Common stalked barnacle Pollicipes mitella is a filter-feeding crustacean that attaches itself to the surfaces of rocks in the ocean intertidal zone. Pollicipes mitella is one of about a dozen species of goose barnacle that are gathered commercially for human consumption. In Portugal and Spain, they are a widely consumed and expensive delicacy known as "Percebes (Pollicipes pollicipes )". However, common stalked barnacle is less popular in Korea, and information on its nutritional component is not yet available. In the present study, we analyzed the proximate and fatty acid compositions and mineral content of common stalked barnacle (muscle and cirrus). The muscle and cirrus of common stalked barnacle were 15.1% and 14.3% protein, and 3.31% and 1.81% lipid, respectively. The prominent fatty acids in the muscle and cirrus were 22:6n-3 (docosahexaenoic acid, DHA), 16:0, 20:5n-3 (eicosapentaenoic acid, EPA), 18:0, 18:1n-9, 16:1n-7 and 14:0. In particular, common stalked barnacle contained high levels of n-3 polyunsaturated fatty acids, such as DHA (25.4-25.6%) and EPA (14.7-15.1%), and low levels of the n-6 fatty acid, 20:4n-6 (1.2-1.3%). Common stalked barnacle contained a relatively large amount of Zn (10-42 mg/100 g) in its muscle and cirrus.
This study screened the antimicrobial and anti-inflammatory activities of three extracts [1% acetic acid (HAc), distilled water (D.W.), and ethanol] from the common stalked barnacle Pollicipes mitella. Among the extracts, the 1% HAc extract showed the strongest antibacterial activity against several bacteria, but exhibited no activity against Candida albicans. To improve the degree of separation of the 1% HAc extract, solid-phase extraction was performed using a C18 cartridge with three solvents (D.W., 60A, and 100A). The 1% HAc 60A eluate showed the strongest antibacterial activity and enzyme, salt, and temperature stability, with no hemolytic activity. In addition, strong DNA-binding ability but no bacterial membrane permeability was observed. These results indicate that the P. mitella 1% HAc 60A eluate may contain antibacterial organic compounds that target intracellular components but not bacterial membranes. In addition, the 1% HAc 60A eluate exhibited potent inhibitory activity to reduce the production of inflammatory mediators (nitric oxide and prostaglandin E2) and pro-inflammatory cytokines (tumor necrosis factor-α, interleukin (IL)-6, and IL-1β) with no cytotoxicity. Therefore, the P. mitella 1% HAc 60A eluate has anti-inflammatory activity. Collectively, our results suggest that the P. mitella 1% HAc 60A eluate can be used as a bioactive source with antibacterial and anti-inflammatory activities.
The whole mitochondrial genome (14,915 nt) of Pollicipes mitella (Crustacea, Maxillopoda, Cirripedia, Thoracica) was sequenced and characterized. It is the shortest of the 31 completely sequenced crustacean mitochondrial genomes, with the exception of a copepod Tigriopus japonicus (14,628 nt). It consists of the usual 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 relatively short non-coding region (294 nt). The thoracican cirripeds apart from Megabalanus volcano have the same arrangement of protein-coding genes as Limulus polypemus, but there are frequent tRNA gene translocations (at least 8). Some interesting translocation features that may be specific to the thoracican cirriped lineage are as follows: 1) trnK-trnQ lies between the control region and trnI, 2) trnA-trnE lies between trnN and trnS1, 3) trnP lies between ND4L and trnT, and 4) trnY-trnC lies between trnS2 and ND1. In P. mitella there are two trnL genes (L1 and L2) in the typical crustacean positions (ND1-L1-LrRNA and CO1-L2-CO2). The present result is compared and discussed with the other three cirriped mitochondrial genomes from one pedunculate (Pollicipes polymerus) and two sessiles (Tetraclita japonica and M. volcano) published so far. Mitochondrial protein phylogenies reconstructed by the BI and ML algorithms show that the thoracican Cirripedia is monophyletic (BPP 100/BP 100) and associated with Remipedia (BPP 98/BP 35). In addition, Oligostraca, including Ostracoda, Branchiura, and Pentastomida, is a monophyletic group (BPP 99/BP 68), and is basal to all the other examined arthropods. Remipedia + Cirripedia appears as an independent lineage within Arthropoda, apart from Thoracopoda (Malacostraca, Branchiopda, and Cephalocarida). The Thoracopoda is paraphyletic to Hexapoda. The present result suggests that the monophylies of Crustacea and Maxillopoda should be reconsidered.
Genetic diversity and gene flow patterns in Pollicipes mitella were investigated with a nucleotide sequence analysis of 514 base pairs from the mitochondrial cytochrome c oxidase subunit I gene (COI) in 124 samples collected from six Korean populations. In total, 59 haplotypes were defined by 40 variable nucleotide sites in the COI region. The haplotypes had shallow haplotype genealogy and no geographic associations. All populations had high haplotype diversity (0.909 to 0.979) and low nucleotide diversity (0.0055 to 0.0098). The haplotypes with recently diverged nucleotides were distributed by long-range larvae dispersal among regional populations. The pairwise fixation indices ($F_{ST}$) estimated with the exact test and migration rates indicate that substantial gene flow has occurred among populations as a result of sea currents, except between the Uljin (East Sea coast) and other Korean populations. This suggests that significant genetic differentiation and low migration rates have affected the Uljin population.
Genomic DNAs were extracted from the turtle leg (Pollicipes mitella, 1798) population of Tongyeong, Yeosu and Manjaedo located in the southern sea of Korea. The turtle leg population from Tongyeong (0.929) exhibited higher bandsharing values than did turtle leg from Manjaedo (0.852). The higher fragment sizes (>1,200 bp) are much more observed in the Yeosu population. The number of unique loci to each population and number of shared loci by the three populations, generated by PCR using 7 primers in the turtle leg (P. mitella) population of Tongyeong, Yeosu and Manjaedo. Genetic distances among different individuals of the Tongyeong population of the turtle leg (lane 1-07), Yeosu population of the turtle leg (lane 08-14) and Manjaedo population of the turtle leg (lane 15-21), respectively, were generated using the CLASSIFICATION option in Systat version 10 according to the bandsharing values and similarity matrix. The dendrogram, obtained by the seven decamer primers, indicated three genetic clusters: cluster 1 (TONGYEONG 01 TONGYEONG 07), cluster 2 (YEOSU 08 YEOSU 14), and cluster 3 (MANJEDO 15 MANJEDO 21). Tongyeong population could be evidently discriminated with the other two Yeosu and Manjaedo populations among three populations. The longest genetic distance (0.305) was found to exist between individuals' no. 02 of the Tongyeong population and no. 13 of the Yeosu population. It seems to the authors that this is a result of a high degree of inbreeding in narrow region for a long while.
A study on the distribution and community structure of the benthic macroinvertebrates on the intertidal zones of Gapa and Mara islets were carried out from March to June 1991. The macroinvertebrates collected and identified were composed of 7 phyla, 14 classes, 29 orders, 68 families and 132 speies in Gapa islet and of 7 phyla, 12 classes, 23 orders, 49 familis and 90 species in Maara islet. The first and second dominant species and the community dominance indices of the upper tidal zones of Gapa and Mara islets were Nodilittorina exigua, Pollicipes mitella and 79.01, 69.77 respectively. However, the dominant species of the middle and lower intertidal zones of the two islets were not identities and different among the tidal zones.
A study on the distribution and seasonal changes of the benthic macroinvertebrate community was performed on the 4 intertidal rocky shores of Biyang-do in Chehu Island throughout four seasons from January 1992 to january 1993. The macroinvertebrates composed of 8 phyla, 15 classes, 31 orders, 71 families, 168 species. The seasonal appearing species were 135 species in summer, 127 species in spring, 124 species in winter and 121 species in autumn, respectively. The dominant species of the upper zones were Nodilittorina exigua, Nerita japonica, Pollicipes mitella, the middle zones were Turbo coronata coreensis, Monodonta neritoides, petrolisthes japonicus and the lower zones were Pagurus samuelis, Chthamalus pilsbryi, Chlorostoma argyrostoma lischkei. The community dominance indices of the upper zones were much higher than the middle and lower zones. The seasonal appearnace in species in the lower zones were remarkable, but individual numbes were not. Species diversity and evenness in all investigated zones were highest in spring and lowest in winter. But species richness was highest in summer, lowest in autumm.
Chthamalus challengeri Hoek was collected from intertidal rocks to rear the larvae from hatching through nauplius to cyprid in the laboratory. Larval development consists of six nauplius stages and a cyprid. Unilobed labrum with a prominent protuberance and the frontolateral horns folded under the anterior cephalic shield margin are diagnostic features through all nauplius stages. The posterior border of the cephalic shield bears no posterior shield spines in nauplius stages IV-Ⅵ. There is a specific hispid seta in the fourth group of the antennal endopodite. Morphological features such as the cephalic shield, labrum, abdominal process, antennules, antennae and mandibles in all nauplius and cyprid stages are illustrated and described. The numerical setations of the antennule are found to aid in the intraspecific identification of barnacle nauplius stages without dissection. The keys to each stage of the barnacle larvae in Korean coastal waters are provided based on the reared nauplii of seven species: Pollicipes mitella Octomeris sulcata, Chthamalus challengeri, Balanus albicostatus B. trigonus, B. amphitrite, and B. improvisus inhabiting Korean coastal waters.
A study on the seasonal changes of the benthic macroinvertebrate community was performed on the intertidal rocky shores ao eastern coast of Cheju island and U-do throughout four seasons from october 1991 to August 1992. The macroinvertebrates were composed of 10 phyla, 18 classes, 39 orders, 94 families and 201 species. the number of speecies were 183 in summer, 147 in autumn, 138 in spring and 94 in winter, respectively. The total wpecies collected throuhout the study period were 193 species in U-do and 151 species in the eastern coast of Cheju Island. Nodilittorina exigua was the most dominant species in all seasons, followed by Monodonta neritoides in spring, autumn and winter, and pollicipes mitella in summer. U-do was higher in species diversity and richness and lower in evenness than the eastern coast. Species diversity and richness were higher in summer than any other seasons in lower intertidal zones. Community dominance indices, however, were significantly higher in upper intertidal zones than in lower intertidal zones.
