Effect of Dietary Calcium, Casein and Suet on Intestinal Absorption of Cadmium in Mice (생쥐에 있어서 카세인, 칼슘 및 우지가 카드뮴의 장흡수에 미치는 영향)

  • This study was carried out to clarify the effect of dietary calcium, casein and suet on intestinal absorption of cadmium in mice. It was performed for 30 days, from April 11, to May 10, 1988. Ninety mice were devided into 4 experimental groups and control group with 6 mice each dietary group, and measured survival ratio, body weight and weight ratio of organ to body. The contents of cadmium in liver, kidney, spleen, muscle and skin with hair, and feces were analyzed by atomic absorption spectrophotometer after sacrifice by anesthesia. The ratio of survival after 30 day breeding was $100\%$ in control group, but $66.7\%$ in group IV(basal diet + Cd + Ca) with $100{\mu}g$ one dose and with 50ppm cadmium containing fluid free-intake, and group V (basal diet + Cd + suet) with free-intake of 50ppm containing cadmium fluid. The ratio of body weight gains in cases of single dose $100{\mu}g$ of cadmium was highest as $42.3\%$ in group IV and lowest as $26.0\%$ in group V, in cases of free-intake of 50ppm containing cadmium fluid was highest as $24.0\%$ in group IV and lowest as $11.6\%$ in group II (basal diet + Cd). The body weight, in the case of single dose of $100{\mu}g$ of cadmium showed no increase untill 5th day from acute poisoning. While in case of free-intake of 50ppm containing cadmium, it showed very slight increase through all breeding period. The weight ratio of organ to body were lowest in liver of group II in both occasions. The most of cadmium administered was excreated in feces within 2 days after single dose $100{\mu}g$ cadmium. The contents of cadmium in all tissues were significantly high in each group comparing to control group as liver, kidney, spleen and muscle of group II which showed the highest level in both occasions of $100{\mu}g$ one dose and free-intake of 50ppm containing cadmium fluid, and higher in group II of the occasion with $100{\mu}g$ single dose, and in group V of the occasion of 50ppm containing cadmium fluid free-inatke for skin with hair. And the content of cadmium in tissues is generally higher in the occasion of 50ppm containing cadmium free-intake than $100{\mu}g$ single dose. From the above results I conclude that the addition of casein, calcium and suet in cadmium containing diet is effective to inhibtion of intestinal absorption of cadmium by particularly, calcium.

A Study on Listeria Strain Species for Fishes and Shellfishes on Sale (시판되는 어 .패류에 대한 Listeria 속균의 조사연구)

  • Listeria spp. from sea water, fishes and shellfishes have been troubled in many countries. So we exam ined its distribution rates, biochemical characteristics of a separated strain, growth curve of pH at set times to 4 species of standard strain, and yes or no of growth inhibition for precautionary measure of food poisoning by L. monocytogenes, garlic, mustard, wasabi, and green tea extracts including sensitivity of antibiotics 10 species. As its results, check numbers of its positivity to Listeria spp. were 32 species in total examination body 200 species, and its isolation rates were 16%, L. innocua was 14.0%, L. monocytogenes 1.0%, and L. seeligeri 1.0% by the strain species. All the standard strain of 4 species showed growth inhibition bellow pH 3.0, its pH conditions of the optimum growth at 7.0∼8.0, and its growth was more active in alkali co]tuition than in acid condition. Its growth inhibition examination by garlic extracts had an the worst effects with O.D values of 0.078∼0.210. But the case of mustard and wasabi had weakened effect, and the case of green tea had some effect as the time went by. The results of sensitivity examination of antibiotics 10 species were as fellows. L. innocua of the 16 cases showed sensitivity of 100% in all 5 species, Ampicillin, etc, and Ciprofloxacin showed sensitivity of 43.7% and gentamicin, 93.7%. But tetracycline showed tolerance of 31.3% , cefotaxine. 75%, nalidixic acid, 100%. L. monocytogenes of the 6 cases showed sensitivity of 100% in all 6 species, ciprofloxacin, etc.

  • This study was carried out to clarify the effect of dietary calcium, casein, and suet on the accumulation of cadmium in mice. After 30 days, the survival rate of control group was loot,, but 66.7% in group IV(basal diet+Cd+Ca) with single dose of $100{\mu}g$ cadmium and with free-intake of water containing 50ppm cadmium, and group V(basal diet+Cd+suet) with free-intake of water containing 50ppm cadmium. The rate of weight gain in the case of single dose of $100{\mu}g$ cadmium was highest in group IV as 42.3% and lowest in group V as 26.0%, whereas in the cases of free intake group W was highest as 24.0% and group II(basal diet+Cd) was lowest as 11.6%. The body weight, in the case of single dose of $100{\mu}g$ cadmium showed no increase until 5th day after acute poisoning. But in the case of free intake group, it showed very slight increase through all the breeding period. The weight ratio of organ to body were lowest in the liver of group II in both occasions. The most of cadmium adminstered. were excreted through feces within 2 days after single dose of $100{\mu}g$ cadmium. The contents of cadmium in each group were significantly higher than those of control group. In the liver, kidney, spleen and muscle, group II showed the highest level in both occasions of $100{\mu}g$ single dose and free intake of water containing 50ppm cadmium. In the skin with hair, group ll of the occasion with single dose of $100{\mu}g$ and group V with free intake showed the highest level. And the contents of cadmium in tissue were markedly higher in the occasion of free intake of water containing 50ppm cadmium. From the above results I would conclude that the addition of casein and calcium are effective in the inhibition of intesitnal absorption of cadmium esp. by calcium.

  • This study was conducted to investigate the inhibitory effect of Ecklonia cava (EC) and Eisenia bicyclis (EB) ethanol extract on histamine production in mackerel. Changes in viable cell counts, histamine contents, pH and VBN of mackerel fillet treated with ethanol extracts during 25 days at 4 were measured. Treatments of EC and EB ethanol extract had reduced growth of viable cells by 2 log cycles during storage. Production of histamine was decreased by EC and EB extracts (115 and 96 ppm) when compared to the control at 5 days (384 ppm). The pH of mackerels treated with EC and EB extracts were no different, while the pH of the control increased during storage. Furthermore, the VBN of mackerels treated with EC and EB extracts were significantly decreased when compared to the control. In conclusion, EC and EB extract may reduce scombroid fish poisoning by decreasing histamine production in mackerel during refrigerated storage.

Preservation of Strawberries and Cucumbers Packaged by Low density polyethylene film impregnated with antimicmbial agent, Scutellariae baicalensis extract (황금추출물을 함유한 항균성 포장필름을 이용한 딸기와 오이의 저장효과)

  • To develop a wrapping film, which suppresses the microbial decay through the storage and prolongs the selflife of fruits and vegetables, the antimicrobial packaging films were prepared and applied to the preservation of strtwberries and cucumbers. Low density polyethylene(LDPE) film of 50㎛ thickness was faricated with 1% of Scutellariae baicalensis extract. The LDPE film impregnated with Scutellariae baicalensis extract showed antimicrobial activity on the disk test against Bacillus cereus, Escherchia coli and Fusarium sp.. The antimicrobial film changed the color and light transmittance, but did not affect heat shrinkage, mechanical tensile strength and wattability. Strawberries and cucumbers were separately wrapped with packaging films in the state of closely-adhered packaging as well as modified atmosphere packaging(MAP). The wrapped strawberries and cucumbers were stored for 21 days at 5$\^{C}$ and for 40 days at l0$\^{C}$, respectively. For the packaged strawberries and cucumbers at 5$\^{C}$ and 10$\^{C}$, the LDPE film impregnated with Scutellariae baicalensis extract showed the reduced growth of total aerobic bacteria, molds and yeasts and did not give any negative effect on other quality attributes during storage in comparison with conttrol film without any additive.

Studies on the Grassland Development in the Forest Ⅷ. Effect of shading degrees on the quality, digestibility and nitrate nitrogen concentration of main grasses (林間草地 開發에 關한 硏究 Ⅷ. 遮光程度가 主要 牧草의 品質, 消化率 및 窒酸態窒素含量에 미치는 影響)

  • A field experiment was conduted to determine the effects of shading degrees (O: full sunlight, 25, 50 and 75%) on the proximate components, cell wall constituents (CWC), digestibility, water soluble carbohydrates (WSC) and nitrate nitrogen ($NO_3$-N) concentration of grasses grown in forest. For the test different artificial shading houses were established and pasutre species used were orchardgrass, timothy, perennial ryegrass and ladino clover. the experiment was performed at LES in Suwon, 1985 1. Considering proximate components, CWC and digestibility of grasses, ladino clover showed the best quality, and then perennial ryegrass. 2. The contents of crude protein, crude ash, and digestibility of grasses were increased with shadking, regardless of pasture species. As the shading degrees are increased, the contents of crude fibe in orchardgrass, perennial ryegrass and timothy were decreased, while that in ladino clover was increased with shading. 3. Grasses grwon in spring showed higher digestibility than those grwon in summer season. 4. The content of WSC was the highest in perennial ryegrass, and then ladino clover, orchardgrass, and timothy, in that order. Also WSC was decreased as the shading degrees are in creased. 5. The content of $NO_3$-N was the highest in perennial ryegrass, and then orchardgrass, ladino clover and timothy, in that order. Also the $NO_3$-N was significantly increased with higher shading level. In the regression equation between shading degrees and $NO_3$-N ($r^2=0.90^{**},\;r^2=0.95^{**}$), shading degree of 43 to 44% was critical level, causing nitrate poisoning to animal. 6. Considering grass quality, dry matter yield and $NO_3$-N, less than 40% of shading degree (over 60% of full sunlight) was desirable for better grassland improvement, management and utilization in the forest.

The Early Prognosis of Burn Patients with Elevated Initial Arterial Carboxyhemoglobin Level (초기 동맥혈 Carboxyhemoglobin 농도가 높았던 화상 환자들의 예후지표에 관한 연구)

  • Background : Smoke inhalation injury is an important determinant of mortality in burn patients. The early detection of inhalation injury in burn patients is important because the incidence of respiratory failure after inhalation injury was known to be high, with hypoxemia, pneumonia, and prolonged ventilatory support being commonplace. Acute carbon monoxide poisoning was one feature of smoke inhalation. The purpose of our study were to investigate the clinical characteristics of burn patients whose initial arterial carboxyhemoglobin (COHb) level had been elevated, to assess the clinical impact of COHb for smoke inhalation injury. Methods : Among 1,416 burn patients had been admitted at our institution from August 1, 2001 to July 31, 2002, 39 patients whose initial arterial COHb level have been more than 5% were included. We compared clinical scoring system for inhalation injury, percent total body surface area (%TBSA) burn, initial chest X-ray findings, APACHE II scores and SAPS II scores between survivors (n=27) and non-survivors (n=12) retrospectively. Results : COHb level were 9.7(5.71% and 10.3(8.81% in survivors and in non-survivors (p>0.05). Mean %TBSA burn of survivors and non-survivors were $16.6{\pm}17.8%$ and $60.7{\pm}28.8%$ (p<0.001). We did not find any difference in clinical scoring system, initial chest X-ray findings in survivors and in non-survivors. But %TBSA burn, APACHE II and SAPS II scores were high in non-survivors than in survivors significantly. Important factors associated with death were %TBSA burn, APACHE II scores, SAPS II scores, and the most important factor in predicting mortality was %TBSA burn. Conclusion : Burn patients with elevated initial arterial COHb level showed poor prognosis, but further study may be performed to know that the effect of COHb on prognosis in burn patients accompanying smoke inhalation.

Measurement of δ-Aminolevulinic Acid in Urine by Fluorometric HPLC and Colorimetric Methods (비색법과 HPLC 법에 의한 요중 δ-Aminolevulinic acid의 측정치 비교)

  • The urinary excretion of ${\delta}$-aminolevulinic acid has been widely used as a measure of the biological effect of lead in lead exposed workers. It is usually measured by colorimetric method based on the color reaction of ALA-pyrrole with Ehrlich's reagent. But the results of ${\delta}$-ALA in urine by this method are somewhat artificially higher than expected due to the urinary ALA-like compound such as aminoacetone. On the other hand, the recently developed fluorometric HPLC method is very sensitive and specific for the measuring urinary ALA. In order to compare the data obtained by two methods and to investigate the interrelation between two methods, 117 lead workers with different lead exposure were checked urinary ${\delta}$-ALA, blood lead and other lead exposure related indices. The results obtained are as follows; 1. Urinary excretion of ${\delta}$-ALA by colorimetric method is 2.15 times higher than HPLC method in overall, revealing 2.47 times in workers of blood lead less than $20{\mu}g/dl$, 2.53 times in workers of blood lead $21-40{\mu}g/dl$ and 1.86 times in workers of blood lead over $41{\mu}g/dl$, respectively. 2. While the correlation coefficients of ${\delta}$-ALA measured by colorimetric method with blood lead and blood ZPP was 0.571 and 0.629, those of ${\delta}$-ALA measured by HPLC with blood lead and blood ZPP were 0.6l0 and 0.637. All the correlation coefficients were statistically significant, but there was no statistical difference of correlation coefficients between two methods. 3. The correlation coefficient of urinary excretion of ${\delta}$-ALA between two method was 0.838 without any correction, but it was 0.852 with the correction of specific gravity 1.024. 4. Simple linear regression of ${\delta}$-ALA measured by HPLC method on ${\delta}$-ALA measured by colorimetric method was (ALA-UPH)=-0.245+0.536 (ALA-UCO) without any correction and it was (SP ALA)=-0.525+0.598 (SP ALA-UCO) with the correction of specific gravity 1.024. With above results, it is recommended that the diagnostic criteria of ${\delta}$-ALA for lead poisoning needed to be revised if ${\delta}$-ALA is measured by HPLC rather than colorimetric method.

Some Physiological Activity of Phenolic Substances in Plant Foods (식물성 식품중 페놀성 물질의 몇가지 생리활성)

  • Nine plant foods (persimmon leaf, perilla seed, Chinese quince, ginger root, walnut, mugwort leaf, arrowroot, buckwheat and sorghum) rich in phenolic substances were examined for their effects on the digestive enzymes, food-poisoning bacteria and mutagenicity/antimutagenicity by Ames test. Among tested samples, Chinese quince significantly inhibited the $\alpha-amylase$ activity (97%), exhibiting an uncompetitive inhibition type. Protease activity was inhibited by Chinese quince (86%), persimmon leaf (51%) and mugwort leaf (20%), in which mugwort extract exhibited a noncompetitive type. Lipase was activated >50% by all samples. The inhibition of $\alpha-amylase$ was highly correlated with the content of condensed tannin (r=0.89) and the inhibition of protease, with total phenolic content (r=0.84). Total phenolies fraction of tested samples showed the growth inhibition toward E. coli. Streptococcus faecalis and Salmonella enteritidis, in which the effect of perilla, sorghum and arrowroot was the highest for E. coli. Standard phenolics and food samples did not show any mutagenicity toward Salmonella typhimurium TA98 and TA100. Tannic acid inhibited the mutation of the two strains by benzo[a]pyrene whereas total phenolics fractions of Chinese quince and walnut exhibited antimutagenicity to a lesser extent.
