A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative (인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구)

  • Here, we conducted the study on the possibility of using FK-13, a short analog of human-derived antibacterial peptide LL-37, as a cosmetic preservative to discover a natural cosmetic preservative that is safe for human body. For the purpose, FK-13 composed of 13 amino acids was synthesized by solid-phase peptide synthesis, and purified using reversed phase-high performance liquid chromatography (RP-HPLC). The purity and molecular weight were confirmed by liquid chromatography-mass spectrometry (LC-MS) analysis. FK-13 showed high antimicrobial activity on the three gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, and Staphylococcus epidermidis), the three gram-negative bacteria (Escherichia coli, Salmonella typhimurium, and Pseudomonas aeruginosa), and also even the fungus Candida glabrata. FK-13 had a broad spectrum of antibacterial activity, showing a suitability as a cosmetic preservative. In addition, FK-13 showed high thermostability and higher antibacterial activity in a comparative test with existing natural herbal cosmetic and chemical preservatives. Therefore, as FK-13 is a safe material and has high antibacterial activity at a low concentration, it is likely to be applied as a peptide natural cosmetic preservative that can replace existing chemical preservatives.

Antimicrobial Plant Extracts as an Alternative of Chemical Preservative: Preservative Efficacy of Terminalia chebula, Rhus japonica (gallut) and Cinnmomum cassia Extract in the Cosmetic Formular (가자, 오배자, 계피 추출물을 이용한 화장품 제형에서의 방부효과)

  • This study was carried to investigate the efficiency of antimicrobial plant extracts as natural preservative in the cosmetic formulations. Ethanol extracts of different plants were tested using the disc diffusion (paper disc) method and the minimum inhibitory concentration (MIC) method for their antimicrobial activity against the common poultry pathogens. Terminalia chebula and Rhus japonica (gallut) extracts exhibited antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. Cinnmomum cassia extract exhibited antifungal activity against Candida albicans and Aspergillus niger while the remaining plant extracts showed no activity. A study of the preservative efficacy of the cosmetic formular containing the T. chebula, R. japonica and C. cassia extracts demonstrated sufficient preservative efficacy against bacteria and eukaryotic test microbes. Also, the cosmetic formulations containing antimicrobial plant extracts more effectively inhibited the microoranisms than the mixture of traditional chemical preservatives. These results suggest that the mixture of antimicrobial plant extracts, T. chebula, R. japonica and C. cassia is incorporated as preservative in the cosmetic formulation and the mixture have considerable effect on its efficacy.

Effects of Earthenware Vase on Vase Life of Cut Gerbera Flowers and Microbial Multiplication (옹기 화병이 절화 거베라의 수명 및 미생물 증식에 미치는 영향)

  • To examine the availability of onggi (earthenware) as a floral vase, we compared the vase life of cut gerberas (Gerbera jamesonii) with generally used vases made of glass and porcelain. After holding cut gerbera 'Honeymoon' and 'Golden Time' in vases made of glass, porcelain, and onggi containing holding solution (tap water or preservative solution), vase life, fresh weight, and water uptake of cut flowers, and total number of microbes in holding solution were collected. In onggi vase, vase life of both the cultivars was longer than the other vases and floral preservative prolonged vase life compared with tap water. Cut gerberas applied with preservative solution in onggi vase maintained their ornamental value for more than 14 days. Cumulative water uptake by cut gerberas was greatest in onggi vase, where 'Honeymoon' and 'Golden Time' absorbed water by 19.2 and 15.8 mL, respectively, for 10 days. Fresh weight of cut flowers in onggi vase was greater than those of the others, and the preservative solution delayed the reduction of fresh weight compared with tap water. Especially, onggi vase with preservative solution maintained 89% of the initial fresh weight at the 8th day of the treatments due to smooth water uptake with comparison with 71% in porcelain. Total number of microbes in the holding solution was smallest in onggi vase. At the 8th day, microbes number in onggi vase was 435 cfu/mL, that was 20-30% level of those of glass and porcelain vases. In conclusion, onggi vase inhibited microbial proliferation in holding solution, maintained water uptake by cut flower, and delayed bending of flower stalks due to vascular blockage, and then prolonged vase life in gerbera. Therefore, onggi with numerous micropores will be a good material for flower vase and preservative solution can improve its function.

Decay Resistance and Effectiveness of CCA Preservative against Decay on the 4 Imported Softwoods (수입침엽수(輸入針葉樹) 4수종(樹種)의 내후성(耐朽性) 및 CCA계(系) 목재방부제(木材防腐劑) 처리(處理)에 따른 방부효과(防腐效果))

  • With the aim to investigation of decay resistance and optimum concentration in chrome-copper-arsenic(CCA) preservative treatment on the imported softwoods from Siberia and North america, preservative absorption after CCA impregnation, weight losses and degradation patterns by decay fungi were examined. The density and latewood rate of Siberia softwoods(Spruce and Larch) were higher than those of North america softwoods(Douglas-fir and Western hemlock), resulting in the decrease of the CCA preservative absorption in the Siberia softwoods. In the case of untreated softwoods, decay resistance against Coriolus versicolor was lower than against Tyromyces palustris. For CCA treated softwoods, preservative effectiveness increased with increase in concentration of CCA solution. When treated with 0.7% CCA solution, efficiency value was more than 80 and 90 for C. versicolor and T. palustris, respectively. From this results, in the CCA preservative treatment for imported softwoods, it can be concluded that optimum concentration of CCA solution is approximately 0.7%. The absorption of CCA preservative distributed in the range of 3.8 and $5.5kg/m^3$. After exposure to testing fungi, in the untreated softwoods, bore holes formed in the cell walls and bordered pits, moreover, bordered pit canals enlarged by the fungi. However, 0.5% CCA treated softwoods was almost no deterioration in the cell walls and bordered pits due to decay.

  • This study was to examine the anti-bacteria effect and the changes of chemical properties of chrome-tanned garment leathers on the preservative treatment. Various test methods, such as investigation of preservative treatment process, resistance test and chemical analysis by cleaning, antbbacterial test by shake flask method are carried out in this study. The results can be obtained as follows: 1. Bacterial reduction percentage of chrome-tanned garment leathers on the preservative treatment was 28.6%. 2. In the antbbacterial effect by dry cleaning, preservative treated leathers has no resistance. 3. Fats content has been removed by dry cleaning using perchloroethylene, so garment leathers properties were altered. 4. PH value was changed by dry cleaning. But once after fats removing, it was changed the little by dry cleaning numbers.

  • Anti-microbial activities of Scutellariae Radix Extracts and Sophora flavecens Extracts was tested by formulation, such as skinsoftener, emulsion for 4 weeks. Control contained no preservative, test group contained 1.00% Scutellariae Radix extracts and 1.00% Sophora flavecens extracts and positive control contained parabens and imidazolidinyl urea. To determine the anti-microbial activity of these extract, the 4 germs such as Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Canida albicans were used. The test groups showed significant anti-microbial activities against the 4 germs at 2 and 3 weeks as compared with control. Anti-microbial activities of these extracts were similar to positive control. Considering that the Scutellariae Radix Extracts and the Sophora flavecens Extracts have a significant anti-microbial activities against 4 germs, it is possible as natural preservative in cosmetics.

Corrosion of Metals in Waterborne Preservative-Treated Wood (수용성(水溶性) 방부처리재(防腐處理材)에서 금속류(金屬類)의 부식(腐蝕))

  • This study was performed to investigate the corrosion of metal fasteners in waterborne preservative-treated wood. Of all the metal tested, steel exhibited the greatest amount of corrosion across all preservatives and exposure conditions whereas stainless steel was totally inert. Galvanized steel corroded at a much lower rate compared to steel and the corrosion of brass was negligible. Among the preservatives, CCA-Type B was the most corrosive system tested. The sequence for the average corrosivity across all metals was: CCA-type B>CCA-Type C${\geq}$ACC${\geq}$CCA-Type A. Across all metals and retentions the salt formulations were more corrosive than the oxide and the corrosion was increased with the increse in the amount of preservative loadings. The amount of corrosion was also increased with the increase in exposure relative humidity (RH) across all metals and presevatives However, at the 60% RH exposure condition, the corrosion of metals was very negligible. Consequently, it can be concluded that across all metals oxide-type preservatives should be used to prevent the corrosion problem of metal fasteners in contact with treated wood and the use of steel nail preservative-treated wood should be avoided without distinction of end-use location, i.e., exterior or interior, and the use of stainless steel or at least brass nail in damp exterior condition was strongly recommended.

  • With the aim to utilize pine wood(Pinus densiflora Sieb. et Zucc.) as an interior building materials, such as flooring material, monoethylene glycol(MEG) resin solution was impregnated into greenwood. Specimens of three different qualities, that is, normal wood, resinous wood and compression wood, were prepared. Distribution of resin element(phosphorus) in MEG resin solution-impregnated woods and preservative effectiveness against brown rot fungi(Tyromyces palustris and Serpula lacrymans) of these woods were investigated. The results were as follows: 1. The concentration of phosphorus into cell walls of resinous wood and compression wood was lower compared to that of normal wood. This shows that the quality of wood has an influence on the penetration of MEG resin solution into the wood. It was shown from a leaching test that MEG resin could be leached out easily from the cell walls. 2. The resinous wood and compression wood, even without MEG resin solution impregnation had high decay resistance. For normal wood, significant improvement of preservative effectiveness was observed after impregnation of MEG resin solution. It was shown that MEG resin was leached out from the woods after leaching test, resulting in the reduction of preservative effectiveness. From this result, suitability of MEG resin solution-impregnated woods as an interior materials was recognized.

Measurment of Copper Concentration in ACQ, CUAZ, and CB-HDO Solutions by Using a Spectrophotometer

  • This research was performed to develop the simple techniques to predict the copper concentration in alkaline copper quat (ACQ), copper azole (CUAZ), and bis-(N-cyclohexyl-diazeniumdioxy)-copper (CB-HDO) solutions. Two simple methods measuring the color due to copper compounds were evaluated by using a spectrophotometer. One is to directly measure the color of the preservative solutions. The other is to measure the color developed on the surface of a treated sample with the preservatives. The $L^*$ of the measured color values appeared to be the most sensitive to the change of copper concentration. The $a^*$ values of the preservative solutions tended to be decreased at above a certain concentration condition, and the $b^*$ values showed no trend with the concentration of copper compounds in preservative solutions. The surface color of the treated samples were changed from bluish to greenish as time passed. Both methods showed the high $R^2$ values of the regression models determined by using the lightness, which suggested that the methods might be applicable in preservative-treatment mills for the easy and fast prediction of the copper concentration.

Effect of Controlled Atmosphere and Modified Atmosphere Storage on the Color and Sensual Properties of Fresh and Red Ginseng (CA 및 MA저장이 수삼 및 홍삼의 색상 및 관능적 특성에 미치는 영향)

  • In order to stabilize the price of fresh ginseng by extension of seasonal variation and marketing structure, and to reduce the cost of production by works of intensive and short term, studies were carried out. As fresh ginseng of 4 years old by the MA and CA was stored for 12 week at 4$^{\circ}C$, samples were collected after every 1, 2, 3, 4, 6, 8, 12 weeks for processing red ginseng. Color and Sensual characteristics on ginseng steamed red for the quality evaluation are summarized as follows. For the time course of storage with red ginseng, which was processed from fresh one after various treatments, L and b values were decreased, however a value was increased. In particular, absorbance was step-wisely increased. Turbidity was somewhat increased, and non-treatment of preservative were shown 2 times higher of absorbance. Extraction ratio of ethanol-soluble extractives slightly increased by the preservation methods, and as storage period was passed. CA storage and preservative treat merit versus MA and non-preservative treatment gave much stability in ginseng quality. In the sensual characters, sour taste and sweet taste were increased, but fresh taste and rice scorched taste were decreased. Bitter taste was not much changed until end of storage. CA was shown smaller differences than MA in the pH change. Addition of preservative extended the storage time.
