  • Reproducible the random amplified polymorphic DNAs(RAPDs) patterns were obtained in the two silkworm strains(J111, Galwon) by adjusting concentration optimized of Taq DNA polymerase(one unit), dNTP(200$\mu$M), MgCl2(1.5mM) and template DNA(30ng). In addition, anealing temperature ranging 35$^{\circ}C$ to 42$^{\circ}C$ by the adjusted condition was investigated and fixed at 35$^{\circ}C$ in this study. Variation among individuals and between male and female of Jam 113 strain was not authorized. DNA polymorhpisms among silkworms were authorized by five RAPD markers using OPM04 random primer. Using the primer showing polymorhpims between parents(J111, Galwon) in thirty three individuals, RAPD-PCR for F2 analysis was performed and segregated 3 : 1 in the F2 population. Consequently, RAPDs detected in the parents were obtained as genetic markers, which can be used for construction of genetic map for this industrially particular insect, silkworm Bombyx mori

  • The need to preserve and use plant genetic resources is widely recognized, and the prospect of dwindling plant genetic diversity, coupled with increased demands on these resources, has made them a topic of global discussion. In the present study, the genetic diversity and population structure of 73 ginseng accessions collected from six regions in China were analyzed using eight simple sequence repeat (SSR) markers. Major allele frequencies ranged between 0.38 ~ 0.78, with a mean allele frequency value of 0.571. The number of alleles discovered ranged from 3 to 10 per accession, with a mean number of 7; 56 alleles were discovered in total. Gene diversity (GD) and polymorphic information content (PIC) values were similar to each other, and they ranged from 0.36 ~ 0.77 (mean 0.588) and 0.33 ~ 0.74 (mean 0.548), respectively. Accessions were divided into three clusters based on their phylogenetic relationships and genetic similarities, and although the populations were similar, they were not classified according to the region. Regional genetic diversity was also similar, with slight differences observed based on the number of accessions per region. It is expected that the findings of the present study can provide basic data for future studies on ginseng genetic diversity and for breeding ginseng cultivars.

  • In this study, we analyzed the microsatellite DNA poly-morphisms for pedigree verification in Kyungju dog (Dongkyung-i) which is one of the Korean breed dogs. A total of 51 Dongkyung-i samples were genotyped using 8 microsatellite markers. The number of alleles observed at single locus ranged from 4 to 12, with average number of alleles per locus of 8.5. The expected heterozygosity and polymorphic information contents (PIC) values of the 8 microsatellite loci were $0.6162{\sim}0.8746$ (mean 0.7587) and $0.5461{\sim}0.8512$ (mean 0.7167), respectively. Of the 8 markers, PEZ3, PEZ6, PEZ12 and FHC2054 loci had relatively high PIC values (>0.7) in Dongkyung-i. Pedigree verification of Dongkyung-i was analyzed based on alleles observed. The results of the parentage testing were noted significant differences compared with breeders. These results show basic information of conservation and research in Dongkyung-i, and further studies of genetic pedigree in Dongkyung-i will be needed.

  • The genus Acorus is known as an indigenous medicinal plant. Genetic diversity of thirteen accessions of A. calamus and eight of A. gramineus, with an accession of Colocasia antiquorum and two of Iris pseudacorus as outgroups, were evaluated using RAPD markers for cluster analysis and principal coordinate analysis, and NIR spectroscopic profiles for principal component analysis.A total of 371 polymorphic bands were obtained by using the selected 12 random primers. The genetic distances were estimated from 0.03 to 0.31 within A. calamus and from 0.03 to 0.51 within A. gramineus. The dendrogram and three-dimensional plot separated the accessions into four distinct groups (A. calamus, A. gramineus, C. antiquorum, and I. pseudacorus). Moreover, for the diversity among genus Acorus, eleven A. calamus accessions, one A. gramineus accession, and two I. pseudacorus accessions were non-destructively analyzed from their leaves by NIR spectroscopy, which discriminated Acorus accessions like the RAPD analysis. Interestingly, thirteen accessions of A. calamus were clustered into two groups based on RAPD and NIR analyses, which indicates that there are two ecotypes of A. calamus in Korea. An accession (CZ) of A. calamus with yellow stripe on leaves was closely grouped with another (CX) at a genetic distance (GD) of 0.03, which shows that the stripe trait might be generated by chimeric mutation. The genetic distance between A. calamus and A. gramineus was revealed to be farthest from 0.80 to 0.88 GD. In genus Acorus the genetic diversity and genetic variation were identified by using RAPD marker technique and non-destructive NIRs.

  • Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.

  • We analyzed seven Y chromosome binary markers (YAP, RPS4Y_711,\;M9,\;M175,\;LINE1,\;SRY_+465$ and 47z) in samples from a total of 254 males from Koreans and tow Mongolian ethnic groups (Buryat and Khalkh) to study the genetic relationship among these populations. We found eight distinct Y haplogroups constructed from the seven binary markers. Haplogroup DE-YAP was present at extremely low frequencies (∼2%) in the Korean and Mongolian populations. This result is consistent with earlier reports that showed the YAP+ chromosomes to be highly polymorphic only in populations from Japan and Tibet in east Asia. The observed high frequency of haplogroup $C-RPS4Y_711$ in the Mongolian populations (∼40%) is concordant with recent findings, showing that the $RPS4Y_711$-T chromosomes were distributed at high frequencies in Siberian and Mongolian populations compared with most other populations from east Asia. Thus, the relatively moderate frequency of haplogroup $C-RPS4Y_711$ in Korean (∼15%) can be seen as genetic evidence for probable interaction with Mongolian and/or Siberian populations. In contrast, the majority (∼75%) of modern Koreans studied here had high frequencies of Y chromosome lineages of haplogroup O-M175 and additional haplogroupts that define sublineage of O-M175, which are most likely related with modern populations in China. In conclusion, our data on the Y chromosome haplogroup distribution may provide evidence for interaction between Korean and Mongolian populations, but Korean tend to be much more related with those from southern-to-northern populations of China than to Mongolians in east Asia.

  • Little millet (Panicum sumatrense) is well known for its salt and drought stress tolerance and high nutritional value, but very limited knowledge of genetic variation and genomic information is available. In this study, a total of 779 primer pairs were designed from the 22,961 EST sequences of switchgrass (Pancium virgatum), of which 48 EST-SSR markers were developed based on the trials of transferability of these primers in little millet. The EST-SSR amplicons showed reproducible single band polymorphism and produced a total of 160 alleles with an average of 3.3 alleles per locus in 37 accessions of little millet. The average values of expected and observed heterozygosities were 0.266 and 0.123, respectively. The polymorphic information content (PIC) values were observed in range of 0.026 to 0.549 with an average of 0.240. The genetic relatedness among the little millet accessions was evaluated by neighbor-joining dendrogram, which grouped all accessions into two distinct groups. The validation thus demonstrated the utility of the switchgrass EST-SSR markers in assessing genomic relationships in little millet. The findings from this study could be useful for designing strategies for the identification of diverse germplasm for conservation and future molecular breeding programs for little millet.

  • Random Amplified Polymorphic DNA (RAPD) analysis was used to investigate the genetic variations of Coreoleuciscus splendidus within and among the West Korea Subdistrict populations (in Han and Geum Rivers) and the South Korea Subdistrict populations (in Seomjin and Nakdong Rivers). Twelve random primers were employed to generate RAPD markers. All primers were produced to identify specific RAPD markers between the West and South Korea Subdistrict populations. Analyses of genetic similarity and distance among the West and South Korea Subdistrict populations of C. splendidus also revealed similar results, with low genetic similarity (0.49~0.53) and high distance value (0.63~0.71). UPGMA dendrogram based on genetic distance was also similar in results. Therefore, the West Korea Subdistrict populations and the South Korea Subdistrict populations vary in genetic structure, and C. splendidus in the South Korea Subdistrict may represent a different species.

  • Four taxa of the genus Phyllostachys were analysed with PCR-based molecular inter-simple sequence repeat (ISSR) determine markers to determine their phylogenetic relationships. Many species of this genus are regarded as economical and ecologically important in the world. With the nine primers screened, ISSR assay generated a total of 64 reproducible bands. Analysis of ISSR from individual plants of genus Phyllostachys resulted in 43 polymorphic bands with 70.49%. When species were grouped by four taxa, within group diversity was 0.092 (Hs), while among group diversify was 0.499 (Gst) on a per locus basis. The estimated gene flow (Nm) between the pairs of species was 0.559. Hence, we can expect weak or low gene flow among species. Total mean genetic diversity ($H_T$) for four taxa was 0.291. The ISSR data allowed us to resolve well-supported clades in the genus Phyllostachys. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic.

  • Beta-carotene producing transformants were produced in the background of 'Nagdongbyeo', a Japonica rice cultivar. Introgression of the carotenoid locus in the transformant, PAC4-2 into the elite cultivar 'Ilpumbyeo' was started. To initiate a backcrossing program, we surveyed 220 SSR markers and found that 38% of them were polymorphic between 'Ilpumbyeo' as a recurrent parent and the PAC4-2 as a recipient parent. The selection strategy comprising foreground and background selection was employed. First, foreground selection was practiced in $BC_1$, $BC_2$, and $BC_3$ generations using the transgene specific PCR-based marker in addition to visual scoring of the seed color. Marker-based background selection combined with phenotypic selection was employed from $BC_3F_2$ to $BC_3F_4$ generations. Blast search indicated that the transgene PAC4-2 was located between SSR markers, RM6 and RM482. 240 $BC_3F_3$ and 63 $BC_3F_4$ lines were evaluated for four agronomic traits including days to heading. Most of the lines were similar to Ilpumbyeo in agronomic traits evaluated. The percentage of PAC4-2 genome ranged from 4% to 21% with a mean of 12.5%, which was higher than the expected for an unselected $BC_3$ backcross population. This could be explained by the fact that two genes for beta-carotene and the stripe virus resistance were targeted in this study. We selected 10 representative $BC_3F_5$ lines from 63 $BC_3F_4$ lines based on agronomic traits and carotenoids content. The selection strategy would be appropriate for the introgression of beta-carotene gene in a breeding program.
