Transcriptomic Approach for Understanding the Adaptation of Salmonella enterica to Contaminated Produce

  • Salmonellosis is a form of gastroenteritis caused by Salmonella infection. The main transmission route of salmonellosis has been identified as poorly cooked meat and poultry products contaminated with Salmonella. However, in recent years, the number of outbreaks attributed to contaminated raw produce has increased dramatically. To understand how Salmonella adapts to produce, transcriptomic analysis was conducted on Salmonella enterica serovar Virchow exposed to fresh-cut radish greens. Considering the different Salmonella lifestyles in contact with fresh produce, such as motile and sessile lifestyles, total RNA was extracted from planktonic and epiphytic cells separately. Transcriptomic analysis of S. Virchow cells revealed different transcription profiles between lifestyles. During bacterial adaptation to fresh-cut radish greens, planktonic cells were likely to shift toward anaerobic metabolism, exploiting nitrate as an electron acceptor of anaerobic respiration, and utilizing cobalamin as a cofactor for coupled metabolic pathways. Meanwhile, Salmonella cells adhering to plant surfaces showed coordinated upregulation in genes associated with translation and ribosomal biogenesis, indicating dramatic cellular reprogramming in response to environmental changes. In accordance with the extensive translational response, epiphytic cells showed an increase in the transcription of genes that are important for bacterial motility, nucleotide transporter/metabolism, cell envelope biogenesis, and defense mechanisms. Intriguingly, Salmonella pathogenicity island (SPI)-1 and SPI-2 displayed up- and downregulation, respectively, regardless of lifestyles in contact with the radish greens, suggesting altered Salmonella virulence during adaptation to plant environments. This study provides molecular insights into Salmonella adaptation to plants as an alternative environmental reservoir.

An Evaluation of the Recognition, Preferences and Quality Factors on Sauces (소스(Sauce)의 인식과 기호도 및 품질요인에 대한 평가분석)

  • This study was performed to investigate and analyze the level of recognition, preferences and quality factors of sauces. A total of 1,340 subjects, 660 males and 680 females in the Daegu and Kyeongbuk areas participated in a questionnaire for this study. The results were summarized as follows: Among the respondents, 41.1% answered that they learned about the sauces from a western style restaurant. 73.4% of the respondents suggested that the taste and the quality of sauces had an influence on their preferences of western dishes. 51.2% of the respondents recognized that sauces are mainly used when baking and broiling; it was widely known that demiglace sauce is used on steaks. The preferred colors of sauces from most popular to least popular were brown, blond, white, yellow and red. The overall favorite stock material of demiglace sauce was beef bone. Foods that were preferred to be served with sauces were found to be beef, pea seafood, fish, shellfish, poultry and cereals from most preferred to least preferred. The favorite sauces were brown stock sauce, tomato sauce, liquor sauce, and cream sauce from most preferred to least preferred. The oil and butter sauces were preferred the least. Using a scale of 1 to 5 with 5 being the highest, the respondents rated the way sauces were cooked as the most important the quality-determining factor, with a mean value of 4.31. The raw ingredient as the most important factor was rated 4.45. Among sensory factors, the respondents agreed taste was that the most important characteristic, with a mean value of 4.65. 36.0% evaluated the overall quality of sauces as good and 47.0% as ordinary when served with western dishes. 44.1% of the respondents stated that the taste is was the topmost characteristic that needed to be improved, and that flavor and color were next in order.

Suggestions for a better inspection method according to the occurrence of fowl typhoid in broiler breeders (육용종계 가금티프스 발생에 따른 검사방법 개선 방안)

  • Pullorum disease and fowl typhoid are septicemic avian diseases transmitting through egg by transovarian infections. A series of tests has been performing in breeding flocks of chickens and test plans for proper inspection have been modified by government veterinary institute to control of such diseases. To improve inspection plans, different test methods were compared using fowl typhoid positive samples from a poultry farm located in Jeonbuk state in 2012. Based on first inspection, 11 samples among total 200 samples were positive by rapid slide agglutination (RSA) test and 7 samples among RSA positive samples were finally diagnosed as Salmonella Gallinarum infection by ELISA, bacterial isolation, PCR, and histopathologic examination. In the second inspection, 20 samples among total 100 samples were positive by RSA test. Among RSA positive ones, 19 samples were positive by ELISA, S. Gallinarm were successfully isolated in 3 samples, and 16 samples were positive by PCR in the cecal tonsils where were not successful for bacterial isolation. Based on histopathologic examination, severe inflammation in the 13 cecal tonsils and infiltration of lymphocytes and heterophils in the 11 livers were observed. Therefore, we suggest that bacterial isolation, PCR, and histopathologic examination methods in the third inspection need to be further used in various tissues for correct diagnosis and for final eradication of pullorum disease and fowl typhoid in breeding flocks of chickens.

Genetic polymorphism in regulatory region of fatty acid binding protein 4 (FABP4) and its effect on carcass weight in Hanwoo steers (한우 지방산결합단백질 4(FABP4) 유전자 조절영역내 단일염기변이(SNP)와 도체형질간 연관성 분석)

  • The aim of this study was to identify the polymorphism on fatty acid binding protein (FABP4) gene promoter region and its association with carcass traits in Hanwoo. We performed PCR-direct sequencing of FABP4 promoter region to identify single nucleotide polymorphism (SNPs) using unrelated 24 Hanwoo bulls. Four SNPs (-298A>G, -472A>G, -887A>G, -862A>G) were detected in the promoter region and genotyped on 583 Hanwoo steers. A linear mixed model revealed an association of three SNPs (-298A>G, -472A>G and -862A>G) with carcass weight and marbling score in dominance model (P<0.05). The animals with AA genotypes for the three SNPs were heavier carcass weight (5 kg) than animals with GG genotypes in the statistical analysis. For the marbling score, the AA genotype was lower effect of marbling score (0.21) than GG genotypes. In conclusion, this study indicates an important role for three SNPs detected in promoter region of FABP4 in determining carcass weight and marbling score in Hanwoo.

Study on Demand Elderly Foods and Food Preferences among Elderly People at Senior Welfare Centers in Seoul (고령자의 식품선호도 및 고령친화식품 요구도에 관한 연구 - 서울시 노인복지관 이용자 중심으로 -)

  • The purpose of this study was to investigate the need to develop foods for the elderly based on their food preferences. A one-to-one survey method was used for data collection, and the survey was conducted on 165 elderly people (male: 47, female: 118) attending senior welfare centers in Seoul. The results of this study show that the preferred cooking method was steaming rather than stir-frying. Vegetables (4.24), fruits (4.22), Kimchi (3.87), fish (3.83), tofu (3.81), rice-cakes (3.65), dairy products/eggs (3.47), meat (3.45), noodles (3.44), seafood (3.44), poultry (3.23), bread/cookies (3.10) showed high preferences in order (p<0.05). The respondent's chewing & swallowing degree were "Not uncomfortable chew" 31.5%, "Hard food, and a big food is a little difficult to intake". 37%, "Hard food, and a big food is difficult to intake" 11.5%, "Small food, and soft foods can intake" 11.5%, and "It is difficult to intake solid food" 1.2%. Foods that were difficult to chew were meat 28.3%, seafood 15.2%, vegetables 15.2%, nuts 14.1%, fruits 12.0%, Kimchi 8.7%, and other 6.5%. For respondents, the most important factors affecting choice of elderly foods were flavor (48.5%), hygiene (43.0%), and nutrition (36.4%). The elderly wanted "Ready to eat (RTE)" 53.9%, "Ready to heat (RTH)" 15.8%, "Ready to cook (RTC)" 21.2%, and 'Fresh cut product" 9.1%.

Nutritional Iron Status in Pre - and Postmenopause Middle - Aged Women in Kangnung Area (일부 폐경 전 , 후 중년 여성의 철분 영양상태에 관한 연구 - 강릉지역을 중심으로)

  • The purpose of this study were to assess iron status and obesity in 82 middle aged women living in Kangnung area. Anthropometric measurements were taken for body weight, height, percentage of body fat and circumferences of waist and hip. Venous blood samples were drawn from subjects for measurement of hemoglobin(Hb), hematocrit(Hct), serum iron(Fe), total iron binding capacity(TIBC), transferrin saturation(TS) and serum ferritin. Dietary intakes of iron(heme iron and nonheme iron), the amounts of MPF(meat, poultry and fish) and ascorbic acid were assessed by modified 24-hr recall method. The results obtained are summarized as follows : Postmenopausal women had more body fat than premenopausal women. That is, postmenopausal women tend to be obeser than premenopausal women. There was no difference in Hb, Hct, Fe, TIBC and TS between pre- and postmenopausal women. But the serum ferritin concentration of postmenopausal women(83.7$\pm$42.1ng/ml) was significantly (p<0.05) higher than premenopausal women(56.4$\pm$41.0ng/ml). Prevalences of iron deficiency (20%, 20.0% and 17.1% respectively) of postmenopausal women. The mean daily intakes of total iron in pre- and postmenopausal women were 17.5$\pm$9.3mg and 15.6$\pm$6.9mg, respectively. Bioavailabilities of dietary iron were 6.5% and 4.5% in pre- and post-groups. These results indicate that individual dietary guidelines should be used to educate middle-aged women different in status of menopause. For example, premenopausal women should increase nutritional iron status and postmenopausal women should try to prevent obesity.

A Study on the Prevalence of Chronic Diseases, Health-related Habits and Nutrients Intakes according to the Quality of Life in Korean Adults (한국 성인의 삶의 질에 따른 만성질환 유병율, 건강관련 생활습관 및 영양섭취 실태 조사 - 2008년 국민건강영양조사를 근거하여-)

  • This study was done to analyze the chronic diseases prevalence and dietary intake status according to the health related quality of life and to identify its significant properties. This study was conducted based on 2906 subjects ranging from 20~64 years old, out of 9704 total subjects from the 4th Korean National Health and Nutrition Survey in 2008. The average quality of life (QL) score was 0.915, where 983 subjects were below the average (low QL group) and 1923 subjects were above (high QL group). In the high QL group, there was more likely to be young, male, and with higher income and education. For high QL group, the self-related health score was high, and the subjective stress level was low. The prevalence of diabetes was significantly higher in the low QL group and anthropometric and biochemical measures were not shown differences between the two groups but waist circumference was significantly higher in the low QL group. The QL score was significantly negative correlated with the waist circumference and the body mass index. As for the recognition rate of Nutrition Guideline, there were no significant differences but high QL group had higher scores in "Eat a variety of foods", "Avoid too much Sodium", and "Enjoy meals and Eating a breakfast". Carbohydrate intake was significantly lower in the high QL group than in low QL group and the high QL group showed higher intake in legumes, meat and poultry, fruits, daily products and beverages. Overall results suggest that increased diabetes prevalence in people with low quality of life might be affected by the diets based on high carbohydrate, increased waist circumference, and lower rate of practice nutritional guideline.

  • The objective of this study were to review and analyze the application technologies of electronics and microprocessor for environment control and automated management systems of livestock production of the advanced countries, and to select the appropriate and applicable technologies for our systems among the analyzed. In this study, the environment control systems were analyzed mainly on the poultry production systems; and the automated management systems on swine and dairy production systems. According to the results, the advanced technologies reviewed and analyzed could be applicable for designing our animal production systems, if those were modified and remodeled for our situation.

Effect of the Adding of Flammulina velutipes Cultivation Media Wastes into Chicken Feed on the Meat Quality and Production Cost of Broiler (팽이버섯 탈병배지의 사료첨가가 육계의 질 및 생산비용에 미치는 영향)

  • To develop environmentally friendly bio-materials for animal feed, the potential of using mushroom was studied. Adding Flammulina velutipes cultivation media wastes (FCMW) has shown beneficial effects on broiler meat quality and production cost because of the $10{\sim}20%$ saving in feeds for broiler chicken. However, differences proximate composition between FCMW and commercial broiler chicken feed were observed. FCMW had higher contents of crude fat (6.2%) and crude fiber (13.7%) than the commercial broiler feed which had crude fat and crude fiber of 4.0%, and 6.0%, respectively. But FCMW crude protein contents were lower (12.2%) than those in commercial broiler (19.0%). Adding $5{\sim}10%$ of FCMW to broiler chicken feed increased amino acid concentration upto $33{\sim}38%$ in the meat, but adding more than 20% decreased amino acid concentration.

Comparison of Physiological Changes in Broiler Chicken Fed with Dietary Processed Sulfur (식이유황 급여에 따른 계육의 생리적 변화)

  • The purpose of this study was to evaluate the mortality rate, weight gain, and protein, lipid cholesterol, and total amino acid contents in processed-sulfur-diet chicken meat. Also investigated were the antioxidant activity and glutathione content of blood plasma. The mortality of the processed-sulfur-diet chickens was relatively reduced, and their weight increased compared to the normal-diet chickens. Although the glutathione content decreased in the processed-sulfur-diet chicken blood plasma, the antioxidant activity increased compared with the normal-diet chickens. Also, there was no difference in the total cholesterol contents of the blood, but the level of HDL cholesterol increased while that of LDL cholesterol decreased. Furthermore, there were no changes in the total amino acid and protein contents, but the fat content was significantly reduced. As no toxicity was found in the DT 40 cells in the MTT assay, it can be concluded that dietary sulfur in chicken feed can improve the quality of poultry products.
