storage temperatures and periods

  • Due to the lack in storage facility, considerable amount of rice is stocked in the open air, which causes increased stale flavor and deteriorates palatability and merchantable quality. Lipoxygenase-3(LOX-3) is involved in the production of volatile constituents in stored rice, and the development of stale flavor is delayed in LOX-3 null rice. LOX activity in the rice grain is localized in bran fraction and the tropical Japonica cultivar 'Dawdam' was reported that the absence of LOX-3 is inherited as a simple recessive trait. Also, it was reported that the peroxidation of unsaturated fatty acids occurs at lower levels in the 'Dawdam' bran fraction during storage than in rice varieties with LOX-3. This study was conducted to develop LOX-3 null rice lines using 'Dawdam' and investigate changes of physicochemical properties of the lines stored at different storage temperatures and periods. So we analyzed texture, toyo glossiness value, germination rate and lipoxygenase activity of 15 LOX-3 null rice lines on the condition of which rough rice had been stored at different temperatures (high temperature condition at $35^{\circ}C$ and low temperature condition at $15^{\circ}C$ for 4months. Hardness and stickiness of the lines tendered to be increased when it was stored at high temperature and adhesiveness, springiness, cohesiveness and chewiness was not considerably different according to storage temperatures and periods. The germination rate of HR29062-B-98-2-1-B among LOX-3 null rice lines was higher than another lines, 99.3, 94.0% after 4months stored at low temperature and high temperature, respectively. The lipoxygenase activity was 3,304, 1,601unit/mg protein after 4months stored at low temperature and high temperatures, respectively. So, it is thought that this line will be useful to breed rice varieties with high storability after tested on agricultural traits.
