Topical or oral treatment of peach flower extract attenuates UV-induced epidermal thickening, matrix metalloproteinase-13 expression and pro-inflammatory cytokine production in hairless mice skin

  • BACKGROUND/OBJECTIVES: Ultraviolet radiation (UV) is a major cause of skin photoaging. Previous studies reported that ethanol extract (PET) of Prunus persica (L.) Batsch flowers (PPF, peach flowers) and its subfractions, particularly the ethylacetate (PEA) and n-butanol extracts (PBT), have potent antioxidant activity and attenuate the UV-induced matrix metalloproteinase (MMP) expression in human skin cells. In this study, we investigated the protective activity of PPF extract against UV-induced photoaging in a mouse model. MATERIALS/METHODS: Hairless mice were treated with PET or a mixture of PEA and PBT either topically or orally along with UV irradiation. Histological changes and biochemical alterations of mouse skin were examined. Major phenolic compounds in PPF extract were analyzed using an ACQUITY UPLC system. RESULTS: The overall effects of topical and oral treatments with PPF extract on the UV-induced skin responses exhibited similar patterns. In both experiments, the mixture of PEA and PBT significantly inhibited the UV-induced skin and epidermal thickening, while PET inhibited only the UV-induced epidermal thickening. Treatment of PET or the mixture of PEA and PBT significantly inhibited the UV-induced MMP-13 expression, but not type I collagen expression. Topical treatment of the mixture of PEA and PBT with UV irradiation significantly elevated catalase, superoxide dismutase (SOD) and glutathione-peroxidase (GPx) activities in the skin compared to those in the UV irradiated control group, while oral treatment of the mixture of PEA and PBT or PET elevated only catalase and SOD activities, but not GPx. Thirteen phytochemical compounds including 4-O-caffeoylquinic acid, cimicifugic acid E and B, quercetin-3-O-rhamnoside and kaempferol glycoside derivatives were identified in the PPF extract. CONCLUSIONS: These results demonstrate that treatment with PET or the mixture of PEA and PBT, both topically or orally, attenuates UV-induced photoaging via the cooperative interactions of phenolic components having anti-oxidative and collagen-protective activities.

The Protective Effect of Chondroitin from Raja kenojei Cartilage on Collagen-induced Arthritis in DBA/1J Mice

  • In this study, we evaluated whether the oral administration of chondroitin from the cartilage of Raja kenojei is effective on the progression of rheumatoid arthritis (RA), using collagen-induced arthritic (CIA) mice. Arthritis development was delayed dose-dependently in the chondroitin-treated groups. The pre- and late-treated groups receiving 1,000 mg/kg of chondroitin had clinical scores that were reduced significantly by 56.9 (p<0.05) and 43.3% (p<0.05), respectively, compared to the vehicle-treated groups. Hematoxylin eosin staining and X-ray radiography showed that the chondroitins reduced the infiltration of inflammatory cells and prevented joint destruction of the knee and paw. Reverse transcription-polyerase chain reaction analysis revealed that chondroitin administration inhibited the expressions of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), $interlukin-1{\beta}$ ($IL-1{\beta}$), and $interferon-{\gamma}$ ($IFN-{\gamma}$) in joints more than the administration of vehicle. Chondroitin treatment also decreased the production of rheumatoid factors (RF), IgG and IgM, in the serum of CIA mice. These results indicate that chrondroitin administration has a protective effect involving the inhibition of pro-inflammatory cytokine production in CIA mice.

강도가 제어된 인공피부 진피를 활용한 기능성 펩타이드의 프로콜라겐 생합성 분석 (Analysis of Procollagen Biosynthesis of Functional Peptides Utilizing Stiffness Controlled Artificial Skin Dermis)

  • 본 연구에서는 가교 분자를 사용하여 0.7 kPa에서 17.7 kPa까지 다양한 강도를 갖는 콜라겐 겔을 성공적으로 제조하였다. 가교된 콜라겐 겔에 다공성 기공을 도입하고 진피세포를 내부에 담지하여, 겔 강도에 따른 세포 성장 및 거동을 확인하였다. 상대적으로 강도가 높은 겔에서 진피세포의 프로콜라겐 생합성이 47 ng에서 32 ng까지 감소하는 것을 확인하였다. 이렇게 제조된 인공피부 진피에 아데노신을 처리하였을 때, 특정 강도를 갖는 콜라겐 겔에서 프로콜라겐 생합성이 감소하는 것을 확인하였다. 반면에 기능성 펩타이드를 처리하였을 때는 프로콜라겐 생합성이 콜라겐 겔의 강도에 크게 영향을 받지 않는 것을 확인할 수 있었다. 이러한 결과는 강도가 제어된 인공피부 제조 및 응용, 나아가 다양한 조직공학 분야의 기반 기술로 활용 가능하리라 기대된다.

Cloning and Expression Analysis of the ${\alpha}$-Subunit of Porcine Prolyl 4-hydroxylase

  • Prolyl 4-hydroxylase (P4H) plays a central role in collagen synthesis by catalyzing the hydroxylation of the proline residue in the X-Pro-Gly amino acid sequence, and controls the biosynthesis of collagen that influences overall meat quality. In order to verify expression level of the catalytic ${\alpha}$ subunit of P4H, a 2.7 kb clone of the ${\alpha}$ subunit gene for P4H was selected from a cDNA library prepared from the muscular tissue of Sancheong berkshire pigs, and the whole gene sequence was determined. As expression level of the ${\alpha}$ subunit of P4H differed between tissues of pigs, we intended to assess more precisely the level of ${\alpha}$-subunit expression between tissues of Sancheong Berkshire pigs by using RT-PCR. Muscular and adipose tissues were taken from each pig grouped by growth stage (weighing 60, 80, and 110 kg) of Yorkshire and Sancheong Berkshire pigs, and the expression levels of the ${\alpha}$-subunit of P4H were examined. Since there were significant differences in the expression level with respect to variation in growth stage (p<0.01), an attempt was made to identify any influences of pig species and tissue variation. The muscular and adipose tissues of pigs weighing 110 kg showed higher expression levels than pigs weighing 60 kg and 80 kg. In general, significantly higher expression levels were found in muscular than in adipose tissue. The expression levels in Sancheong Berkshire were significantly higher than in Yorkshire pigs (p<0.01 or p<0.05). Since expression level of the ${\alpha}$-subunit of P4H affects the activity of P4H and is connected to the biosynthesis of collagen and increased collagen can improve meat texture, this finding may explain why meat quality of the Sancheong Berkshire pig is acclaimed in Korea. Given the higher expression levels of the ${\alpha}$-subunit gene in adipose than in muscular tissue, and also in the heavier pigs, more intensive studies are required to assess the correlation between expression level of the ${\alpha}$ subunit gene and overall meat quality.

유자 부산물 에탄올 추출물의 항노화 및 미백효과 (Antiaging and Whitening Activities of Ethanol Extract of Yuza (Citrus junos SIEB ex TANAKA) By-product)

  • 유자(Citrus junos Sieb ex TANAKA)는 북동아시아 지역에서 재배되는 감귤류 종이며, 비타민 C, 플라보이드, 리모노이드를 풍부히 함유하고 있다. 한국에서 재배되는 유자는 차나 음식료품으로 사용되고, 부산물인 씨와 과피는 전량 폐기되고 있다. 이 연구에서는 유자 부산물을 기능성 화장품 소재로서 이용 가능성을 평가하였다. 씨와 과피 에탄올 추출물에서 DPPH (2,2-diphenyl-1-picrylhydrazyl) 라디칼 소거활성 효과는 크지 않았지만 고농도의 phenolic compounds를 함유하고 있다. 각질형성세포(HaCaT Keratinocyte)와 섬유아세포(Normal Fibroblast)에 UVB를 각각 12.5 $mJ/cm^2$, 15 $mJ/cm^2$를 조사한 후 항염효과와 Collagen 합성량, Collagen 분해효소인 MMP-1 (matrix metalloproteinase)의 발현량을 확인하였다. 과피 추출물에서 UVB에 의해 증가된 TNF-$\alpha$를 상당량 줄여주었다. 마찬가지로 과피 추출물은 MMP-1의 발현량을 줄여주고 Collagen의 합성량도 증가시켰다. 한편 유자 추출물의 $\alpha$-MSH (Melanocyte stimulating hormone) 처리에 의해 유도된 멜라닌 생합성 억제효능과 타이로시나제 발현량 감소를 확인하였다. 결론적으로 유자 부산물의 에탄올 추출물질은 항노화, 미백 기능성 화장품 소재로 가치가 기대된다.

당귀작약산의 창상 회복에 대한 효과 (The Effect of Dangguijakyak-san on Wound Healing)

  • Objectives We evaluated the wound healing effects of Dangguijakyak-san (DJ) using C57BL/6 mice that were generated open wound. Methods The study was conducted with seven C57BL/6 mice assigned to each group, divided into the normal group, control group, vitamin E group, DJ low-dose group, DJ high-dose group. We measured total polyphenol, flavonoid contents, the size of the wound, liver function, pro-inflammatory cytokine activity in serum, inflammation-related proteins, adhesion molecules and chemokine proteins, collagen-related proteins in skin tissue and histopathological changes by H&E and Masson's staining. Results DJ treatment significantly reduced the area of the wound compared to the control group. Also, inflammatory cytokines were reduced and the expression of anti-inflammatory-related factors (interleukin-4 [IL-4] and IL-10) was significantly increased in the DJ treatment group. We identified that DJ treatment inhibits both pathways of inflammation, the mitogen-activated protein kinases and nuclear factor-κB pathway. Moreover, the protein expressions of Sirt1 (sirtuin 1), MCP-1 (monocyte chemoattractant protein 1), ICAM-1 (intercellular adhesion molecule 1), and VCAM-1 (vascular cell adhesion molecule 1) were decreased by DJ administration. Also, the expression of α-smooth muscle actin and collagen type I alpha 1, collagen-related proteins, that help skin recovery was significantly increased in the DJ treatment group. Histopathologically, a relatively thin epithelial layer could be observed in the DJ administration group, as well as an increase in fibroblasts and collagen fibers. Conclusions These data suggest that DJ treatment is effective in wound healing, suppressing inflammatory proteins, increasing skin repair factors and improving histopathological changes caused by wounds.

KCl Mediates $K^+$ Channel-Activated Mitogen-Activated Protein Kinases Signaling in Wound Healing

  • Background Wound healing is an interaction of a complex signaling cascade of cellular events, including inflammation, proliferation, and maturation. $K^+$ channels modulate the mitogen-activated protein kinase (MAPK) signaling pathway. Here, we investigated whether $K^+$ channel-activated MAPK signaling directs collagen synthesis and angiogenesis in wound healing. Methods The human skin fibroblast HS27 cell line was used to examine cell viability and collagen synthesis after potassium chloride (KCl) treatment by Cell Counting Kit-8 (CCK-8) and western blotting. To investigate whether $K^+$ ion channels function upstream of MAPK signaling, thus affecting collagen synthesis and angiogenesis, we examined alteration of MAPK expression after treatment with KCl (channel inhibitor), NS1619 (channel activator), or kinase inhibitors. To research the effect of KCl on angiogenesis, angiogenesis-related proteins such as thrombospondin 1 (TSP1), anti-angiogenic factor, basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF), pro-angiogenic factor were assayed by western blot. Results The viability of HS27 cells was not affected by 25 mM KCl. Collagen synthesis increased dependent on time and concentration of KCl exposure. The phosphorylations of MAPK proteins such as extracellular-signal-regulated kinase (ERK) and p38 increased about 2.5-3 fold in the KCl treatment cells and were inhibited by treatment of NS1619. TSP1 expression increased by 100%, bFGF expression decreased by 40%, and there is no significant differences in the VEGF level by KCl treatment, TSP1 was inhibited by NS1619 or kinase inhibitors. Conclusions Our results suggest that KCl may function as a therapeutic agent for wound healing in the skin through MAPK signaling mediated by the $K^+$ ion channel.

바이러스 안전성이 보증된 돼지유래 골 이식재 제조 공정 개발 (Process development of a virally-safe dental xenograft material from porcine bones)

  • 동물유래성분을 원재료로 사용하는 의료기기는 원료물질 유래 바이러스가 오염될 가능성이 있기 때문에 생산과정 중 바이러스가 오염되지 않도록 하여야 한다. 또한 생산공정은 오염될 가능성이 있는 바이러스들을 불활화하거나 제거하는 공정을 포함하여야 한다. 본 연구를 통해 돼지유래 이종골을 원재료로 사용한 바이러스 안전성이 보증된 치과용 골이식재(THE Graft$^{(R)}$) 제조공정을 개발하였다. THE Graft$^{(R)}$ 제조공정은 30% 과산화수소수와 80% 에탄올을 각각 처리하여 지방을 제거하는 공정과 $1,300^{\circ}C$ 열처리 공정을 통해 콜라겐과 유기물을 제거하는 공정을 포함한다. 또한 최종적으로 생산된 hydroxyapatite 성분의 골이식재에 25 kGy의 감마선을 조사하여 감염성 위해인자를 불활화하는 공정을 포함한다. THE Graft$^{(R)}$의 형태학적 특성을 소유래 hydroxyapatite 성분의 골이식재인 Bio-Oss와 SEM과 TEM을 이용하여 비교한 결과 구조적 특성이 유사함을 확인하였다. $1,300^{\circ}C$ 열처리 공정과 25 kGy 감마선 조사 공정의 바이러스 불활화 효과를 평가하기 위해 transmissible gastroenteritis virus (TGEV), pseudorabies virus (PRV), porcine rotavirus (PRoV), porcine parvovirus (PPV)를 모델 바이러스로 선정하였다. $1,300^{\circ}C$ 열처리 공정에서 TGEV, PRV, PRoV, PPV 모두 검출한계 이하로 불활화되었으며, 바이러스 로그 감소 값은 각각 ${\geq}4.65$, ${\geq}5.81$, ${\geq}6.28$, ${\geq}5.21$이었다. 또한 감마선 조사 공정에서도 TGEV, PRV, PRoV, PPV 모두 검출한계 이하로 불활화되었으며, 바이러스 로그 감소 값은 각각 ${\geq}4.65$, ${\geq}5.87$, ${\geq}6.05$, ${\geq}4.89$이었다. 두 공정에서 TGEV, PRV, PRoV, PPV의 누적 바이러스 로그 감소 값은 각각 ${\geq}9.30$, ${\geq}11.68$, ${\geq}12.33$, ${\geq}10.10$이었다. 이상의 결과에 의하면, THE Graft$^{(R)}$ 제조공정은 바이러스 안전성 보증을 위한 충분한 바이러스 불활화 능력을 가지고 있는 것으로 판단된다.

Calamagrostis arundinacea (실새풀) 추출물의 항노화 및 항염증 활성 (Anti-aging and Anti-inflammatory Activities of the Extracts of Calamagrostis arundinacea)

  • 본 연구에서는 실새풀(Calamagrostis arundinacea)의 열수 추출물(Ca-HW)과 70% 에탄올에 추출된 추출물(Ca-E70)을 이용한 항노화 및 항염증 활성을 조사하였고, 또한 이들의 생리 활성 능력을 무세포와 세포 기반 시스템으로 조사하였다. 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical decolorization 방법으로 항산화 활성 평가를 실시한 결과, Ca-HW와 Ca-E70은 각각 27%와 48%의 DPPH 라디칼 소거능으로 항산화 활성을 나타냈다. 각 추출물에서는 RAW 264.7, B16F10 및 CCD986-sk 세포에서 독성, 세포증식 및 증진 효과는 없는 것으로 확인되었다. 멜라닌 생성과 관련 있는 B16F10 세포주에서 100 mg/ml 농도의 Ca-HW와 Ca-E70 처리구는 abutin 처리구에 비해 멜라닌 생성 억제 효과가 높은 것을 확인하였다. 인간의 섬유아세포인 CCD986-sk를 통한 pro-collagen 생합성에 미치는 Ca-HW와 Ca-E70의 영향을 조사하였더니 각각 24.69%와 12.55%가 증가하는 것으로 나타났다. LPS를 처리하여 염증반응을 유도 한 RAW264.7 세포에서 실새풀 추출물의 효과를 조사하였다. Ca-E70은 LPS에 의한 산화질소의 생성과 전염증성 사이토카인인 IL-6의 생성을 억제함으로써 염증 반응에 관여하는 것으로 보인다. 이러한 결과는 실새풀 추출물이 피부와 관련하여 항노화 및 항염증 능력이 있는 것을 제시한다.

승마, 황련, 황백, 후박 추출 혼합물의 항노화 및 멜라닌 생성 저해효과 (Anti-aging and Anti-melanogenesis Effect of Cimicifuge dahurica, Coptis chinensis, Phellodendri amurense and Magnol obovata Extracts Mixture)

  • 한의학에서 승마(Cimicifuge dahurica, C. dahurica), 황련(Coptis chinensis, C. chinensis), 황백(Phellodendri amurense, P. amurense) 추출물은 해독작용을 하며 후박(Magnol obovata, M. obovata) 추출물은 항균 효과가 있다고 알려져 있다. 본 연구에서는 승마, 황련, 황백, 후박 추출물을 혼합하여 기능성 화장품 소재로써의 이용 가능성을 검토하였다. MTT assay를 이용한 세포 독성평가와 DPPH와 ABTS를 이용한 항산화 효과를 측정한 결과, 단일 추출물보다 추출혼합물이 더 안전함을 확인 하였으며 농도 의존적으로 항산화 활성이 증가됨을 확인하였다. 뿐만 아니라 NO 생성을 억제함으로써 우수한 항염증 효과가 있을 것으로 예상되며, 섬유아세포에서 UVB로 유도된 세포 손상을 보호하면서 type 1 pro-collagen 합성량이 증가되는 것을 관찰하였다. 또한 추출 혼합물은 melanin 합성과 tyrosinase 활성 및 melanin 합성 관련 인자 및 효소들(MITF, tyrosinase, TRP-1, TRP-2)의 발현을 효과적으로 억제하였다. 이러한 연구결과로 보아 승마, 황련, 황백, 후박 추출 혼합물은 항노화 및 미백 등 기능성 화장품 소재로써 개발 가능성이 높을 것으로 사료된다.
