Effect of Pluronic F-68 on the Post-thaw Growth of Cryopreserved Transgenic Nicotiana tabacum Cells (Pluronic F-68이 동결보존된 형질전환 담배세포의 해동 후 세포생장에 미치는 영향)

  • To enhance the growth of cryopreserved cells of transgenic Nicotiana tabacum, Pluronic F-68 was supplemented in a recovery medium during post-thaw period. As cryoprotective agents, 1 M sucrose, 0.5 M glycerol and 0.5 M dimethyl sulfoxide (DMSO) were added before freezing steps. The post-thaw growth of the cells was improved with Pluronic F-68, ranged from 0.1 to 10 g/L. The interactions of Pluronic F-68 with the cells were confirmed by the changes of hydrophobicity or permeability of the cells. Pluronic F-68 did not show any effect on the activity of $\beta$-glucuronidase (GUS) in all treatments. Therefore, the addition of Pluronic F-68 in a recovery medium was found to be beneficial to enhance the post-thaw growth of cryopreserved transgenic tobacco cells without affecting the production of recombinant protein.

Effects of Diluent Component, Freezing Rate, Thawing Time and Thawing Temperature on Acrosome Morphology and Motility of Frozen-thawed Boar Sperm

  • This study was carried out to obtain informations regarding the effect of N-acetyl-D-glucosamine in the LEY (lactoseegg yolk) diluent according to incubation time in 5 ml maxi-straw and the effects of freezing rate, thawing temperature and thawing time in the LEN (lactose-egg yolk and N-acetyl-D-glucosamine) diluent on acrosome morphology and motility of frozen-thawed boar sperm. The study showed that the LEN diluent was higher post-thaw NAR (normal apical ridge) acrosome than the LEY diluent for 0.5 h incubation at 37$^{\circ}C$. However, there were no differences between the LEN and LEY diluents on post-thaw sperm motility according to incubation time. The straws frozen from 5.0 cm (20$^{\circ}C$/min) to 17.0 cm (1$^{\circ}C$/min) above the liquid nitrogen surface did not show any significant differences on post-thaw sperm motility. However, the straws frozen above 5.0 cm from the liquid nitrogen surface were higher NAR acrosome than those frozen above 17.0 cm. The post-thaw percentages of motile sperm and NAR acrosome were significantly higher (p<0.05) for the maxi-straws submerged for 40 or 45 sec in a 52$^{\circ}C$ water bath than for 30, 35, 50 or 55 sec. The mean sample temperatures of maxi-straws after 40 or 45 sec submersion were 20.7 or 26.4$^{\circ}C$. In conclusion, the sample temperature of the thawed semen was very important for post-thaw sperm survival in the LEN diluent of 5 ml maxi-straw. When the temperature of the thawed semen was 20.7$^{\circ}C$, the percentages of motile sperm and NAR acrosome were highest.

  • In this study, we developed a "Korean-style button mushroom cultivation Compost post-fermentation system." The purpose is to increase farm income by reducing the labor force of button mushrooms farmers and shortening the production cycle. The "Korean-style button mushroom cultivation Compost post-fermentation system" was designed to reflect the reality of domestic button mushroom farmers. By reducing the temperature difference of the fermentation Compost in the fermentation system, the company produces a button mushroom Compost that ensures uniform quality. As a result of the performance experiment, the working time of the Compost post-fermentation system was shortened by 40 hours. The number of aerobic bacteria and actinomyces that help the button mushrooms Compost increased. Filamentous bacteria that deteriorate the quality of mushrooms have been sterilized.

Seminal plasma modulates post-thaw longevity and motility of frozen sperm in dromedary camel

  • Objective: This study investigated the effect of adding seminal plasma to frozen-thawed semen on the quality of sperm and pregnancy following insemination in dromedary camels. Methods: In experiment 1, the frozen-thawed semen from 9 collections (3 bulls) was further diluted with either the base extender or homologous seminal plasma (HSP). In the second experiment, a pooled sample of frozen-thawed semen was diluted with either seminal plasma from another three bulls. Live percentage, total and progressive motility, functional and acrosome integrity, and sperm kinematics were evaluated at 15, 60, and 120 minutes post-thawing and compared to the non-treated control. In experiment 3, frozen semen was used to inseminate camels in the following experimental groups: 1-Single insemination with double dose undiluted frozen semen (n = 9); 2-Re-insemination in 6 hours with undiluted semen (n = 13); 3-Single insemination with HSP treated sperm (n = 14). Results: Frozen-thawed sperm diluted in HSP or the non-homologous seminal plasma from Bull C indicated an improvement in all parameters after 1 hour post-thawing incubation (p<0.05). The proportion of total and progressively motile sperm did not drop significantly at 60 minutes post-thawing when diluted with the seminal plasma of Bull C (p>0.05). Double insemination with nontreated sperm and single insemination with HSP-treated sperm resulted in similar pregnancy rates (15.3% vs 21.4%, p>0.05). None of the camels conceived with double-dose single insemination of nontreated sperm. Conclusion: Seminal plasma improves sperm longevity and motility after thawing in dromedary camel with a significant between-bull variation in effect. Low post-thaw sperm longevity might be the cause behind the low pregnancy rates in frozen semen insemination of dromedary camels.

Effects of Varying Creep Feed Duration on Pre-weaning and Post-weaning Performance and Behavior of Piglet and Sow

  • 32 sows (Landrace${\times}$Yorkshire) and their litters were used to evaluate the effects of varying creep feed duration on pre-weaning, post-weaning performance of piglets and sows. Sows were randomly assigned with 1, 2 or 3+ parities into 1 of 4 treatments. Creep feeding was initiated at day 5, 10 and 15 from birth for treatment 1 (TRT1), 2 (TRT2) and (TRT3), respectively, with a control group provided no creep feed. In this study, TRT1 and TRT2 diets had reduced (p<0.05) the post-weaning diarrhea scores in piglets and the weaning-to-estrus interval and cortisol concentration in sows at weaning time compared with other treatments. Dietary TRT1 led to a higher (p<0.05) epinephrine and norepinephrine concentrations than other treatments. No differences (p>0.05) were noted in suckling, sleeping, fighting frequency and mortality in piglet and eating, standing times, backfat and body weight loss in sows. In conclusion, creep feed initiated from day 5 and 10 reduce diarrhea scores in piglets and benefit the estrus interval in sows compared with those initiated from day 15 and no-creep feeding diets, indicating creep feeding could improve the pigs and sows performance, especially those initiated from day 5 and 10.

Spray Dried Animal Plasma as an Alternative to Antibiotics in Weanling Pigs - A Review -

  • Piglet health at weaning is compromised due to several stress factors. Following the ban of antibiotic growth promoters new alternatives are required to control these problems. This paper reviews the evidence available for the use of spray dried animal plasma (SDAP) as an alternative to antibiotics in weaning pigs. Data from 75 trials in 43 publications involving over 12,000 piglets (mean values) have been used to calculate the performance responses of piglets according to several factors including SDAP origin, protein source from the control diet being replaced, dose of inclusion, age and weight of the piglets at weaning, sanitary conditions and simultaneous use or not of medication. Although the use of SDAP of all origins results in positive responses, it appears that plasma from porcine origin has the highest efficacy. This could be explained by the specificity of its IgG against porcine pathogens. During the first week post-weaning the response to plasma appears to increase with the inclusion dose, although over the two-week pre-starter period an optimal inclusion level of 4-8% is suggested. SDAP improves feed efficiency more markedly when the piglets are challenged with an experimental infection or when feed does not contain medication, which could be indicative of a lower expenditure of energy and nutrients to build an immune response against the challenge. There is evidence supporting that SDAP IgG and other bioactive substances therein prevent the binding of pathogens to the gut wall and reduce the incidence of diarrhoea in the post-weaning phase. Overall, plasma can be postulated as an excellent alternative to in-feed antimicrobials for piglets in the post-weaning phase.

  • Selection of oocyte cryopreservation method is a prerequisite factor for developing an effective bank system. Compared with slow freezing method, the vitrification has various advantages such as avoiding intracellular ice crustal formation. In our previous, we attempted to employ a vitrification method using ethylene glycol and an electron microscope grid for cryopreservation of mouse oocytes. However, A high incidence of spindle and chromosome abnormalities was detected in thawed oocytes after vitrification. We examined whether the addition of a cystoskeleton stabilizer Taxol $^{TM}$, to the vitrification solution could promote the post-thawed survival and subsequent development of stored oocytes. More oocytes developed to the 4-cell (44.7% vs. 69.7%), 8-cell (31.8% vs. 64.2%), morula (24.7% vs. 54.3%), and blastocyst (20.3% vs. 49.2%) stages after the addition of Taxol$^{TM}$ to the cryoprotectant than after no addition. 21 and 26 mouse pups were born after transfer of blastocyst derived from oocytes vitrified without and with Taxol. The addition of Taxol to vitrification solution greatly promoted post-thaw preimplantation development of ICR morose oocytes.tes.

  • This stud tires to present a general picture about Alessandro Mendini's unique idea of redesign. One of Italian post-war redical architect and designer mendini gives us a special opportunity to appreciate the social cultural and political context where the post-war Italian design lies. Also known as "banal design" Mendin's design revolutionized the way in which we practice utilize and think about design itself. It is my opinion that the idea of his redesign can be best understood when we consider the social contest of Italian design. Unlike that of other European countries post-war Italian design gave special emphasis on how design can or should be more than a simple activity of making aesthetic or industrial products. In terms of these possibilities Mendini was never optimistic : today we are completely controlled by dehumanized mass production and it is impossible for design to take a special role for a social change Then Mendini's pessimism is bound up with the spirit of this age widely known as postmodernism. Even though Mendini himself never characterized his design as postmodern it is not difficult to identify various postmodern elements in his idea and practice of redesign. Thus in the final section of this study I shall investigate the postmodern elements in this design and furthermore such an idea as what makes Mendini postmodern.

  • Industrialization and modernization of the beginning of the IT industry is growing very fast. Since telecommunications industry was developed rapidly, technologies about miniaturization and high-precision of parts have been actively developed to lead information revolution. generally, the entire shear surface of the product applying fine blanking technology must be very precise. Fine blanking is used to save cost by avoiding post-processing of the product. When using press blanking, it spends a lot of money on the production by using many post-processing. Fine blanking typically used in 0.5~18 mm thick steel plate. Because a lot of post-processing cost can be used to process, except for fine blanking. In order to develop components "CHANCE CONTENTS" in the fine blanking process, the purpose of this study is to minimize the edge of the bridge, secured 95% of the material thickness of the shear surface using the 1.6 mm thickness of the material SPCC. Blanking process by introducing after changing thickness through forging process, due to change in vee-rring force and counter force, the experimental amount of depressions and flatness and the shear surface were analyzed.

Effect of Cryopreservation by Vitrification on Viability of Nuclear Transplant Rabbit Embryos (Vitrification 동결보존이 토끼 핵이식 수정란의 생존성에 미치는 영향)

  • For a large sclase production of genetically identical or cloned animals, the effect of cryopreservation by vitrification on the post-thaw viability of nuclear transplant rabbit embryos were investigated. The embryos of 16-cell stage were collected from the mated does at 48 hours post-hCG injection, and they were synchronized to G1 phase of 32-cell stage were injected into enucleated recipient cytoplasms by micromanipulation. After culture until 20h post-hCG injection, the nuclear transplant oocytes were electrofused and activated by electrical stimulation. After in vitro culture for 48h, the nuclear transplant embryos developed to morula stage were cryoperserved with EFS solution by vitrification method. The forzen nuclear transplant embryos were thawed and cultured for 72h and the nuclear transplant of blastomeres under a fluorescence microscopy. The in vitro development to blastocyst of intact-fresh and intact-frozen 16-cell embryos was found to be 96.9 and 63.9%, respectively. The in vitro development to blastocyst of nuclear transplant and frozen-thawed nuclear transplant embryos was found to be 74.5 and 42.9%, respectively. Also, their mean blastomere numbers and mean cell cycles/day was 153 and 105, 145 and 1.34, respectively. From the above results it was concluded that the present cryopreservation by vitrification of nuclear transplant rabbit embryos might be useful though was decreased significantly.
