Studies on Setting up of Radioimmunoassay System of Thyroid Stimulating Hormone (갑상선자극(甲狀腺刺戟)호르몬의 방사면역측정법(放射免疫測定法) 확립(確立)에 관한 연구(硏究))

  • Various TSH RIA kit components were prepared. Conditions for $^{125}I$ labelling of h-TSH were optimized by diminishing the amount of chloramine-T, ertending reaction time and lowering reaction temperature. Yield, specific activity, and immunological activity could be maintained moderately under such mild reaction conditions. The mixture of polyethyleneglycol(PEG) and second antibody worked effectively as a B/F separation agent. Even though the mixture was made with more diluted PEG and second antibody than those of using the sole component separately, the tine required for the B/F separation was shorter in case of using the mixture. The sequential saturation technique was efficient than those of applying ordinary equilibrium saturation technique in assay sensitivity and assay precision points of view.

Blood Eosinophil Counts in Chronic Obstructive Pulmonary Disease: A Biomarker of Inhaled Corticosteroid Effects

  • Blood eosinophil counts have emerged as a chronic obstructive pulmonary disease (COPD) biomarker that predict the effects of inhaled corticosteroids (ICS) in clinical practice. Post-hoc and prospective analysis of randomized control trials have shown that higher blood eosinophil counts at the start of the study predict a greater response to ICS. COPD patients with frequent exacerbations (2 or more moderate exacerbations/yr) or a history of hospitalization have a greater response to ICS. Ex-smokers also appear to have a greater ICS response. Blood eosinophil counts can be combined with clinical information such as exacerbation history and smoking status to enable a precision medicine approach to the use of ICS. Higher blood eosinophil counts are associated with increased eosinophilic lung inflammation, and other biological features that may contribute to the increased ICS response observed. Emerging data indicates that lower blood eosinophil counts are associated with an increased risk of bacterial infection, suggesting complex relationships between eosinophils, ICS response, and the airway microbiome.

HisCoM-GGI: Software for Hierarchical Structural Component Analysis of Gene-Gene Interactions

  • Gene-gene interaction (GGI) analysis is known to play an important role in explaining missing heritability. Many previous studies have already proposed software to analyze GGI, but most methods focus on a binary phenotype in a case-control design. In this study, we developed "Hierarchical structural CoMponent analysis of Gene-Gene Interactions" (HisCoM-GGI) software for GGI analysis with a continuous phenotype. The HisCoM-GGI method considers hierarchical structural relationships between genes and single nucleotide polymorphisms (SNPs), enabling both gene-level and SNP-level interaction analysis in a single model. Furthermore, this software accepts various types of genomic data and supports data management and multithreading to improve the efficiency of genome-wide association study data analysis. We expect that HisCoM-GGI software will provide advanced accessibility to researchers in genetic interaction studies and a more effective way to understand biological mechanisms of complex diseases.

  • To confirm the accuracy of the prediction of parturition day, the actual parturition days were compared with each day of the prediction of parturition day (n = 80). The accuracy of the prediction of parturition day was 80.0% (64/80) with a precision of ± 2 days from the first day of estrus after the first vaginal discharge, 97.5% (78/80) from the day when plasma progesterone concentrations increase above 4.0 ng/ml, and 72.5% (58/80) from the first day of diestrus, respectively. The accuracy of the prediction of parturition day by plasma progesterone concentration was higher than that by the first day of estrus and diestrus after the first vaginal discharge. These results indicated that the determination of plasma progesterone concentrations at estrus were a useful method for estimating of parturition day and for the reproductive management in pregnant bitches.

Photo-triggered Theranostic Nanoparticles in Cancer Therapy

  • In cancer therapy, it is often desirable to use precision medicine that involves treatments of high specificity. One such treatment is the use of photo-triggered theranostic nanoparticles. These nanoparticles make it possible to visualize and treat tumors specifically in a controlled manner with a single injection. Several novel and powerful photo-triggered theranostic nanoparticles have been developed. These range from small organic dyes, semiconducting and biopolymers, to inorganic nanomaterials such as iron-oxide or gold nanoparticles, carbon nanotubes, and upconversion nanoparticles. Using photo-triggered theranostic nanoparticles and localized irradiation, complete tumor ablation can be achieved without causing significant toxicity to normal tissue. Given the great advances and promising future of theranostic nanoparticles, this review highlights the progress that has been made in the past couple of years, the current challenges faced and offers a future perspective.

Determination of Liraglutide in Rat Plasma Using Selective Liquid Chromatography-Tandem Mass Spectrometry

  • Liraglutide is a medication prescribed for the management of type 2 diabetes and chronic obesity. A simple, sensitive, and selective liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the quantitative analysis of liraglutide in rat plasma. After a simple protein precipitation step, liraglutide was chromatographically separated using the ACQUITY Premier Peptide BEH C18 Column with mobile phases comprising 50% acetonitrile and 50% methanol, and water with 0.3% FA. Positive ion electrospray ionization in multiple reaction monitoring mode was used to achieve detection. Good linearity was observed in the 5-600 ng/mL concentration range (R2 > 0.99). Liraglutide had intra- and inter-day precision values of 2.13%-9.86% and 4.14%-8.36%, respectively. The accuracy ranged from -2.36% to 2.58%. The recovery and matrix effect were within acceptable limits. This selective LC-MS/MS method was used to study the pharmacokinetic properties of liraglutide after subcutaneous administration in rats.

  • Prediction is a significant topic in clinical research. The development and validation of a prediction model has been increasingly published in clinical research. In this review, we investigated analytical methods and validation schemes for a clinical prediction model used in digestive cancer research. Deep learning and logistic regression, with split-sample validation as an internal or external validation, were the most commonly used methods. Furthermore, we briefly introduced and summarized the advantages and disadvantages of each method. Finally, we discussed several points to consider when conducting prediction model studies.

Evaluation of HbA1c Levels Via the Latex Immunoturbidimetric Method by Using Chemistry Autoanalyzer (자동화학분석기에서의 라텍스 면역비탁법의 Autolab HbA1c 평가)

  • Background: Measurement of HbA1c levels is widely used to diagnose diabetes mellitus and to evaluate and monitor plasma-glucose concentrations over 6-8 weeks. In this study, we evaluated the diagnostic performance of the newly developed latex immunoturbidimetric method by using Autolab HbA1c. Methods: We analyzed and compared the diagnostic performance of Autolab HbA1c with that of Toshiba 200FR between April 2009 and July 2009. According to guidelines (EP5-A2, EP6-P, EP9-A2) of the clinical and laboratory standards institute (CLSI), we compared linearity, precision and correlation of Autolab HbA1c with those of G7 (Tosoh Corp., Kyoto, Japan) by using high-performance liquid chromatography (HPLC) method. Results: Data obtained using Autolab HbA1c showed good linearity in mixtures of samples with low (3.1%) and high (15.1%) levels of HbA1c (r2=0.9997). In the analysis of within-run precision of the samples with HbA1c levels of 5.1% and 12.1%, the SDs were 0.04 and 0.06 and covariances of these samples were 0.8% and 0.5%, respectively. In the Deming regression model, the regression equation was as follows: Autolab HbA1c=1.0859×Tosoh HPLC-0.6957. Conclusions: In this study, Autolab HbA1c method showed better performance characteristics than Tosoh G7 did. In reference review, there was no interference of variant hemoglobin. The data acquisition time of Autolab HbA1c was lower than that of Tosoh G7. The advantages of Autolab HbA1c are that it can be used as an autoanlyzer in routine chemical analysis, it does not require pre-analytical treatment, and the samples are automatically treated with distilled water for hemolysis.

  • This study investigated changes of mechanical characteristics in the trabecular and cortical bone of the lumbar vertebrae of the ovariectomised(OVX) rat. In previous researches, there were many studies for morphology of osteoporotic bones based on Micro-CT (Micro - Computed Tomography). However, there were few studies for detecting and tracking changes of mechanical characteristics in the trabecular bone and cortical bone of the lumbar vertebrae for the OVX rat. For this study, one female Sprague-Dawley rat was used: an OVX rat. The 4th lumbar of the OVX rat was utilized as a specimen. Detecting and tracking changes of mechanical characteristics could be investigated in the trabecular and cortical bone of the lumbar vertebrae for an OVX rat using In-vivo Micro-CT. An OVX rat was scanned at week 0 (just before surgery), at week 4, and week 8 after surgery. Micro finite element(${\mu}\;FE$) analysis was used to investigate mechanical characteristics in the trabecular and cortical bone of the lumbar vertebrae for an OVX rat.

Development of Bio-Check Unit and Health Index for Measuring Health Degree through Noninvasive Examination (비침습적 검사를 통한 건강 정도 측정을 위한 바이오체크 유닛과 건강지수의 개발)

  • A bio-check unit and health index were developed to provide information on personal health state with easily available noninvasive measurements and surveys. Four health indices were defined such as cardiovascular index, stress index, obesity index, and management index. Methods were developed to calculate health index scores from measured physiological signals and answer of survey questions. In order to evaluate effectiveness of the health indices, a clinical trial was conducted for 362 persons who visited general hospital for annual health inspection. The cardiovascular index showed a good correlation coefficient of 0.685 with the cardiovascular health graded by a medical doctor. The stress index showed a good correlation coefficient of 0.638 with the results of stress questionnaires being used in the public health center. Once the health index function is added in the bio-check unit, the unit may provide useful contents for personal health management.
