The Movement of Boron Compound by Infusion Method and Combination of Injection and Bandage-Wrapping

  • The existing preservation methods are difficult to be applied to a large dimension log which is needed for making traditional wooden ship 'Jalur' in Riau Province. Novel techniques to provide the use of readily available species to replace traditional species alternative were investigated. These included infusion and a combination of injection and bandage-wrapping methods for preserving living trees of Balam (Macaranga conifera (Rchb.f. & Zoll.) Müll.Arg.) and Bintangor (Calophyllum soulattri Burm.f.). Water-based boron compounds were applied as wood preservatives. In total, 18 discs from the bottom, middle, and top of four trees and two controls were used. Trees undergoing treatment were also used to see how wood anatomical structure might affect the boron penetration. The overall aim was to identify the best method for use in Jalur manufacturing. The results showed that in infused Balam tree where the hose position for the preservative intake was deep (10-15 cm from the bark), no boron compound was observed in the outer sapwood. Combination of injection and bandage-wrapping method gave higher percentage of boron penetration at bottom and middle of Balam tree. However, infused Bintangor showed 100% boron penetration. The larger vessel diameter, the absence of tyloses, and the simple perforation plates in Bintangor wood were likely to have contributed to the higher penetration of boron. The combination of bandage-wrapping and infusion, or alternatively by infusing the living trees close to the bark, and at as low as position in the stem gives better protection when treatments are applied to living trees.

Exper mental Studies of Fowl Pox Vaccine (계두예방약(鷄痘豫防藥)의 실험적연구(實驗的硏究))

  • This experiment was conducted on the fowl pox embryo vaccine for the production immunity, and stability, using an attenuated fowl pox virus (Nakano strin). Burnet's window method was applied, that is, 0.1 ml. of seed virus was inoculated on the chorioallantoic membrane of 12-day old chicken embryos, and incubated for 5 to 6 day, and then the result were read. Four kinds of suspensions of different embyo tissue were prepared and tested for the infectivity in chickens. Finally the suspension of chorioallantoic membrane was used as the vaccine throughout the experiment. Results obtained in this experiment are summarized as follows: (1) Of embryo tissue infected with the vaccine virus, chorioallantoic membrane had the highest virus titer of $10^{-5.4}$ $EID_{50}$, and albumen the lowest titer of $10^{-0.7}$ $EID_{50}$. (2) Suspensions of infected whole embryo with or without saline, and de-embryonated whole egg had about the same virus titer of $10^{-4.4}$ $EID_{50}$, whereas the chorioallantoic membrane had $10^{-5.7}$ EID 50 or higher. The virus titer droped one log from $EID_{50}$ when inoculated into chickens. Takes were observed 35.6% of 500 chickens by stick method and 89% of 500 chickens by brush method. (3) The chorioallantoic membrane conferred almost perfect immunity for chickens by 10 days after vaccination. (4) Satisfactory immunity was observed in the chickens when eruption in a single follicle. (5) Eight of 10 vaccinated chickens revealed durable immunity for 307 days following vaccination. (6) The vacuum-dried vaccine maintained its infectiviy for 899 days at $5^{\circ}C$ or below and maintained the vius titer of $10^{-3.6}$ $EID_{50}$. On the other hand, non-desiccated wet vaccine maintained the titer of $10^{-3.0}$ $EID_{50}$ for 50 days of preservation period at $5^{\circ}$. However, in 50% glycerin-saline the infectivtiy of the same wet vaccine dropped to $10^{-1.5}$ $EID_{50}$ (7) The vartation of virus titer of the vaccine before and after desiccation was $10^{-0.5}$ $EID_{50}$ on the average. (8) As suspending media, 0.85 per cent saline and distilled water showed nearly the same effect on the infectivity of the vaccine by retaining the titer $10^{-3.0}$ $EID_{50}$ after 50 days of preservation both at $5^{\circ}C$ and $20^{\circ}C$, while 50 percent cent glycerine-saline dropped the titer to $10^{-2.5}$ EID and $10^{-1.5}$ $EID_{50}$ respectively at $5^{\circ}C$ and $2^{\circ}C$ after the same period.

Effects of Packaging Method on the Quality of Blanched Namul during Storage (포장방법에 따른 데침나물의 저장중 품질변화)

  • The objective of this study was to investigate the storage effects of the packaging method of blanched namul(Gosari, Torandae, Chwinamul and Siraegi). The samples were packaged with three packaging types (Vinyl packaging, sealing packaging and vacuum packaging) and were stored for 10 days at $10^{\circ}C$. The quality characteristics were evaluated via a microbiological test, hardness, pH and flavor patterns analysis. The pH values of the samples were not affected by packaging method. The total aerobic and coliform plate counts were high, in the order of vacuum packaging < sealing packaging < vinyl packaging. Vacuum packaging resulted in the highest hardness value. The flavor patterns of blanched namul by packaging type were analysed with electronic nose system equipped with 12 metal-oxide sensors, and the storage shelf life of namul was evaluated by measuring the change in volatile production. As a result, it was shown that namul in vacuum packaging had few volatile production changes with higher storage time.

Physico-Chemical Changes in Pork Bellies with Different Cooking Methods (조리방법에 따른 삼겹살의 물리화학적 특성 변화)

  • This study was carried out to suggest an effective cooking method for pork bellies to decrease intake of animal fat. The physico-chemical characteristics of pork bellies cooked by different methods (boiling, steaming, baking and frying) were investigated. The moisture contents of cooked meats decreased but crude lipid contents increased. The cooking losses, moisture drain rates, and lipid drain rates were high after frying and boiling. The pH values increased markedly with boiling and both the acid value and the refraction index of the fat significantly increased with frying. The hardness, gumminess, and chewiness of the meat increased considerably with boiling, but decreased notably after frying. The springiness decreased very much with boiling and cohesiveness greatly increased with steaming. The CIE $L^*$ (lightness) value increased notably with steaming but decreased markedly with frying. The CIE $a^*$ (redness) value decreased markedly with all cooking methods, especially boiling, and the CIE $b^*$ (yellowness) value decreased with both boiling and steaming but increased with both baking and frying. The fatty acids of fat from the raw pork bellies were primarily oleic acid (42.4%), palmitic acid (23.9%), and linoleic acid (16.1%). The ratio of total monounsaturated fatty acids to total saturated fatty acids was 1.190 and the ratio of total polyunsaturated fatty acids to total saturated fatty acids was 0.446. In addition, the composition of fatty acids was not significantly changed with any cooking method except frying. Therefore, boiling is the effective cooking method for pork bellies to decrease intake of animal fats.

Sensory properties of oriental melon (Cucumis melo var. makuwa) byquantitative descriptive analysis and consumer test (묘사분석 및 소비자 조사에 의한 참외의 관능특성)

  • This study was carried out to determine the descriptive sensory profile of oriental melon. The sensory profile of oriental melons (cultivated in Seonju, Kyungsangbuk-do) were used quantitative descriptive analyses and twelve trained panel developed the total forty sensory attributes related appearance, aroma, texture etc. Six appearances attributes and two aromas attributes were derived from whole oriental melon with skin. Five aromas, six tastes and six textures were derived from the mesocarp of oriental melon. And also, oriental melons were analyzed for pH, titratable acidity, solid soluble contents and weight. The weight, titratable acidity and soluble solid content of oriental melons showed significant differences according to samples. And also, in the sensory attributes of these samples by trained panel and consumer test, there were significant differences between organic cultivated samples and conventional cultivated samples. Based on principal component analysis of sensory attributes data by trained panel and consumer, oriental melons were primary separated along the first principal component, which accounted for 27.73% (trained panel test) and 19.82% (consumer test) of total variance (trained panel test; 58.36%, consumer test; 46.18%) between the samples farm, cultivation method, total acidity, sweet aroma, bitterness, etc. Generally, whole oriental melon with skin showed significant differences in the sensory attributes according to cultivation method and farms. But, the mesocarp and endocarp part showed not certainly differences in the sensory attributes between organic cultivated samples except conventional cultivated samples.

Biological activities in Aronia melanocarpa depending on drying methods (건조방법에 따른 아로니아의 생리활성)

  • To investigate biological activities in Aronia melanocarpa various drying methods were employed such as vacuum freeze drying, hot air drying and cold air drying. DPPH radical scavenging activity and ABTS radical scavenging activity of vacuum freeze dried Aronia melanocarpa was higher than hot and cold air dried Aronia melanocarpa. Vacuum freeze drying method showed the greatest contents of total phenol (15.34 g GAE/100 g), flavonoid (3.10 g GE/100 g) and tannin (2.46 g TE/100 g). Total anthocyanin content decreased to 163.52 mg C3G/100 g and 50.15 mg C3G/100 g for hot and cold air drying, respectively. Vacuum freeze-dried method increased the total anthocyanin content (743.09 mg C3G/100 g) when compared with fresh Aronia melanocarpa (163.52 mg C3G/100 g). Total proanthocyanidin content of vacuum freeze dried Aronia melanocarpa has increased to 6.21 g CE/100 g more than eight times compared with fresh Aronia melanocarpa (0.71 g CE/100 g). Chlorogenic acid and neochlorogenic acid content of vacuum freeze dried Aronia melanocarpa were higher than hot air dried and cold air dried Aronia melanocarpa, increasing about three times compared with fresh Aronia melanocarpa. These results suggested that vacuum freeze drying is optimal drying method to enhance biological activities in Aronia melanocarpa.

Analysis of Antimicrobial Substance isolated from Scutellariae Radix Extract using LC-MS (LC-MS를 이용한 황금추출물의 항균물질 검색)

  • It was carried out for development of natural antimicrobial on Scutellariae Radix by measuring the general nutritional composition and mineral contents. This study analyzed it with LC-MS to identify antimicrobial activity substance in the Scutellariae Radix extract, its content was analyzed using HPLC. The searching substance was examined antimicrobial activity by disk method. The approximate composition of Scutellariae Radix was $8.93\%$ of moisture, $4.13\%$ of crude ash, $15.30\%$ of crude protein, $3.71\%$ of crude fat, $67.93\%$ of carbohydrate, respectively. Scutellariae Radix was contained high contents of minerals such as K(12,050 mg/kg),. Mg(4,295 mg/kg), remarkably. It was identified that molecular weight 445' peak was baicalin, molecular weight 283' peak was wogonin and molecular weight 269' peak was baicalein on Scutellariae Radix extract Baicalin showed remarkable antimicrobial activity against V. parahaemolyticus and L monocytogenes when examined by disk method, which was estimated antimicrobial activity substance in the Scutellariae Radix extract. The content of baicalin was $0.75\%$(with D.W.) and $0.82\%$(with methanol) in the Scutellariae Radix extract using HPLC.

2D Industrial Image Registration Method for the Detection of Defects (결함 검출을 위한 2차원 산업 영상 정합 기법)

  • In this paper, we propose 2D industrial image registration method for the detection of defects. Proposed method performs preprocessing to smooth the original image with the preservation of the edge for the robust registration against general noise. Then, x-direction gradient magnitude image and corresponding binary image are generated. Density analysis around neighborhood regions per pixel are performed to generate feature image for preventing mis-registration due to moire-like patterns, which frequently happen in industrial images. Finally, 2D image registration based on phase correlation between feature images is performed to calculate translational parameters to align two images rapidly and optimally. Experimental results showed that the registration accuracy of proposed method for the real industrial images was 100% and our method was about twenty times faster than the previous method. Our fast and accurate method could be used for the real industrial applications.

EVALUATION OF THE VIABILITY OF PERIODONTAL LIGAMENT CELL IN RAT TEETH USING SLOW CRYOPRESERVATION METHOD WITH MAGNETIC FIELD (자기장 저속 냉동보관법을 이용한 쥐 치아 치주인대세포의 활성도 검사)

  • The purpose of this study was to evaluate the viability of periodontal ligament cell in rat teeth using slow cryopreservation method with magnetic field through MTT assay and TUNEL test. For each group, 12 teeth of 4 weeks old white female Sprague-Dawley rat were used for MTT assay, and 6 teeth in TUNEL test. The Maxillary left and right, first and second molars were extracted as atraumatically as possible under tiletamine anesthesia. The experimental groups were group1 (immediately extraction), group 2 (cold preservation at 4$^{\circ}C$ for 1 week), group 3 (rapid cryopreservation in liquid nitrogen), group 4 (slow cryopreservation with magnetic field of 1 G), and group 5 (slow cryopreservation). F medium was used as preservation medium and 10% DMSO as cryoprotectant. After preservation and thawing, the MTT assay and TUNEL test were processed. One way ANOVA and Scheffe method were performed at the 95% level of confidence. The value of optical density obtained after MTT analysis was divided by the value of eosin staining for tissue volume standardization. In both MTT assay and TUNEL test, it had showed no significant difference among group 3, 4, and 5. And group 3 had showed higher viability of periodontal ligament cell than group 2. From this study, slow cryopreservation method with magnetic field can be used as one of cryopreservation methods.

Grid-based Semantic Cloaking Method for Continuous Moving Object Anonymization (이동 객체 정보 보호를 위한 그리드 기반 시멘틱 클로킹 기법)

  • Location privacy has been a serious concern for mobile users who use location-based services to acquire geographical location continuously. Spatial cloaking technique is a well-known privacy preserving method, which blurs an exact user location into a cloaked area to meet privacy requirements. However, cloaking for continuous moving object suffers from cloaked area size problem as it is unlikely for all objects travel in the same direction. In this paper, we propose a grid-based privacy preservation method with an improved Earth Mover's Distance(EMD) metric weight update scheme for semantic cloaking. We also define a representative cloaking area which protects continuous location privacy for moving users. Experimental implementation and evaluation exhibit that our proposed method renders good efficiency and scalability in cloaking processing time and area size control. We also show that our proposed method outperforms the existing method by successfully protects location privacy of continuous moving objects against various adversaries.
