익수영진고가미방의 지황박, 복령피 배합과 발효에 따른 제제가 면역활성에 미치는 영향 (The effect of prescriptions prepared by adding medicinal herbs(Rehmannia glutinosa Residue, Poria cocos Bark) and fermenting herbal materials based on formulas Iksuyoungjingo on immunological activity)

  • Objectives : This study was carried out to enhance the activity of Iksuyoungjingo, which has the tonify Qi, nourish Yin effect of oriental medicine, and to study the effect of the prescriptions on the immunological activities. Methods : Immunosuppression was induced by methotrexate (MTX) 2 mg / kg, the experimental group was divided into IYGgami-I, IYGgami-I-F, IYGgami-II and IYGgami-II-F groups. Each prescription was administered with drinking water for 20 days, and body weight was measured every 5 days during this period. Leukocyte, $TNF-{\alpha}$, IL-2, IL-6, IgE, spleen weight and body weight were measured Results : In the changes of $TNF-{\alpha}$, IL-2 and IL-6 as pro-inflammatory elements, all of the experimental groups showed a significant increase compared to the control group. In the IgE changes, the IYGgami-I-F, IYGgami-II and IYGgami-II-F groups showed a significant decrease compared to the control group. In the changes of spleen weight, the IYGgami-II-F group showed a significant increase compared to the control group. In the changes of WBC and lymphocytes, the IYGgami-I-F group showed a significant increase compared to the control group. Conclusions : From the above results, it can be observed that the efficacy against immunity is exerted in all of the preparations, and it was confirmed that the efficacy was maintained constant even when utilizing the Rehmannia glutinosa Residue and Poria cocos Bark, and that a more beneficial effect can be exerted in the effectiveness when the fermentation is carried out.

복령(茯岺)의 인공배양(人工培養)에 관한 연구(硏究) (Studies on Artificial Cultivation of Poria cocos)

  • 소나무(Pinus densiflora)에 기생(寄生)하는 복령(茯岺)(Poria cocos) 의 생육(生育)과 배양(培養)에 관한 성질(性質)을 조사(性質)하기 위하여 1979년(年) 5월(月) 1일(日) 부터 1980년(年) 10월(月) 30일(日)까지 연구(硏究) 결과(結果)는 다음과 같다. 1. 최적(最適) pH 는 5.0 이고 최저(最適) 3.0 에서는 성장(成長)이 부양(不良)하며 최고(不良) 7.0 이상에서는 생육(不良)되지 않았다. 2. 적온도(適溫度)는 $25{\sim}27^{\circ}C$이며, $20^{\circ}C$ 이하(以下)에서는 생육(生育)이 불량(不良)하고 $5^{\circ}C$에서는 생장(生長)하지 않았다. 3. Robbins와 Hervey(1958)의 고체배지(固體培地)에서는 malt extract(2% malt extract 에서 집락(集落)의 지름 90mm), tomato extract(8% extract 에서 집락(集落)의 지름 90mm)의 순으로 생장(生長)하였다. 4. Badcock의 방법(方法)에서는 P.D.A.+5%accelerator (집락(集落)의 지름 85mm)로 가장 좋은 생육(生育)을 하였고, 2% malt extract+5% accelerator (균사의 건량 410mg)이었으며, wood extract agar 에 5% accelerator를 첨가(添加)한 배지(培地)에서는 집락(集落)의 지름 36mm로 생육상태(生育狀態)가 불량(不良) 하였다. 5. Wood extract 첨가배지(添加培地)는 20%씩 첨가(添加)했을 때 참나무(Querous acutisssima)의 균사건량 305.3mg, 아카시아나무(Robinia pseudoacasia) 256.3mg., 오리나무(Alnus juponica) 135mg, (Pinus densiflora) 134mg., 밤나무(Meliosma myriantha) 122mg.의 성장(成長)을 나타냈다. 6. amino acid 첨가배지(添加培地)에서 는 L-aspargine 이 균사건량 44.5mg.으로 성장(成長)이 가장 양호(良好)하였고, ammonium sulfate 가 15.5mg 으로 가장 저조(低調)하였다.

복령 균핵의 알칼리추출물에서 정제한 면역활성 증강물질의 작용과 화학구성 (Chemical Composition and Biological Activities of Immunostimulants Purified from Alkali Extract of Poria cocos Sclerotium)

  • 복령 균핵을 여러단계로 추출하여 얻은 다당류들의 면역 증강활성을 생쥐 비장세포에서 혼합임파구 반응으로 측정하였다. 이들 중 탄산나트륨 추출물의 활성이 가장 높았고 이를 DEAE-셀롤로오스와 Sephadex G-50 크로마토그래피로 면역활성증강물질을 정제하여 PCSC22를 얻었다. PCSC22는 탄수화물과 단백질이 약 78:22의 비율로 이루어진 단백다당체이었으며 GP-HPLC로 측정된 평균분자량은 약 8 kDa이었다. PCSC22 분획은 단당들의 구성성분들 중 mannose(92%)가 가장 많고 galactose(6.2%)와 약간의 arabinose(1.3%)도 함유하고 있는 heteromannan 이었다. 단백질 부분은 총 15종의 아미노산으로 구성되어 있었고 그 중 aspartic acid, serine, valine 등의 합이 약 47%로 주요 성분이었다. PCSC22는 B 임파구의 항체형성능력과 macrophage의 일산화질소 분비능력을 함께 증강시켰다.

삼음교(三陰交)(SP6)·족삼리(足三里)(ST36) 백복령약침(白茯笭藥鍼)이 만성 스트레스 모델 흰쥐에 미치는 영향 (The Effect of Poria Cocos Pharmacopuncture at SP6(Sameumgyo) and ST36(Joksamni) on Chronic Stress Model(CSM) Rats)

  • Objectives : The purpose of this study is evaluating the effect of Poria cocos pharmacopuncture (PCP) at $SP_6$(Sameumgyo) and $ST_{36}$(Joksamni) on chronic stress model(CSM) rats. Methods : The experimental rats were assigned to five groups: normal, CSM(control), PCP1X, PCP2.5X and PCP5X. The experimental rats in CSM(control), PCP1X, PCP2.5X and PCP5X were put through a timetable to cause CSM. PCP($20{\mu}l$) was administered at $SP_6$, $ST_{36}$ to groups PCP1X, PCP2.5X and PCP5X once every three days for a total of six times. The weight of the rats was measured by an electronic scale after PCP injection, once every three days for a total of six times. Blood samples were taken from the rats for analysis of aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Serotonin and norepinephrine were measured by enzyme-linked immunosorbent assay(ELISA) kit. Results : The experimental groups(PCP1X, 2.5X, 5X) showed significant body weight increase compared to the control group. Regarding AST and ALT changes in serum, the experimental groups(PCP2.5X, 5X) showed a significant decrease compared to the control group. For Serotonin changes in serum, the experimental group(PCP5X) showed a significant increase compared to the control group. For Norepinephrine changes in serum, the experimental groups(PCP2.5X) showed a significant decrease compared to the control group. Conclusions : These data suggest that PCP at $SP_6$, $ST_{36}$ has therapeutic effects on CSM rats.

재료 배합비를 달리한 복령병의 품질특성 (Quality Characteristics of Bokryung-Byung Prepared with Bokryung (Poria cocos Wolf) Powder)

  • 멥쌀가루에 복령가루를 0, 5, 10, 15, 20% 첨가하여 제조한 복령병의 품질특성을 조사하였다. 저장기간 중 복령병의 수분함량은 42.89~44.85%로 대조구가 44.85%로 가장 높았으며, 복령가루 함량이 증가할수록 유의적으로 감소하였다. 저장기간이 경과할수록 pH는 낮아졌으며, 산도는 증가하였다. 총균수는 제조직후 2.70에서 3.00 log CFU/g이었으며, 저장 4일에는 7.00 log CFU/g 이상으로 급격히 증가하였다. 그러나 대조구와 복령병 간의 차이는 없었다. 제조직후에는 복령가루 첨가량이 증가할수록 명도는 유의적으로 감소하였으며, 적색도와 황색도는 유의적으로 증가하였다. $20^{\circ}C$에서 저장하는 동안 색도는 영향을 받지 않았다. 복령병의 경도, 강도, 검성, 응집력은 대조구에 비해 증가하였으며, 부서짐성은 증가하였다. 설기떡에 복령가루를 첨가하는 것은 기호도가 감소하였으며 그 결과 탄력성이 있고 쫄깃한 떡을 선호하는 것을 알 수 있었다. 전반적인 기호도에서는 대조구가 높게 평가되었으나, 복령병 제조시 복령가루 5% 첨가가 높은 기호도를 나타내어 기능성 떡으로 개발 가능성이 있는 것으로 판명되었다.

사군자탕(四君子湯), 사물탕(四物湯), 팔물탕(八物湯) 및 십전대보탕(十全大補湯)의 처방 구성 및 용량 용법에 관한 연구 (A Study On Compositions, Dosages and Usages of Sagunjatang, Samultang, Palmultang, Sipjeondaebotang in Literature)

  • The result is the followings after investigating composition, dosage and usage of Sagunjatang(四君子湯), Samultang(四物湯), Palmultang(八物湯), Sipjeondaebotang(十全大補湯) in literature by country and Era 1. Sagunjatang has become equal to Panax ginseng(人蔘), Atractylodes japonica(白朮), Poria cocos(복령) each for 2 Jeon(錢), Glycyrrhiza uralensis(甘草) 1 jeon since the Ming Dynasty in China and it is standardized by Panax ginseng, Atractylodes japonica, Poria cocos and Glycyrrhiza uralensis each for 1 Jeon 2 Pun(分) 5 Ri(里) since Dongeuibogam in Korea. 2. Samultang has become equal to Angelica gigas(當歸) and Rehmannia glutinosa(地黃) each three Jeon, Cnidium officinale(川芎) is 1 Jeon 2 Pun or 1 jeon and Paeonia lactiflora(芎藥) 2 Jeon since the Ming Dynasty in China and it is standardized by Angelica gigas, Cnidium officinale, Paeonia lactiflora and Rehmannia glutinosa each for 1 Jeon 2 Pun 5 Ri since Dongeuibogam in Korea. 3. In the case of Palmultang, compositional usage of medicine was different in the Ming Dynastry and the Ching(靑) Dynastry in China. Total dosage was increased and ratio of Angelica gigas and Rehmannia glutinosa was increased comparatively in the Ching Dynasty. All prescription of medicine was consisted of 1 Jeon 2 Pun and specific dosage was presented except Euirimchwalyo(醫林撮要) that dosage was not recorded in Korea. 4. Sipjeondaebotang tended to increase total dosage in the latter part in China. Dosage was mostly used 1 Jeon thus Dongeuibogam but Bangyakhap(方藥合編) used 1 Jeon 5 Pun differentially.

Streptozotocin 유도 당뇨 흰쥐에서 복령약침의 ${\beta}$-cell 손상 방지 효과 (Poria cocos Herbal Acupuncture Prevents ${\beta}$-cell Damage on Streptozotocin-induced Diabetic Rat)

  • 목적 : 정상 췌장조직 속에 존재하는 췌장 소도세포들을 파괴시켜 고혈당을 유발시키고 복령 물추출물로 약침을 시술하여 췌장 조직의 보호효과와 항당뇨 효과를 살펴보고자 실험을 진행하였다. 방법 : 5주령의 Sprague-Dawley rat을 통제된 실험실 환경에 적응시킨 후 1주일간 복령약침액(125mg/kg 복령약침군 및 250mg/kg 복령약침군)을 좌우 신수($BL_{23}$)에 교대로 각각 피하에 약침하고 streptozotocin을 복강내 주사하여 3일 후 diabetes mellitus 유도 정도를 평가하고 2주일간 추가 치료를 진행 한 뒤, 혈액지표(plasma glucose, insulin, TG, TC, NEFA, sGOT, sGPT, ALP, BUN, CRE)와 췌장조직의 형태학적 분석 및 염증 관련 단백질의 발현을 평가하였다. 결과 : 복령약침군(125mg/kg 복령약침군 및 250mg/kg 복령약침군)에서 insulin과 triglyceride, NEFA 수치가 유의하게 감소하였으며 간 기능 효소수치인 sGOT가 감소하는 경향을 나타내었으나, 신장기능지수는 유의한 감소를 나타내지 않았다. 특히 250mg/kg 복령약침군에서 streptozotocin 투여로 인한 pancreatic islet의 형태학적 변성이 현저하게 개선되었다. Western blot 결과 JNK-2, P-JNK-2, P-JNK-1, ERK1/2 및 phosphorylated ERK1의 발현이 감소되었다. 결론 : 복령약침이 고인슐린혈증과 고지질질혈증을 개선시키고 streptozotocin에 의한 pancreatic islet의 파괴를 억제하며, 이는 inflammation-related transcription factor인 NF-kB와도 관련이 있는 것으로 판단된다. 향후 복령약침의 항당뇨 효과와 그 기전에 관한 추가 연구가 필요할 것으로 사료된다

재배 복령(Poria cocos Wolf)의 채취시기에 따른 화학적 품질과 기능적 특성 (Quality and Functional Characteristics of Cultivated Hoelen (Poria cocos Wolf) under the Picking Date)

  • The nutritional quality and physiological activity of cultivated hoelen from 13 months to 24 months were examined to compare with natural hoelen. General compositions of hoelen were as follows re spectively; crude protein 1.80~2.50%, crude fat 0.68~1.23%, crude ash 0.12~0.43%, crude fiber 6.30~7.14%, nitrogen free extract 89.25~90.44%. The major free sugar was glucose, and the content of 13 months cultivated hoelen was higher than any other cultivated hoelen. The major fatty acids of hoelen were linoleic acid, palmitic acid, stearic acid and oleic acid. The ratio of unsaturated fatty acids was 62.62~77.96% and the content was higher in cultivated hoelen than in natural hoelen. The contents of amino acid were high in the order of tyrosine, aspartic acid, glutamic acid and serine, and increased as the day of cultivation became longer. The contents of mineral components were higher in the order of Ca, K, Mg and the contents of Na and Ca were higher in natural hoelen than in cultivated hoelen. The contents of pachymic acid and dehydropachymic acid was similar in the natural and cultivated hoelen. On the results of antimicrobial test the ethanol extract showed a stronger effect than water extract, and had an excellent antimicrobial activity on Micrococcus luteus and Escherichia coli. The ethanol extract of hoelen showed comparatively strong electron donating ability. The ethanol extract of hoelen showed a high inhibition activity on the growth of lung cancer, ovary cancer, skin cancer, central nerve cancer and rectum cancer, especially the activity of 19 months cultivated hoelen was the highest. On the above results of nutritional quality and physiological activity of hoelen, it is supposed that the picking date of cultivated hoelen was suitable over than 19 months.

불면증에서 산조인의 유무에 따른 약물배오의 경향성 고찰 (Combination Tendency Analysis on Herbal Formula to Treat Insomnia Focused on Zizyphi spinosi Semen)

  • Objectives : Zizyphi spinosi Semen is widely used for treating insomnia in korea and other oriental country. This study is analyzing combination tendency of Zizyphi spinosi Semen in herbal formula for treating insomnia. Methods : Herbal formulas for treating insomia were searched in "The Prescription Dictionary of Traditional Chinese Medicine", and checked in each literary source. Then, the herbal formula was divided in two groups : FCZ(The formula group containing Zizyphi spinosi Semen) and FWZ(The formula group without Zizyphi spinosi Semen). FCZ and FWZ were compared by following criteria; i) Prevalence of herbal formula by period ii) Prevalence of medicinal material's effects iii) Prevalence of medicinal material iv) Prevalence of the nature of midicinals v) Prevalence of flavor of medicinals vi) Prevalence of meridian entry. Results : 116 Herbal formula met our criteria, and herbal formulas were divided in two groups : FCZ(n=59), FWZ(n=57). The result of prevalence in FCZ is following; Herbal formula prevalence increased as time passed to the Qing(淸) dynasty. Tonifying and replenishing medicinal(補益藥), tranquillizing medicinal (安神藥), and inducing diuresis to drain dampness medicinal(利水滲濕藥) are the three most prevalent types of materia medica. In specifically, Panax ginseng radix(人蔘), Liriopis Tuber(麥門冬), Angelicae Gigantis Radix(當歸), Poria Cocos(茯神), Glycyrrhizae Radix(甘草), Poria cocos(茯神), Polygalae Radix(遠志), Thuja Orientalis L.(柏子仁), Paeonia Japonica(白芍藥), and HgS(硃砂) are combined frequently. The most prevalent nature of medicinals were 'warm microwarm(溫 微溫)' and flavor of medicinals was normal(平). Meridian entrys that frequently combined were heart(心), spleen(脾), and liver(肝). Conclusion : We found combination tendency of Zizyphi Semen in herbal formula for treating insomnia.

Pachymic Acid Enhances Pentobarbital-Induced Sleeping Behaviors via GABAA-ergic Systems in Mice

  • This study was investigated to know whether pachymic acid (PA), one of the predominant triterpenoids in Poria cocos (Hoelen) has the sedative-hypnotic effects, and underlying mechanisms are mediated via ${\gamma}$-aminobutyric acid (GABA)-ergic systems. Oral administration of PA markedly suppressed locomotion activity in mice. This compound also prolonged sleeping time, and reduced sleep latency showing synergic effects with muscimol (0.2 mg/kg) in shortening sleep onset and enhancing sleep time induced by pentobarbital, both at the hypnotic (40 mg/kg) and sub-hypnotic (28 mg/kg) doses. Additionally, PA elevated intracellular chloride levels in hypothalamic primary cultured neuronal cells of rats. Moreover, Western blotting quantitative results showed that PA increased the amount of protein level expression of $GAD_{65/67}$ over a broader range of doses. PA increased ${\alpha}$- and ${\beta}$-subunits protein levels, but decreased ${\gamma}$-subunit protein levels in $GABA_A$ receptors. The present experiment provides evidence for the hypnotic effects as PA enhanced pentobarbital-induced sleeping behaviors via $GABA_A$-ergic mechanisms in rodents. Taken together, it is proposed that PA may be useful for the treatment of sleep disturbed subjects with insomnia.
