Physiological Characteristics and Antioxidant Enzyme Activity of Populus euramericana and Populus alba x Populus glandulosa under Livestock Waste Leachate Treatment (돈분침출수 처리에 의한 이태리포플러와 현사시나무의 생리적 특성 및 항산화효소 활성)

  • We examined two Populus species (Populus euramericana and Populus alba ${\times}$ Populus glandulosa) trees grown under livestock waste leachate treatment in the field. We investigated their physiological properties (photosynthetic rate, chlorophyll contents, transpiration rate and intercellular $CO_2$ concentration) and antioxidant enzyme activities. Ascorbate peroxidase and glutathione reductase were analyzed. According to our measurements, P. euramericana at control site showed a lower total chlorophyll contents, photosynthetic rate, intercellular $CO_2$ concentration, transpiration and stomatal conductance than those of trees at treatment site. P. alba ${\times}$ P. glandulosa showed low stomatal conductance and low photosynthetic rate.

Sapflow of Two Poplars under Swine wastewater Irrigation (양돈폐수 처리할 때 포플러의 수액이동)

  • We examined sapflow of two populus tree species (Populus alba ${\times}$ Populus glandulosa and Populus euramericana) grown under Swine wastewater treatment, and measured relationship between sapflow and climate facters. Sapflow of P. euramericana was 1.5 times greater than that of P. alba ${\times}$ P. glandulosa from September to late December 2007. However, sapflow of P. euramericana and P. alba ${\times}$ P. glandulosa was similar after late January. The relationship between the mean temperature and sapflow was relatively high. There was a decrease in sapflow tendency of the two poplar species during the late October when the mean temperature was low. Trends on relationship between the relative humidity and sapflow showed low relationship. However, data showed weak relationship between sapflow and precipitation. During the period of less precipitation, sapflow of P. alba ${\times}$ P. glandulosa has no relationship. Sapflow of two populus species was still active during non-growing season. P. euramericana was considered as a better species than P. alba ${\times}$ P. glandulosa, in terms of phytoremediation.

Visual and Physiological Characteristic Changes of Five Tree Species Exposed to SO2

  • After long exposure to low-density $SO_2$ the five tree species showed different changes in their visible damage: Ailanthus altissima showed no visible damage; Acanthopanax sessiliflorus, Populus alba ${\times}$ Populus glandulosa, and Platanus orientalis showed typical damage of yellow spots on their leaves; and Liriodendron tulipifera showed serious damage on the end of leaf tip. The photosynthesis rate of Liriodendron tulipifera and Acanthopanax sessiliflorus was usually lower than the control group: Ailanthus altissima, Populus alba ${\times}$ Populus glandulosa, and Platanus orientalis showed no difference from the control group, while Acanthopanax sessiliflorus and Liriodendron tulipifera showed large changes. In regard to the light compensation points, Ailanthus altissima showed similar values, Populus alba ${\times}$ Populus glandulosa and Platanus orientalis exposed to $SO_2$, showed 3 to $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ higher values than the control group, and Acanthopanax sessiliflorus and Liriodendron tulipifera exposed to $SO_2$ showed approximately twice higher light compensation points.

Molecular Characterization and Expression of CuZn-superoxide Dismutase (PSOD1) from Populus alba${\times}$Populus glandulosa

  • A cDNA, PSOD1, encoding cytosolic copper/zinc superoxide dismutase (CuZn-SOD) was cloned and characterized from a full length cDNA library prepared from Populus alba${\times}$Populus glandulosa cultured in vitro. A PSOD1, is 725 nucleotides long and has an open reading frame of 459 bp with 152 amino acid residues (pI 5.43). The deduced amino acid sequence of PSOD1 perfect matched to the previously reported CuZn-SOD (CAC33845.1). Consensus amino acid residues (His-45, -47, -62, -70, -79, -119) were involved in Cu-, Cu/Zn-, and Zn- binding ligands. The deduced amino acid sequence of PSOD1 exhibited the high level of similarity from 100 to $85\%$ among previously registered SOD genes. The expression of PSOD1 in poplar increased at the 1 mM $H_{2}O_2$ and drought stress during 30 min and 60 min, but the ozone treated poplar increased at 30 min in the early time and then decreased at 60 min.

  • The hybrid poplar, populus alba ${\times}$ P.glandulosa $F_1$ shows a strong ability to produce root suckers. The relationship between crown density and root sucker was studied and the other factors that may be related to production of root sucker were also discussed. The possibility was also discussed to use its root suckers for fuelwood forests and vegetation in the area that erosion control is needed.

  • A type 3 metallothionein cDNA (PMT3a) from ozone-treated Populus alba${\times}$Populus glandulosa cDNA library has been isolated and characterized. A PMT3a cDNA is 459 nucleotides long and has an open reading frame of 201 bp with a deduced amino acid sequence of 66 residues (pI 4.94). The deduced amino acid sequence of PMT3a matched to the previously reported metallothionein genes. The deduced amino acid sequence of PMT3a showed the $86\%$ identity with P. balsamifera ${\times}$P. deltoides. Expression of PMT3a by the RT-PCR was increased 60 min than 30 min after drought treatment. The ozone treated poplar increased at 30 min in the early time and then decreased at 60 min.

  • This experiment has been made to find out the influence of soil bulk density on growth and root development of Populus alba ${\times}$ P. glandulosa in Buyeo-kun, Chungnam. Bulk density and porosity significantly change according to slope position and soil depth and have a significant effect on tree height, DBH, biomass, and the distribution of root development. The results extracted from the experiments are as follows; 1) Bulk density in average changes from $1.17g/cm^3$ (1.05 ~ 1.40) in lower slope position to $1.43g/cm^3$ (1.36 ~ 1.60) in upper slope position, and porosity from 52.65% (55.05 ~ 45.50) in lower slope position to 41.20% (43.81 ~ 37.21) in upper slope position. 2) Bulk density increases significantly with soil depth. 3) Tree height, DBH, and total biomass decrease with bulk density. 4) Foliage, branch, bolewood, bolebark, and root also significantly have a negative correlation with hulk density. 5) Populus alba ${\times}$ P. glandulosa has a shallow root system chiefly distributing only in AI horizon, and critical bulk density is $1.43g/cm^3$. Therefore, slope position, hulk density, and porosity are considered to influence on the growth, biomass, and root development of Populus alba ${\times}$ P. glandulosa.

  • Two fast-growing clones 65-29-19 and 66-6-8 and two less fast-growing clones 66-14-29 and 66-14-99 were compared at intensively cultured plots to identify what morphological characters are related to fast growth and high biomass yield of Populus alba ${\times}$ Populus glandulosa $F_1$. The plantations were established in the spring of 1982 and 1983 at the Seoul National University nursery in Suweon. Differences in dry matter production were found among two-year-old clones but not, among one-year-old clones. Highly significant correlations were found between total leaf area per tree and total dry weight per tree and between total branch length per tree and total dry weight per tree for two-year-old clones. This relation was more obvious during the late growing season. Morphological characters desirable for producing high biomass identified in fast-growing clones were as follows; high shoot-root ratio, high leaf surface area per tree, long leaf retention period, and many small leaves and compact crown architecture in the upper portion of true crown.

  • To sled the superior clones of Populus alba ${\times}$ Populus glandulosa $F_1$, growth and some growth-related enzyme activities were examined for thirteen, two-year-old and fifteen, three-year-old trees at Seoul National University nursery in Suwon. Clonal differences in total dry weight per tree and leaf surface area per tree were significant at the 5% level. Significant correlations were found between total dry weight per tree and leaf surface area per tree (r=0.875), between leaf peroxidase activity per tree and total dry weight per tree (r=0.854), and between leaf nitrate reductase activity per tree arid total dry weight per tree (r=0.914). Leaf peroxidase and nitrate reductase activities per unit fresh weight of one-year-old tree increased with increasing leaf order numbers basipetally, reaching maximum values in the eighteenth and thirty-third leaves, respectively, and decreased gradually from those leaves to basipetal lower leaves. Clones 65-29-19, 66-15-3, 65-22-11, 66-14-93, and 66-26-55 among two-year-old trees, and clones 64-6-44, 66-14-29, 66-26-55, 65-22-11, and 68-1-54 among three-year-old trees showed greater leaf surface areas, peroxidase and nitrate reductase activities per unit leaf fresh weight than other clones, Growth of Populus alba ${\times}$ Populus glandulosa $F_1$ clones might be estimated from either leaf surface area per tree or peroxidase and nitrate reductase activities per tree. Therefore, measurements of leaf surface area and leaf enzyme activities appear useful to select superior Populus clones at early growth stages.
