Studies on the Utilization of Agricultural Wastes (Part 2) Isolation and Identification of Cellulose Utilizing Bacteria. (농산폐자원의 이용에 관한 연구(제이보) 섬유소자화세균의 분리 및 동정)

  • For the purpose of producing cellulosic single-cell protein from the agricultural wastes, 172 strains of cellulose-assimilating bacteria were isolated from 102 samples of rotten woods, compost soils, soils and so on by the enrichment culture technique. The isolates were examined for their ability to utilize cellulose as carbon source, and then six strains were screened by their strong cellulose assimilating ability and identified as follows: 1. Among six strains of bacteria screened, five strains were identified as species belonged to the genus Cellulomonas and the remainder to the genus of Sporocytophaga. 2. The isolated Sporocytophaga species was identified as S. ellipsosporn because it has a ellipsoidal microcyst. 3. The isolated Cellulomonas species were identical to a strain of C. fimi, C. aurcgena, C. gelida, respectively and two strains to C. flavigena. 4. The isolated C. aurogena was proved to be a new variety becauuse it has different characteristics of assimilating pentoses such as arabinose and xylose from the strain discribed in Bergey's Manual.

Effects of Phloretin, Cytochalasin B, and D-Fructose on 2-deoxy-D-Glucose Transport of the Glucose Transport System Present in Spodoptera frugiperda Clone 21-AE Cells

  • The baculovirus expression system is a powerful method for producing large amounts of the human erythrocyte-type glucose transport protein, heterologously. Characterization of the expressed protein is expected to show its ability to transport sugars directly. To achieve this, it is a prerequisite to know the properties of the endogenous sugar transport system in Spodoptera frugiperda Clone 21 (Sf21) cells, which are commonly employed as a host permissive cell line to support the baculovirus replication. The Sf21 cells can grow well on TC-100 medium that contains 0.1% D-glucose as the major carbon source, strongly suggesting the presence of endogenous glucose transport system. However, unlike the human glucose transport protein that has a broad substrate and inhibitor specificity, very little is known about the nature of the endogenous sugar transport system in Sf21 cells. In order to characterize further the inhibitor recognition properties of the Sf21 cell transporter, the ability of phloretin, cytochalasin B and D-fructose to inhibit 2-deoxy-D-glucose (2dGlc) transport was examined by measuring inhibition constants $(K_i)$. The $K_i's$ for reversible inhibitors were determined from plots of uptake versus inhibitor concentration. The 2dGlc transport in the Sf21 cells was very potently inhibited by phloretin, the aglucone of phlorizin with a $K_i$ similar to the value of about $2{\mu}M$ reported for inhibition of glucose transport in human erythrocytes. However, the Sf21 cell transport system was found to differ from the human transport protein in being much less sensitive to inhibition by cytochalasin B (apparent $K_i$ approximately $10\;{\mu}M$). In contrast, It is reported that the inhibitor binds the human erythrocyte counterpart with a $K_d$ of approximately $0.12\;{\mu}M$. Interestingly, the Sf21 glucose transport system also appeared to have high affinity for D-fructose with a $K_i$ of approximately 5mM, contrasting the reported $K_m$ of the human erythrocyte transport protein for the ketose of 1.5M.

Isolation, Identification, and Characterization of Pichia guilliermondii K123-1 and Candida fermentati SI, Producing Isoflavone β-Glycosidase to Hydrolyze Isoflavone Glycoside Efficiently, from the Korean Traditional Soybean Paste

  • A total of 155 microbial strains were isolated from the Korean traditional soybean paste based on their morphological features on the growth of agar plate. Among the isolated strains, a total of 28 strains were capable of hydrolyzing isoflavone glycoside to isoflavone aglycone efficiently in the soybean paste. Finally, two strains, K123-1 and SI, were selected because of their resistance to 15% NaCl and ability to convert isoflavone glycoside to isoflavone aglycone efficiently during the fermentation of soybean paste. The isolated strains K123-1 and SI were identified to be Pichia guilliermondii and Candida fermentati, respectively, using the partial 26S rDNA sequence analysis and phylogenic analysis. Pichia guilliermondii K123-1 and Candida fermentati SI converted daidzin to daidzein up to 96% and 95%, respectively, and genistin to genistein up to 92% when soybean pastes were fermented at $30^{\circ}C$ for 20 days with a single isolated strain. Pichia guilliermondii K123-1 and Candida fermentati SI were able to grow in the presence of 15% NaCl on both liquid medium and agar plate. We think that Pichia guilliermondii K123-1 and Candida fermentati SI might be one of good candidates for making functional soybean paste because they are isolated from the Korean traditional soybean paste and have a good ability to convert isoflavone glycosides to isoflavone aglycones and a high salt tolerance.

  • Internet is taking up as a new culture on the cyber-space beyond a simple hobby as its utilization ratio is rapidly being spread throughout the world. However most education for internet media has been focused on cyber ethics education and internet addiction. Therefore it arises to need to educate internet literacy for positive and desirable usage of internet media raises. Previous research on media eduction doesn't have been analyzed the outcome of media education. In this study, we show the result of internet media education which performed for fostering internet literacy. For the analysis of education result we observed emotional attitude about internet, internet media recognition, efficacy of using internet, and consciousness of internet ethics for middle school student. As a result, there are many changes for emotional attitude about internet, ability of using internet media, and consciousness of internet ethics.

Effect of Shape and Flow Rate on T10 in Clearwell (정수지의 형상과 유입 유량이 T10에 미치는 영향 연구)

  • To guarantee the disinfection ability in clearwell, a value of CT is considered where C[mg/l] is disinfectant residual at the exit of clearwell and T[min] means $T_{10}$, the contact time when 10% of tracer is out of clearwell after introducing the tracer at the inlet. To meet a CT value required, increasing the C value is not recommended because high C value can increase potential of producing disinfection by product like THMs. Increasing the hydraulic efficiency surrogated by $T_{10}$ is thus an option widely recommended. Right now, it is widely adopted estimating $T_{10}$ considering LW ratio only due to the suggestions of previous researches. The authors think however there are other factors to consider including shape, flow rate, configuration of inlet and outlet, and the existence of intra basin. This study is initiated to closely look at the effects of two factor on hydraulic efficiency. The factors are shape and inlet flow velocity, i.e., inflow. For that, computational fluid dynamics (CFD) model is developed and pilot test is also carried out. The results show that at a L/W ratio, disinfection ability is overestimated with larger length in shape and higher inlet flow velocity. This suggests that in determining $T_{10}$, the shapes of clearwell and inlet flow velocity should also be considered as well as L/W ratio.

  • The objectives of this study are to define the fundamental notions of collaboration in design field. The results of this study are summarized as follows: Firstly, collaboration means a cooperative work which are cooperatively done by collabarator and collaboratee on the equal status within a certain period of time, based on core ability of each of them. This cooperative work is presented with a visual image based on the identity of participants, and it creates an added value. Collaboration is different from convergence and hybrid in the methodological aspects and from brand alliance and strategic alliance in the aspects of the subject of project. Secondly, collaboration has been developing through step of using the specific ability, step of sharing each core abilities, and step of fusing their identities, in a view of the sphere of cooperative work and the intimate relations in reciprocity. Thirdly, according to strategic purpose, collaboration can be classified into value ascending collaboration, image changing collaboration, business-field extension collaboration, and event collaboration. Fourthly, production methods of collaboration consist of limited edition method, premium line method, common line method, producing a collaborative product, and so on. Fifthly, differentiation effect, image upgrade effect, production of higher value-added products, sale increase, diversification of business, sharing the targets, and promotion effect are achievable through collaboration. Sixthly, to make progress in collaboration successfully, the compatibility of combination between collaborator and collaboratee, the superiority of collaboratee, definitude of collaboration, the fit method of collaboration, the novelty of collatoration, possibility of issuing of collaboration, and perfection of collabo-product must be the points which deserve our attention.

Optimization of Extraction Conditions of Antioxidants from Sporophyll of Undaria pinnatifida by Response Surface Methodology (반응표면분석법에 의한 미역 포자엽의 항산화물질 추출 조건 최적화)

  • We selected a solvent for producing antioxidants from sporophyll of Undaria pinnatOptimum extraction condition for antioxidant ability(Y) of sporophyll of U. pinnatifida was investigated using response surface methodology(RSM). A compound central design was used with variables ($X_1$)temperature($^{\circ}C$) (3.4; 20; 60; 100; 116.6); ($X_2$) pH(1.8; 3; 6; 9; 10.2) and ($X_3$) treatment time(min) (1.7; 10; 30; 50; 58.3). Antioxidant activities were increased with decreasing pH and increasing temperature. The optimal extraction conditions for antioxidant ability(Y) of sporophyll of U. pinnatifida were found to be temperature $51.55^{\circ}C$, pH 4.2 and treatment time 28.2 min. Among the variables tested, pH showed greater significant facin extraction of antioxidants from sporophyll of U. pinnatifida.

Isolation of Mutants Overproducing Amylase from Nuruk Fungi by NTG (NTG에 의한 Amylase활성이 높은 누룩사상균의 변이주의 분리)

  • Aspergillus coreanus NR 15-1, Asp. oryzae NR 15-3 and Asp. oryzae NR 2-5 isolated from traditional Korean nuruk were screened as parental strains producing starch hydrolyzing enzymes. They were mutagenized by N-methyl -N'-nitro-N-nitrosoguanidine (NTG) and mutants were isolated for analysis of various amylase activities and the ability of acid production. Among them, the mutants harboring high saccharogenic activity, dextrinogenic activity, and the ability of acid production were selected. Fifteen, six, and five strains of mutants were isolated from Asp. coreanus NR 15-1, Asp. oryzae NR 2-5, and Asp. oryzae NR 15-3, respectively followed by NTG mutagenesis. Among these mutants, thirteen strains were identified as auxotrophic mutants. \ulcorner (Arg. ̄) mutant from Asp. coreanus NR 15-1 showed high glucoamylase activity and total acid productivity. Z6 (Ade. ̄) mutant from Asp. oryzae NR 2-5 showed the highest $\alpha$-amylase activity, therefore \ulcorner and Z6 mutant were selected.

A Cellulolytic and Xylanolytic Enzyme Complex from an Alkalothermoanaerobacterium, Tepidimicrobium xylanilyticum BT14

  • A cellulolytic and xylanolytic enzyme complex-producing alkalothermoanaerobacterium strain, Tepidimicrobium xylanilyticum BT14, is described. The cell was Grampositive, rod-shaped, and endospore-forming. Based on 16S rRNA gene analysis and various lines of biochemical and physiological properties, the strain BT14 is a new member of the genus Tepidimicrobium. The strain BT14 cells had the ability to bind to Avicel, xylan, and corn hull. The pH and temperature optima for growth were 9.0 and $60^{\circ}C$, respectively. The strain BT14 was able to use a variety of carbon sources. When the bacterium was grown on corn hulls under an anaerobic condition, a cellulolytic and xylanolytic enzyme complex was produced. Crude enzyme containing cellulase and xylanase of the strain BT14 was active in broad ranges of pH and temperature. The optimum conditions for cellulase and xylanase activities were pH 8.0 and 9.0 at $60^{\circ}C$, respectively. The crude enzyme had the ability to bind to Avicel and xylan. The analysis of native-PAGE and native-zymograms indicated the cellulosebinding protein showing both cellulase and xylanase activities, whereas SDS-PAGE zymograms showed 4 bands of cellulases and 5 bands of xylanases. Evidence of a cohesinlike amino acid sequence seemed to indicate that the protein complex shared a direct relationship with the cellulosome of Clostridium thermocellum. The crude enzyme from the strain BT14 showed effective degradation of plant biomass. When grown on corn hulls at pH 9.0 and $60^{\circ}C$ under anaerobic conditions, the strain BT14 produced ethanol and acetate as the main fermentation products.

Functions of Virginiae Butanolide C(VB-C) and Receptor in Virginiamycin Production (Virginiamycin 생산유도에 관여하는 Virginiae Butanolide C(VB-C) 및 Receptor의 기능)

  • Streptomyces virginiae produces a set of autoregulators termed virginiae butanolide A-E(VB-A-E) which trigger virginiamycin production, and possesses a high-affinity virginiae butanolide receptor. To elucidate the functions of VB-C and VB-C receptor, we isolated two mutants from S. virginiae by N-methyl-N'-nitro-N-nitrosoguanidine and hydroxylamine. The characteristics of the mutants showed that the producing time of antibiotics was very delayed due to a slower production of VB-C receptor than that of VB. In S. ostreogriseus(VB', receptor -) and S. graminofaciens(VBU, receptor+), which produce the virginiamycin, the addition of synthetic VB-C repressed the production of antibiotics in S. ostreogriseus but induced tbe production in S. graminofaciens. HPLC analysis of S. graminofaciens suggested that the VB-C might have an ability to induce the production of virginiamycin and other antibiotics. These results imply that the VB-C has an ability to trigger the production of other secondary metabolites as well as virginiamycin under VB-C receptor existence.
