Characteristics and Inhibition of Polyphenol Oxidase from Fuji Apples (후지 사과 Polyphenol Oxidase의 특성 및 활성억제)

  • As a basic research for inhibition of enzymatic browning of apples during dehydration or processing, polyphenol oxidase was extracted from Fuji apples to investigate heat inactivation, chemical inhibition and other properties. Polyphenol oxidase showed the highest activity at $20^{\circ}C$ and pH 5.5 with catechol as substrate, and the Michaelis constant of 0.14 M under the same condition of substrate and pH. The thermal inactivation followed pseudo first-order kinetics to have activation energy of 23.0 kcal/mol and z value of $19.7^{\circ}C$. As for substrate specificity the polyphenol oxidase showed high affinity toward the o-diphenolic compounds, particularly chlorogenic acid. Neither the m- and p-dihydroxy phenols nor monophenols were attacked. Browning by polyphenol oxidase was completely inhibited at the concentrations of 10mM for potassiummetasulfite and thiourea and 1mM for L-cysteine, ascorbic acid and sodium diethyldithiocarbamate.

Purification and some properties of polyphenol oxidase from Spuriopimpinella bracycarpa (참나물로부터 추출한 polyphenol oxidase의 부분정제 및 성질)

  • Three polyphenol oxidase(polyphenol oxidase I, II and III ) were isolated from the crude extract of a Spuriopimpinella bracycarpa by $(NH_4)_2SO_4$ precipitation and subsequent Sephadex G-150 chromatography. The final preparation thus obtained showed three peaks of enzyme activity. Optimum pH and temperature for the activity of polyphenol oxidase were 7.5 and $30^{\circ}C$, respectively. The enzyme was completely inactivated when i4 was treated at$70^{\circ}C$ for 30min and at $80^{\circ}C$ for 5min at pH 6.5. The enzyme was partially inactivated by ascorbic acid, glutathione and potassium cyanide (0.1mM), and was completely inhibited by L-cysteine, ascorbic acid, glutathione and potassium cyanide(0.5 and 1.0mM). The enzyme has good activity on catechol and 3,4-dihydroxytoluene but was strongly inactivated on pyrogallol, dopamine and DL-dopa. The Michaelis cons4ant of the enzyme was 86.5mM with catechol as a substrate.

Effect of Inhibition on Glucosultransferase and Antimicrobial Activity of Polyphenol Fraction of Gallnut and Red Grape Husk (오배자와 적포도과피 폴리페놀 분획물의 항균성 및 Glucosyltransferase 저해효과)

  • The antimicrobial and glucosyltransferase(GTase) inhibition activity were searched for 30 species of various folk drugs and by products of food industry. Among them, two species, gallnut and red grape husk water extracts, were selected for the powerful antimicrobial and GTase inhibition activity. The polyphenol fractions of gallnut and red grape husk were showed very greater antimicrobial activity on both Gram(+) and (-), B. subtilis and E. coli. The minimum antimicrobial activity of gallnut polyphenol fraction were 1.0mg for B. subtilis and 3,0mg for E. coli. Red grape husk was 2.0mg for B. subtilis and 3.0mg for E coli. The polyphenol fractions of gall nut and red grape husk were showed powerful GTase inhibition activity. The concentrations of these fractions for 80% inhibition of GTase activity were 1.08$\times$10$\^$-3/mg/㎖ and 1.08$\times$10$\^$-2/mg/㎖, respectively. The most abundant compound in these fraction seems to be polyphenol derivatives. From these results, we think that the gallnut and the red grape husk polyphenol fraction had more antimicrobial and anti-plaque activities than artificial synthetic preservatives as an economic point of view.

Antioxidant and Anti-inflammatory Activity of Six Halophytes in Korea

  • The aim of this study was to measure and compare polyphenol content, antioxidant, and anti-inflammatory activity of six halophytes (Limonium tetragonum, Suaeda glauca, Suaeda japonica, Salicornia europaea, Triglochin maritimum, and Sonchus brachyotus). Depending on the total polyphenol content, the plants were categorized into two groups: (1) a high total polyphenol content group that included L. tetragonum, S. brachyotus, and S. europaea, and, (2) a low total polyphenol content group consisting of S. glauca, T. maritima, and S. japonica. Antioxidant activity was evaluated using DPPH and hydroxyl radical scavenging assays, and by measuring ROS. Anti-inflammatory activity was evaluated by measuring NO and $PGE_2$. L. tetragonum and S. brachyotus, that have high polyphenol content, also showed strong antioxidant activity. In addition, L. tetragonum, S. brachyotus, and S. europaea showed good anti-inflammatory activity. Consequently, the total polyphenol content was thought to be related to antioxidant and anti-inflammatory activity. Therefore, S. brachyotus and L. tetragonum are good candidates for use in pharmaceuticals and functional foods.

Changes in Total Flavonoid and Total Polyphenol Contents of Leafy Vegetables(Spinach, Chard and Whorled Mallow) by Blanching Time (데치는 시간에 따른 엽채류(시금치, 근대, 아욱)의 총 플라보노이드 및 총 폴리페놀 함량 변화)

  • This study was conducted to investigate the changes in contents of total flavonoid and total polyphenol of three leafy vegetables, spinach(Spinacia oleracea L.), chard(Beta vulgaris L.), whorled mallow(Malva verticillata L.) by various blanching times. Total flavonoid and total polyphenol contents of spinach were 21.01 and 25.58 mg/g, those of chard were 26.02 and 35.14 mg/g, and those of whorled mallow were 16.80 and 19.94 mg/g, respectively. Total flavonoid and total polyphenol contents of chard were the highest among the leafy vegetables. Total flavonoid and total polyphenol contents of leafy vegetables were decreased according to blanching time. Especially, total flavonoid content of chard showed significant loss of $15-30\%$ by blanching time. Total polyphenol content of whorled mallow showed loss of $37-39\%$, but the decrease was not significant.

  • Dipalmitoyl phosphatidyl choline(DPPC), Polyphenol Derivatives, and Hematoxylin-Eosin were directly sonicated in acidic condition for 6 minutes to give clear stock solutions. Absorbtion properties of Polyphenol Derivatives in lecithin vesicle of Diphalmitoyl phosphatidyl choline system at $25^{\circ}C$ have been studied by absorbtion spectroscopy. The equilibrium of Polyphenol Derivatives between monomer and dimer in lecithin vesicles have been existed at low concentration of Polyphenol Derivatives, but oligomer has been formed in vesicle at high concentration of lecithin vesicles. By adding Bacteriorhodopsin(BR) to constant concentration of Polyphenol Derivatives decreased the absorbtion ratio(${\alpha}/{\beta}$) of Polyphenol Derivatives was increased during phase transition of dipalmitoyl phosphatidyl choline. In the presence of column eluted lamella vesicle and mixture of uni- and multilamella aggregates. The differences of rate between column eluted- and mixture were observed, therefore column eluted lamella reaction was represented more catalytic effect. The phase transition temperature of hydrolysis on Dipalmitoyl phosphatidyl choline and Polyphenol Derivatives were measured higher than it of Dipalmitoyl phosphatidyl choline and no Polyphenol Derivatives.

Hibernation of Mammalian Cells at a Living Body Temperature

  • The present study revealed that polyphenol induces the hibernation of mammalian cells at the living body temperature. It was found that polyphenol is a cytostatic sleeping agent for mam-malian cells, where almost all cells resume proliferation after the hibernation period and cell death seldom occurs. By changing the concentration for polyphenol, various mammalian cells can be stored under different conditions, such as temporary sleep, and hibernation condi-tions.

  • This study was conducted to investigate the potential of polyphenols and polyphenol oxidase activity on the induction of rusty symptom development in ginseng root. When rusty inducing bacteria were inoculated on fresh ginseng root, the hue value of the inoculated root increased from 101.2 (white yellow) at 1 day after innoculation to 60.9 (brownish red) at 30 days after innoculation. Lysobacter gummosus, Pseudomonas veronii and Agrobacterium tumefaciens enhanced the accumulation of total phenolics. Along with the increase of total phenolics, total activity of polyphenol oxidase concomitantly increased but the specific activity of the enzyme was not.

Studies on the Distribution of Polyphenols in the Parts of Quercus acutissima (상수리 나무중 Polyphenol 성분들의 분포에 관한 연구)

  • Distribution of polyphenolic compounds in oak tree (Quercus acutissima, three years old) collected from Forest Research Institute located in Kwang Leung, Kyeonggi-do, Korea, was investigated using chromatographic studies. Total 25 polyphenolic fractions were separated from an oak tree, of which 15, 11, 7, 7, and 4 were in leaf, stem, root, bark, and seed, respectively. Catechins are predominant compounds in the polyphenols and some flavonoids were also identified. Distribution of polyphenols was relatively different in each part. Polyphenols in all of the part studied, except leaf where polymer was not detected, were existed as polymeric, oligomeric, and monomeric forms. Relative contents of total polyphenols in Quercus acutissima were the highest in bark, followed by root, leaf, acorn, and stem. Monomeric polyphenols were the predominant compounds present in all of the part of the oak tree.
