The gamma irradiation was on to Poly(butylene sebacate-co-terephthalate) (PBSeT), Poly(butylene adipate-co-terephthalate) (PBAT), Poly(lactic acid) (PLA) and casting polypropylene (CPP) at dose levels from 0 to 50 kGy. The properties of gamma irradiated samples were analyzed using DSC, TGA, UTM and FT-IR spectra. The mechanical and thermal properties of PBSeT and PBAT after gamma irradiation were less affected than CPP. The tensile strength and elongation of PBSeT was not affected by gamma irradiation, while these of PBAT, PLA and CPP were significantly decreased at 50 kGy gamma-ray dose. The thermal stability of PBSeT, PBAT, PLA and CPP showed a similar tendency to tensile strength. The glass transition temperature(Tg) and melting temperature(Tm) of PBSeT and PBAT were not altered by increasing gamma-ray dose, while these of PLA and CPP decreased. The chemical composition of all samples was not modified by gamma irradiation, and it was confirmed by FT-IR spectra. Based on mechanical and thermal stability studies of gamma irradiation on bioplastics, tested biodegradable packaging materials showed a potential to be used in sterilization process up to 35 kGy.
Bioabsorbable devices have been utilized and experimented in many aspects of orthopaedic surgery. Depending upon their constituent polymers, these materials can be tailored to provide sufficient rigidity to allow bone healing, retain mechanical strength for certain period of time, and then eventually begin to undergo degradation. The objective of this study was to estimate extent in which Poly-L-latic acid (PLLA) implants had bioabsorbability and biocompatibility with bone and soft tissue in dogs and also to develop bioabsorbable, biocompatible materials with the appropriate strength and degradation characteristics to allow for regular clinical use for treating orthopedic problems in humans as well as animals. Eighteen dogs were used as experimental animals and were inserted two types of PLLA implants. PLLA rods were inserted into subcutaneous tissue of back or the abdomen wall. And the rods were tested for material properties including viscosity, molecular weight, melting point, melting temperature, crystallinity, flexural strength, and flexural modulus over time. PLLA screws were inserted through cortical bone into bone marrow in the femur of the dogs and stainless steel screw was inserted in the same femur. Radiographs were taken after surgery to observe locations of screw. Histological variations including cortical bone response, muscular response, bone marrow response were analyzed over the time for 62weeks. The physical properties of PLLA rods had delicate balances between mechanical, thermal and viscoelastic factors. PLLA screws did not induce any harmful effects and clinical complications on bone and soft tissue for degradation period. These results suggest that PLLA implants could be suitable for clinical use.
Solutions of $Cd^{2+}$, $Co^{2+}$ and $Ni^{2+}$ were mixed with the solutions of hydroxycarboxylic acids such as salicylic, lactic and mandelic acids in the presence of cation exchange resin at room temperature. The distribution ratios of the metal ions between resin and solution were measured, using radioactive metal ions as tracer. From the observed variation of the distribution ratios with the acid anion concentrations, it was concluded that $Cd^{2+}$, $Co^{2+}$ and $Ni^{2+}$ formed the one-to-one complexes with salicylate, lactate and mandelate ions in aqueous, 20 % ethanol-water and 20 % acetone-water solutions. The results of the present study indicated that the relative stabilities of the metal-acid complexes in solution increased in the order: $Cd^{2+}$ <$Co^{2+}$ <$Ni^{2+}$ complexes. Salicylate
This study was carried out to suggest the optimal spinning process condition which provides a proper range of tenacity and biodegradability as textile fibers. The effects of the melt spinning speed and heat treatment on the mechanical property and biodegradability of polylactic acid fiber were investigated. Polylactic acid(PLA) was spun in a high spinning speed of $2000{\sim}4000m/min$. Each specimen was heat-treated at $100^{\circ}C$ during 30min. Mechanical properties such as breaking stress and the degree of crystallinity were evaluated using WAXS. Biodegradability was estimated from the decrease of breaking stress, weight loss, and the degree of crystallinity after soil burial. Experimental results revealed that heat treated specimens showed higher breaking stress than untreated specimens, but the increase was not so high as was expected from the remarkable change of crystallinity by heat treatment. It was concluded that breaking stress was more influenced by spinning speed than heat treatment. In the soil burial test, however biodegradability calculated from weight loss was more influenced by heat treatment than spinning speed.
본 실험은 bioceramic을 첨가하여 만든 다공성 poly D,L-lactic-co-glycolic acid(PLGA)-scaffold가 인간 지방조직에서 유래된 중간엽 줄기세포(human adipose tissue derived mesenchymal stem cells, ATMSCs)의 골 형성과정에 효과적인지를 알아보고자 수행하였다. ATMSCs를 well plate에 접종하여 골형성 유도(osteogenic induction, OI) 배양액으로 28일 동안 배양하였다. OI배양액군의 증식률은 세포접종 후 14일까지는 세포증식이 활발하게 진행됐지만, 21일 이후 세포의증식이 둔화되는 양상을 보였다. 반면, 기본배양액군은 꾸준한 세포 증식을 보이며 21일 이후에는 OI배양액군보다 더 높은 증식을 나타냈다. OI배양액군의 alkaline phosphatase(ALP) 활성은 세포배양 21일까지는 증가했지만, 28일에는 감소한 반면에 기본배양액군은 계속 감소하는 양상을 띠었다. OI배양액군의 세포는 배양 21일에는 뚜렷한nodule의 형성을 관찰할 수 있었고, nodule에 칼슘의 축적이 일어남을 확인하였다. ATMSCs를 scaffold에 접종하여 OI배양액으로 배양하였다. Scaffold 내 골아세포 분화에 따른 ALP 활성은 PLGA scaffold와 Bioceramic-PLGA scaffold 모두에서 세포 배양 21일에 급격히 증가하였고, Bioceramic-PLGA scaffold의 ALP 활성이 PLGA scaffold보다 크게 증가하였다. 칼슘과 인의 함량 역시 Bioceramic-PLGA scaffold에서 높게 나타났으며, Bioceramic-PLGA scaffold의 Ca/P ratio가 PLGA scaffold보다 높게 나타났다. 생체내에 이식된 scaffold의 생분해성과 광물화는 bioceramic-PLGA scaffold에서 더욱 뚜렷하게 관찰되었다. 본 실험의 결과들을 종합해 볼 때 ATMSCs의 골형성능은 well plate보다 scaffold가 더 효과적이며, bioceramic이 scaffold의 세포 부착률과 ALP 활성을 증가시켜 골형성능에 효과적으로 작용하는 것으로 생각된다. 또한 bioceramic이 ATMSCs의 골형성 분화에 따른 광물화단계의 scaffold 내 칼슘과 인의 함량을 증가시키는 것으로 사료된다.생체 내 scaffold의 생분해성은 PLGA scaffold보다 Bioceramic-PLGA scaffold가 빠른 분해를 나타내며, 광물화에 따른 칼슘 침착이 더 활발한 것은 scaffold에 포함된 bioceramic이 생체 내 세포의 부착, 증식, 분화를 증가시켜 골형성을 촉진시키는 물질로 작용한 것으로 사료된다.
In this study, we investigated whether PLGA microspheres in combination with hyaluronic acid (HA) gel have appropriate properties as a bulking agent for urologic injection therapies and whether the implantation of PLGA microspheres and HA gel induces angiogenesis in the newly formed tissues. In order to investigate whether this bulking agent is injectable, this material was injected through 24-gauge needles into the subcutaneous dorsum of the mouse. The bulking agent was easily injected without needle obstruction. Histological analyses of the hybrid tissues at 2 weeks showed that host cells at the surrounding tissues migrated into the spaces between the implanted PLGA microspheres and formed tissue-like structures. An inflammatory response to the implants was mild at 2 weeks and diminished at 8 weeks. Importantly, extensive ingrowth of blood vessels was observed in the hybrid tissues formed by the injection of PLGA microspheres and HA, whereas blood vessels rarely formed in the hybrid tissues formed by the injection of PLGA microspheres only. The implant volume was conserved for almost the entire implantation period. Histological analyses of the distant organs of the bulking agent-implanted animals, such as the lungs, liver, heart, brain, kidney, and spleen, showed no evidence of the injected microsphere migration. These results show that PLGA microspheres in combination with HA possess the appropriate characteristics for a bulking agent for urologic injection therapies and induce extensive blood vessel formation in the hybrid tissues.
황칠나무 추출물의 순차적 복합발효를 통한 복합 기능성 물질들을 강화시킨 발효소재를 개발하였다. 황칠 추출물에 MSG와 glucose를 첨가한 후 1차 B. subtillis HA 발효를 통해 γ-PGA와 단백질 분해효소를 생성시켰다. 이후 skim milk와 glucose, yeast extract를 추가적으로 첨가하여 30℃에서 L. plantarum KS2020에 의한 2차 복합 발효를 통해서 기능성 물질인 GABA를 생산하였다. 황칠 추출물의 고초균 발효에서 생성된 점질물 및 점조도은 발효 2일 4.21±0.04 Pa·s, 4.1%±0.48%로 높은 점조도 값과 점질물을 함량을 보였다. pH 및 산도는 각각 8.9±0.01, 0.00±0.00이었으며 생균수는 7.27 log CFU/mL에서 발효 2일에 9.50 log CFU/mL로 높은 생균수를 보였다. 순차적 복합 발효가 진행되면서 고초균의 생균수는 4.30 log CFU/mL로 크게 감소하였으며 젖산균은 9.12 log CFU/mL로 증가하였다. Protease 활성 및 tyrosine 함량은 각각 0.8 unit/g, 57 mg%로 측정되었다. 이후 연속적 복합 발효 과정에서 발효 5일 차 106 mg% 정도로 증가되었으며, SDS-PAGE를 이용하여 casein 단백질이 분해된 것을 확인하였다. GABA 정성분석 결과 skim milk 5%만을 첨가한 조건을 제외하고 glucose, yeast extract가 추가로 첨가된 모든 조건에서 전구물질 MSG가 소진되면서 GABA로 전환되었다. 이중 최적 조건을 골라 정량분석을 진행하였으며 18.29 mg/mL의 GABA 함량을 나타내었다. 최종 황칠 복합 발효물은 γ-PGA, peptides, 고농도 GABA를 함유한 발효소재로써 다양한 식품의 원료 활용이 기대된다.
조직공학에 이용되는 고분자인 poly(lactic-co-glycolic acid)(PLGA)는 높은 생체적합성 및 생분해성의 장점을 지니지만, 생체활성물질의 결여와 소수성으로 인해 세포의 부착에 어려움을 가진다. 또한 PLGA의 가수분해 과정 중 생성되는 산 분해물이 조직주변의 pH를 감소시켜 염증을 유발하는 단점을 가지고 있다. 따라서 이러한 단점을 보완하기 위한 방법으로 천연고분자인 케라틴을 첨가한 PLGA필름을 제작하였으며 조직공학적 디스크재생에 응용하기 위하여 케라틴/PLGA 복합체필름의 원통형 지지체를 제작하였다. 이 원통형 지지체의 내부는 케라틴/PLGA 필름을 말아서 사용하였으며 외부는 PLGA 다공성 지지체 링을 제작하여 사용하였다. 본 연구에서는 이 지지체에 섬유륜세포를 파종하여 PLGA 필름과 케라틴/PLGA 필름을 이용한 원통형 지지체에서의 세포의 부착 및 생존율을 비교하고 케라틴을 첨가한 지지체의 우수성을 확인하였으며 강도의 우수성 또한 확인하였다. 이 결과는 디스크재생을 위한 지지체의 연구에 유용한 정보를 제공할 것이다.
본 연구에서는 생분해성 고분자인 poly(latic acid) (PLA), poly(butylene adipate-co-terephthalate) (PBAT)와 첨가제로 hydrogenated castor oil (HCO) powder를 이용하여 친환경 생분해성 소재를 제조하였다. 제조한 PLA/PBAT/HCO 블렌드 필름의 특성은 scanning electron microscope (SEM)와 fourier-transform infrared spectroscopy (FT-IR)를 이용하여 분석하였다. SEM 분석 결과 HCO가 첨가된 PLA/PBAT (8 : 2) 블렌드 필름은 12-hydroxy stearic acid (12HSA)와 cellulose가 첨가된 필름과 비교했을 때, 안정적인 표면을 나타내었다. FT-IR 결과는 PLA/PBAT 블렌드 필름에서 HCO의 특성 피크의 나타냈으며, HCO 함량이 0에서 0.2 wt%까지 증가함에 따라 intensity가 증가함을 알 수 있었다. 또한, 제조한 PLA/PBAT/HCO 블렌드 필름의 물리적 특성, 열 분석을 수행하였다. 그 결과 HCO 첨가에 의해 물리적 특성과 열 안정성이 3배 이상 향상되었음을 확인하였다. 제조한 생분해성 소재의 토양에서 생분해 정도는 90 days 동안 6~20% 분해됨을 확인하였다.

Avian influenza is a serious threat to both public health and the poultry industry worldwide. This respiratory virus can be combated by eliciting robust immune responses at the site of infection through mucosal immunization. Recombinant probiotics, specifically lactic acid bacteria, are safe and effective carriers for mucosal vaccines. In this study, we engineered recombinant fusion protein by fusing the hemagglutinin 1 (HA1) subunit of the A/Aquatic bird/Korea/W81/2005 (H5N2) with the Bacillus subtilis poly γ-glutamic acid synthetase A (pgsA) at the surface of Lactobacillus casei (pgsA-HA1/L. casei). Using subcellular fractionation and flow cytometry we confirmed the surface localization of this fusion protein. Mucosal administration of pgsA-HA1/L. casei in mice resulted in significant levels of HA1-specific serum IgG, mucosal IgA and neutralizing antibodies against the H5N2 virus. Additionally, pgsA-HA1/L. casei-induced systemic and local cell-mediated immune responses specific to HA1, as evidenced by an increased number of IFN-γ and IL-4 secreting cells in the spleens and higher levels of IL-4 in the local lymphocyte supernatants. Finally, mice inoculated with pgsA-HA1/L. casei were protected against a 10LD50 dose of the homologous mouse-adapted H5N2 virus. These results suggest that mucosal immunization with L. casei displaying HA1 on its surface could be a potential strategy for developing a mucosal vaccine against other H5 subtype viruses.
