Novel Sulfonated Poly(arylene ether ketone) Containing Benzoxazole Membranes for Proton Exchange Membrane Fuel Cell

  • Novel sulfonated poly(aryl ether ketones) containing benzoxazole were directly synthesized by aromatic nucleophilic polycondensation using various ratios of 2,2'-bi[2-( 4-flurophenyl)benzoxazol-6-yl]hexafluoropropane to sodium 5,5'-carbonylbis(2-fluorobenzenesulfonate). The copolymers were soluble in polar aprotic solvents such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide, and N,N-dimethylformamide at a relatively high solid composition (>15 wt%) and formed tough, flexible and transparent membranes. The membranes exhibited a degradation temperature of above $290^{\circ}C$. The exact dissolution times of these membranes at $80^{\circ}C$ in Fenton's reagent (3 wt% $H_2O_2$ containing 2 ppm $FeSO_4$) were undetectable, confirming their excellent chemical stability in fuel cell application. The membranes showed a moderate increase in water uptake with respect to increasing temperature. The proton conductivities of the membranes were dependent on the composition and ranged from $1.10{\times}10^{-2}$ to $5.50{\times}10^{-2}Scm^{-1}$ at $80^{\circ}C$ and 95% relative humidity (RH). At $120^{\circ}C$ without externally humidified conditions, the conductivities increased above $10^{-2}Scm^{-1}$ with respect to increasing benzoxazole content, which suggested that the benzoxazole moieties contributed to the proton conduction.

Caenimonas aquaedulcis sp. nov., Isolated from Freshwater of Daechung Reservoir during Microcystis Bloom

  • A Gram-stain-negative, white-coloured, and rod-shaped bacterium, strain DR4-4T, was isolated from Daechung Reservoir, Republic of Korea, during Microcystis bloom. Strain DR4-4T was most closely related to Caenimonas terrae SGM1-15T and Caenimonas koreensis EMB320T with 98.1% 16S rRNA gene sequence similarities. The average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between strain DR4-4T and closely related type strains were below 79.46% and 22.30%, respectively. The genomic DNA G+C content was 67.5%. The major cellular fatty acids (≥10% of the total) were identified as C16:0, cyclo C17:0, summed feature 3 (C16:1ω7c and/or C16:1ω6c), and summed feature 8 (C18:1ω7c and/or C18:1ω6c). Strain DR4-4T possessed phosphatidylethanolamine, diphosphatidylglycerol, and phosphatidylglycerol as the main polar lipids and Q-8 as the respiratory quinone. The polyamine profile was composed of putrescine, cadaverine, and spermidine. The results of polyphasic characterization indicated that the isolated strain DR4-4T represents a novel species within the genus Caenimonas, for which the name Caenimonas aquaedulcis sp. nov. is proposed. The type strain is DR4-4T (=KCTC 82470T =JCM 34453T).

Mucilaginibacter aquariorum sp. nov., Isolated from Fresh Water

  • A Gram-stain-negative, rod-shaped bacterial strain, JC4T, was isolated from a freshwater sample and determined the taxonomic position. Initial identification based on 16S rRNA gene sequences revealed that strain JC4T is affiliated to the genus Mucilaginibacter with a sequence similarity of 97.97% to Mucilaginibacter rigui WPCB133T. The average nucleotide identity and digital DNA-DNA hybridization values between strain JC4T and Mucilaginibacter species were estimated below 80.92% and 23.9%, respectively. Strain JC4T contained summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) and iso-C15:0 as predominant cellular fatty acids. The dominant polar lipids were identified as phosphatidylethanolamine, one unidentified aminophospholipid, one unidentified phospholipid, and two unidentified lipids. The respiratory quinone was MK-7. The genomic DNA G+C content of strain JC4T was determined to be 42.44%. The above polyphasic evidences support that strain JC4T represents a novel species of the genus Mucilaginibacter, for which the name Mucilaginibacter aquariorum sp. nov. is proposed. The type strain is JC4T (= KCTC 92230T = LMG 32715T).

Prediction of Parthenogenetic Developmental Potential by Polar Body Extrusion and First Cleavage on In Vitro Maturation and Development of Porcine Follicular Oocytes (돼지 난모 세포의 Ethanol 처리에 의한 단위 발생에 있어서 극체 방출란과 분할란 선별에 따른 배발달율 비교)

  • The objective of this study was carried out to examine the selection effects of in vitro matured porcine follicular oocytes with polar body extrusion and early cleavage as non-invasive marker to know the developmental competence in advance. The porcine oocytes matured for 48 h were examined the polar body extrusion. The examined oocytes were matured for additional $16{\sim}18h$ and activated with 7% ethanol and cultured in $5{\mu}g/ml$ cytochalasin B for 5 h for diploid formation. The treated oocytes were cultured and examined the cleavage after 48 h and continued culturing for 5 days. The oocytes of 21.9% were discarded in morphological selection and 32.1% oocytes were discarded by failure of first polar body extrusion. The selected oocytes were matured and activated and then after 48 h the cleavage rates were examined. In morphologically selected oocytes, 15.8% oocytes were not cleaved and 52.6% oocytes were normally cleaved and 31.6% oocytes were hyper-cleaved over 8-cell stage. However in the first polar body extruded oocytes, 7.1% oocytes were not cleaved and 73.1% oocytes were normally cleaved and 19.8% oocytes were hyper-cleaved. The morphologically selected embryos that not cleavage-selected were developed in 16.7% up to blastocyst and the morphologically selected and cleavage-selected embryos were developed in 31.7%. The polar body extruded oocytes that were not carried out cleavage selection were developed in 39.0% and the polar body extruded and cleavage-selected embryos were developed 49.0%. The first cleavage timing was examined with 12 h interval after activation. In $0{\sim}12,\;12{\sim}24,\;24{\sim}36,\;and\;36{\sim}48h$ intervals, 4.1%, 68.6%, 19.1%, and 2.3% oocytes were cleaved and 5.9% oocytes were not cleaved until 48 after activation. The cleaved oocytes in each interval were cultured and developed upto blastocyst with 0, 39.1, 9.5, and 0%, respectively. This results suggests that polar body extruded and cleaved at $12{\sim}36h$ embryo has higher developmental potential than the others.

Inhibition of Melanogenesis by Ramalin from the Antarctic Lichen Ramalina terebrata (남극 지의류 Ramalina terebrata로부터 분리된 라말린의 미백효과)

  • Ramalin (${\gamma}$-glutamyl-N'-(2-hydroxyphenyl)hydrazide) isolated from the Antarctic lichen Ramalina terebrata has been shown to have strong antioxidant activities in the previous study. To investigate additional activities of ramalin, we studied the effects of ramalin on melanogenesis in melan-a cells, a non-tumorigenic melanocyte cell line. At a non-cytotoxic concentration, ramalin dramatically decreased melanin synthesis in melan-a cells in a dose-dependent manner, which was more potent than arbutin, a well-known tyrosinase inhibitor. Ramalin inhibited cell-free tyrosinase activity directly and intracellular tyrosinase activity as well. Its inhibitory mechanisms on melanin production were further assessed, and we found that ramalin significantly decreased the protein levels of melanogenic enzymes such as tyrosinase, tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2). However, the mRNA levels of these enzymes were not altered. In a clinical study, application of 0.2 % ramalin on human skin significantly improved the degree of skin brightness after 3 weeks. In conclusion, ramalin has strong anti-melanogenic activity that is exerted both by the direct inhibition of tyrosinase activity and by down-regulation of melanogenic proteins. Furthermore, ramalin showed skin brightening effect in a clinical study. Collectively, these results suggest that ramalin may be a useful inhibitor for melanogenesis in skin.

Effect of Supplementation of Vitamin E on In Vitro Maturation and Activation of Bovine Oocytes

  • This study was carried out to assess the effect of vitamin E against the reactive oxygen species (ROS) on chemical activation of in vitro matured oocytes. Bovine oocytes were aspirated from slaughtered ovaries and transferred to maturation medium with or without vitamin E ($100\;{\mu}M$). After 22 hours of culture, oocytes with polar bodies were selected and submitted to activation treatments with or without vitamin E. After activation, oocytes were cultured in mSOF medium and rate of development was monitored. For ROS ($H_2O_2$) detection, in vitro matured and activated oocytes were selected and stained with DCFDA and observed under fluorescence microscope. The ROS contents were not significant differences in IVM rate, activation process and embryonic development to blastocysts with or without vitamin E. The cell number of blastocyst showed significant difference (p<0.05) in embryos matured and activated with vitamin E. The results of the present study demonstrated that the exposure of vitamin E in IVM and activation process improved the quality of embryos evaluated by the cell number of blastocysts.

  • Forty herbal drugs which are described to have potential antitumor activity were solvent-fractionated with petroleum ether, ether and ethyl acetate in sequence. The cytotoxic activity was mostly shown in the ether fraction(40.54%) and petroleum ether fraction (35.15%), but scarcely in the water phase (10.8%), meaning that most of the active components had less polar property. Twenty-seven percent of the drugs tested were active, which is higher value than 10.4% of the random sampled drugs The drugs possessing the $ED_{50}$ values less than $10{mu}g/ml$ were the roots of Lithospermum erythrorhizon, Curcuma domestica, Salvia miltiorrhiza, Astragalus membraneceus and Scutellaria indica, the leaves of Panax ginseng, S. indica and Liriodendron tulipifera, the barks of Picrasma ailanthoides and Rhus vernifera, the herbs of Agrimonia pilosa and Siegesbeckia pubescens the seeds of Tricosanthes kirilowii, P. ailanthoides, and the stem of P. ginseng.

The Suppression of Maturational Competence by Streptomycin during In vitro Maturation of Goat Follicular Oocytes

  • Antibiotics are common additives in culture media during in vitro embryo development, but their effects on oocyte maturation in vitro have not been tested. The effects of penicillin, streptomycin and gentamicin on the maturational competence and subsequent development potential of goat follicular oocytes were examined after parthenogenetic activation in vitro. Maturation rates at 24 h after in vitro maturation, and parthenogenetic development at 48 h after activation, were evaluated by observing the protruding first polar body and the 4 cell stage cleavage, respectively. When streptomycin was present in the maturation medium, the percentages of matured oocytes 24 h after activation were significantly (p<0.01) lower than those from the other groups (42.5-45.7% vs. 69.1-73.8%). Penicillin and gentamicin treatment did not affect the maturation rates or the percentages reaching the 4 cell stage 48 h after activation. There was no significant difference in cleavage rates among the different antibiotic treatments 48 h after activation. Therefore, streptomycin suppresses the in vitro maturation of immature goat oocytes, but does not influence their subsequent development.

Non-polar Solvents (Toluene and Styrene) Enhance Methanol Skin Absorption

  • The quantitative assessment of the penetration of organic solvents through skin is necessary for the evaluation of health hazards in occupational environments. We investigated the rate of dermal penetration when mixed or single forms of organic solvents were placed into a diffusion cell in vitro or into an experimental animal in vivo. The diffusion rates of methanol. toluene, and styrene were 6.07, 0.129, and 0.046 mg/$cm^2$/h, respectively. When skin was exposed to the mixed solvent of methanol and toluene, the penetration rate of toluene did not change significantly (0.110 mg/$cm^2$/h). However, the rate of methanol penetration increased to 43.90 mg/$\textrm{cm}^2$/h. The penetration rate of methanol also increased significantly to 54.69 mg/$cm^2$/h by mixing it with styrene. The concentration of methanol in the blood was monitored during the epicutaneous exposure in rats. The blood concentration of methanol was increased by mixing methanol with toluene as seen in the in vitro experiments. These results showed that the penetration rate of organic solvents would be enhanced by mixing them with other solvents.

  • Gambierdiscus toxicus, Adachi et. Fukuyo, is a benthic ciguatoxin-producing armored dinoflagellate, often attached to macroalgae. This organism is the primary causative agent of ciguatera fish poisoning which occurs in tropical and subtropical regions. However, regardless of the fact that the population of Gambierdiscus spp. has expanded to such temperate areas from sub-trophic and trophic areas, monitoring of G. toxicus has been lacking in the Korean coastal waters of temperate areas. This study was performed at the uninhabited Baekdo Islands off the southern coast of Korea and at Seopsom Island in the vicinity of Seogwipo, Jeju Province during April and May, 2011. Cell densities of Gambierdiscus spp. on macroalgae at Baekdo and Jeju Island ranged from zero to 56.4 cells $g^{-1}$. Maximum density was recorded on the brown alga Cladophora japonica at St. 3 of Jeju Island. In particular, the cell densities of Gambierdiscus spp. were influenced by the substrate characteristics of macroalgae. In the future, the continuous monitoring of toxic benthic dinoflagellate is necessary to predict and prevent ciguatera poisoning in Korean coastal waters.
