  • Lactobacillus casei YIT 9018 was infected with phage J1 and subjected to grow in $Ca^{++}$ -free MRT (spell out) medium under the presence of four different types of phosphates, sodium-metaphosphate,-pyrophos-phate,-dibasic phosphate, and potassium-phosphate. Among the phosphates tested, sodium pyrophosphate showed sufficient inhibition on the lytic activity of the phage at 0.1% level whereas other phosphate needed more than 0.2% for the same effect. When the concentration of sodium pyrophosphate increased to 0.17%, the bacteria could be protected from lysis until the second succeeding transfer.

  • To test for the effect of applied fertilizer and nutrients on uptake and transport for paddy rice, two paddy field trials were conducted with Dongjinbyeo in degenerated salt paddy field of Jeonbuk series from 1999 to 2000. After experiment, soil acidity, content of organic matter phosphate, silicate, potassium, calcium, and total nitrogen was increased by application of fresh cattle manure(FCM). Content of Nitrogen in soil layer leached inorganic nitrogen $NO_3$ was higher that that of $NH_4$ and was high in treatment of FCM. Content of $PO_4$ was higher in FCM than other treatments. But content of potassium was in high control. During the growth of rice plant, the amount of water consumption was 477mm. The amount of supplied nitrogen was high in treatment of no nitrogen(NN), 20% reduced application of LCU(LCU-20%), and no fertilizer. In case of phosphate, the supplied amount was more than the consumed amount with the exception of treatment "no phosphate(NP) and no fertilizer(NF)". In case of potassium, the consumed amount was more than the supplied amount in all treatments. The amount of applied nitrogen in the nutrient infiltrated water was high in treatment soil test(ST), C+FCM+Si(Silicate) and the ratio of recovered nitrogen was high in 20% reduced application of LCU. The amount of applied phosphate in the nutrient infiltrated water was high in FCM and that of applied potassium was high in 20% reduced application of LCU. Nitrogen use efficiency of paddy rice was high in 20% reduced application of LCU and use efficiency of phosphate and potassium was high in C+Si(Silicate). Grain yield of rice was high in order of 20% reduced application of LCU>C+Si=C+FCM+Si>C+FCM.

  • The objective of this study was to determine phosphorus effects on drought stress-induced oxidative stress in Kentucky bluegrass. Drought stress was induced by reducing of water to plants in pots. Two types of phosphorus were applied as potassium phosphate (P) or potassium phosphonate (PA). Application of phosphorus was efficient to ameliorate the adverse effects of drought. Osmotic potential, total chlorophyll and carotenoid content were significantly decreased by drought stress, but was relieved by P or PA application. Superoxide (O2•-) concentration was significantly increased more than 14-fold under drought-stressed plants, was accompanied with increase of hydrogen peroxide (H2O2) and lipid peroxidation (MDA). However, malondialdehyde (MDA) was much less in P or PA applied plants under drought stress condition. Activities of catalase (CAT), ascorbate peroxidase (APX) and guaiacol-peroxidase (GPX) were largely increased by drought stress and its increase rate was much higher in P or PA applied plants except APX. These results indicate that drought stress-induced oxidative stress is alleviated by P or PA application due to the increase of activities of antioxidant enzymes.

  • The monitoring of chemical dynamic changes in controlled horticultural lands is very important for agricultural sustainability. Field monitoring was performed to evaluate the soil chemical properties of 200 controlled horticultural soil samples in Gyeongnam province every 4 years from 2000 to 2012. Soil chemical properties such as pH, amount of organic matter, available phosphate, nitrate nitrogen, and exchangeable potassium, calcium, magnesium, and sodium were analyzed. The amount of exchangeable calcium and soil pH were significantly higher in 2012 than in 2000. In 2012, the frequency distribution for values of pH, organic matter, available phosphate, and exchangeable potassium, calcium, and magnesium that were within the optimum range was 16.0%, 22.5%, 11.5%, 3.5%, 2.5%, and 5.0%, respectively. Especially, available phosphate and exchangeable calcium were excess level with portions of 76.0% and 96.5%, respectively. These results indicated that a balanced management of soil chemical properties can reduce the amount of fertilizer applied for sustainable agriculture in controlled horticultural lands.

  • This study examined the effect of water-to-binder ratio (W/B) and phosphate-to-binder ratio (P/B) on the flow, setting time, compressive strength development, and pH variation of magnesium-potassium phosphate composites, MKPC mortars. Ten mortars mixtures were prepared with the W/B varying from 20% to 40% at each P/B of 0.3 or 0.5. The hydration products and microstructural pore distribution of the MKPC pastes were investigated using X-ray diffraction (XRD), scanning electron microscope (SEM) and mercury intrusion porosimetry (MIP). The initial flow and setting time of MKPC mortars tended to decrease with an increase of P/B, indicating that the final setting time was shortened by approximately 24% when P/B increased from 0.3 to 0.5. The slope of the early-strength development measured in the MKPC mortars was considerably higher than that of cement concrete specified in code provisions. For obtaining a relatively good 28-day strength (above 30 MPa) and a near neutral pH (below 9.0) in MKPC mortars, the P/B and W/B need to be selected as 0.5 and 30%, respectively. The strubite-K crystal increased with the increases of P/B and W/B, which leads to the decrease of the macro-capillary pores.

  • 1. Kum Nam Poong which is highly susceptible to bacterial leaf blight was used as the host plant throughout this experiment. Xanthomonas oryzae 6526 was inoculated on the top of upper leaves by single needle inoculation method. After 14 days, the enlarged spots were examined in the experimental pots. Each of 3 levels of nitrogen, phosphate and potassium pots were arranged at random with three replications. 2. The amounts of nitrogen fertilizer applied and the lesion development of bacterial leaf blight were positively correlated regardless of application of phosphate and potassium fertilizers. 3. The effect of phosphate fertilizer on the lesion development was not significantly different from standard level. The lesion development was stimulated when the amount of phosphate fertilizer was increased as twice as standard level. 4. The inhibitory effect of potassium fertilizer on leaf blight was maximum by applying standard level. The stimulative effect of potassium fertilizer on the lesion development, however, was noticed. when potassium fertilizer applied was increased as twice as the standard level. 5. The heading date and spike number of rice plant were significantly correlated with the lesion development, and such phenomena were depended on the amount of nitrogen fertilizer applied regardless of the other fertilizers applied in this experiment.

  • Production of human granulocyte-macrophage colony stimulating factor (hGM-CSF) by Nicotiana tabacum cell suspension culture was studied in Murashige and Skoog (MS) medium with sucrose as a carbon source, ammonium nitrate and potassium nitrate as nitrogen sources, potassium dihydrogen phosphate and sodium dihydrogen phosphate hydrate as phosphate sources, respectively. Optimum concentrations for carbon, nitrogen, phosphate was determined to enhance the production of hGM-CSF. Cell growth was better at high initial sucrose concentration (60 g/L), high initial nitrogen concentration (121.04 mM). Maximum cell density (18.28 g/L) was obtained at 60 g/L of sucrose after 14 days. Cell growth was not so good at low initial sucrose concentration 00 g/L), but the highest hGM-CSF production was obtained at the latter half of exponential phase. hGM-CSF production increased about 3 fold at initial phosphate concentration of 4.96 nM

  • The purpose of this study was to compare the retention and fitness of crown treated with various desensitizing agents on natural teeth. Varnish, dentin primer, bonding agent and potassium oxalate treated groups and control group with no treatment were used. The retention test was executed with the Instron test machine(Instron Co.,U.S.A.) on 24 hours after cementation with zinc phosphate cement. After retention test, the teeth and crowns were washed and recemented. The cemented crowns were embedded in clear resin and sectioned with a diamond saw. The occlusal and marginal fitnesses, were measured indirectly with a stereomicroscopic photographs. The result of this study were obtained as follows : 1. The retention of control group was higher than any other desensitizing agent treated group. 2. The differences of retention among varnish, dentin primer, bonding agent and potassium oxalate treated groups were not statistically significant. 3. There were no statistically significant difference in fitness at occlusal surface and margin among control, varnish, dentin primer, bonding agent and potassium oxalate treated groups. According to these results, desensitizing agents used to protect the pulp from irritation of luting agents may adversely affect the retention of castings. Therefore, desensitizing agents should be used in limited areas carefully.
