한약재의 전혈혈소판응집억제 (Inhibition of Whole Blood Platelet aggregation from Traditional medicines)

  • To evaluate anti-aggregatory activity of traditional prescriptions and medicines for treatment of Ohyul symptom, 70% methanol extracts were examined using collagen stimulated in vitro platelet aggregation by impedance method in rat whole blood. The crude extracts from DoHaekSeungKiTang, BoYangHwanOhTang, Caesalpinia sappan, Rhus verniciflua, Rheum palmatum, Polygonum cuspidatum, Salvia miltiorrhiza were found to inhibit platelet aggregation. The effective crude extracts of traditional medicine were fractionated to dichloromethane, ethyl acetate, butanol and aqueous layer. Polygonum cuspidatum, Caesalpinia sappan aud Rhus verniciflua ethyl acetate fractions concentration-dependently $(250-50{\mu}m/ml)$ inhibited the aggregation of platelet in whole blood induced by collagen. These results suggested that ethyl acetate fractions of Polygonum cuspidatum, Caesalpinia sappan and Rhus verniciflua have potent anti-aggregatory activity.

수종의 한약재 분획물의 생리활성 효과 (The Biological Activities of Extracts and Fractions of Herbal Plants)

  • Objectives : This study was conducted to identify the antimicrobial anti-oxidative and tyrosinase inhibitory effect of three kinds of herbal plants, Polygonum cuspidatum, Chelidonii herba, Pharbitis nil L. naturally grown across the nation. Methods : To investigate in vitro anti-oxidative activity assay, antimicrobial effect and tyrosinase inhibitory activity, MeOH 80% extract, n-hexane, chloroform, ethyl acetate, n-butanol and water fractions of herbal plants were tested by diphenylpicrylhydrazyl(DPPH) method. Results : Bsutanol fractions of each plants showed about 90% on anti-oxidative effect. In case of tyrosinase activity, ethlyacetate fraction of Polygonum cuspidatum showed a potent inhibition effect. In the antimicrobial effect, ethylacetate fraction of Polygonum cuspidatu, hexane fractions of Chelidonii herba and the methanol 80% extracts of Pharbitis nil L. exhibited inhibition effect significantly. Conclusions : In MeOH 80% extracts, we couldn't found any extracts or fractions which have antimicrobial, anti-oxidative and tyrosinase inhibitory activity, however ethylacetate fractions of Polygonum cuspidatum showed effects commonly in these three assay system.

  • To develop natural antimicrobial agents for keeping qualities of postharvested greenhouse produce the antimiocrobial actions of Polygonum cuspidatum Sieb. et Zucc. extract , which showed remarkable antimicrobial effects against microorganism causing the postharvest decay of greenhouse produce, were investigate. In the inhibitory experiment of enzymes related to energy production metabolism hexokinase activities decreased to 73% and 68% by treating with Polygonum cuspidatum Sieb. et.Zucc. extract and Eugenia caryophyllata Thumnberg extract in comparison with control, respectively. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed that microbial cell membrane was destroyed by treating with the dilute extract solution. this change of celluloar membrane permeability could be identified in the experiment that 0-nitrophenyl-${\beta}$-D-galactopyrano-side(ONPG), the artificial substrate of ${\beta}$-galactosidase, was hydrolyzed in the presence of the extract, indicating that the membrane was perturbed. The separation and identification of the most antimicrobialo substances isolated from Polygonum cuspidatum Sieb et. Zucc. extract and Eugenia caryophyllata Thunberg extract were carried out by using gas chromatography and mass spectrometry 9GC/MSD), which were identified as eugenol. As a result, the functionality of Polygonum cuspidatum Sieb. et Zucc. extract and Eugenia caryophyllata Thunberg extract as antimicrobial agents for keeping qualities of postharvested greenhouse produce may be recommended.

  • In activated macrophage, large amounts of nitric oxide (NO) are generated by inducible nitric oxide synthase (iNOS), resulting in acute or chronic inflammatory disorders. In Raw 264.7 cells stimulated with lipopolysaccharide (LPS) to mimic inflammation, water extracts from the fruit of Chaenomeles sinensis, the root of Polygonum cuspidatum and Boswellia carterii inhibited the LPS-induced NO production in a parallel dose-dependent manner. To investigate the mechanism by which those extracts inhibits NO production, we examined the expression of iNOS and COX-2 in both mRNA and protein levels. We observed a significant change in the iNOS expression between LPS alone and LPS plus those extracts-treated cells. However, water extracts from Chaenomeles sinensis, Polygonum cuspidatum and Boswellia carterii did not inhibit COX-2 expression which was induced by LPS treatment in Raw 264.7 cells. These data suggest that water extracts from Chaenomeles sinensis, Polygonum cuspidatum and Boswellia carterii can modulate anti-inflammatory immune response, which may be in part associated with the regulation of NO synthesis through the regulation of iNOS expression in mouse macrophage cells.

Anticandidal Effect of Polygonum cuspidatum on C. albicans Biofilm Formation

  • Candida albicans is a common opportunistic pathogen and is frequently associated with biofilm formation occurring on the surfaces of host tissues and medical devices. On account of the distinct resistance of C. albicans biofilms to the conventional antifungal agents, new strategies are required to cope with these infections. The root of Polygonum cuspidatum has been used for medicinal purposes in East Asia. The aim of this study was to assess the anticandidal potential of the P. cuspidatum ethanol extract by evaluating biofilm formation, integrity of the cell membranes of C. albicans and adhesion of C. albicans cells to polystyrene surfaces. The growth and development of the biofilm was assessed using an XTT reduction assay, and the extract (0.39 mg/ml) significantly reduced ($41.1{\pm}17.8%$) biofilm formation of 11 C. albicans strains. The extract damaged the cell membranes of C. albicans and remarkably inhibited cell adhesion to polystyrene surfaces. The plant extract displayed fungistatic activity without significant hemolytic activity. Based on the results of this study, the P. cuspidatum extract has promising potential for use in treating biofilm-associated Candida infection.

  • Free amino acids and mineral substance in radix of Polygonum cuspidatum, Polygonum elliptica, and Polygonum sachalinense were analyzed by amino acid autoanalyzer and atomic absorption spectrophotometer. Gross contents of free amino acids in root of P. cuspidatum and P. elliptica are 12.99mg/g and 9.58mg/g respectively. The analysis of inorganic constituents of Polygonum sp. showed that it generally contains copper, iron, manganese, zinc, potassium and potassium salt.

Effect of Polygonum cuspidatum on renal function

  • Polygonum cuspidatum has been used as treatments of dermatitis, inflammation, hyperlipidemia and diuretics in folk remedies. In order to evaluate the urinary effect of Polygoni cuspidati Radix, its MeOH extract was administerd in rats. We determined the total urin volume, chemical parameters(urea nitrogen, creatinine, uric acid), electrolytes(natrium, potassium, chloride) ) in serum and urin. Polygoni cuspidati Radix showed increase in urin volume and electrolytes.

호장근으로부터 분리된 emodin의 혈관신생 억제 활성 (Emodin from Polygonum cuspidatum showed Angiogenesis Inhibiting Activity in vitro)

  • 호장근에서 분리한 emodin은 VEGF로 유도된 혈관신생의 한 단계인 혈관내피세포의 이동을 강하게 억제하였다. 또한 emodin은 혈관내피세포 이동을 억제시킨 $0.1\;{\mu}g/ml$ 농도에서 시험관내 혈관신생을 억제하였으며, 그 효과는 농도의존적인 양상을 보였다. 생체내 혈관신생 모델인 CAM assay에서도 emodin은 혈관신생을 억제하였다. 이러한 결과는 emodin이 현재까지 보고된 여러 가지 생리활성 이외에 혈관신생 억제활성을 가지고 있다는 사실을 보여주는 것이다. 따라서 emodin을 함유하고 있는 호장근은 혈관신생 관련 질환에 대한 천연물 유래 치료제의 개발을 위한 중요한 식물자원으로 활용될 수 있을 것이다.

  • During the screening for inhibitors of nitric oxide production in LPS-activated macrophage, RAW 264.7 cells, two coumarins were isolated from chloroform extract of the root of Polygonum cuspidatum. They were identified as decursin (1) and decursinol angelate (2) on the basis of spectroscopic methods. The $IC_{50}$ values of these compounds on NO production were $0.76\;{\mu}M$ (1) and $2.6\;{\mu}M$ (2), respectively. However, the iNOS activity was not inhibited by treatment with these compounds. Their inhibitory effect on NO production seems to result from the suppression of iNOS induction through the suppression of NF-kB activity.

  • Matrix metalloproteinase-9 (MMP-9) is considered to be an important component in the progression of inflammation. Monocytes/macrophages are prominent at inflammation sites, and activation of these cells by stimulants such as lipopolysaccharide (LPS) leads to the production of significant amounts of MMP-9. Here, we show that LPS-induced MMP-9 production and activation was inhibited by the water extract from the fruit of Chaenomeles sinensis (CS), the root of Polygonum cuspidatum (PC), but increased by the extract from Boswellia carterii (BC). To investigate the mechanism by which those extracts inhibits MMP-9 activation, we examined the level of MMP-9 mRNA expression. We observed a significant change in the MMP-9 expression between LPS alone and LPS plus Chaenomeles sinensis and Polygonum cuspidatum extracts-treated cells. In addition, LPS significantly up-regulated MMP-9 promoter activity in Raw 264.7 cells, which was attenuated by the CS and PS extracts. However, water extracts from Boswellia carterii increased MMP-9 expression and MMP-9 promoter activity which were induced by LPS treatment in Raw 264.7 cells. These data suggest that water extracts from Chaenomeles sinensis and Polygonum cuspidatum can modulate anti-inflammatory immune response, which may be in part associated with the regulation of MMP-9 production and/or activation through the regulation of MMP-9 expression in mouse macrophage cells.
