In Vitro Performance Assessment of a Bioartificial Liver Packed with Polyurethane Foam (다공성 폴리우레탄 충진형 생인공간의 체외 성능 평가)

  • Recently hepatocyte-based bioartificial liver (BAL) and hepatocyte transplantation have been actively investigated to treat acute hepatic failure. The BAL acts as a bridge to provide patients with more time until a donor organ becomes available for transplantation or until their own liver can be regenerated. In this study, we manufactured a polyurethane foam (PUF) using 15% NCO-prepolymer with a pore opening that allows it to be used as a hepatocyte immobilizing material. Cubes of PUF (3 mm dim.) were seeded with rat primary hepatocytes at a density of 5.5$\pm$1.1$\times$ $10^6$ cells/$cm^3$ PUF by centrifuging them together. The cell laden PUF cubes were packed into a prototype reactor and perfused with a hormonally defined medium for a week. Hepatocytes in the pores of the PUF formed spheroids that showed stable ammonia removal and urea synthesis activities. The albumin production level was comparable to other BAL systems. The PUF packed hepatocyte bioreactor has the potential to be used as a BAL.

Simultaneous Determination of Isocyanates by High Performance Liquid Chromatography (고속액체크로마토그래피를 이용한 이소시아네이트의 동시 정량)

  • This study was designed to identify and quantitate airborne isocyanate simultaneously by HPLC. These samples were collected using 1-(2-pyridyl)piperazine(1-2PP) coated glass fiber filter from polyurethane painting works at 8 wood furniture factories in Kimpo and Inchun. The results obtained were as follows : 1. The most suitable mobile phase condition of simultaneously analyzing isocyanates was 0.01 M ammonium acetate buffer ACN(70/30) adjusted to pH 6.2 from the beginning of the analysis to 20 min and 0.01 M ammonium acetate buffer/ACN(50/50) adjusted to pH 6.2 from 21 min to 40 min using the gradient mode. The peaks of isocyanates were able to obtain within 30 min. 2. The recovery efficiencies for 2,6-TDI, 2,4-TDI, HDI and MDI urea derivates spiked at the target concentration on coated glass fiber tillers were 91.00, 93.42, 91.31 and 94.21 %, respectively. 3. The qualitative analysis of the isocyanates samples from polyurethane spray painting works in wood furnture factories identified Ihree isocyanates, 2,6-TDI, 2,4-TDI and MOI. And their concentration ranges were 0-312.6, 0-56.3 and $0-62.1{\mu}g/m^3$, respectively. A disadvantage of using the colorimetric method for isocyanate analysis is its inability of separating isocyanates. This study identified such three isocyanates as 2,6-TDI, 2,4-TDI and MDI from polyurethane spray painting works in wood furniture factories. These isocyanates were successfully quantitated by HPLC by modifying the mobile phase condition and switching to gradient mode.

Preparation and Comparison the Physical Properties of Polyurethane-Urea Using Biomass Derived Isosorbide (바이오매스 유래 이소소르비드를 이용한 폴리우레탄-우레아의 제조 및 특성 비교)

  • Polyurethane-ureas(PUUs) were prepared from 4,4'-methylenebis(cyclohexyl isocyanate) and various diols including isosorbide. Isosorbide is starch-derived monomer that exhibit a wide range of glass transition temperature and are therefore able to be used in many applications. PUU was synthesized by a pre-polymer polymerization using a catalyst. Successful synthesis of the PUU was characterized by fourier transform-infrared spectroscopy. Thermal properties were determined by differential scanning calorimetry, thermogravimetric analysis, and dynamic mechanical analysis. It was found that by tuning isosorbide content in the resin, their glass transition temperature(Tg) slightly decreased. Physical properties were also determined by tensile strength and X-ray diffraction. There is no significant differences between petroleum-derived diol and isosorbide in XRD analysis. Moreover, their physical and optical properties were determined. The result showed that the poly(tetramethylene ether glycol)/isosorbide-based PUU exhibited enhanced tensile strength, transmittance, transparency and biodegradability compared to the existing diols. After 11 weeks composting, the biodegradability of blends increased in ISB-PUU. The morphology of the fractured surface of blend films were investigated by scanning electron microscopy.

  • A novel approach of microbiologically-enhanced crack remediation (MECR) has been initiated and evaluated in this report. Under the laboratory conditions, Bacillus pasteurii was used to induce $CaCO_3$ precipitation as the microbial urease hydrolyzes urea to produce ammonia and carbon dioxide. The ammonia released in surroundings subsequently increases pH, leading to accumulation of insoluble $CaCO_3$. Scanning electron micrography (SEM) and x-ray diffraction (XRD) analyses evidenced the direct involvement of microorganisms in $CaCO_3$ precipitation. In biochemical studies, the primary roles of microorganisms and microbial urease were defined. Furthermore, the role of urease in $CaCO_3$ precipitation was characterized utilizing recombinant Escherichia coli that encoded B. pasteurii urease genes in a plasmid. Microorganisms immobilized in polyurethane (PU) polymer were applied to remediate concrete cracks. Although microbiologically- induced calcite precipitation enhanced neither the tensile strength nor the modulus of elasticity of the PU polymer, cement mortar whose crack was remediated with the cemaden polymer showed a significant increase in compressive strength. Through detailed investigation, MECR showed an excellent potential in cementing cracks in granite, concrete, and beyond.

Synthesis of Melamine Phosphate-Polyurethane Composite Foam Blown by Water and Characterization of Its Thermal Properties (H2O로 발포된 멜라민포스페이트-폴리우레탄폼 복합체 합성과 열적 특성 분석)

  • Polyurethane/melamine phosphate composite foam (MP-PUF) was prepared from poly(adipate)diol/melamine phosphate composite (f=2), polyether-polyol (f=4.6), and PMDI (f=2.5). The thermal properties of MP-PUF such as morphology, closed-cell content, thermal conductivity, and thermal stabilities were characterized. Water was used as a blowing agent, and the composition of melamine phosphate (MP) was maintained at $1.43{\pm}0.3wt%$ of MP-PUF. As the content of water increased, the thermal conductivity of pure polyurethane foam (PUF) decreased, whereas the thermal conductivity of MP-PUF increased. The thermal stabilities of the PUF and the MP-PUF were maximized at 5 php $H_2O$, and then decreased at the higher $H_2O$ contents. The thermal stabilities of MP-PUF were greatly enhanced due to the synergetic effect of MP and urea, which was generated during the blowing process. The temperature of 50% residual mass of MP-PUF increased to $370{\sim}450^{\circ}C$ and the temperature of 30% residual mass exceeded over $700^{\circ}C$. Compared to the PUF, the temperature of 50% residual mass and 30% residual mass were higher than 25 and $70^{\circ}C$, respectively.

Cell Opening of High Resilience Polyurethane foam II. Structure Effect of Polyether Type Cell Opener (고탄성 폴리우레탄 발포체의 기포개방 II. 폴리에테르형 기포개방제의 구조 영향)

  • For the preparation of high resilience polyurethane (PU) foams with polyether type cell openers which have different ethylene oxide (EO) content, molecular weight and chain structure, the influences of tell opener structure on the kinetics, rheology, structural stability, open cell content and mechanical properties of the obtained foam were investigated. It was observed that urea formation reaction was delayed with the increase of EO content and incorporation of ester linkage in cell opener molecule and was relatively independent on the molecular weight. With the rheological studies, the decreases of viscosity and storage modulus were confirmed for the increase of EO content and molecular weight, so that the resulted foam had low structural stability and high open cell content. The cell opener having ester linkage in molecule exhibited the lowest values of viscosity and storage modulus and the obtained foam has high open cell content. However, the structural stability increased due to the larger intermolecular interaction of ester linkage. The hardness, tensile strength, tear strength and elongation of foam were deteriorated with increase of EO content and molecular weight of tell opener. On the other hand, the cell opener having ester linkage in molecule improved the values of tensile strength, tear strength and elongation.
