  • From the root bark of Acanithopanax koreanum, two polyacetylene compounds and one lignan compound were isolated and identified as falcarinon, falcarindiol and ariensin. Furthermore the steam bark also afforded methyl n-hexacosanoate, methyl linoleate and coniferin.

Studies on the Extraction of Polyacetylene from Korean Ginseng Using Supercritical Carbon Dioxide (초임계 $CO_2$를 이용한 고려인삼으로부터 Polyacetylene 추출에 관한 연구)

  • Polyacetylenes were extracted from Korean ginseng using supercritical $CO_2$ Yield of extraction of panaxydol and panaxynol was increased as the pressure of supercritical $CO_2$ increased at the range from 200 to 300 bar. The optimal yields of panaxydol and panaxynol was achieved at 65 and $55^{\circ}C$, respectively. Methanol was applied as a modifier. The highest yield of panaxydol and panaxynol were 0.230 and 0.054 mg/g-dry weight at modifier concentration of 10%(w/w), 300 bar, and $65^{\circ}C$. When these results were compared to that of methanol-extraction, the recoveries of panaxydol and panaxynol by supercritical $CO_2$ extraction in SFE were 37.8 and 55.1%, respectively.

Molecular Mechanisms of Cell Cycle Arrest and Apoptosis by Dideoxypetrosynol A, a Polyacetylene from the Sponge Petrosia sp., in Human Monocytic Leukemia Cells

  • Dideoxypetrosynol A, a polyacetylene from the marine sponge Petrosia sp., is known to exhibit significant selective cytotoxic activity against a small panel of human tumor cell lines, however, the mechanisms of which are poorly understood. In the present study, it was investigated the further possible mechanisms by which dideoxytetrosynol A exerts its anti-proliferative action in cultured human leukemia cell line U937. We observed that the proliferation-inhibitory effect of dideoxypetrosynol A was due to the induction of G1 arrest of the cell cycle and apoptosis, which effects were associated with up-regulation of cyclin D1 and down-regulation of cyclin E without any change in cyclin-dependent-kinases (Cdks) expression. Dideoxypetrosynol A markedly induced the levels of Cdk inhibitor p16/INK4a expression. Furthermore, down-regulation of phosphorylation of retinoblastoma protein (pRB) by this compound was associated with enhanced binding of pRB and the transcription factor E2F-1. The increase in apoptosis was associated with a dose-dependent up-regulation in pro-apoptotic Bax expression and activation of caspase-3 and caspase-9. Dideoxytetrosynol A decreased the levels of cyclooxygenase (COX)-2 mRNA and protein expression without significant changes in the levels of COX-1, which was correlated with a decrease in prostaglandin E2 (PGE2) synthesis. Furthermore, dideoxytetrosynol A treatment markedly inhibited the activity of telomerase, and the expression of human telomerase reverse transcriptase (hTERT), a main determinant of the telomerase enzymatic activity, was progressively down-regulated by dideoxytetrosynol A treatment in a dose-dependent fashion. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of dideoxytetrosynol A.

  • Panax ginseng, a medicinal plant, has been used as a blood-nourishing tonic for thousands of years in Asia, including Korea and China. P. ginseng exhibits adaptogen activity that maintains homeostasis by restoring general biological functions and non-specifically enhancing the body's resistance to external stress. Several P. ginseng effects have been reported. Korean Red Ginseng, in particular, has been reported in both basic and clinical studies to possess diverse effects such as enhanced immunity, fatigue relief, memory, blood circulation, and anti-oxidation. Moreover, it also protects against menopausal symptoms, cancer, cardiac diseases, and neurological disorders. The active components found in most Korean Red Ginseng varieties are known to include ginsenosides, polysaccharides, peptides, alkaloids, polyacetylene, and phenolic compounds. In this review, the identity and bioactivity of the non-saponin components of Korean Red Ginseng discovered to date are evaluated and the components are classified into polysaccharide and nitrogen compounds (protein, peptide, amino acid, nucleic acid, and alkaloid), as well as fat-soluble components such as polyacetylene, phenols, essential oils, and phytosterols. The distinct bioactivity of Korean Red Ginseng was found to originate from both saponin and non-saponin components rather than from only one or two specific components. Therefore, it is important to consider saponin and non-saponin elements together.

  • Polyacetylenes (conjugated poly-ynes) were Isolated from the roots of Panax Ginseng C.A. Meyer by the low pressure preparative liquid chromatography and characterized by spectral analyses. The advantages of the isolation method are the simplicity of the isolation procedure and the much higher yield of poly-ynes compared to the old method.

Bioceramic Effects to Enhance Secondary Metabolites Production in Tissue Culture of Some Medicinal Plants

  • We have investigated that a couple of soft ferrite ceramic powders having a spinal structure have shown the effect on growth and secondary metabolites production of some medicinal plants cultured in vitro. The addition of the ceramic powders as bare state to culture medium has stimulated the growth of Achyranthes japonica callus and plantlet, adventitious root of Hyoscyamus niger and Platycodon grandiflorum hairy root about 65, 75, 150 and 50%, respectively. Whereas Centella asiatica callus and plantlet, Scopolia parviflora hairy root, and Hyoscyamus albus adventitious root were not affected markedly. Moreover, the ceramic powder has enhanced the growth of H. niger adventitious roots even under conditions of irradiating alone without any direct contact between ceramic powder and media. Based on growth stimulation effect, the ceramic powders have enhanced the gross production of tropane alkaloid in H. niger adventitious root, and polyacetylene in P. grandiflorum hairy root about 35 and 30%, respectively.

  • 아세틸렌 화합물의 삼중결합은 2개의 $\pi$-결합을 가지고 있는 전자풍부 화학종으로서 다양한 촉매 system에 의해서 비교적 쉽게 부가중합반응이 진행되며 이를 통하여 공액구조 고분자를 쉽게 제조할 수 있다. 가장 간단한 삼중결합 화합물인 아세틸렌은 다양한 촉매 system에 의해서 쉽게 부가중합반응이 진행되어 분말상의 폴리아세틸렌이 합성되었으며 1974년 일본의 Shirakawa 등은 Ziegler-Natta 촉매의 일종인 Ti(OC$_4$H$_{9}$)$_4$-Al(C$_2$H$_{5}$)$_3$(Al/Ti = 3-4) 촉매 system을 사용하여 실험한 결과 은빛의 필름형 폴리아세틸렌의 합성에 성공하였다.   • From the rhizome of Atractylodes japonica Koidzumi (Compositae) which is the original plant of oriental medicine 'Cang-Zhu', four essential oil compounds were isolated. Three of them were identified as atractylon, hydroxyatractylon and $5{\alpha}H,\;10{\beta}-selina-4(14),\;7(11)-diene-8-one$, which were already known as the constitutents of Atractylodes Rhizoma. The fourth is a novel polyacetylene type compound, and the structure is postulated as 1, 4-diacetoxytetradeca-6, 12-diene-8, 10-diyne by the spectral data.
