The present study was carried out to investigate the hypoglycemic effect of a polyherbal aqueous extract (Curcuma longa Linn., Emblica officinalis Gaertn., Trigonella foenum-graecum Linn., Enicostemma littorale Blume) in alloxan-induced diabetic rats. Short term experiments showed a decrease in blood glucose levels at $2^{nd}\;hr$ of administration of the aqueous extract in alloxan-induced diabetic rats with increase in serum insulin levels. The extract did not show any effect on blood glucose or serum insulin levels in normoglycaemic rats. Treatment with the extract (1.5 g dry plant equivalent extarct/100 g body weight/day) for 20 days in diabetic rats showed a significant decrease in blood glucose and glycosylated haemoglobin levels and an increase in serum insulin levels. The aqueous extract also showed an enhanced glucose-induced insulin release at 11.1 mM glucose from isolated rat pancreatic islets. The extract did not show any toxicity at the particular dose used.
The object of this study was to evaluate the single dose toxicity of Yukmijihwangtanggamibang (YMJHTGMB), a polyherbal formula have been traditionally used as prevention or treatment agent for various lung diseases including chronic obstructive pulmonary disease (COPD), in male and female mice. Aqueous extracts of YMJHTGMB (Yield = 16.33%) wasadministered to female and male ICR mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changesin the body and organ weight except for soft feces restricted to YMJHTGMB 2,000 mg/kg treated two male mice (2/5; 40%) at 1 day after administration. In addition, no YMJHTGMB-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of YMJHTGMB aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines.
In this research, the genotoxic effects of Kong-Jin-Dan(KJD), a polyherbal formula were evaluated using the mouse micronucleus test. KJD was administered once a day for 2 continuous days by oral gavage to male ICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxic agent in a positive control. The appearance of a micronucleus is used as an index for genotoxic potential. In addition, the changes on the total white blood cells and differential counts on the lymphocytes, neutrophils, eosinophils, basophils and monocytes in the prepared blood smears were also conducted to observe the possible immunosuppress. The results obtained indicated that KJD shows no genotoxicity effects up to 2000 mg/kg dosing levels, but KJD shows slight increased trends in the blood total leukocyte numbers as pharmacological effects of immune stimulation. In addition, it is also considered that there were no problems from cytotoxicity of KJD tested in this study because the polychromatic erythrocyte ratio was detected as > 0.42 in all tested groups.

The object of this study was to evaluate the single dose toxicity of Kong-Jin-Dan (KJD), a polyherbal formula in male and female mice. KJD was administered to female and male ICR mice as an oral dose of 2000, 1000 and 500 mg/kg (body wt.) according to the recommendation of KFDA Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy, organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changes in the body and organ weight except for increases of lymphoid organ weights in KJD-dosing groups. These increases of lymphoid organ weights considered that related to the immune modulate effect of KJD not toxicological signs. In addition, no KJD-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. The results obtained in this study suggest that the KJD does not cause any toxicological signs. The $LD_{50}$ and approximate LD of KJD extracts in both female and male mice were considered as over 2000 mg/kg.

The effect of arogh, a polyherbal formulation-PHF [each 3 g powder contained Nelumbo nucifera G. (0.24 g), Hemidesmus indicus R. (0.24 g), Zingiber officinale R. (0.24 g), Terminalia chebula R. (0.24 g), Quercus infectoria O. (0.12 g), Hibiscus rosa-sinensis L. (0.24 g), Rosa damascene M.(0.24 g), Eclipta alba H.(0.24 g), Glycyrrhiza glabra L. (0.24 g)] was investigated in various experimental models of pain and inflammation. Analgesic activity of PHF was studied in mice using acetic acid induced writhing, tail immersion and hot plate methods. Anti-inflammatory activity of PHF was studied in rats using carrageenan induced hind paw edema and formalin induced rat paw edema methods. PHF significantly (P < 0.05) reduced the number of writhings, increased latency to flick tail in tail immersion method and elevated the mean basal reaction time in hot plate method. PHF significantly (P < 0.05) inhibited carrageenan induced hind paw edema and formalin induced rat paw edema. The PHF was tested at dose of 30, 100, 300 and 500 mg/kg.

Aqueous extracts of CWS (yield = 11.00%) was administered to female and male mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 14 principle organs were also examined. As results, we could not find any CWS treatment related mortality and clinical signs, changes in the body and organ weights, gross findings and changes in histopathology of principle organs, except for some dose-independent accidental findings. The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of CWS aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines, and can be safety used in clinics.

The effect of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for various respiratory disorders were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Two different dosages of DHU001 (300 and 150 mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference material, dexamethasone (15 mg/kg, intraperitoneal treatment). End of 7 days oral administration of DHU001 extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these contact dermatitis signs induced by DNFB treatment were reduced by treatment of the both different dosages of DHU001 and dexamethasone, respectively. The results obtained in this study suggest that oral treatment of DHU001 extracts also has relatively favorable effects on contact dermatitis.
An experiment was conducted to study the protective role of polyherbal feed supplement (Growell) during induced mycotoxicosis in broilers. A total of 240 Vencobb broilers were divided at day old stage into eight equal groups. Group A served as control and was given plain feed, group B, D, F and H were given Growell at 0.35 g/kg of feed. Group C, D, G and H were given dietary aflatoxin $B_1$ at 0.2 ppm and groups E, F, G and H were given ochratoxin A at 0.2 ppm in feed to study effect of Growell on individual aflatoxicosis, ochratoxicosis and combined mycotoxicosis of broilers. The chicks were given their respective feeds from 1st day to 6th week of age and were vaccinated at 7th and 28th day of age with Lasota strain of Newcastle disease virus. There was no statistically significant effect of mycotoxins individually or in combination on body weight of broilers. However, body weights were highest in group B and lowest in co-mycotoxicated group G. Feed conversion ratio was best in group B followed by A, D, F, E, H and G. Significant improvement in haemoglobin values was observed in broilers due to feeding of Growell in ochratoxin and co-mycotoxicated groups. There was no significant effect of mycotoxin treatment on PCV, TEC and TLC of broilers. Due to single and combined mycotoxicosis, there was reduction in serum total protein, albumin, cholesterol and triglyceride and rise in alkaline phosphatase, creatinine and uric acid levels. Supplementation of diets with Growell reduced the alterations induced due to mycotoxins. There was a significant rise in per cent organ weight of liver and reduction of that of spleen, bursa of Fabricius and thymus of broilers fed mycotoxins. Protection from alteration in per cent organ weight of these organ by supplementation of Growell was recorded. The observed impaired immune response and histopathological changes in liver, kidney, spleen, bursa of Fabricius and thymus of broilers given mycotoxins were protected by supplementation of Growell.
Objectives : In this study, we investigated the effect of KOB, a polyherbal medicine for allergic rhinitis and its main herb, Astragali Radix on allergic responses in ovalbumin (OVA)-induced Allergic rhinitis(AR) mice. Methods : Sprague Dawley (SD)-rats were orally administrated with KOB (500 mg/kg), Astragali Radix water extract (ARW, 100 mg/kg) or anti-histamine drug, dosodium cromoglycate (50 mg/kg) as a reference drug, and then intraperitoneally injected with compound 48/80 (8 mg/kg). Rats were measured the mortality and serum levels of histamine. BALB/c mice were orally administrated with KOB (500 mg/kg), ARW (100 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured the serum levels of histamine and IgE, and observed histopathological changes of nasa mucosa H&E staining. Results : KOB and ARW significantly decreased the mortality and the serum levels of histamine in compound 48/80-induced anaphylatic rats. KOB and ARW also decreased the serum levels of histamine and IgE in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that KOB has a strong anti-allergic effect through the inhibitory property of Astragali Radix, the main component of KOB against allergic responses in allergic rhinitis.
