Pattern Clustering of Symmetric Regional Cerebral Edema on Brain MRI in Patients with Hepatic Encephalopathy (간성뇌증 환자의 뇌 자기공명영상에서 대칭적인 지역 뇌부종 양상의 군집화)

  • Purpose Metabolic abnormalities in hepatic encephalopathy (HE) cause brain edema or demyelinating disease, resulting in symmetric regional cerebral edema (SRCE) on MRI. This study aimed to investigate the usefulness of the clustering analysis of SRCE in predicting the development of brain failure. Materials and Methods MR findings and clinical data of 98 consecutive patients with HE were retrospectively analyzed. The correlation between the 12 regions of SRCE was calculated using the phi (φ) coefficient, and the pattern was classified using hierarchical clustering using the φ2 distance measure and Ward's method. The classified patterns of SRCE were correlated with clinical parameters such as the model for end-stage liver disease (MELD) score and HE grade. Results Significant associations were found between 22 pairs of regions of interest, including the red nucleus and corpus callosum (φ = 0.81, p < 0.001), crus cerebri and red nucleus (φ = 0.72, p < 0.001), and red nucleus and dentate nucleus (φ = 0.66, p < 0.001). After hierarchical clustering, 24 cases were classified into Group I, 35 into Group II, and 39 into Group III. Group III had a higher MELD score (p = 0.04) and HE grade (p = 0.002) than Group I. Conclusion Our study demonstrates that the SRCE patterns can be useful in predicting hepatic preservation and the occurrence of cerebral failure in HE.

Probing into the optimum preparation and the chemical durability of Sr0.5Zr2(PO4)3-SmPO4 dual-phase ceramics for nuclear waste forms via in-situ synthesis

  • In this work, Sr0.5Zr2(PO4)3-SmPO4 dual-phase ceramics were prepared via in-situ synthesis process, which is a potential novel nuclear waste form for immobilizing the fission product 90Sr and the trivalent actinide radionuclides in high-level waste (HLW). And the preparation technology, microstructure and chemical durability of Sr0.5Zr2(PO4)3-SmPO4 dual-phase ceramics were systematically investigated. It was confirmed that the optimum microwave-sintering temperature (1050 ℃) and heat preservation time (1.5 h) is estimated by Archimedes method. Besides, the as-prepared samples that were consisted of strontium zirconium phosphate (SrZP) and monazite showed the remarkable densification, in which the two crystalline phases were intermixed well with each other. Meanwhile, the formation and evolution of microstructure was also consistent with the variational rule of Sr0.5Zr2(PO4)3/SmPO4, indicating that there was not mutual reaction during the in-situ synthesis process. The PCT and MCC-1 experimental results demonstrated that the elemental normalized leaching rates of tested samples are all at a low level (LRSr ~10-4 g·m-2·d-1, LRZr ~10-8-10-6 g·m-2·d-1, LRSm ~10-7-10-5 g·m-2·d-1 and LRP ~10-4 g·m-2·d-1). It is indicated that Sr0.5Zr2(PO4)3-SmPO4 dual-phase ceramics possesses excellent chemical durability for HLW disposal.

Comparative Study on the Viability of Frozen-thawed Primordial Germ Cells using Vitrification in Chicken Breed (초자화 동결법을 이용한 닭 품종간의 원시생식세포 동결성적의 비교)

  • This study was conducted to establish the method for preserving PGCs that enables long-term storage in liquid nitrogen for developmental engineering or preservation of species. The purpose of this study is to clarify the effects of freeze-thaw treatment on viability of PGCs in chickens. PGCs were collected separately from a germinal gonad of an early embryo of 5.5~6 day (stage 28) of Isa brown, Korean Oge (KO), White Leghorn and Commercial breeds. PGCs separated from a germinal gonad of an early embryo of 5.5~6 day (stage 28) are suspended in a freezing medium containing a freezing and protecting agents (e.g. dimethyl sulfoxide (DMSO), ethylene glycol (EG) and glycerol). The PGCs were then purified using magnetic activated cell sorting (MACS) method. The viability of PGCs after thawing was $87.4{\pm}0.4%$ and $89.4{\pm}0.2%$ with the 10% EG treatments with no significant difference between the Isa brown and Commercial breeds. The viability of PGCs after freeze- thawing was significantly higher for Isa brown ($87.4{\pm}0.4%$) and Commercial breeds ($89.4{\pm}0.2%$) than Korean Oge (KO) ($77.6{\pm}1.1%$) and White Leghorn ($76.2{\pm}0.9%$)(p<0.05) using 10% EG cryoprotectant. This study established a method for pre- serving chicken PGCs that enables systematic storage and labeling of cryopreserved PGCs in liquid ($LN_2$) at agermplasm repository and ease of entry into a data base. In the future, the importance for this new technology is that poultry lines can be conserved while work is being conducted on improving the production of germline chimeras.

A Study on the Structure and Function of the Underground Storage Facility in Baekje (백제 지하저장시설(地下貯藏施設)의 구조와 기능에 대한 검토)

  • Increasing discovery cases of underground storage facilities made of earth, wood, or stone are being reported from the recent excavation survey of the Baekje relics. Accordingly, the purpose of this study is to examine the structure and function of the underground storage facilities of Baekje following a classification made by the type and building method as follows: plask shape, wooden box shape, and stone box shape. The plask shape storage is the most representative underground storage of Baekje that has been found in numerous relics more than 600 sets around Hangang(Han River) and Geumgang(Geum River) from the Hansung period to Sabi period in Baekje Dynasty. It is a historical artefact as a part of the unique storage culture of Baekje around Hangang and Geumgang from the 3rd to 7th Century. Considering its structure and the example of Chinese one, it might had been used for a long-term storage of grains and various other items including earth wares. The storage facility in wooden box shape and stone box shape are found mostly in the relics Of Sabi period. Thus it might had taken some functions of the storage in traditional pouch shape which had decreased after the 6th Century. In particular, the wooden box shape and stone box shape storage required enormous labor force to build owing to their structure and building method. Thus, they were considered to had been used for official purposes in province fortress and citadel artefact. The wooden box shape storage facility is classified into flat rectangular type and square type based on the structure, and into Gagu type(架構式) and Juheol type(柱穴式) based on the building method. It might had been decided according to the geography and geological feature of the place where the storage was to be built. Considering the examples of Gwanbuk-ri relics and Weolpyong-dong relics, the wooden box shape storage facility might had been used for various items depending on the needs, including foods such as fruits and essential provisions at the military base. Considering the long-term food storage, the examples in Japan, and the functional characteristics of the underground storage facility, there is a possibility that the wooden and stone box shape storage facilities had been built so as to safely store important items in case of fire. This study is only a rudimentary examination for the storage facility in Baekje. Thus further studies are to be made specifically and comprehensively on the comparison with other regions, distribution pattern, discovered relics and artefacts, and functions.

Studies on the Virus in Silkworm Bombyx mori L. -Resistance to Virus Disease- (가잠 Virus에 관한 연구 -저항성에 관한 기초조사-)

  • 1. Objectives and Importance. Many silkworms have been damaged by nuclear polyhedrosis virus diseases thoughout the country every year causing a decease in cocoon production by approximately ten per cent per year. The damage caused by the infections virus has occured in spite of complete disinfection. In this respect, it is well known it is impossible, at the present time, to protect the silkworm from these virus infections through chemical and physical control methods. Therefore, this author has attempted to solve this urgent problem from the view point of heredity and breeding, discovering the different resistances and heritabilities among 120 stains collected from throughout the country, and selecting the ones with highest resistance for the basic materials in the silkworm breeding. 2. Results of work 1) The strains with strong resistance to the nuclear polyhedrosis virus diseases are N$_4$, N$\sub$15/, N$\sub$48/, C$\sub$55/ and Em. the log ED$\sub$50/ values of them vary between 0.799 and 1.611. The susceptible strains are N$\sub$20/, C$\sub$62/, N$\sub$76/, N$\sub$79/ and C$\sub$108/, the log ED$\sub$50/ values of them vary between 5.159 and 7.258. (Reference Table 4) The Japanese strain with a log ED$\sub$50/, value of 3.770 is the strongest, followed by the Chinese strain with a log ED$\sub$50/ value of 3.564. The weakest is the European strain with a log ED$\sub$50/ value of 3.3381. The direction coefficient of the regression equation of the susceptibility varies between 0.1 and 0.6, the uniformness of the resistance of the preserved strains of this country is comparatively low. The hereditary henomena of the resistance of each strain and the conerete method of its application for silkworm breeding main the subjects for later studies. 2) The content of water and ash in silkworm has not been correlated with the capability for resistance to the virus diseases(Reference. Table 8). but it is very significantly correlated with mortality rate (in common reaning). In the case of the silkworms which have just completed the fourth moulting the content of water and ash is not related to the mortality rate. In the case of the silkworms which have just completed the third moulting, however, the water( +0.326) and ash (+0.362) registered a high significance. The ash content in the first ($\div$0.520) and second ($\div$0.386) moults is highly significant but water content in both cases is not significant (Reference Table 7). 3) The No. 205 strain proved to be the best in character among the various F$_1$ hybrids. No. 204 was very good in strength but a little lower in cocoon character than the control. No. 212 was a little low in coccon character and mortality was average, but the cocoon harvest was the best among all the varieties offered for (Reference Table 9). 4), In short, the above mentioned strains which are known to have strong resistance to the virus disease are expected to provide basic data for breeding strong varieties. The Effect of the Histidine-Tryptophan-Ketoglutarate (HTK) Solution on Myocardial Protection in Isolated Rat Heart (흰쥐의 적출심장에서 HTK 용액의 심근보호 효과)

  • Background: The Histidine-Tryptophan-Ketoglutarate (HTK) solution has been shown to provide the excellent myocardial protection as a cardioplegia. The HTK solution has relatively low potassium as an arresting agent of myocardium, and low sodium content, and high. concentration of histidine biological buffer which confer a buffering capacity superior to that of blood.. Since HTK solution has an excellent myocardial protective ability, it is reported to protect myocardium from ischemia for a considerable time (120 minutes) with the single infusion of HTK solution as a cardioplegia. The purpose of this study is to evaluate the cardioprotective effect of HTK solution on myocardium when the ischemia is. exceeding 120 minutes at two different temperature (10 to 12$^{\circ}C$, 22 to 24$^{\circ}C$) using the Langendorff apparatus, Material and Method: Hearts from Sprague-Dawley rat, weighing 300 to 340 g, were perfused with Krebs-Henseleit solution at a perfusion pressure of 100 cm $H_2O$. After the stabilization, the heart rate, left ventricular developed pressure (LVDP), and coronary flow were measured. Single dose of HTK solution was infused into the ascending aorta of isolated rat heart and hearts were preserved at four different conditions. In group 1 (n=10), hearts were preserved at deep hypothermia (10∼12$^{\circ}C$) for 2 hours, in group 2 (n=10), hearts were preserved at moderate hypothermia (22∼24$^{\circ}C$) for 2 hours, in group 3 (n=10), hearts were preserved at deep hypothermia for 3 hours, and in group 4 (n=10), hearts were preserved at moderate hypothermia for 3 hours. After the completion of the preservation, the heart rate, left ventricular developed pressure, and coronary flow were measured at 15 minutes, 30 minutes, and 45 minutes after the initiation of reperfusion to assess the cardiac function. Biopsies were also done and mitochondrial scores were counted in two cases of each group for ultrastructural assessment. Result: The present study showed that the change of heart rate was not different between group 1 and group 2, and group 1 and group 3. The heart rate was significantly decreased at 15 minutes in group 4 compared to that of group 1 (p<0.05 by ANCOVA). The heart rate was recovered at 30 minutes and 45 minutes in group 4 with no significant difference compared to that of group 1. The decrease of LVDP was significant at 15 minutes, 30 minutes and 45 minutes in group 4 compared to that of group 1 (p < 0.001 by ANCOVA). Coronary flow was significantly decreased at 15 minutes, 30 minutes, and 45 minutes in group 4 compared to that of group 1 (p < 0.001 by ANCOVA). In ultrastructural assessment, the mean myocardial mitochondrial scores in group 1, group 2, group 3, and group 4 were 1.02$\pm$0.29, 1.52$\pm$0.26, 1.56$\pm$0.45, 2.22$\pm$0.44 respectively. Conclusion: The HTK solution provided excellent myocardial protection regardless of myocardial temperature for 2 hours. But, when ischemic time exceeded 2 hours, the myocardial hemodynamic function and ultrastructural changes were significantly deteriorated at moderate hypotherma (22∼ 24$^{\circ}C$). This indicates that it is recommended to decrease myocardial temperature when myocardial ischemic time exceeds 2 hours with single infusion of HTK solution as a cardioplegia.

Bladder Preserving Treatment in Patients with Muscle Invasive Bladder Cancer (근침윤성 방광암 환자의 방광 보존적 치료 결과)

  • [ $\underline{Purpose}$ ]: This study analyzed the tumor response, overall survival, progression free survival and related prognostic factors in patients with muscle invasive bladder cancer subjected to bladder preserving treatment. $\underline{Materials\;and\;Methods}$: Between August 1995 and June 2004, 37 patients with muscle invasive (transitional cell carcinoma, clinically stage T2-4) bladder cancer were enrolled for the treatment protocol of bladder preservation. There were 33 males and 4 females, and the median age was 67 years (range $38{\sim}86\;years$). Transurethral resection of the bladder (TURB) was performed in 17 patients who underwent complete resection. The median radiation dose administered was 64.8 Gy (range $55.8{\sim}67\;Gy$). The survival rate was calculated by the Kaplan-Meier method. $\underline{Results}$: An evaluation of the response rate was determined by abdomen-pelvic CT and cystoscopy at three months after radiotherapy. A complete response was seen in 17 patients (46%). The survival rate at three years was 54.7%, with 54 months of median survival (range $3{\sim}91$ months). During the study, 17 patients died and 13 patients had died from bladder cancer. The progression free survival rate at three years was 37.2%. There were 24 patients (64.9%) who had disease recurrence: 16 patients (43.2%) had local recurrence, 6 patients (16.2%) had a distant recurrence, and 2 patients (5.4%) had both a local and distant recurrence. The survival rate (p=0.0009) and progression free survival rates (p=0.001) were statistically significant when compared to the response rate after radiotherapy. $\underline{Conclusion}$: The availability of complete TURB and appropriate chemoradiotherapy were important predictors for bladder preservation and survival.

The Method of Selecting Landscape Control Points for Landscape Impact Review of Development Projects (개발사업의 경관영향 검토를 위한 주요 조망점 선정 방법에 관한 연구)

  • The Natural Landscape Rating System was introduced in the amendment of the NATURAL ENVIRONMENT CONSERVATION ACT in 2006. For landscape preservation, the system aims to consider the effects of development projects or plans implemented in a natural landscape on skylines, scenic resources, and view corridors. Currently, a lack of consistency in standards for determining Landscape Control Points (LCP) to assess landscape impact lowers the accuracy and reliability of the assessment results. As the perception of and the impact on a landscape varies, depending on the location of the LCP, it is necessary to establish a reasonable set of criteria to select viewpoints and avoid unreliability in the assessment due to unclear criteria. The intent of this study is to propose an objective and reasonable set of criteria for LCP selection to effectively measure the impact on the landscape from development projects that anticipate a change in the landscape and, ultimately, to suggest basic analysis methods to assess the landscape impact of development projects and to monitor the landscape in the future. Among the development projects affecting natural landscapes, as reported in the statement of the environmental impact assessment, cases of construction of a single building or other small-scale development projects were studied. Four spot development projects were analyzed in depth for their landscape impacts, in order to make recommendations for the LCP selection procedure, which aims to widen the scope of selection according to the direction of viewpoints from the target site. The existing results of analysis based on LCP have limitations because they failed to cover the viewshed of the target buildings when there are topographical changes in the surroundings. As a solution to this problem, a new viewshed analysis method has been proposed, with a focus on the development site and target buildings, rather than viewpoints, as used in past analysis.

Effects of Maturation Duration and Activation Treatments on Activation and Development of Porcine Follicular Oocytes (돼지 난모세포의 단위발생에 있어서 성숙시간과 활성화 처리가 활성화와 발달에 미치는 영향)

  • This study is a part of research that development of effective genetic resources preservation system using the in vitro spermatogenesis, in vitro insemination and culture system. We aimed for establishment of in vitro culture system with in vitro activated porcine oocytes. The porcine oocytes were matured for 48 hours in $TCM199+10\%$ FCS and activated with $7\%$ ethanol. The activated oocytes were cultured for 7 days in $TCM199+10\%$ FCS or $NCSU23+0.4\%$ BSA medium. The activated oocytes were not developed to the blastocyst stage in $TCM199+10\%$ FCS medium. However in $NCSU23+0.4\%$ medium, those were developed to blastocyst with $3\%$ of treated oocytes. We extended maturation duration of porcine follicular oocytes fur 48, 52, 56, 60, 64, 68, and 72 hours and activated with $7\%$ ethanol and cultured using $NCSU23+0.4\%$ BSA medium. The six percents of activated oocytes were developed to blastocyst in 48 hours and $10\%$ in 52 hours with comparatively low rates suggested to be not fully activated by regenerated MPF. Maturation durations from 56 hours to 68 hours supported to develop upto $11.9\~18.3\%$ of blastocysts. However the developmental rate was declined to $7.2\%$ at 72 hours of maturation duration because of cytoplasmic deterioration. The assumed time window for activation will be $56\~68$ hours of maturation duration. When the matured oocytes were activated with electric pulse of 1, 1.2, 1.4, 1.6, 1.8 and 2.0kV/cm for $80{\mu}s$, although appling the electric current once was not enough for activation, appling twice with 1.6kV/cm for $80{\mu}s$ was shown the highest developmental rate with $11.3\%$. When those were compared with activating methods, $15.7%$ of blastocyst rate was obtained in the $7\%$ ethanol. That was higher than those in electric pulse with $9.5\%$ and calcium ionophore method with $5.8\%$. In this experimental condition, the $7\%$ ethanol treatment was the most effective method for activating porcine oocytes.

Distribution and remediation design of heavy metal contamination in farm-land soils and river deposits in the vicinity of the Goro abandoned mine (고로폐광산 주변 농경지 토양 및 하천 퇴적토의 중금속 오염 분포 및 복원 대책 설계)

  • River deposits and farmland soils were analyzed to investigate the pollution level of heavy metals in the vicinity of the Goro abandoned Zn-mine. Surface (0-40 cm) and subsurface (40-100 cm) soils were collected around a main river located at the lower part of the Goro mine, and analyzed by ICP-MS for Cd, Cu, Pb, Zn and Cr after 0. 1N HCI extraction and by AAS for As after IN HCI extraction. Concentrations of cadmium and lead at the surface river deposits close to the mine were over the Soil Pollution Warning Limit (SPWL), and 43% of sample sites (6 of 14 samples) were over SPWL for As suggesting that river deposits were broadly contaminated by arsenic. Results from farmland soil analysis showed that surface soils were contaminated by heavy metals, while only arsenic was over SPWL at 50% of sampling sites. Main pollution mechanism around the Goro mine was the discharge of mine tailing and waste rocks from the storage site to the river and to adjacent farmland during flood season. Pollution Grades for sample locations were prescribed by the Law of Soil Environmental Preservation, suggesting that the pollution level of heavy metals around the Goro mine was serious, and the remediation operation fur arsenic and the isolation of mine tailing and waste rocks from river and farmland should be activated to protect further contamination. The area needed to clean up was estimated from pollution distribution data and the remediation methods such as a soil washing method and a soil improvement method were considered as the further remediation operation for arsenic contaminated soils and river deposits around the Goro abandoned mine.
