Changes of DNA fragmentation by Irradiation Doses and Storage in Gamma-irradiated Meats and Poultry (감마선 조사 육류, 가금류에서 저장전과 후의 조사선량에 따른 DNA fragmentation의 변화)

  • The changes in DNA damage were investigated during storage after irradiation. Beef, pork and chicken were irradiated at 1.0, 3.0 and 5.0 kGy and stored for 6 months at $-20^{\circ}C$. The comet assay was applied to the sample muscles at the beginning of irradiation and at the end of storage. Muscles were isolated, sliced, and the suspended cells were embedded in an agarose layer. After lysis of the cells, they were electrophoresed for 2 min. and then stained. DNA fragmentation in tissues caused by irradiation was quantified as tail length and tail moment (tail length ${\times}$ % DNA in tail) by comet image analyzing system. Right after irradiation, the differences in tail length between unirradiated and irradiated muscles were significant(p<0.05) in beef, pork and chicken. With increasing the increasing doses, statistically significant longer extension of the DNA from the nucleus toward anode was observed. Similarly even 6 months after irradiation, all the irradiated muscles significantly showed longer tail length than the unirradiated controls. The results represented as tail moment showed similar tendency to those of tail length, but the latter parameter was more sensitive than the former. These results indicate that the comet assay could be one of the simple methods of detecting irradiated muscles. Moreover, this method suggest that using comet assay, we were able to detect DNA damage differences even after 6 months after irradiation.

Isolation and Characterization of Feather Keratin-Degrading Bacteria and Plant Growth-Promoting Activity of Feather Hydrolysate (우모 케라틴 분해세균의 분리, 특성 및 우모 분해산물의 식물 생육촉진 효과)

  • This study was conducted to isolate and characterize a novel feather-degrading bacterium producing keratinase activity. A strain K9 was isolated from soil at poultry farm and identified as Xanthomonas sp. K9 by phenotypic characters and 16S rRNA gene analysis. The cultural conditions for the keratinase production were 0.3% fructose, 0.1% gelatin, 0.04% $K_2HPO_4$, 0.06% $KH_2PO_4$, 0.05% NaCl and 0.01% $FeSO_4$ with an initial pH 8.0 at $30^{\circ}C$ and 200 rpm. In an optimized medium containing 0.1% chicken feather, production yield of keratinase was approximately 8-fold higher than the yield in basal medium. The strain K9 effectively degraded chicken feather meal (67%) and duck feather (54%), whereas human nail and human hair showed relatively low degradation rates (13-22%). Total free amino acid concentration in the cell-free supernatant was about 25.799 mg/l. Feather hydrolysate produced by the strain K9 stimulated growth of red pepper, indicating Xanthomonas sp. K9 could be not only used to increase the nutritional value of chicken feather but also a potential candidate for the development of natural fertilizer applicable to crop plant soil.

Prolonged Expression of Exogenous GFP Gene in the Porcine Embryos generated by Intracytoplasmic Sperm Injection-Mediated Gene Transfer

  • Understanding the behavior of transgenes introduced into oocyte or embryos is essential for evaluating the methodologies for transgenic animal production. To date, many studies have reported the production of transgenic pig embryos with, however, low efficiency in environment of blastocyst production. The aim of present study was to determine the expression and duration of transgene transferred by intracytoplasmic sperm injection-mediated gene transfer (ICSI-MGT). Embryos obtained from the ICSI-MGT procedure were analysed for the expression of GFP and then for the transmission of the transgene. Briefly, fresh spermatozoa were bound to exogenous DNA after treatment by Triton X-100 and Lipofectin. When ICSI-MGT was performed using sperm heads with tails removed, the yield of blastocyst (25.3%), treated with Lipofectin (18.8%) and Triton X-100 (19.2%) were observed. Treatments of Lipofectin or Triton X-100 did not further improve the rates of blastocysts. Moreover, the apoptosis rates of embryos were obtained from the control and LIpofectin groups (8.7%, 9.7%, respectively), but were significantly higher in the Triton X-100 group (13.0%). Our results demonstrated that ICSI-MGT caused minimal damage to oocytes that could develop to full term. Moreover, the embryos derived by ICSI-MGT have shown prolonged exogenous DNA expression during preimplantation stage in vivo. However, more efforts will be required to improve the procedures of both sperm treatments cause of high frequency of mosaicisms.

In-transit development of color abnormalities in turkey breast meat during winter season

  • Background: The poultry industry suffers losses from problems as pale, soft and exudative (PSE), and dark, firm and dry (DFD) meat can develop in meat as a result of short- and long-term stress, respectively. These abnormalities are impacted by pre-slaughter animal welfare. Methods: This work evaluated the effects of open vehicle container microclimate, throughout the $38{\pm}10km$ journey from the farm to the slaughterhouse, on commercially turkey transported during the Brazilian winter season. The journey was initiated immediately after water bath in truck fitted with portable Kestrel anemometers to measure air ventilation, relative humidity, temperature and ventilation. Results: The inferior compartments of the middle and rear truck regions showed highest temperature and relative humidity, and lower air ventilation. In addition, the superior compartments of the front truck regions presented lower temperature and wind chill, and highest air ventilation. The breast meat samples from animals located at the inferior compartments of the middle and rear truck regions and subjected to with water bath (WiB) treatment presented highest DFD-like and had lowest PSE-like meat incidence than those from animals located at other compartments within the container. Lower incidence of PSE-like meat was observed in birds without water bath (WoB). Conclusions: Assessment on turkeys transported under Brazilian southern winter conditions revealed that breast meat quality can be affected by relative humidity, air ventilation, temperature, and transport under subtropical conditions promoting color abnormalities and the formation of simultaneously PSE-like and DFD-like meat.

Household food insecurity, diet quality, and weight status among indigenous women (Mah Meri) in Peninsular Malaysia

  • BACKGROUND/OBJECTIVES: This cross-sectional study assessed household food security status and determined its association with diet quality and weight status among indigenous women from the Mah Meri tribe in Peninsular Malaysia. SUBJECTS/METHODS: The Radimer/Cornell Hunger and Food Insecurity Instrument and the Malaysian Healthy Eating Index (HEI) were used to assess household food security status and diet quality, respectively. Information on socio-demographic characteristics and 24-hour dietary recall data were collected through face-to-face interview, and anthropometric measurements including weight, height, and body mass index (BMI) were obtained from 222 women. RESULTS: Majority of households (82.9%) experienced different levels of food insecurity: 29.3% household food insecurity, 23.4% individual food insecurity, and 30.2% fell into the child hunger group. The food-secure group had significantly fewer children and smaller household sizes than the food-insecure groups (P < 0.05). The mean household income, income per capita, and food expenditure significantly decreased as food insecurity worsened (P < 0.001). The food-secure group had significantly higher Malaysian HEI scores for grains and cereals (P < 0.01), as well as for meat, poultry, and eggs (P < 0.001), than the food-insecure groups. The child-hunger group had significantly higher fat (P < 0.05) and sodium (P < 0.001) scores than the food-secure and household food-insecure groups. Compared to the individual food-insecure and child-hunger groups, multivariate analysis of covariance showed that the food-secure group was significantly associated with a higher Malaysian HEI score while the household food-insecure group was significantly associated with a higher BMI after controlling for age (P < 0.025). CONCLUSIONS: The majority of indigenous households faced food insecurity. Food insecurity at the individual and child levels was associated with lower quality of diet, while food insecurity at the household level was associated with higher body weight. Therefore, a substantial effort by all stakeholders is warranted to improve food insecurity among poorer households. The results suggest a pressing need for nutritional interventions to improve dietary intake among low income households.

Evaluation of Compost Maturity by Physico-chemical Properties and Germination Index of Livestock Manure Compost (축분 퇴비의 이화학적 특성과 발아지수를 이용한 부숙도 평가)

  • This study was conducted to evaluate the humification grade of compost, based on physico-chemical property and phyto-toxicity during the composting with three kinds of livestock manures and saw dust. The ratios of the compost, which was mixed with pig manure(P) and sawdust(S) were 4 : 6(PS-1), 5 : 5(PS-2) and 6 : 4(PS-3); poultry manure(PO) and sawdust(S) were 4 : 6(POS-1), 5 : 5(POS-2) and 6 : 4(POS-3); cow manure(C) and sawdust(S) were 4 : 6(CS-1), 5 : 5(CS-2) and 6 : 4(CS-3) by volume to volume, and they were decomposed for 60 days. In the result, the temperature in all treatments during composting rapidly increased above $65.4^{\circ}C$, and then gradually decreased to around $40^{\circ}C$. At 60 day, after the treatment, pH 5.9 ~ 8.0 at the incipient stage increased to 7.6 ~ 8.5, and the C/N ratio was 13.8 ~ 21.1 at the final composting stage. Germination Index(G.I.) showed in the range of 75.1 ~ 94.6 in all treatments at day 60. Therefore, it is likely recommended to take the best humification grade when the temperature maintains above $65^{\circ}C$ longer than a day at minimum, in the range of pH 6.5 ~ 8.5 for the final compost, under 20 of C/N ratio, and G.I. above 80. The level of G.I. above 80 should be the mature compost which could be used in the field without gas demage to crops.

Pathological observations on diseased cockerels in rural areas of Bangladesh

  • The poultry farmers of rural area in Bangladesh usually prefer raising cockerel to broiler due to availability, low price of chicks, requirement of less space and feed, and high price of meat and the farmers believe that the cockerels are less susceptible to diseases in comparison to broilers. This study was carried out to observe the pathology of diseased cockerel in 3 farms of rural area in Bangladesh. A total of 974 birds were examined and the diagnoses of different disease/conditions were based on the history, clinical signs, characteristic gross, tissue alterations, clinical pathology and isolation and identification of the pathogenic organisms. The diseases in this study included infectious bursal disease, yolk sac infection, vitamin E deficiency, coccidiosis, and other diseases. The proportionate mortality rate were $7.29\%,\;0.62\%,\;0.72\%,\;0.21\%\;and\;0.10\%$, respectively, which indicated that most of the fatal causes of death were due to infectious bursal disease. Age group of 2-8 week old were the most susceptible to this disease and E coli was suggested as a cause of yolk sac infection. The data also suggested that the coccidiosis in rural areas of Bangladesh has decreased due to awareness of the farmers and routine use of coccidiostates.

Assessment of probiotic potential of Lactobacillus reuteri MD5-2 isolated from ceca of Muscovy ducks

  • Lactic acid bacteria (LAB) are commonly used as probiotics in poultry. The present study employed in vitro and in vivo methods to select and test LAB isolated from Muscovy duck ceca as potential probiotics. In the in vitro study, 50 LAB were isolated from Muscovy duck ceca and tested for growth inhibition against Salmonella (S.) Enteritidis. Eleven isolates strongly inhibited S. Enteritidis and only 1 isolate (MD5-2) showing the strongest inhibition was selected for identification. This isolate was called as Lactobacillus (L.) reuteri MD5-2. For the in vivo investigation, 90 1-day-old Muscovy ducks were randomly assigned into three groups of 30 animals each (group 1, control; group 2, treated with $10^8$ colony-forming unit (CFU) of L. reuteri MD5-2 orally once on day 1; and group 3, treated with $10^8CFU$ of L. reuteri MD5-2 orally once daily from days 1 to 5). The ducks were housed in three large cages and raised for 50 days, after which body weight, duodenal villus height and crypt depth were measured. Both villus height and villus height to crypt depth ratio were significantly greater in group 3 than in groups 1 and 2. In conclusion, further investigation of L. reuteri MD5-2 as a potential probiotic strain is warranted.

Effect of Organic Fertilizer and Mulch Sources on Growth and CO2 Assimilation in MM.106 Apple Trees (유기질 비료와 멀칭자재가 MM.106 사과 대목의 생장과 광합성에 미치는 영향*)

  • The study was conducted to investigate the effects of organic fertilizers and mulches on the growth and $CO_2$ assimilation in MM.106 apple trees. Growth and $CO_2$ assimilation of MM.106 apple trees grown in a greenhouse were affected by the nutrient concentrations and carbon (C) and nitrogen (N) ratio in the raw materials of organic fertilizers and mulches. The optimum C:N ratios, which makes microorganism convert the organic N into the inorganic N, were obtained in the organic fertilizer, poultry litter, green compost, and grass clippings, resulting in increasing single shoot height, SPAD, and $CO_2$ assimilation. The SPAD and $CO_2$ assimilation were affected by the treatments 5 weeks after the treatments, and then the tree growth was affected by the treatments 6 weeks later. The most efficient tree growth and development were observed in the 10 to $15\;mg{\cdot}kg^{-1}$ of the inorganic N in a soil, and the N was strongly related to the tree growth and development.

Comparison of Meat Characteristics between Korean Native Duck and Imported Commercial Duck Raised under Identical Rearing and Feeding Condition

  • This research was conducted to compare the meat characteristics of Korean native duck and imported commercial duck. The Korean native ducks and imported commercial ducks (broiler duck: Grimaud) were raised under identical rearing and feeding conditions for 8 wk and 6 wk, respectively. At the end of the rearing period, ten ducks from each group were slaughtered, and breast and leg meat were obtained 24 h after slaughtering for analyses of meat characteristics. The results showed that the breast of Korean native ducks contained lower moisture and fat, and higher protein and water holding capacity (WHC) than those of imported commercial ducks (p<0.05). The breast of Korean native ducks also had higher CIE $a^*$ and lower CIE $L^*$ (p<0.05). After cooking, the breast meat of Korean native ducks had higher shear-force, sensory attributes of texture, taste and overall likeness (p<0.05). Also, the breast meat of Korean native ducks contained a higher percentage of palmitic acid ($C_{16:0}$) and arachidonic acid ($C_{20:4}$) (p<0.05) than those of imported commercial ducks. Furthermore, the leg meat of Korean native ducks contained higher percentages of total unsaturated fatty acid and lower percentages of total saturated fatty acid (p<0.05). It is concluded that the meat from Korean native ducks, especially breast meat, had better quality parameters and contained higher amounts of unsaturated fatty acids.
