Properties of a Fibrinolytic Enzyme Secreted by Bacillus amyloliquefaciens RSB34, Isolated from Doenjang

  • Nine bacilli with fibrinolytic activities were isolated from doenjang, a traditional Korean fermented soy food. Among them, RSB34 showed the strongest activity and was identified as Bacillus amyloliquefaciens by 16S rRNA and recA gene sequencing. During growth on LB up to 96 h, RSB34 showed the highest fibrinolytic activity ($83.23mU/{\mu}l$) at 48 h. Three bands of 23, 27, and 42 kDa in size were observed when the culture supernatant was analyzed by SDS-PAGE and 27 and 42 kDa bands by fibrin zymography. The gene encoding the 27 kDa fibrinolytic enzyme AprE34 was cloned by PCR. BLAST analyses confirmed that the gene was a homolog to genes encoding AprE-type proteases. aprE34 was overexpressed in Escherichia coli BL21(DE3) using pET26b(+). Recombinant AprE34 was purified and examined for its properties. The $K_m$ and $V_{max}$ values of recombinant AprE34 were $0.131{\pm}0.026mM$ and $16.551{\pm}0.316{\mu}M/l/min$, respectively, when measured using an artificial substrate, N-succinyl-ala-ala-pro-phe-p-nitroanilide. aprE34 was overexpressed in B. subtilis WB600 using pHY300PLK. B. subtilis transformants harboring pHYRSB34 (pHY300PLK with aprE34) showed higher fibrinolytic activity than B. amyloliquefaciens RSB34.

Characterization of a Glutamate Decarboxylase (GAD) from Enterococcus avium M5 Isolated from Jeotgal, a Korean Fermented Seafood

  • To develop starters for the production of functional foods or materials, lactic acid bacteria producing ${\gamma}-aminobutyric$ acid (GABA) were screened from jeotgals, Korean fermented seafoods. One isolate producing a high amount of GABA from monosodium $\text\tiny{L}$-glutamate (MSG) was identified as Enterococcus avium by 16S rRNA gene sequencing. E. avium M5 produced $18.47{\pm}1.26mg/ml$ GABA when incubated for 48 h at $37^{\circ}C$ in MRS broth with MSG (3% (w/v)). A gadB gene encoding glutamate decarboxylase (GAD) was cloned and overexpressed in E. coli BL21 (DE3) using the pET26b (+) expression vector. Recombinant GAD was purified through a Ni-NTA column and the size was estimated to be 53 kDa by SDS-PAGE. Maximum GAD activity was observed at pH 4.5 and $55^{\circ}C$and the activity was dependent on pyridoxal 5'-phosphate. The $K_m$ and $V_{max}$ values of GAD were $3.26{\pm}0.21mM$ and $0.0120{\pm}0.0001mM/min$, respectively, when MSG was used as a substrate. Enterococcus avium M5 secretes a lot of GABA when grown on MRS with MSG, and the strain is useful for the production of fermented foods containing a high amount of GABA.

미국내 청소년기 여학생과 어머니의 신체이미지 개념에 대한 연구 (Body Image Perceptions of Adolescent Daughters and Their Mothers in U.S.)

  • 미국내 9살과 14살 사이의 여학생과 어머니와의 관계가 여학생의 신체이미지와 육체만족도(Body Satisfaction)에 미치는 영향에 관한 연구이다. 총 41쌍의 여학생과 어머니가 미국의 노스 캐로라이나에 위치한 $[TC^2]$에 의뢰하여 리쿠르트 되었다. 참가자들은 우선 미국 질병관리 및 방지국(CDC)의 자료를 바탕으로 그들의 BMI 점수를 바탕으로 보통체중과 과체중으로 나누어졌다. Stunkard, Sorenson, and Schulsinger (1983)의 9개의 신체이미지 그림을 사용하여 어머니와 딸의 각자의 이상화된 신체이미지(Ideal Body Image), 상대방의 이상화된 신체이미지, 각자의 신체에 대한 만족도, 그리고 서로의 신체에 대한 만족도를 연구하였다. 논의 및 결론부분으로 과체중 여학생과 그들의 어머니의 서로간의 신체이미지 만족도가 보통체중의 여학생과 그들의 어머니의 신체이미지 만족도에 비해 현저히 낮았고, 상대방에 대한 신체이미지 만족도 또한 현저히 낮았다.

유아복과 아동복의 치수 규격에 관한 연구 (A Study on Garment Sizing Systems for Infants and Children)

  • For the purpose of standardizing sizing systems for infants and children's clothing, the researchers surveyed 20 different apparel manufacturers in Korea. The apparel production sizing systems used in the industry were analysed. The researchers also compared standard apparel sizing systems developed for infants and children's clothing in Korea. ISO, Japan, and U.S.A. The followings are the results of this study. 1. As survey findings of companies's views about sizing systems, boys or girls sizing system needed to be developed for 3 years old and over. The plus sizes are needed to be developed and children 9 years old and over need the plus size. 2. The body size specification in most sizing systems collected shows that chest circumference was increased 2cm for every infants sizes, and 3cm for boys and girls'sizes, from size 100 to size 130. However, the chest and waist circumference measurements were increased 4 to 8cm for large sizes, from size 130 to size 170. 3. There are similarity and dissimilarity among the standard sizing systems of ISO, Japan(JIS), U.S.A(ASTM), and Korea(KS). The ASTM developed infants'garment sizes for the babies who are 24months old and under. The most national standards, however, developed infant apparel sizing system for the babies under 105cm tall or 4 years old. 4. The range of body measurements covered by boys/girls sizing system were different by the nation. The U.S. system (ASTM) is for boys and girls who are taller than 120cm. The Japanese system(JIS) is for the children who are taller than 90cm.

플러스사이즈 여성 온라인 의류 쇼핑물의 정장 바지 사이즈 현황 및 맞음새 분석 (An Analysis of the Present Condition of Sizing System and Fitting on Formal Pants of the On-line Shopping Mall for Plus-sized Women)

  • The purpose of this study was to recognize of the sizing system and analyze the fitting on formal pants of the on-line shopping mall for plus-sized women. To pursue this purpose, the investigation of the sizing system was focused on 22 online companies. The formal pants of 7 companies was selected among them. And actual sizes were compared with the guidance sizes of goods. For the assessment about the fitting on formal pants of 7 online companies, these came to make on to 16 obese women with 'A' body shaped and 'O' body shaped. A data analysis used the SPSS 14.0 statistics program. To verify the difference, this study used the variance analysis and Duncan's test for the postmortem verification. The result was as follows. 1. All of 22 companies were using the symbol such as 88, 99, 100 and 110 in the present condition of sizing system. The information about products size was presenting the products size not to be the body size. the guidance sizes of products were different from actual sizes of 7 companies to be assorted. Therefore, the guidance sizes of goods could not accomplish the facility. 2. According to the fitting-tests by the sensory assessment, the obese women with 'A' body shaped, did the dissatisfaction at the items of the hip circumference, abdominal region, crotch deep and pants length. The obese women with 'O' body shaped, did the dissatisfaction at the the items of crotch deep and pants length.

GMA 용접공정에서 적외선 온도 센서를 이용한 용융지 크기 예측 (Weld pool size estimation of GMAW using IR temperature sensor)

  • A quality monitoring system in butt welding process is proposed to estimate weld pool sizes. The geometrical parameters of the weld pool such as the top bead width and the penetration depth plus half back width are utilized to prove the integrity of the weld quality. The monitoring variables used are the surface temperatures measured at three points on the top surface of the weldment. The temperature profile is assumed that it has a gaussian distribution in vertical direction of torch movement and verify this assumption through temperature analysis. A neural network estimator is designed to estimate weld pool size from temperature informations. The experimental results show that the proposed neural network estimator which used gaussian distribution as temperature information can estimate the weld pool sizes accurately than used three point temperatures as temperature information. Considering the change of gap size in butt welding, the experiment were performed on various gap size.

Properties of a Bacteriocin Produced by Bacillus subtilis EMD4 Isolated from Ganjang (Soy Sauce)

  • A Bacillus species, EMD4, with strong antibacterial activity was isolated from ganjang (soy sauce) and identified as B. subtilis. B. subtilis EMD4 strongly inhibited the growth of B. cereus ATCC14579 and B. thuringiensis ATCC33679. The antibacterial activity was stable at pH 3-9 but inactive at pH 10 and above. The activity was fully retained after 15 min at 80℃ but reduced by 50% after 15 min at 90℃. The activity was completely destroyed by proteinase K and protease treatment, indicating its proteinaceous nature. The bacteriocin (BacEMD4) was partially purified from culture supernatant by ammonium sulfate precipitation, and Q-Sepharose and Sephadex G-50 column chromatographies. The specific activity was increased from 769.2 AU/mg protein to 8,347.8 AU/mg protein and the final yield was 12.6%. The size of BacEMD4 was determined to be 3.5 kDa by Tricine SDS-PAGE. The N-terminal amino acid sequence was similar with that of Subtilosin A. Nucleotide sequencing of the cloned gene confirmed that BacEMD4 was Subtilosin A. BacEMD4 showed bactericidal activity against B. cereus ATCC14579.

동체시력 검사기의 광학계 설계 (Optical Design of an Inspection Apparatus for Dynamic Visual Acuity)

  • 현재 국내외적으로 야간에서의 차량운전자의 차량사고와 동체시력과의 관계가 관심이 되고 있다. 이에 본 연구에서 우리는 동체시력을 측정할 수 있는 검사기의 광학계를 설계해 보았다. 시표 표준은 란돌트 환으로 하였는데, 30m에서 1.0의 시력을 유지하기 위해 란돌트 환의 직경이 45mm이고 gap이 9mm인 크기를 기준으로 하였다. 광학적 구성은 관찰자, 플러스 굴절력 렌즈, 마이너스 굴절력 렌즈, 시표의 순서로 이루어졌는데, 먼 거리에서 가까운 거리로의 시표 이동에 따른 마이너스 굴절력의 렌즈에 의한 시표의 제1 허상의 크기가 점점 커지는 방향으로 배치되어졌고, 또한 이 제1 허상은 플러스 굴절력 렌즈의 초점 안에 배치되어 크기가 점점 커지면서 플러스 굴절력렌즈의 초점에서 렌즈 쪽으로 이동되도록 하였다. 이렇게 하여 플러스 굴절력 렌즈에 의한 제 2허상의 크기를 일정하게 유지시키면서 최종 제 2허상의 위치를 50m에서 3m의 범위로 이동시킬 수 있는 광학계를 완성하였다.

취반 재고미를 청가하여 제조한 저지방 분쇄우육의 관능검사 및 물성학적 특성 (Sensory and Instrumental Texture Characteristics of Low Fat Ground Beef Manufactured with the Addition of Cooked Old Rice)

  • 10% 지방육은, 풍미, 단단함을 제외하고는 대체로 좋지 않은 것으로 나타났고 10% 지방에 재고미 5%를 첨가한 육은 풍미, 입자 크기, 단단함, 전반적 적성은 좋게, 다즙성, 연도는 재고미의 첨가량이 더 많은 육과 30% 지방육에 비해 낮은 것으로 나타났다. 10% 지방에 10% 또는 20%의 재고미가 첨가된 육은 다즙성, 연도, 입자 크기, 전반적 적성은 좋게, 풍미, 단단함 등은 낮게 나타났으며 30% 지방육은 풍미, 다즙성, 연도, 단단함은 좋게, 입자 크기, 전반적 적성은 좋지 않게 나타났다. 결론적으로 재고미를 다량(20%) 혼입하는 것은 전반적 적성을 감소시켰지만 5% 혹은 10% 수준의 재고미를 혼합하는 것은 재고미의 적절한 소비를 꾀하면서 열량 감소, 지방 제거시 잃기 쉬운 육의 풍미, 다즙성, 전반적 적성의 유지 등 좋은 효과를 얻을 수 있었다. 관능 검사 결과 10% 지방에 5%와 10%의 재고미가 첨가된 우육이 좋은 평가를 받은 것은 근래 지방 섭취량을 줄이고자 하는 추세에 맞추어 큰 호응이 있을 것으로 기대된다.
